副溶血弧菌(Vibrio parahaemolyticus)是多种水产养殖动物的主要病原菌,尤其是可引起凡纳滨对虾幼虾呈毁灭性死亡。本研究基于gyrB和toxR两种基因序列设计2对特异性引物,建立一种副溶血弧菌快速、准确的双重PCR检测方法,扩增目的片段大小分别为285 bp和368 bp。结果表明在同一PCR反应体系中副溶血弧菌可同时扩增大小分别为285 bp和368 bp的2种基因片段,两种引物对4种其他水产动物病原菌无交叉反应;敏感性检测结果显示,该双重PCR最低能检测8.867 2×103 CFU/mL菌体浓度的副溶血弧菌,对副溶血弧菌模板DNA的检出极限为0.029 3 mg/L;对发病中国对虾糠虾幼体、水产品及虾池养殖用水进行双重PCR检测,呈阳性反应的样品可分离出优势生长的副溶血弧菌。该实验所建立的基于gyrB和toxR两种基因的双重PCR检测方法可用于副溶血弧菌引起的水产动物疾病的快速诊断及分子流行病学的调查研究。  相似文献

本研究将交叉引物恒温扩增技术(cross priming amplification,CPA)与核酸试纸条相结合建立一种副溶血性弧菌(Vibrio parahaemolyticus)快速可视化检测方法。针对副溶血性弧菌特有的不耐热溶血素基因tlh的六个不同区域设计两对特异性引物和一对检测探针,通过优化反应条件确定了最佳反应体系。CPA-核酸试纸条方法对副溶血性弧菌的检测具有较强的特异性,对纯培养物的检测灵敏度达到58 cfu/m L,对污染牡蛎中副溶血性弧菌的检测灵敏度为5.2 cfu/g,比传统的PCR技术灵敏度提高了10倍,且具有较高的稳定性。交叉引物恒温扩增技术与核酸试纸条相结合的方法操作简便、特异性强、灵敏度高且能有效防止污染,可用于现场及基层单位副溶血性弧菌的快速检测。  相似文献

利用十二烷基磺酸钠抽提并结合超速离心的方法提取了鳗弧菌(Vibrio anguillarum)、河口弧菌(V.aestuarianus)、霍乱弧菌(V.cholerae)和副溶血弧菌(V.parahaemolyticus)4种致病性弧菌的主要外膜蛋白,通过SDS-PAGE分析比较4种弧菌主要外膜蛋白的组分。结果表明:4株弧菌的外膜蛋白电泳图谱一般可得到5~10条蛋白带,分子量多数集中在26~40 kD和48~85 kD。对所提取的4种弧菌主要外膜蛋白进行SDS-PAGE后,分别与自制的兔抗鳗弧菌血清、抗河口弧菌血清、抗副溶血弧菌血清进行Western-blotting印迹分析。结果显示每种全菌抗血清可与相应菌的外膜蛋白部分组分发生免疫反应,并与其他3种弧菌外膜蛋白的部分组分产生交叉免疫反应,这些反应条带分子量主要集中于30~48 kD之间。本研究为进一步研究致病性弧菌外膜蛋白免疫学特性提供参考。  相似文献

提要应用BLAST程序和DNAstar软件,比较分析了5株副溶血弧菌和其他细菌的16S—23S rDNA间区序列,选取IGSIA作为扩增靶区,结合相邻的16S rDNA序列,设计合成了一对特异性引物,通过优化扩增条件,建立了快速检测副溶血弧菌的PCR方法,对其特异性和敏感性进行了探讨,并初步进行了应用研究。结果表明,新建的PCR方法能特异性地扩增出大小为306bp和552bp的2条带,分别对应于副溶血弧菌的IGS0和IGSIA,检测灵敏度为5.6×102CFU/ml,半个工作日即可得到准确的结果,能有效检测出在杂色鲍、凡纳滨对虾和各种水质中的副溶血弧菌,适用于海洋水产动物副溶血弧菌病的早期快速诊断、水产品的检疫及水质环境的监测。  相似文献

Macroalgae contribute to intertidal food webs primarily as detritus, with unclear implications for food web studies using stable isotope analysis. We examined differences in the thallus parts of two South African rhodophytes (Gelidium pristoides and Hypnea spicifera) and changes in overall δ13C, δ15N signatures and C:N ratios during degradation in both the field and laboratory. We hypothesized that both degrading macroalgal tissue and macroalgal-derived suspended particulate material (SPM) would show negligible changes in δ13C, but enriched δ15N signatures and lower C:N ratios relative to healthy plants. Only C:N laboratory ratios conformed to predictions, with both species of macroalgae showing decomposition related changes in δ13C and significant depletions in δ15N in both the field and laboratory. In the laboratory, algal tissue and SPM from each species behaved similarly (though some effects were non-significant) but with differing strengths. Gelidium pristoides δ13C increased and C:N ratios decreased over time in tissue and SPM; δ15N became depleted only in SPM. Hypnea spicifera, δ13C, δ15N and C:N ratios all decreased during degradation in both SPM and algae.  相似文献

石斑鱼及暂养环境中副溶血弧菌分离鉴定与毒力评价   总被引:1,自引:1,他引:0  
副溶血弧菌(Vibrio parahaemolyticus,VP)存在于海洋环境和海产品中,可引起人的食物中毒。本研究的目的是对某地石斑鱼暂养环境的副溶血弧菌的致病风险进行评价。结合弧菌选择性培养基TCBS、gyr B基因和16Sr RNA特定片段的序列分析方法对分离菌株进行鉴定;通过PCR检测毒力基因,血琼脂平板溶血实验、细胞感染实验、KM鼠灌胃实验分别检测各菌株的致病性、溶血性、细胞毒性与肠毒性。共鉴定出4株具有毒力的副溶血弧菌,分别为HS51-5、HS54-4、MJ01-1和MJ03-1。所有菌株都检测到溶血素基因(tdh和tlh)、三型分泌系统(T3SS)基因簇1(T3SS-1)的4个效应蛋白基因(vop Q、vop R、vop S和vpa0450)、和基因簇2(T3SS-2)的1个效应蛋白基因(Vop A)的分布,同时在HS51-5和HS54-4还检测到T3SS-2中的其他3个效应蛋白基因(vop C、vop T和vop L)。所有菌株都具有溶血现象、细胞毒性以及对小鼠的致死能力,而HS54-4、MJ01-1、MJ03-1对小鼠的毒力明显强于HS51-5。因此,从毒力基因检测、神奈川溶血、细胞毒性和对小鼠的毒力等4个方面的结果都表明副溶血弧菌具有致病性,说明石斑鱼暂养环境的副溶血弧菌具有致病风险,应加强对这些环境的监测。  相似文献

The potential role of generalist herbivores to serve as a source of biotic resistance against algal invasion in marine ecosystems has been poorly examined. The present study investigates the capacity of Mediterranean herbivorous fishes to consume three of the most invasive seaweeds of the Western Mediterranean (Caulerpa racemosa, Lophocladia lallemandii and Womersleyella setacea) and examines vertical and temporal variations of such consumption. Our results show that although fish feed throughout the depth gradient examined (5–35 m), they concentrate in shallow waters, and can consume high amounts of C. racemosa. Such high ingestion of C. racemosa does not appear to be random, since this alga is consistently chosen when offered in pairs with several native species. Conversely, L. lallemandii and W. setacea are barely eaten by fish even though they can be very abundant in the field throughout the year. Our results suggest that fish could be an important controlling agent that has been overlooked in temperate marine invasions, and they may be able to provide certain resistance to C. racemosa invasion. In contrast, they are unlikely to exert any important control effects on L. lallemandii or W. setacea.  相似文献

Populations dominated by Cystoseira zosteroides, an endemic and threatened Mediterranean seaweed, colonize deep-water rocky habitats down to more than 50 m depth. Assemblages dominated by this species display high algal and invertebrate species richness. Algal biomass averages 1134 g dw m−2. Erect and turf algae account for only 25% of total algal dry weight, while encrusting corallines are responsible for the remaining 75%. Sponges, bryozoans and ascidians constitute the dominant sessile macrofauna. Cystoseira zosteroides is the dominant erect algae, with a mean biomass of 60.6 g dw m−2, and densities ranging from 4 to 7 plants m−2. The alien turf alga Womersleyella setacea has a biomass of 104.2 g dw m−2 and covers most of the understory substrate. The size-frequency distribution of C. zosteroides populations shows differences over time. Mean annual growth of the main axis is around 0.5 cm and mean annual mortality rate is lower than 2%. Recruitment was almost nil during the studied period of time (10 years). Processes structuring these deep-water Cystoseira stands must be driven by episodic disturbances, after-disturbance recruitment pulses, and long periods of steady growth that last at least 10 years. However, it is also possible that recruitment is irreversibly inhibited by the alien alga W. setacea in which case these old-growth stands are faced with extinction. The highly diversified assemblages and the low growth and low mortality rates of C. zosteroides indicate high vulnerability to natural and anthropogenic disturbances, and call for effective measures to ensure their conservation.  相似文献

Chemotactic motility is involved in the virulence of many pathogenic bacteria. In order to understand the role of chemotactic motility of Vibrio harveyi in cellular processes and virulence, mini-Tn10/Kan transposon-induced mutants with deficient chemotactic motility were constructed, screened, and identified. Sequence analysis revealed that the 465-bp fragment (GenBank accession number HM630274) flanking the transposon insertion site in mutant TS-CM1 had the highest identity (96.9%) with a hypothetical protein gene of V. harveyi ATCC BAA-1116 and the second-highest identity (91.8%) with the pgk gene of V. parahaemolyticus RIMD 2210633. In another mutant, TS-CM2, 356 bp of transposon-flanking sequence (GenBank accession number HM630275) also showed the highest identity (94.6%) with a hypothetical protein gene of V. harveyi ATCC BAA-1116 and the second-highest identity (92.4%) with the flaB gene of V. alginolyticus HY9901. Studies on virulence-related biological characteristics such as growth,motility, adhesion, and infectivity of themutants showed that disruption of either the flagellin gene or energymetabolism gene led to subsequent loss of chemotactic motility and changes in growth, motility, adhesion, and virulence of the pathogenic V. harveyi. Hence, the flagellin gene and crucial energy metabolism gene played an important role in the chemotacticmotility of V. harveyi.  相似文献

本文应用免疫组织化学方法,对健康大黄鱼和患溶藻弧菌病大黄鱼组织中巨噬细胞移动抑制因子(macrophage migration inhibitory factor,MIF)蛋白进行了检测,以研究溶藻弧菌病大黄鱼组织中MIF的表达情况及其对预后的影响。结果表明,MIF在健康大黄鱼头肾、脾、肠各组织中均无表达;MIF在溶藻弧菌病大黄鱼各组织中高表达,表达强度从弱到强依次是肠、脾、头肾,MIF表达阳性率分别为:56%、64%、92%。推测MIF在大黄鱼溶藻弧菌病发病中发挥重要作用。  相似文献

本实验以新月菱形藻为受试生物,研究了低浓度不同粒径TiO2颗粒(21nm、60nm和400nm)对海洋微藻生长、抗氧化酶活性(超氧化物歧化酶SOD、过氧化氢酶CAT和过氧化物酶POD)、脂质过氧化产物(MDA)含量的影响,并测定了相应的活性氧自由基(ROS)的含量,初步探讨了TiO2颗粒对海洋微藻的作用机制。结果表明,1mg/L TiO2颗粒对新月菱形藻生长的抑制作用随着粒径的减小而逐渐增强,第48h、72h、96h呈现出显著的纳米效应。TiO2颗粒可以诱导藻细胞内ROS的含量增加,对藻细胞产生氧化胁迫,新月菱形藻的抗氧化酶活性发生应激响应,以清除过量的ROS,但剩余的ROS对藻细胞产生氧化损伤,导致MDA含量升高,并且纳米级TiO2颗粒对新月菱形藻的氧化损伤大于微米级颗粒。在不同粒径TiO2颗粒的胁迫下,藻细胞SOD和CAT活性的响应也存在差异。本研究将为开展人工纳米材料对海洋生态系统影响的潜在风险评估提供科学依据。  相似文献

副溶血弧菌广泛分布于海洋环境中,虽然大多数环境菌株并非致病菌,但它们常常携有毒力基因,从而具有潜在的致病性。本文从黄海(山东青岛)海水中分离到一株副溶血弧菌D3112,对其进行了全基因组测序、溶血实验和人工感染实验等。基因组测序分析发现D3112并不含有副溶血弧菌的致病性标志溶血素基因tdh和trh,这与PCR检测结果一致,但含有其他溶血素基因以及多种毒力相关基因。生物学实验显示,该菌可产生明显的溶血现象,而且具有蛋白酶、明胶酶、脂酶和淀粉酶活性,但是缺少卵磷脂酶活性。人工感染实验表明副溶血弧菌D3112具有致病性,感染斑马鱼的半致死剂量约为5×105CFU。药物敏感实验证明了D3112具有多重耐药性。本文对海洋环境中的副溶血弧菌D3112同时开展了基因型和表型研究,为准确评价环境细菌的潜在致病性提供了有用的数据信息。  相似文献

Simple sequence repeat (SSR) markers were obtained for the large yellow croaker Pseudosciaena crocea using 1 205 expressed sequences tags (ESTs) from the NCBI database. Primers for 48 ESTSSR loci were designed and screened with 30 P.crocea specimens captured from Guanjingyang sea area in Fujian Province of China. Sixteen of the loci were polymorphic, which were amplified with 3 to 11 alleles per locus and the mean of 6.13. The observed and expected heterozygosity per locus ranged from 0.091 to 0.844 (mean 0.544) and from 0.118 to 0.892 (mean 0.644), respectively. Polymorphic information content (PIC) ranged from 0.115 to 0.866 (mean 0.593). The results for cross-species amplification of the 16 large yellow croaker EST-SSRs on P. polyactis, C. niveatus, C. lucidus, A. argentatus and J. belengeri revealed that 14, 12, 11, 7 and 6 loci were successfully amplified with 1 to 10 alleles with an average of 4.5 per locus, respectively, which are suitable for population genetics studies of these species and useful for phylogenetic relationship analysis among these species. Overall, this study provides a set of type I markers for population genetics studies and genome mapping for large yellow croaker and its closely related species.  相似文献

研究了石斑鱼肠道原籍嗜冷杆菌SE6在体外的抑菌特性及对人工胃肠液的耐受特性,在此基础上研究了饲料中添加SE6(108CFU/g)对斜带石斑鱼生长性能和免疫功能的影响。体外试验结果表明:SE6对副溶血弧菌、哈维氏弧菌、梅氏弧菌和金黄色葡萄球菌等常见水产致病菌均有一定的抑制作用。pH为4.0的人工胃液能促进SE6的生长,处理2和4 h存活率分别为110.7%和173.0%;pH为3.0的人工胃液处理2 h后活菌数为1.59×105 CFU/cm3,存活率为1.36%,但SE6不能耐受pH为2.0的人工胃液。经浓度分别为0.1%,0.3%,0.5%和0.7%的胆盐处理3 h后,SE6的存活率分别为89.17%,88.33%,77.17%和12.33%。动物试验结果显示:投喂SE6 60 d后试验组饵料系数(0.95±0.01)显著低于对照组(1.03±0.02)。试验组血清补体C3在试验全期均高于对照组,补体C4含量在投喂SE6 60 d时显著上升。血清IgM含量在投喂30 d时显著上升。总之,嗜冷杆菌SE6能抑制多种常见病原菌的生长,而且对模拟胃肠道环境具有一定的耐受能力,添加到饲料中能显著提高石斑鱼饵料利用率,增强动物的免疫功能。  相似文献

克隆了三疣梭子蟹TLR家族的一个新基因,将其命名为PtToll6。该基因全长为4 034 bp,预测分子量为109.078 kDa,理论等电点为6.06,共编码977个氨基酸。SMART预测显示, PtToll6具有Toll样受体典型的结构域,包括前端的序列区(LRR)、跨膜区(TM)和胞内(TIR)区,进化树分析表明其与同属节肢动物门的果蝇Dmtoll9聚为一支。组织表达分布结果显示,PtToll6在肝胰腺中特异性地高表达,其次表达于肠道,在鳃中的表达量最低。采用3种PAMPs进行注射刺激,发现PtToll6对脂多糖的响应最为强烈,在感染48 h后的表达量为对照组的上千倍,推测其可能为PtToll6基因主要识别的病原相关分子模式。PtToll6在人工感染副溶血弧菌后的12 h开始上调表达,至24 h达到峰值(上调9.02倍)。采用RNAi敲降PtToll6后, MyD88的表达量相应下调,同时发现感染副溶血弧菌后的死亡率显著提高,为阴性对照组的1.8倍。实验结果表明:PtToll6基因在抗副溶血弧菌中发挥重要功能。本实验对解析三疣梭子蟹抵御病原入侵的分子机制具有重要指导作用。  相似文献
