The esterification of two model vertebrate steroid hormones - estradiol (E2) and dehidroepiandrosterone (DHEA) - was studied in the oyster Crassostrea virginica. The activity of acyl-CoA:steroid acyltransferase was characterized in microsomal fractions isolated from oyster digestive glands. The apparent Km and Vmax values changed with the fatty acid acyl-CoA used (C20:4, C18:2, C18:1, C16:1, C18:0 or C16:0), and were in the range of 9-17 microM, and 35-74 pmol/min/mg protein for E2, and in the range of 45-120 microM, and 30-182 pmol/min/mg protein for DHEA. Kinetic parameters were also assessed in gonadal tissue. The enzyme saturated at similar concentrations, although conjugation rates were lower than in digestive gland. Preliminary data shows that tributyltin (TBT) in the low microM range (1-50) strongly inhibits E2 and DHEA esterification, the esterification of E2 being more sensitive to inhibition than that of DHEA. Overall, results indicate that apolar conjugation occurs in oysters, in both digestive gland and gonads, at a very similar rate to mammals, suggesting that this is a well conserved conjugation pathway during evolution. Esterification, together with other mechanisms, can modulate endogenous steroid levels in C. virginica, and might be a target for endocrine disrupters, such as TBT.  相似文献

Cholinesterases (ChEs) have been characterized in marbled sole (Limanda yokohamae) for use as a possible biomarker of pollution exposure. In brain, ChEs existed almost exclusively (>95%) as acetylcholinesterase (AChE) whereas in muscle, about 20-30% of ChE activity was in the form of butyrylcholinesterase (BChE; pseudocholinesterase). Acetylthiocholine and butyrylthiocholine (identified in mammalian studies as diagnostic substrates for AChE and BChE respectively) were hydrolyzed mainly, but not exclusively, by these enzymes. The inhibitors BW284C51 and iso-OMPA (identified in mammalian studies as diagnostic inhibitors of AChE and BChE respectively) were not specific for these enzymes in marbled sole. Brain AChE and muscle AChE and BChE were characterized in terms of their kinetic properties (KM etc.) and optimal conditions (substrate concentration, protein concentration, pH etc.) were established to allow routine assays of ChE activity to proceed under pseudo-first order conditions. The sensitivity of ChEs to a locally significant pesticide, iprobenfos (IBP; kitazin) was established in terms of IC50 concentrations. Brain AChE was relatively insensitive to IBP, but muscle AChE and BChE were sensitive to IBP concentrations in the high nM range. However, ambient IBP concentrations in Korean coastal waters are usually not high enough to cause detectable ChE inhibition in this species.  相似文献

Cholinesterases (ChE) from brain, muscle and liver in Nile tilapia (Oreochromis niloticus) were characterized using three substrates: acetylthiocholine iodide, propionylthiocholine iodide, and butyrylthiocholine iodide. Eserine was used as a total ChE inhibitor; BW284c51 and iso-OMPA were used as selective inhibitors for acetylcholinesterase (AChE) and butyrylcholinesterase (BuChE), respectively. The results indicate that AChE is the enzyme present in brain, whereas in both liver and muscle, the presence of atypical ChEs are suggested. These findings indicate that characterization of ChE is necessary prior to use in monitoring programs.  相似文献

Intestinal metabolism plays a significant role in the bioavailability of ingested environmental toxicants. In this study, the potential for first pass, phase 2 biotransformation of benzo[a]pyrene-7,8-dihydrodiol (BaP-7,8-diol) in intestinal mucosa was examined. Sulfotransferase and Uridine 5'-Diphospho-Glucuronyl-transferase activity were measured in cytosol, and microsomes respectively. Radiolabeled conjugation products were analyzed by TLC and high-performance liquid chromatography (HPLC). The results indicated that BaP-7,8-diol was a poor substrate for intestinal sulfotransferase. Vmax for the sulfation of BaP-7,8-diol was 0.002 nmol mg-1 min-1, which is at least three orders of magnitudes lower than the Vmax for phenolic BaP metabolites. Studies with 3'phosphoadenosine-5' phosphosulfate (PAP)-35S as co-substrate showed that an unidentified compound in the reaction mixture was sulfated, dependent on the BaP-7,8-diol concentration. This could indicate that BaP-7,8-diol was interacting with a regulatory site on the enzyme and stimulated sulfation of an endogenous molecule in cytosol. Kinetic analysis of microsomal glucuronidation resulted in a Vmax of 0.30 nmol mg-1 min-1 (+/- 0.06 S.D., n = 4), with a Km of 23.39 microM (+/- 2.66 S.D.). The Km for the co-substrate UDP-glucuronic acid was approximately 43 microM. The slow rates for sulfation and glucuronidation of BaP-7,8-diol may explain its relatively high systemic availability when ingested or produced by intestinal phase 1 enzymes.  相似文献

Polar bears bioaccumulate lipophilic pollutants, including polychlorinated biphenyls (PCBs), into their bodies from their exclusive diet of marine organisms. Hydroxylated PCB metabolites (OH-PCBs) have been found in plasma, presumably due to CYP-dependent biotransformation of PCBs in liver. Little is known about the phase 2 metabolism of hydroxylated xenobiotics in polar bears. The objective of this study was to examine UDP-glucuronosyltransferase (UGT) activity with OH-PCBs and a hydroxylated polycyclic aromatic hydrocarbon, 3-hydroxy-benzo(a)pyrene (3-OH-BaP), in polar bear liver. Samples of frozen polar bear liver were used to prepare microsomes. UGT activity with 3-OH-BaP in Brij-treated microsomes, measured by a fluorescence assay, was readily measurable with protein concentrations in assay tubes of up to 10 g/ml, but dropped off very sharply at higher protein concentrations. The apparent Km for 3-OH-BaP was 1.71 +/- 0.04 microM, and Vmax 1.26 +/- 0.16 nmol/min/mg protein (mean +/- SD, n=3). UGT activities with a model tetrachloro-OH-PCB (4'-OH-CB72) and a model hexachloro-OH-PCB (4'-OH-CB159) were assayed with [14-C]-UDPGA and separation of the [14-C]-glucuronide by ion-pair extraction and thin-layer chromatography. [14-C]-glucuronide conjugates were readily formed by polar bear liver microsomes in the absence of added substrate, apparently from contaminants present in liver. This phenomenon was not observed using hepatic microsomes from laboratory-held catfish. Glucuronidation efficiency was much higher with 4'-OH-CB72 (Km 7.3 microM; Vmax 1.55 nmol/min/mg) than 4'-OH-CB159 (Km 16.1 microM; Vmax 0.46 nmol/min/mg). The identities of the aglycones present in polar bear liver are not known, but could include OH-PCBs or hydroxylated metabolites of other persistent organic pollutants. This study demonstrates that UGT with high activity for 3-OH-BaP and other substrates is present in polar bear liver.  相似文献

This laboratory study describes phenanthrene (Ph) and/or anthracene (An) ability to alter the total thiol redox status (TRS), via depletion of protein free thiols (PSH) and glutathione (GSH) levels, in gills of mussel Mytilus galloprovincialis, after a short-term (7 days) exposure period to each contaminant (at a final concentration of 0.1 mg L−1) or in a mixture (ration 1:1, at a final concentration of 0.2 mg L−1). A number of observable changes, like lysosomal membrane impairment (as detected via the neutral red retention time assay, primarily performed in haemocytes), enhancement of lipid peroxidation byproducts, increased nuclear abnormalities, inhibition of AChE and ALP activity, as well as a significant depletion of PSH and GSH were detected in gills of exposed mussels, in any case. Significant relationships occurred among TRS parameters with each change/stress indices measured in tissues of mussels, could reinforce the use of PSH as a potent biomarker.  相似文献

用紫外分光光度法及荧光分光光度法测定中国对虾肝胰腺及鳃氨基比林-N-脱甲基酶(APND)、乙氧基香豆素-O-脱乙基酶(ECOD)、谷光甘肽-S-转移酶(GST)几种药物代谢酶的活性,探讨黄芩苷对上述几种酶活性的影响.实验结果表明,中国对虾肝胰腺和鳃丝APND活性分别为3.172和2.242 nmol/(min·mgprotein~(-1))、ECOD活性分别为0.325 0和0.357 9 pmol/(min·mgprotein-~(-1))、GST活性分别为70.59和13.97 nmol/(min·mgprotein~(-1)).中国对虾连续7 d投喂黄芩苷药饵(100 mg/kg)后,黄芩苷对几种酶均有不同程度的诱导作用,酶的活性均呈现先升高后降低的趋势.肝胰腺和鳃丝APND活性均在3 h左右达到最高,ECOD活性均在1 h左右达到最高,而GST活性迟于CYP450酶被诱导,2种组织GST活性分别在6,12 h达到最高水平.在临床用药中应考虑到黄芩苷对药物代谢酶的诱导作用,适当用药,合理配伍.  相似文献

The neutral red lysosomal retention assay (NRR) of the haemocytes, and the acetylcholinesterase activity (AChE) in the haemolymph, the digestive gland, the gills and the mantle/gonad complex have been evaluated on mussels Mytilus galloprovincialis collected from Thermaikos and Strymonikos gulfs (northern Greece) in June and October 2001. The validity of performing the above core biomarkers is supported, firstly by their ability to respond to different pollution levels and, secondly, by the significant linear correlation among them. The evaluation of the micronuclei frequency (MN) has been performed in gill tissue and haemocytes of the same mussels and, according to the results, it needs more research in order its use as stress indices to be validated. In addition, the first results on cAMP levels in the gills, the mantle/gonad complex and the digestive gland, whose concentrations correlated to both, NRR and AChE introduce this signal transduction molecule as a new, promising biomarker.  相似文献

Brain acetylcholinesterase (AChE) of some fishes from the coast of Rio de Janeiro State was studied as a possible pesticide biomarker in marine environmental monitoring. AChE specific activity in brain varied from 145 to 530 U/g of proteins and the Michaelis-Menten constant (K(M)) for acetylthiocholine varied from 104 to 291 microM among the 20 species studied. The enzyme sensitivity to methyl paraoxon, evaluated by the inhibition kinetic constants, shows that some species (Paralonchurus brasiliensis and Genidens genidens) are more sensitive (IC50-30 min=455 and 468 nM, respectively). The less sensitive Merluccius hubbsi and Percophis brasiliensis (IC50-30 min=3339 and 3259 nM, respectively) belong to the super-order Paracanthopterygii, which includes the more ancient species. On the other hand, more susceptible species belong to the super-order Acanthopterygii, which includes more recent species. These results suggest a possible evolutionary linkage for AChE sensitivity to methyl paraoxon. The application of inhibition kinetic constants for fish brain AChE in phylogenetic studies is still being investigated. The results have shown that a fish sentinel species should have the highest brain AChE level among the more sensitive ones.  相似文献

经Tris-HCl缓冲液浸提、硫酸铵分级沉淀盐析、DEAE-Sepharose Fast Flow离子交换层析和Sephadex G-75凝胶过滤层析等步骤,从菲律宾蛤仔内脏团中分离得到碱性磷酸酶,SDS-PAGE显示为单一条带,提纯倍数为14.42,比活力达到131.08U/mg。酶学性质研究表明:该酶亚基分子量约44kD,最适反应温度为45℃,最适pH值为9.0,米氏常数Km为0.098mmol/L,最大反应速度Vmax为1.01μmol/(mL·min)。Ca2+、Mg2+对酶活力表现出显著的激活作用。  相似文献

The effects of 7 days’ exposure to differing Triclosan (TCS) concentrations (300, 600, and 900 ng/L) were investigated in the clam Ruditapes philippinarum. Vitellogenin (Vg)-like protein levels in haemolymph and digestive gland from males and females, gill acetylcholinesterase (AChE) activity, superoxide dismutase (SOD) and catalase (CAT) activities in gills and digestive gland, and gill lipid peroxidation (LPO) were measured. The highest TCS concentrations decreased significantly Vg levels in male haemolymph and digestive gland, whereas no significant variations were found in females. The highest TCS concentrations increased significantly SOD activity in gills, but decreased it in digestive gland. No changes in CAT activity were observed. In gills, TCS reduced significantly AChE activity, but it did not induce significant variations in LPO. Our study demonstrates that TCS alters biochemical parameters in R. philippinarum, even at environmentally realistic concentrations, and suggests differing modes of action of the contaminant, in clams at least.  相似文献

Metals are natural components in ecosystems; however, if these elements are in excess, they can have adverse effects on living organisms. This study analyzes the interference of copper, lead, iron and cadmium in acetylcholinesterase (AChE) and carboxylesterase (CbE) activities in zebrafish. AChE was significantly inhibited in vitro by copper, iron, lead and cadmium at higher concentrations (10 and 20 mmol/L), whereas CbE was inhibited only at a concentration of 20 mmol/L. In vivo, only lead and cadmium were able to cause AChE inhibition at higher concentrations, while iron didn't cause any changes, and copper promoted an increase in AChE activity at a concentration of 0.06 mg/L. CbE activity did not change at any of the times (two and seven days) and concentrations tested, except in the case of copper exposure, which resulted in a decrease in CbE activity. Indeed, iodoacetamide treatment didn't changed AChE neither CbE activities, results which indicate that the metal inhibiting effect is probably not due to its biding to thiol groups close the active site of the enzyme. This outcome reveals that metals are important esterase inhibitors in zebrafish, and should be considered in environmental monitoring studies that use esterase inhibition as exposure biomarkers of organophosphate and carbamate pesticides.  相似文献

The results of an intercalibration exercise among the laboratories participating in the MED POL program for monitoring biological effects of pollutants along the Mediterranean coasts are presented. Three established biomarkers, i.e. lysosomal membrane stability, metallothionein concentration and ethoxyresorufin-O-deethylase (EROD) activity, were intercalibrated. The stability of lysosomal membranes in mussels (Mytilus galloprovincialis Lam.) was assessed with a cytochemical method. The four participating laboratories were able to discriminate between control animals (membrane labilization times ranging from 21 to 35 min) and Cu-exposed animals (40 micrograms/l Cu for 3 days) (labilization times ranging from 4.5 to 7.4 min). The metallothionein concentration was evaluated in digestive gland homogenates of control mussels and of animals exposed to 200 micrograms/l Cd for 7 days. The eight participating laboratories were able to discriminate between controls and treated samples using a spectrophotometric method. The EROD activity was evaluated by 11 laboratories. All laboratories were able to discriminate between liver microsomal preparations obtained from control and from benzo-a-pyrene exposed fish (Dicentrarchus labrax), with values ranging from 0.5 to 15.88 pmol/min/mg protein in controls and from 5.41 to 165.13 pmol/min/mg protein in treated animals. Using S9 fractions, it was possible to correctly identify control and treated fish, with a variation similar to that found using microsomal fractions, albeit with an inevitable difference in specific activity. As a corollary, all laboratories involved produced comparable data and were able to identify pollutant-induced stress syndromes in sentinel organisms. Thus, intercalibration enables the use of biomarkers in large biomonitoring programs.  相似文献

In Toulon Bay (France), very high phosphatase activities have been found in the zooplankton fraction>90 microm. This work was intended to specify their origin. For that purpose, larvae, juvenile and adult Crustacea (Copepods: Calanoids, Cyclopoids, Branchiopods: Cladocera, and Cirripeds) were isolated. Their activities were measured using paranitrophenyl phosphate dissolved in sea water in order to calculate Km (the enzyme half saturation concentration) and Vmax (the reaction rate when the enzyme is saturated with substrate). Vmax were referred to protein contents of the isolated organisms to calculate specific activities. For all zooplankton groups high and low affinity phosphatase activities were found. The low affinity enzyme was responsible for at least 70% of the total phosphatase activity. Its specific activity was higher for larvae than for copepodites and adults. In Cirriped nauplii this activity was particularly high with values which were several hundred times higher than that in other Crustacea. These enzymes had optimum pH close to 8.4, magnesium requirement and were competitively inhibited by orthophosphate. Experiments with intact and lysed Cirriped nauplii confirmed that living organisms had only a weak external activity and showed that most of the activity of these larvae was primarily intracellular.  相似文献

利用综合生物标志物响应(IBR)指数对北部湾潮间带沉积环境中多氯联苯(PCBs)、石油烃(TPH)、壬基酚(NP) 和苯并芘(B[a]P)的综合污染水平进行评价。在9个站位采集波纹巴非蛤(Paphia undulata)和沉积物样品,测定沉积物和生物软组织中有机污染物含量,同时对鳃、内脏中与细胞防御系统有关的各7种标志物(包括还原型谷胱甘肽(GSH)、氧化型谷胱甘肽(GSSG)、谷胱甘肽过氧化物酶(GPx)、过氧化氢酶(CAT)等)进行分析。利用相关性分析法筛选出对1~2种有机污染物响应敏感的生物标志物共8种,分别为内脏中的GPx、GSH、GSSG、CAT和鳃中的GPx、GSH、GSSG、CAT。以筛选的8种生物标志物为基础, 9个站位IBR指数范围为0.27~1.48,其中站位S2(湛江市乐民镇码头)、站位S9(湛江市江洪码头)的IBR较小,显示污染程度最轻;站位S4(防城港西湾)、站位S3(钦州湾老人沙浅滩) IBR较高,显示生物体因有机污染产生氧化逆境。不同站位的IBR值与污染源分布、化学污染物分析结果基本一致,表明该方法可用于北部湾潮间带沉积物有机污染程度的综合评价。  相似文献

Brown bullheads (Ameriurus nebulosus) are a demersal freshwater species that can be found in a number of polluted ecosystems. The purpose of the present study was to determine the overall capacity for in vitro glutathione S-transferase (GST) detoxification by brown bullheads, and to see if bullhead GST catalysis was altered in bullheads from a polluted site. Brown bullhead liver cytosolic GSTs catalyzed the conjugation of 1-chloro-2,4-dinitrobenzene (CDNB) over a large range of substrate concentrations, with apparent Km and Vmax for CDNB at fixed nucleophile (glutathione, GSH) concentrations of 1.8-1.9 mM and 12.1-14.6 mumol CDNB conjugated/min/mg, respectively. Bullhead GSTs were also highly active toward other substrates such as ethacrynic acid (ECA), delta 5-androstene-3,17-dione (ADI), and nitrobutyl chloride (NBC). Initial rate GST catalytic activities toward CDNB, NBC, ECA, and ADI were significantly lower in female bullheads from a contaminated lake (Lake Apopka Marsh) as compared to female bullheads inhabiting a nearby control site (Lake Woodruff). No site differences were observed with respect to male bullhead GST activities. These studies suggest that brown bullheads efficiently carry out GST conjugation of diverse electrophilic substrates. However, bullhead GST catalysis may be compromised in bullheads inhabiting polluted ecosystems.  相似文献

有机磷农药(OPs)对乙酰胆碱酯酶(AChE EC 3.1.1.7)具有专一性抑制作用,因此AChE常作为淡水和土壤OPs污染的特异性标识物(biomarker).本文在海水背景下,通过测定两种有机磷农药(马拉硫磷和甲基对硫磷)对海洋鱼类-鲈鱼[Lateolabrax japonicus(cuvier)]脑组织AChE的抑制效应,分析了海鱼AChE监测海水OPs的可行性.结果显示,抑制时间为20min时,抑制程度达到相对稳定;在低浓度OPs抑制下,纯酶液比粗酶液表现出更好的敏感性,对马拉硫磷和甲基对硫磷的灵敏度分别达到0.28μg*L-1和0.2μg*L-1;在一定浓度范围内,AChE活力对OPs具有显著的线性响应.研究表明,利用鲈鱼AChE指示和监测海水OPs的微污染是完全可行的.  相似文献

对两种丝状蓝藻(钝顶节旋藻和盐泽螺旋藻)在基因工程中常用作选择试剂的7种抗生素——氯霉素、氨苄青霉素、红霉素、链霉素、卡那霉素、庆大霉素和新霉素的敏感性作比较实验.结果表明,两种蓝藻对抗生素的敏感性既有共同的特点,也有明显的差异.它们对红霉素、氯霉素和链霉素最敏感,致死浓度分别为0.1,0.5和5μg/cm3.两种蓝藻对氨苄青霉素比较敏感,1μg/cm3的氨苄青霉素即可抑制Arthrospira.341和Spirulina.351的生长,但6d后生长恢复.Arthrospira.341和Spirulina.351对卡那霉素、庆大霉素和新霉素均有抗性,而且存在很大差异:300μg/cm3的卡那霉素对Arthrospira.341的生长仍然没有影响,但对于Spirulina.351,50μg/cm3的卡那霉素即对其生长有明显抑制作用;200μg/cm3的卡那霉素即可将其全部致死.200μg/cm3的庆大霉素和300μg/cm3的新霉素不能抑制Arthrospira.341和Spirulina.351的生长,但在这两种抗生素环境中两种藻的生长状态有很大差异.并验证了氯霉素、红霉素和链霉素是节旋藻和螺旋藻基因转化过程中的有效的抗性选择剂,也从对抗生素敏感性方面表明节旋藻和螺旋藻两个属的遗传差异.  相似文献

三价砷和五价砷对鱼类乙酰胆碱脂酶的体外抑制比较   总被引:1,自引:0,他引:1  
为开展水产品中三价砷的酶法快速检测工作,文中研究了三价砷(As3 + )、五价砷(As5+ )对8种鱼脑中乙酰胆碱酯酶(AChE)的抑制率。结果发现,在10 -7~10 -4mol/L范围内,草鱼和鲫鱼脑中AChE的抑制率与三价砷的浓度负对数值有较好的线性关系,相关系数都在0 .97以上;在10 -6~10 -4mol/L范围内,黑、马鲛、鲈鱼脑中AChE的抑制率与三价砷的浓度负对数值的线性相关系数大于0 .99。对于三价砷,由IC50 得到AChE的敏感性为:美国红鱼>草鱼,鲫,鲈鱼>黑,马鲛>鲤,白鲢。鱼种不同,AChE受到的三价砷、五价砷的抑制作用也不同。在浓度为10 -6~10 -4mol/L范围内,三价砷、五价砷对8种鱼脑中AChE的抑制作用存在极显著差异(p <0 .0 1) ,三价砷对AChE的抑制作用远大于五价砷的抑制作用。  相似文献

Metals are known to influence lipid peroxidation and oxidative status of marine organisms. Hydrothermal vent mussels Bathymodiolus azoricus live in deep-sea environments with anomalous conditions, including high metal concentrations. Although B. azoricus are aerobic organisms they possess abundant methano and thioautotrophic symbiotic bacteria in the gills. The enzymatic defences (superoxide dismutase (SOD), catalase (CAT), total glutathione peroxidase (Total GPx) and selenium-dependent glutathione peroxidase (Se-GPx)) and lipid peroxidation were determined in the gills of B. azoricus exposed to Cd (0.9 microM), Cu (0.4 microM) and Hg (0.1 microM) with different times of exposure. The experiments were performed in pressurized containers at 9+/-1 degrees C and 85 bars. Results show that vent mussels possess antioxidant enzymatic protection in the gills. Cd and Cu had an inhibitory effect in the enzymatic defence system, contrarily to Hg. These enzymatic systems are not completely understood in the B. azoricus, since reactive oxygen species might be produced through other processes than natural redox cycling, due to hydrogen sulphide and oxygen content present. Also the symbiotic bacteria may play an important contribution in the antioxidant protection of the gills.  相似文献
