A new genus and a new species have been established on the basis of three specimens collected from Fujian's coastal zone in China. This new genus is named Laevispinereis. Its brief characters are as follows: oral ring of proboscis has papillae, maxillary ring has conical paragnaths; the peristomium greatly expands into a collar; notosetaes are homogomph spinigers, neurosetaes are homogomph spinigers and homogomph falcigers; tip of spinigers setae with minute serratures and smooth on side. The type of genus is Laevispinereis fujianensis sp. nov., it has conical paragnaths on maxillary ring of proboscis: I has Ⅰ; Ⅱ has 6-7: Ⅲ has 12;Ⅳ has 6-7.  相似文献

于1988年,中国科学院南海海洋研究所在南沙群岛进行综合考察,在采集到的多毛类标本中,发现了背褶沙蚕Tambalagamia的一新种,其与代表种背褶沙蚕T.fauveli和东方背褶沙蚕T.orientalis在眼、双背须出现的刚节数以及背须的形状等有明显的区别,故定名为中华背褶沙蚕Tambalagamia sinica,模式标本保存于中国科学院南海海洋研究所。  相似文献

A molecular phylogeny is presented for the subfamily Dorippinae (including 9 individuals, representing 5 species and 4 genera), based on the sequence data from 16S rRNA gene. Two-cluster test between lineages in these phylogenetic trees has been performed. On the basis of rate constancy, the rate of nucleotide substitutions of 16S rDNA sequence data is estimated as 0.27% per million years. The analysis strongly supports the recognition of the Dorippinae as a monophyletic subfamily. Phylogenetic tree indicates that the subfamily Dorippinae is divided into two main clades, and genus Dorippe appears basal in the subfamily, diverging from other species 36.6 Ma ago. It is also clear that the Heikea is closely related to the genus Neodorippe. The divergence time between them is 15.8 Ma.  相似文献

传统的形态鉴定主要依赖于分类者的经验和水平,容易产生分类错误。为提高物种鉴定的准确性,作者采用形态学与线粒体16S rRNA基因序列分析相结合的方式对广西北海涠洲岛采获的3种珍珠贝进行了鉴定。研究结果表明,基于形态学可将3种珍珠贝分别鉴定为企鹅珍珠贝(Pteria penguin)、宽珍珠贝(P.loveni)和中国珍珠贝(P.chinensis),但基于16S r RNA基因序列,形态学鉴定为P.loveni的标本应为P.lata。P.loveni和P.lata是两个完全独立的种,并非为同物异名。珍珠贝属16S rRNA基因序列的种间遗传距离明显大于种内遗传距离,适宜作为该属种类鉴定的分子标记。在基于16S r RNA基因序列构建的系统树中,相同种类的不同个体均形成单系群,并获得很高的支持率。分布于中国沿海的珍珠贝属种类应包括P.lata,是否有P.loveni的分布尚需进一步证实。  相似文献

以南极海洋细菌为研究对象,采用2216E和MMC液体培养基结合的方法,从菌种库里筛选南极低温降解菌,进行石油烃降解率测定、表型特征确定和16SrRNA分子鉴定,构建系统发育树,并对南极低温降解菌NJ49的低温降解适应性进行了初步研究。结果表明,3株南极海洋细菌NJ41、NJ49和NJ289可以在以柴油为惟一碳源和能源的培养基中生长,被视为南极低温降解菌;16SrRNA分子鉴定结果表明,南极低温降解菌NJ41、NJ49和NJ289分别属于Planococcus、Shewanella和Pseudoalteromonas属;NJ49通过改变脂肪酸组分的膜修饰方式适应低温降解。  相似文献

对古氏非螺(Afer cumingii (Reeve,1844))的齿舌形态以及线粒体CO1和16S rRNA基因序列进行了分析,并以犬齿螺科种类为外群,采用邻接法(NJ)和最大简约法(MP)构建了系统发育树。结果表明,古氏非螺的齿舌具有典型的蛾螺科种类特征;古氏非螺在系统树上与蛾螺科种类聚在一起,而不与犬齿螺科种类聚类。齿舌形态和线粒体基因分析的结果都表明古氏非螺属于蛾螺科,进一步证实了将非螺属修订到蛾螺科的结论。  相似文献

三疣梭子蟹线粒体DNA 16S rRNA和COI基因片段序列的比较研究   总被引:19,自引:2,他引:19  
本文采用 PCR扩增和序列测定等技术 ,对三疣梭子蟹 (Portunustrituberculatus)线粒体DNA1 6Sr RNA和 COI基因片段进行了初步研究。经 PCR扩增和序列测定 ,分别得到 1 6Sr RNA和 COI2个基因片段的碱基序列 ,其中 1 6Sr RNA基因片段的大小为 566bp,碱基 A,T,G,C的含量分别为 35.1 6% ,34.45% ,1 2 .37%和 1 8.0 2 % ;COI基因片段的大小为 658bp,碱基 A,T,G,C的含量分别为 36.63% ,2 6.44% ,2 0 .52 %和 1 6.41 %。在 2种基因片段中 ,AT含量均明显高于 GC含量 ,这与果蝇、虾类、蟹类等无脊椎动物的 1 6Sr RNA和 COI基因片段研究结果相似。通过对三疣梭子蟹 1 6S r RNA和 COI2个基因片段遗传特征的研究 ,发现其种内变异较低 ,在 3个样本中 1 6Sr RNA基因片段序列完全一样 ,COI基因片段中也只有 2个 T/ C位点转换。另外 ,比较本研究所得序列与 Gen Bank中梭子蟹科 5个属的 1 6Sr RNA基因片段序列后 ,发现其聚类结果与传统分类相一致。  相似文献

长臂虾科(Caridea:Palaemonidae)是真虾类中最大的科级单元之一,为了探索长臂虾科内部的系统演化关系,作者采用从Genbank中下载的长臂虾科两亚科(Palaemoninae,Pontoniinae)14属30种长臂虾的线粒体16s rRNA基因序列片段,以鼓虾科(Alpheidae)细足鼓虾(Alpheus gracilipes)为外群分析长臂虾亚科和隐虾亚科之间的系统发育关系。在获得的466个序列位点中,变异位点263个,简约信息位点213个。DNA序列分析显示,隐虾亚科的德曼隐虾属(Manipontonia)与长臂虾亚科的遗传距离(0.211)要小于其与所在亚科的遗传距离(0.245);此属在基于贝叶斯法(BI)和邻接法(NJ)构建的系统发育树中也与长臂虾亚科的种属聚合为一支,然后与隐虾亚科种属聚合的一支成为姊妹群。结合此属与长臂虾亚科尾瘦虾属(Urocaridella)形态相似的特征,建议对德曼隐虾属在长臂虾科中的分类地位进行重新评估。除隐虾亚科德曼隐虾属聚合到长臂虾亚科聚类簇外,两亚科所属的其他种属各自聚合为一支,在系统树中互为姊妹群,支持将长臂虾分为两亚科的分类系统。  相似文献

A new classification of the Korean pleuronectids was proposed based on a molecular phylogeny using specimens collected from Korea (including some Japanese specimens) between 2008 and 2013. A molecular phylogeny based on partial sequences of the two mitochondrial DNA regions (COI and 16S rRNA) supported the reciprocal monophyly of the three genera, Cleisthenes, Pleuronectes and Pseudopleuronectes. We also found that the genus Poecilopsetta is clearly distinct from Pleuronectidae at the family level. Therefore, the previous classification of the Korean pleuronectids should be changed as follows; two families (Pleuronectidae and Poecilopsettidae), 18 genera, and 26 species. Further research is required to resolve the taxonomic uncertainty of the five species in the genus Limanda, which clustered into two clades in our analysis.  相似文献

对3种蛏类大竹蛏(Solen grandis),长竹蛏(Solen strictus)和小刀蛏(Cultellus attenuatus)的线粒体16SrRNA和COI基因片段序列进行了比较并对其系统学进行了初步研究。得到的序列总长度分别为472-481bp(16S)和658bp(COI)。3种蛏序列的碱基组成均显示出较高的A+T比例(16SrRNA基因62.1%;COI基因62.8%)。对位排序比较表明,16SrRNA片段种内个体间变异较小,3种类间存在128个碱基变异位点(其中包括127个简约信息位点)和5个插入/缺失位点,总共12个碱基长;COI片段有200个碱基存在变异,其中包括191个简约信息位点,不存在任何插入/缺失位点。数据分析结果表明16SrRNA和COI基因片段大竹蛏与长竹蛏两片段的遗传距离分别为0.0856和0.1712,两竹蛏类与小刀蛏的遗传距离分别为0.3054,0.2798和0.2662,0.2933。作者认为小刀蛏与竹蛏之间的遗传距离已达到科之间的水平,结果支持将其提升为刀蛏科的分类观点。3种蛏类线粒体16SrRNA和COI基因在种间明显的多态性,证实了16SrRNA和COI基因序列均普遍适用于蛏类种及以上阶元的系统学分析。  相似文献

通过纯培养和16SrRNA基因序列的相似性比对,并构建系统发育树对威海海域柄海鞘共附生细菌的多样性进行了研究。用2216E培养基从柄海鞘样品中分离到78株细菌,通过形态学特征和TCBS培养基选择性筛选后,选取30株具有代表性的菌株进行了16SrRNA基因测序和基于16SrRNA基因序列的系统发育多样性分析。结果表明,30株菌株分别属于细菌域的4个系统发育类群(Actinobacteria,Bacteroidetes,Firmicutes,Gro-teobacteria)的11个科,11个属。根据16SrRNA基因序列,大多数菌株与其系统发育关系最密切的模式菌株之间存在一定的遗传差异(16SrRNA基因序列相似性为91.82%~98.93%)。其中菌株HQW7,HQYD1,HQWD4,HQE1和HQE2与相关模式菌株的16SrRNA基因最高相似度仅分别为91.82%,92.51%,92.99%,93.52%,93.87%,这5株菌可能代表新的分类单元。威海海域柄海鞘共附生细菌存在着较为丰富的物种多样性和系统发育多样性,并可能存在新的物种。  相似文献

Fresh and preserved (type material) specimens of the black ghost chimaera Hydrolagus melanophasma were compared for morphometric characteristics. A molecular comparison was also performed on two mitochondrial gene sequences (12S rRNA and 16S rRNA gene sequences). While significant differences in measurements were found, the differences were not attributable to sexual dimorphism or the quality of the specimens, but to the sample size and the type of statistical tests. The result of the genetic characterization showed that 12S rRNA and 16S rRNA genes represented robust molecular markers that characterized the species.  相似文献

We report on a hexactinellid sponge new to science, Walteria demeterae sp. nov., which was collected from the northwestern Pacific seamounts at depths of 1271–1703 m. Its tubular and basiphytous body, extensive lateral processes, numerous oval lateral oscula which are irregularly situated in the body wall, the presence of microscleres with oxyoidal, discoidal and onychoidal outer ends, and the absence of anchorate discohexasters, indicate it belongs to the genus Walteria of family Euplectellidae, which is also supported by molecular phylogenetic evidence from 18S, 28S, 16S rRNA and cytochrome c oxidase subunit I (COI) gene sequences. The unique morphotype, which is structured by a thin and rigid framework of body wall and lateral processes consisting of diactins, characterizes it as a new species. Local aggregations of individuals of this new species coupled with their associated macrofauna in the Suda Seamount are reported, highlighting its functional significance in providing biogenic microhabitats in the deep sea.  相似文献

采用线粒体COⅠ和12S rRNA基因片段作为DNA条形码,分析珠江口春季鱼卵和仔稚鱼种类组成和分布特征,并探究两种条形码在鱼卵和仔稚鱼种类鉴定中的适用性。研究共扩增样本391个,成功鉴定的鱼卵和仔稚鱼共7目25科42属60种(2种未鉴定到种)。其中,以鲈形目(Perciformes)种类和数量最多,种类数占比为51.6%,数量占比为47.91%;其次为鲱形目(Clupeiformes),种类数占比为25%,数量占比为34.56%。优势种10种,其中凤鲚(Coilia mystus)优势度最高,为0.071;棘头梅童鱼(Collichthys lucidus)最低,为0.014。COⅠ和12S rRNA基因片段扩增结果显示,鱼卵和仔稚鱼12S rRNA基因片段扩增成功率(95.60%)明显高于COⅠ基因(43.22%)。遗传距离和ABGD分析显示,COⅠ基因种内遗传距离为0~0.005(平均0.003),种间遗传距离为0.061~0.376(平均0.253),两者间存在明显的“条形码间隙”,ABGD划分结果与数据库比对结果一致;12S r RNA基因种内遗传距离为0~0.011(平均0...  相似文献

DNA条形码不仅为物种鉴定提供了有效方法,而且也有助于分类学和生物多样性研究。本研究旨在探讨将COI和16S rRNA基因序列应用于中国沿海蜑螺科贝类物种鉴定的可行性,获得了该科3属7种贝类61个个体的COI和16S rRNA基因序列。基于COI基因序列的种内遗传距离为0.00—1.29%,平均为0.67%;属内种间遗传距离为4.62%—19.25%,平均为13.02%;基于16S rRNA基因序列的种内遗传距离为0.00%—0.48%,平均为0.23%;属内种间遗传距离为2.47%—8.48%,平均为6.37%。两种基因序列在所研究的蜑螺中,种内遗传差异均小于种间遗传差异,存在明显的条形码间隙,所有物种在系统发生树上都表现为独立的单系群。结果表明,线粒体COI和16S rRNA基因序列可以作为DNA条形码标准基因对蜑螺科贝类进行有效地物种鉴定。  相似文献

利用线粒体16SrRNA基因片段及核糖体DNA转录间隔区ITS1序列分析的方法,以近缘属的青蛤(Cyclinasinensis)为外群,对文蛤属的文蛤(M.meretrix)、丽文蛤(M.lusoria)、帘文蛤(M.lyrata)和斧文蛤(M.lamarckii)4种贝类进行了系统学研究。经ClustalX多重比对及DNAsp软件分析后,用PAUP4.0分析软件,计算出种间序列的碱基转换/颠换频率,利用邻接法NJ构建系统发育树。结果表明,帘文蛤与该属其他三种的分歧时间较早,两类序列与其他种类间的相对遗传距离分别在0.25866—0.28218和0.15644—0.20104之间。文蛤与丽文蛤间的16SrDNA及ITS1序列间的遗传距离仅为0.04805和0.04201,拓扑结构图显示关系很近,并且二者的分布区大面积重叠,其序列间的转换/颠换频率接近种内单元型(haplotype)间的序列变化,鉴于它们的贝壳形态有一定差别,应归为同一个种内的不同地理亚种比较恰当。  相似文献

A systematic assessment of Planctomycetales diversity in a South China Sea, deep‐sea sediment (1657 m) was conducted using the 16S rRNA gene analysis approach. PCR amplification of the samples from seven sediment layers (0.1, 1, 3, 5, 7, 9 and 11 m below the surface sediment) using the primer set Pla‐46‐F/1392‐R showed that the Planctomycetales existed within a limited range of sediment depths (≤ 5 m), and had a decreasing trend in diversity with increasing depth. The majority of the retrieved Pla‐46‐F/1392‐R sequences belonged to Pirellula‐related Planctomycetales, and two sequences retrieved from the 0.1‐m layer (GenBank accession numbers: DQ996944 and DQ996945 ) shared the same anammox‐related signature oligonucleotides and were closely related to commonly recognized anammox organisms. To identify new anammox‐related biomarkers, three primer sets were designed for amplifying the fragments of hydroxylamine oxidoreductase and S‐adenosylmethionine radical enzyme genes, but no related sequences were found. Our multiple 16S rRNA gene primer sets (Journal of Rapid Methods and Automation in Microbiology, 2008, in press) revealed even an higher diversity of Planctomycetales in the 0.1‐m layer of the sediment, especially at genus level. Our data profiled the distribution pattern of Planctomycetales diversity along sediment depths, and provided molecular evidence for the existence of anammox‐related bacteria in a new location, which broadens our understanding of Planctomycetales diversity in deep sea sediments.  相似文献

2013年青岛输油管道爆炸,大量石油污染了附近海岸。课题组采集了污染的沉积物样品,以原油为唯一碳源和能源,富集了四个石油降解菌群。生物多样性和群落分析表明,Luteibacter、Parvibaculum 和属于食烷菌科的一个属是降解菌群的主要优势菌,都属于变形菌门。从石油降解菌群中分离筛选,获得了9株具有不同16S rRNA基因序列的降解菌,分别属于8个属。重量法测定降解菌的石油降解率,其中5株的石油降解率大于30%。GC-MS分析结果表明,石油降解菌多倾向于降解烷烃,对多环芳烃的降解能力较差,其中5株细菌的烷烃降解率较大,仅1株菌D2对多环芳烃的降解率较大,其降解率在34.9%到77.5%。通过对高通量数据的分析,表明食烷菌属是菌群A和菌群E的主要降解菌群,其中筛选获得的菌株E4可能是菌群E的一株优势降解菌。本研究所筛选菌株证明了其石油降解潜力,为油污染海滩生物修复提供了菌株资源。  相似文献

One new species of free-living marine nematodes Enoplus taipingensis sp.nov.from the rocky intertidal seaweeds in the Taiping Bay,Qingdao is described and illustrated with scanning electron microscopy observation and 18S rRNA gene sequence analysis.Enoplus taipingensis is characterized by special arrangement of lateral setae on the tail,three cervical setae,distal structure on the supplement,and much longer body length with bigger De Man ratio "a".Type specimens are deposited in the College of Marine Life Science,Ocean University of China.  相似文献
