Design and application of the method for isolating magnetotactic bacteria   总被引:1,自引:0,他引:1  
A simple apparatus was designed to effectively isolate magnetotactic bacteria from soils or sediments based on their magnetotaxis. Through a series of processes including sample incubation, MTB harvesting, isolation, purification and identification, several strains of bacteria were isolated from the samples successfully. By Transmission Electron Microscopy (TEM) and Energy-Dispersive X-ray Analysis (EDXA), these bacteria were certificated to be magnetotactic bacteria. The phylogenetic relationship between the isolated magnetic strains and some known magnetotactic bacteria was inferred by the construction of phylogenetic tree based on 16SrDNA sequences. This apparatus has been proven to have the advantages of being inexpensive, simple to assemble, easy to perform and highly efficient to isolate novel magnetotactic bacteria. The research indicated that the combined approach of harvesting MTB by home-made apparatus and the method of plate colony isolation could purify and isolate magnetotactic bacteria effectively.  相似文献

Geotopes or geological sites provide information on the evolution, structure and properties of the Earth‘s crust and on the geological past. They also represent the great variety of features that characterize a landscape, For this reason, their preservation is of special concern to geosciences. As early as in the 19th century, outstanding rock formations became important tourist sites although the modern terms ““geosites““ and ““geotourism““ were unknown. Although magnetotelluric sounding method applied to the land is advanced, there are many difficulties when it is applied to marine environment, one of which is how to lay magnetic field sensors down to the seafloor to complete measurements. To protect the magnetic field sensors from intense erosion and high pressure, suitable high-pressure sealed cabins must be designed to load them. For the consideration of magnetic measurement and marine operation, the sealed pressure cabin should be nonmagnetic and transportable. Among all optional materials, LC4 super-hard aluminum alloy has the highest performance of price/quality ratio to make the sealed pressure cabin. However, it does not mean that the high-pressure sealed cabin made using LC4 will be perfect in performance. In fact, because of its weak magnetism, the pressure cabin made using LC4 has distorting effect on frequency responses of the magnetic field sensors sealed in it. This distorting effect does not affect the use of the magnetic field sensor, but if we want to eliminate its effect, we should study it by experimental measurements. In our experiment tests, frequency sweep magnetic field as excitation signal was used, and then responses of the magnetic field sensor before and after being loaded into the high-pressure sealed cabin were measured. Finally, normalized abnormal curves for the frequency responses were obtained, through which we could show how the high-pressure sealed cabin produces effects on the responses of the magnetic field sensor. Experimental results suggest that the response distortion induced by the sealed pressure cabin appears on mid- andhigh-frequency areas. Using experimental results as standardization data, the frequency responses collected from seafloor magnetotelluric measurements can be corrected to restore real information about the seafloor field source.  相似文献

Research on international student’s post migration plans treats migration as a binary stay-return category and focuses on push-pull factors as the cause of this migration. In this paper we expand the definition of migration and consider the role of life experiences and aspirations, particularly the concept of home. We ask, what are the different conceptualizations of home and how are these tied to differential migratory plans? We analyze data from 232 interviews with international students from more than 50 countries who attended a flagship public university in Canada from 2006–2013.We find that students have four ways of thinking about home: as host, as ancestral, as cosmopolitan, and as nebulous. These understandings of home correspond to particular post-migration plans. While students who view home as a host plan to stay, and those who view home as ancestral plan to return, those with cosmopolitan and nebulous conceptions of home have more open migration plans.  相似文献

A Dirichlet–Neumann representation method was recently proposed for upscaling and simulating flow in reservoirs. The DNR method expresses coarse fluxes as linear functions of multiple pressure values along the boundary and at the center of each coarse block. The number of flux and pressure values at the boundary can be adjusted to improve the accuracy of simulation results and, in particular, to resolve important fine-scale details. Improvement over existing approaches is substantial especially for reservoirs that contain high-permeability streaks or channels. As an alternative, the multiscale mixed finite-element (MsMFE) method was designed to obtain fine-scale fluxes at the cost of solving a coarsened problem, but can also be used as upscaling methods that are flexible with respect to geometry and topology of the coarsened grid. Both methods can be expressed in mixed-hybrid form, with local stiffness matrices obtained as “inner products” of numerically computed basis functions with fine-scale sub-resolution. These basis functions are determined by solving local flow problems with piecewise linear Dirichlet boundary conditions for the DNR method and piecewise constant Neumann conditions for MsMFE. Adding discrete pressure points in the DNR method corresponds to subdividing faces in the coarse grid and hence increasing the number of basis functions in the MsMFE method. The methods show similar accuracy for 2D Cartesian cases, but the MsMFE method is more straightforward to formulate in 3D and implement for general grids.  相似文献

The Zermatt–Saas-Fee ophiolites (ZSFO) are one of the best preserved slices of eclogitic oceanic crust in the Alpine chain. They formed during the opening of the Mesozoic Tethys and underwent subduction to HP/UHP conditions during Alpine compression. A cathodoluminescence-based ion microprobe (SHRIMP) dating of different zircon domains from metagabbros and oceanic metasediments was carried out to constrain the timing of formation and subduction of this ophiolite, two fundamental questions in Alpine geodynamics. The formation of the ophiolitic sequence is constrained by the intrusion ages of the Mellichen and the Allalin metagabbros (164.0 ± 2.7 Ma and 163.5 ± 1.8 Ma) obtained on magmatic zircon domains. These data are in line with the maximum deposition age for Mn-rich metasediments which overlie the mafic rocks at Lago di Cignana (161 ± 11 Ma) and at Sparrenflue (ca. 153–154 Ma). An Eocene age of 44.1 ± 0.7 Ma was obtained for whole zircons and zircon rims from an UHP eclogite and two metasediments at Lago di Cignana. One of the Eocene zircons contains a rutile inclusion indicating formation at HP conditions. As the temperature and pressure peak of these rocks nearly coincide, the Eocene zircons probably constrain the age for the deepest subduction of the ZSFO. This Eocene age for the UHP metamorphism implies that the ZSFO were subducted later than the Adriatic margin (Sesia-Lanzo Zone) and before the Late Eocene subduction of the European continental crust below Apulia. A scenario with three subduction episodes propagating in time from SE to NW is proposed for the geological evolution of the Central and Western Alps. We consider linear and nonlinear spectral-correlational and kinetic models for fluctuations in the motion of the Earth based on the Fokker-Planck-Kolmogorov and Pugachev equations. Climate change is projected to increase the frequency and severity of extreme weather events. As a consequence, economic losses caused by natural catastrophes could increase significantly. This will have considerable consequences for the insurance sector. On the one hand, increased risk from weather extremes requires assessing expected changes in damage and including adequate climate change projections in risk management. On the other hand, climate change can also bring new business opportunities for insurers. This paper gives an overview of the consequences of climate change for the insurance sector and discusses several strategies to cope with and adapt to increased risks. The particular focus is on the Dutch insurance sector, as the Netherlands is extremely vulnerable to climate change, especially with regard to extreme precipitation and flooding. Current risk sharing arrangements for weather risks are examined while potential new business opportunities, adaptation strategies, and public–private partnerships are identified.  相似文献

Establishing the age and crustal nature of exotic terranes and their underlying basements helps to determine their paleogeographic origin and tectonic histories. We present U–Pb ages of zircons and Sm–Nd whole rock isotopic data for volcanic and plutonic rocks of the Carolina terrane, one of several peri-Gondwanan terranes that were accreted to the margins of the circum-Atlantic continents during the Paleozoic. Volcanism in this subduction-related arc culminated in the eruption of the Morrow Mountain rhyolite, at ca. 540 Ma; thus, magmatism in the Carolina terrane ceased at the beginning of the Cambrian. The presence of inherited zircons and non-juvenile depleted mantle model ages of Carolina slate belt rocks favor a basement that is, at least in part, composed of evolved continental crust. Ages of inherited xenocrystic zircons cluster at ca. 1000, 2100 and 2500 Ma. These ages, in addition to volcanism at ca. 618–540 Ma, correlate best with well-known tectonic events in present-day northern South America. Specifically, the Orinoquian-Sunsas, the Trans-Amazonian and the Central Amazonian orogenic zones are likely candidates for potential basement correlatives to the Carolina terrane. Sm–Nd isotopic signatures vary significantly, but permit assimilation of Orinoquian age (1000 Ma) crust by magmas derived from the depleted mantle in a subduction (arc-related) setting. Our findings are also consistent with proposed correlations between the Carolina terrane and Avalonia which is likewise believed to have formed along the northern margin of present-day South America.  相似文献

Polynuclear aromatic hydrocarbons (PAHs) and black carbon (BC) in sediments are powerful tools in the identification of the combustion process throughout geologic history. In this study, combustion-derived PAHs and BC were carefully investigated in sediments from the Global Stratotype Section and Point of the Permian-Triassic (P/Tr) boundary in Meishan, China. Quantitative analyses of combustion-derived PAHs and BC demonstrate anomalously high concentrations in the boundary event beds that coincide with the mass extinction horizon. The prevalence of parent polynuclear aromatics (e.g., phenanthrene) in PAHs, together with non-metric multidimensional scaling analysis, confirms that the PAHs are mainly derived from vegetation burning instead of having a coal and/or oil origin. BC detected in sediment occurs in various forms from large irregular charcoal particles to fine aciniform soot, with an equivalent reflectance of up to 3.5%. The results strongly suggest that a wildfire occurred during the P/Tr boundary, which served as one of the possible triggers of mass extinction on land. The wildfire occurrence indicates that the O2 concentration of the atmosphere during (or before) the P/Tr mass extinction was probably >17%. The temporal coincidence of the mass extinction with intensive volcanic eruption, marine anoxia and wildfire events in the region of the Meishan section provides new insight into the mechanisms of the P/Tr biotic crisis. Our results show that wildfires could have played an important role in the collapse of the ecosystem in the Meishan P/Tr events.  相似文献

For the semi-arid environment of southern California, a reach-scale classification system and conceptual model were created to synthesize the observed floodplain forms into three basic floodplain continuums: armored, non-armored, and active-regional alluvial fan. These continuums are comprised of three to five alluvial floodplain forms (cascade, step-pool, plane-coarse-bed, plane-mixed-bed, plane-fine-bed, pool-riffle, braided, and dune-ripple). For the non-armored floodplain continuum, a floodplain state diagram has been generated to quantitatively describe the natural downstream progression of floodplain forms and geometry, using specific stream power and the width-to-depth ratio as the state and shape metrics, respectively. Based on field data for 91 cross sections along 14 watercourses, this floodplain state diagram provided the basis to define geomorphic thresholds for floodplain instability and braiding using logistic regression analyses. This method for defining floodplain instability thresholds provides a unique management tool by providing both a systematic and a quantitative means to assess the stability state of watercourses and, thereby, provide a means for assessing the potential for the hazards associated with watercourse instability.  相似文献

GPS results from 25 stations in Macedonia measured in 1996 and 2000 show that Macedonia moves SSE relative to Eurasia essentially as a single crustal piece along with parts of westernmost Bulgaria. Geological studies show active N–S normal faults and two NNW-striking right-lateral faults in western Macedonia, and NW-trending left-lateral faults SE Macedonia, with a region in central Macedonia essentially devoid of active faults. Distribution of seismic activity supports the geological studies. However, the GPS results cannot discriminate the active faulting, except perhaps in the northern part of Macedonia in the Skopje and adjacent areas, where active ~NS extension occurs. Slip-rates on the strike-slip faults must be low, in the range of 0–2 mm/year. There is a progressive increase in GPS velocities southward in northern Greece toward the North Anatolian fault zone, across which the velocities increase and change direction dramatically.  相似文献

The observed mass distribution for the compact remnants of massive stars (neutron stars and black holes) and its relationship to the possible mechanism of ejection of the envelopes of type II and Ib/c supernovae are analyzed. The observed lack of compact remnants with masses 1.5–3 M suggests a magneto-rotational mechanism for the supernovae, and a soft equation of state for neutron stars with limiting masses near 1.5 M . The observational consequences of this hypothesis are discussed.  相似文献

The western Ireland Ordovician stratigraphy has been previously used to constrain the timing of docking of an island arc and its fore‐arc basin with the margin of Laurentia for the British and Irish Caledonides. New field relationships and age data indicate that one of the key formations, the Rosroe Formation (459.2 ± 0.8 and 465.1 ± 2.1 Ma), and its supposed lateral equivalent, the Maumtrasna Formation are younger than previously interpreted. New age data for a tuff band in the Maumtrasna Formation (468.9 ± 1.3 Ma) also support previous studies showing it can be correlated to the adjacent Mweelrea Formation. The new field evidence, age data and geochemistry contradict some previous studies and show that the Maumtrasna, Rosroe and Derrylea formations can no longer be considered lateral equivalents. Based on the new stratigraphy a revised tectonic model is required with sedimentation in this part of the Caledonides taking place in a fore‐arc basin outboard of a continental arc and the oceanic arc was an along‐strike equivalent of this arc situated in an embayment of the Laurentian margin.
