The accumulation of cadmium from seawater by the blue crab, Callinectes sapidus, was studied as a function of metal concentration and exposure time, with special emphasis on cadmium-binding proteins. Cadmium was found, in decreasing order of magnitude, in gills, digestive gland and hemolymph. When exposed to 0·5 ppm cadmium for 2–24h, virtually all of the cadmium in the cytosolic fraction of the gill was associated with a low molecular weight (LMW) cadmium-binding protein (MW 8000). However, after 48h of exposure only 50% of the cadmium in the cytosol was bound to this protein. The rest wasfound to be associated with proteins of a molecular weight of 300 000 and 60 000. This pattern of cadmium distribution did not change over a 12-day depuration period. Similar results were obtained upon exposure to 0·1 ppm cadmium. The pattern of cadmium accumulation in the cytosolic fraction of the digestive gland was in marked contrast to that observed for the gill. Initially, the cadmium was distributed over three low molecular weight fractions. During depuration the distribution of cadmium changed and all of the metal became bound to a low molecular weight protein (MW 9000). The cadmium concentrations in the gill and digestive gland remained essentially constant during depuration (12 days). The LMW cadmium-binding proteins were purified by a combination of gelpermeation and ion-exchange chromatography. Their molecular weight, spectral properties and amino acid composition are characteristic of the vertebrate metallothioneins. During exposure to cadmium the metal rapidly appeared in the hemolymph, mainly associated with hemocyanin. During depuration cadmium was transferred from the hemolymph to the digestive gland, demonstrating that hemocyanin acts as a carrier in trace metal transport.  相似文献

This paper examines the potential availability to a demersal fish of cadmium and zinc associated with digested sewage sludge, via a food chain as well as directly from the sludge, and the tissue distribution and possible excretion of any accumulated cadmium and zinc. Radioactive tracer techniques were used in order to follow the food chain transfer of the metals. Flounder (Platichthys flesus) accumulated cadmium both directly from sludge (delivered in newly ingested unassimilated gutfuls in the amphipods) and in assimilated form from the tissues of the amphipod crustacean Corophium volutator that had been feeding on a sludge–sediment mixture. Cadmium from both sources could be excreted by the fish. The concentration of cadmium within the whole fish increased as the input of cadmium in the diet increased. Zinc, similarly, is available to the flounder both directly from sludge and from zinc accumulated in C. volutator. Only a limited increase in net accumulation of zinc by the flounder was observed upon increased inputs of zinc through the diet, perhaps indicating some regulation of body zinc concentration by the flounder.  相似文献

The processes of bioaccumulation and elimination of cadmium by tissues of Styela clava (Herdman 1881) were investigated based on a semi-static two-compartment model. The kinetic parameters (accumulation rate constant k 1, elimination rate constant k 2, bioconcentration factor BCF, biological half-life t 1/2 and maximum equilibrium concentration C Amax) were obtained by non-linear regression. The results showed that S. clava accumulates cadmium from its aquatic environment. BCF decreased with increasing metal concentration in the water, and when the accumulation achieved balance, C Amax correlated positively with metal concentrations in the water. Concentrations of cadmium in different tissues of S. clava were: gonad > digestive gland ≈ other parts > tunic. The t 1/2 values for cadmium were 15.54-50.40 days in the accumulation process and 11.53-24.55 days in the elimination phase. The high rate of accumulation and elimination of cadmium from the body reveals the potential of the organism to be used as a biomonitor of short-term cadmium fluctuation in marine systems.  相似文献

Laboratory experiments were carried out to study cadmium (Cd) toxicity in an intertidal gastropod, Turbo intercostalis. Tolerance studies revealed an LC50 of 6.18 ppm for 96 h. Experiments were also conducted to determine oxygen consumption and bioaccumulation of the metal by exposing the snails to a sublethal concentration of Cd (1.24 ppm) for 20 days. A significant fall in oxygen consumption was observed from 48 h onwards with maximum decrease (65.5%) at 20 days. The accumulation of Cd was determined in gonad-digestive gland (GDG) complex, mantle and foot at different intervals. There was a gradual increase in accumulation with exposure time in all tissues. The accumulation of Cd was: GDG complex>mantle>foot. The percentage body burden of the metal showed a decreasing trend in the mantle and an increasing trend in GDG complex. The fractional retention coefficients increased with exposure time. The snails exhibited a tissue-specific depuration of the metal and the trend was similar to that of accumulation. The results suggest that T. intercostalis may be a useful species for biomonitoring studies.  相似文献

Following the in vivo exposure of dab (Limanda limanda L.) to cadmium chloride, kidney phagocytes were collected and their respiratory burst measured in vitro using chemiluminescence. Fish were exposed to mean measured concentrations of 1.3, 2.7 and 5.5 mg Cd litre−1 (as total cadmium ion) for a total of nine weeks, followed by a three week depuration period in clean sea water. Compared with control fish, the respiratory burst of kidney phagocytes from dab sampled after six weeks was significantly reduced in the 2.7 and 5.5 mg Cd litre−1 treatments (Steel's test, p < 0.05). Significant reductions were observed in the respiratory burst of phagocytes from all cadmium exposed fish compared with control fish after nine weeks (Steel's test, p < 0.05). After a further three week depuration period in clean sea water, the respiratory burst of phagocytes from fish previously exposed to 1.3 and 2.7 mg Cd litre−1 were still significantly less than in the control group (Steel's test, p < 0.05). Muscle tissue cadmium concentrations were also analysed, although there was no clear relationship between the muscle total cadmium levels and kidney phagocyte chemiluminescence. The results are discussed with respect to the possible mechanism(s) of cadmium immunotoxicity in dab and recommendations made for future work.  相似文献

Metallothionein (MT) has been proposed as a specific biochemical probe for metal exposure of aquatic organisms. It is recognized that its usefulness as a monitoring tool depends on the full understanding of its function and on the possibility of measuring its concentration in tissues. Therefore the study of MT in crustaceans is interesting from two different points of view: the need to understand mechanisms associated with the toxicology of metals, and the potential use of this protein for monitoring metal-contamined environments. Several methods have been developed for quantitative measurements of MT in biological samples. Immunochemical and electrochemical procedures are reliable techniques but they require sophisticated equipment and highly trained staff. In the present work, modifications to the silver-saturation method have been developed for the quantitative determination of MT in crustacean tissues, estimating silver concentrations by flame in an atomic absorption spectrophotometer. Cadmium and MT were measured both in Artemia and Procambarus clarkii after sublethal exposure to cadmium. An increase in MT content was recorded in crustacean tissues after 12 h of cadmium exposure. A high responsiveness was found in MT induction by cadmium in the two crustaceans and there was a clear relationship between cadmium concentration in water and MT levels in tissues. The silver-saturation assay may be a rapid assay technnique for biomonitoring purposes.  相似文献

The population dynamics and morphological and functional characteristics (the concentration of hemocytes in the hemolymph and the level of histopathology) of the bivalve Anadara sp. cf. Anadara inaequivalvis were studied in 2005–2007 on the North Caucasian coast of the Black Sea. A drastic decline in the abundance of the bivalves simultaneously with cadmium’s exceedence in their soft tissues and bottom sediments were recorded in May of 2007. Six months before the manifestation of the population effects, morphological and functional changes (a decrease in the content of hemocytes in the hemolymph and an increase in the content of brown cells in the connective tissue and lipofuscin-like pigment granules in the digestive gland epithelium) in the bivalves were detected that were the consequences of an external effect (presumably, cadmium intoxication). Thus, the morphological and functional changes of the individual Anadaras may be a promising tool for the early detection of the impact of pollutants on benthic ecosystems.  相似文献

Organisms have evolved a cellular response called stress protein response that increases their tolerance in adverse environmental conditions. Well known stress proteins that bind essential and toxic metals are metallothionein (MT). The scallop Mizuhopecten yessoensis is the most interesting organism because it is able to accumulate toxic cadmium in its digestive gland. However, in the tissue of the digestive gland of Mizuhopecten yessoensis MT (metallothioneins) have not been found. Eastern scallops, Mizuhopecten yessoensis, were collected from two locations ?? one clean and one polluted site. The concentrations of cadmium (Cd), copper (Cu) and zinc (Zn) were measured in the digestive gland. There was a significant increase in Cd concentrations in this studied tissue. We found that in the presence of cadmium Mizuhopecten yessoensis can induce high molecular proteins. The results of experiments have shown that Cd-binding ligands have a number of properties similar to MT: acetone and temperature stability; the ability to bind some metals, including Cd, Cu and Zn. Protein chromatography (FPLC, Superosa 12) from the digestive gland of scallop M. yessoensis has shown that cadmium is associated with high molecular weight Cd-binding proteins (72 kDa and 43 kDa). The major cadmium-binding protein 72 kDa is glycoprotein. In experiments we have demonstrated that Cd-binding proteins can be induced when there is cadmium exposure. The results of this study strongly suggest that the far eastern scallop Mizuhopecten yessoensis has a unique and well-developed system for the detoxification of heavy metals and it allows for biochemical systems to be maintained in a relatively stable manner in the presence of heavy metals.  相似文献

通过暴露染毒方法研究了栉孔扇贝对镉离子(Cd2+)的35d富集与30d释放规律。结果表明:栉孔扇贝对镉有很强的富集性,体内的镉含量与暴露溶液里的Cd2+浓度呈正相关;栉孔扇贝对镉的释放速率随着暴露水体中镉浓度的增加而减慢。应用双箱动力学模型,通过非线性拟合得到了栉孔扇贝对Cd2+的吸收速率常数k1随着外部水体中镉浓度的增大而减小,生物富集系数BCF则随着外部水体中镉浓度的增大而减少。当镉浓度为0.05mg/L时BCF仅为3 730,而当镉浓度为0.000 5mg/L时BCF达到1.79×1011。平衡状态下栉孔扇贝体内镉含量(CAmax)随着水体中镉浓度增加急剧下降,与水体中镉浓度呈负相关;排出速率常数k2、生物学半衰期B1/2与低浓度镉无明显相关性,随着镉浓度的增大k2显著降低而B1/2显著增大。验证了双箱动力学模型适用于栉孔扇贝对Cd2+的生物富集与释放规律研究。  相似文献

Sequestration of large quantities of vitellogenin (VTG) is critical for proper oocyte development in most oviparous vertebrates. While previous studies have shown a general correlation between oocyte growth and the accumulation of various exogenous and endogenous ligands, few studies have attempted to elucidate the role VTG plays in this maternal transfer. In the present study, we have demonstrated that oocytic accumulation of [3H]-2,3,7,8-tetrachlorodibenzo-p-dioxin (3H-TCDD) and [14C]-benzo(a)pyrene (14C-BaP) by gravid Fundulus heteroclitus is directly correlated with oocyte maturational status. A positive correlation was observed between oocyte maturational state (size) and both total quantity (total pmoles) as well as concentration (pmoles/g tissue) of TCDD and BaP. Further, the bi-phasic accumulation of both TCDD and BaP mirrors that previously observed for protein sequestration by cultured Fundulus oocytes. Additionally, both TCDD and BaP were associated with VTG in vivo. HPLC analysis of serum extracts has shown that VTG associates with both parent BaP and BaP metabolites. Studies with 125I-T4 (thyroxine) also suggest that this critical hormone also associates with VTG in vivo. Ongoing studies are examining the VTG-associated transport and oocytic sequestration of 125I-T4 in gravid Fundulus.  相似文献

Metallothioneins and lysosomes are known to be involved in cellular detoxication and sequestration of certain metals1–3 and both have been identified in this role in elimination of copper from marine mussels (Mytilus edulis/galloprovincialis).3 Cadmium (Cd), however, has been shown to persist in the cells of the digestive gland for long periods with only minimal elimination. An experiment was designed to test the effects of Cd on the fragility of lysosomal membranes in the digestive cells as a measure of cellular injury,4,5 metallothionein content of the digestive gland and cadmium concentration in this organ. Phenanthrene was used also to destabilise lysosomal membranes6 in order to test if increased lysosomal fragility interfered with cadmium metabolism and detoxication. The results demonstrated that Cd induced metallothionein synthesis and that elimination of Cd was minimal after 28 days in clear seawater. Lysosomal fragility was initially increased but this effect was soon reversed, even with continued exposure to Cd. The lysosomal destabiliser, phenanthrene, did not appear to affect accumulation of Cd or levels of metallothionein.  相似文献

茎柔鱼(Dosidicus gigas)是重要的大洋性经济头足类,其性腺组织发育滞后于个体生长,但在性腺发育开始后生殖能量积累显著。为研究茎柔鱼脂肪积累对组织能量积累的贡献,采用组织能量密度测定技术和组织脂肪萃取技术对茎柔鱼卵巢、输卵管卵子、肌肉和消化腺4个组织脱脂前后的组织能量密度、单位质量脂肪含量及脂肪能量进行测定和分析。结果显示,脱脂前各组织的能量密度以消化腺最大,为(27.34±0.85) kJ/g;脱脂后的能量密度则以输卵管卵子的最大,为(20.01±0.56) kJ/g。各组织脱脂后的能量密度值均下降显著,尤以消化腺的下降最大,达31%。消化腺的单位质量脂肪含量最高,为(413.83±96.81) mg/g,其次为输卵管卵子和卵巢,肌肉组织的最低。在生理性成熟后,卵巢组织脂肪积累较大,输卵管卵子、肌肉和消化腺等组织则维持较稳定的脂肪积累水平。4个组织的脂肪能量占比的平均值在10%~32%之间。综上研究表明,脂肪是茎柔鱼卵巢、输卵管卵子、肌肉和消化腺等组织的重要能量物质,在组织能量积累过程中起着重要的作用。  相似文献

Changes in the concentration of lipovitellin, a lipoprotein that is the major protein in mature oocytes of crustaceans, were determined during ovarian development in female blue crabs, Callinectes sapidus. To test the effects of reproductive toxins on vitellogenesis, crabs were fed food containing cadmium or injected with the juvenile hormone antagonist, precocene II. Monoclonal antibodies prepared against one of the lipovitellin peptides (peptide A—107 kDa) were used in an enzyme linked immunosorbent assay to determine lipovitellin concentrations. Recently molted female crabs fed food containing cadmium (0.02 mg/g) for 18 days had oocytes with the same lipovitellin concentration as controls, but after 24 days the oocytes in cadmium treated crabs had significantly lower lipoprotein contents than controls (30 ± 4 ng/oocyte—cadmium treated; 45 ± 4 ng/oocyte—control). Precocene II-treated crabs had significantiy lower lipovitellin concentrations after 18 and 24 days. Diameters of oocytes were the same in both treated and control crabs. The results suggest that some pollutants may affect reproduction by acting during secondary vitellogenesis i.e., period when there is a large increase in lipovitellin synthesis. Since lipovitellins serve many of the nutritional needs of recently emerged crab embryos, poor larval survival may occur as a result of lower lipovitellin content.  相似文献

Interactions between cadmium and sulfhydryl-containing compounds in striped mullet, Mugil cephalus, were examined. Exposure to 10 mg Cd/liter for up to 6 weeks caused an increase in the acid-soluble thiol (AST) content of the liver. Although the AST concentration was elevated at 1·4 weeks, increases in the glutathione and cysteine contents were not observed until after 4 weeks of exposure. Homogenates of livers were fractionated on a Sephadex G-75 column. A homogenate of livers of mullet injected with 109Cd 2 days prior to sampling had an elution profile for thiols similar to controls. After 3 weeks exposure to 10 mg Cd/liter the 9400 MW thiol peak increased 3·6-fold and the peak with the same retention volume as glutathione increased 4·3-fold. Greater than 70% of the thiols in these two peaks were acid-soluble. In both experiments, almost all the cadmium was associated with the 9400 MW peak. The ratio in the 9400 MW peak declined from 14·1 (controls) to 3·2 after 3 weeks exposure. No cadmium was associated with glutathione in the bile.  相似文献

An interlaboratory calibration experiment was conducted at three laboratories to test two sources of variation associated with bioassay experiments, variation due to the experimenter and to the natural seawater. Twenty-eight day static (with frequent media renewal) bioassays exposing the polychaete Capitella capitata to cadmium were conducted with synthetic and natural seawaters. Test results varied between the three laboratories; however, the variations are most probably explained by the shipment of the experimental animals to the participating laboratories.  相似文献

To establish a base line against which future pollution may be measured, eight common commercial species of New Zealand sea fish were analysed for cadmium, chromium, copper, iron, lead, manganese, nickel, and zinc. One sample of edible muscle tissue was analysed for each of 70 samples of each species. The internal organs of up to five specimens of each species were also analysed. The fish studied were: kahawai, Arripis trutta, trevally, Caranx lutescens, tarakihi, Cheilodactylis macropterus, snapper, Chrysophrys auratus, moki, Latridopsis ciliaris, hapuku, Polyprion oxygeneios, kingfish, Seriola grandis and gurnard, Trigla kumu.

Although, none of the edible parts of the fish appeared to have trace element levels likely to be a public health problem (cadmium 0.002–0.02 ppm, chromium 0.01–0.04 ppm, copper 0.04–0.95 ppm, iron 0.9–13.5 ppm, lead ‘ 0.14–0.95 ppm, manganese 0.04–2.00 ppm, nickel 0.01–0.08 ppm, zinc 2.0–36.0 ppm), some of the organs (particularly the liver) had relatively high concentrations of elements such as cadmium (up to 54 ppm). Only if some industry were to seek to exploit internal organs of fish for human consumption would such levels become important. There was some evidence for a relationship between trace element concentrations and fish size for copper in kingfish and snapper, iron in hapuku, manganese in gurnard, and for zinc in kingfish and tarakihi. Some element pairs such as copper and zinc, iron and manganese, appeared to have concentrations which were mutually related. It was assumed that the elemental concentrations reported represent natural levels and are not the result of significant man‐made pollution because there are no major industries or large urban centres adjacent to the fishing grounds.  相似文献

为探究光裸星虫卵子发生的细胞学特点,利用电镜技术观察了光裸星虫体腔液中卵子发生过程的超微结构变化。结果表明:(1)光裸星虫体腔液中存在游离的卵原细胞和卵母细胞,卵母细胞的发育分为卵黄形成前期、卵黄形成初期、卵黄旺盛合成期和成熟期4个阶段。(2)卵原细胞的细胞器少,核质比大。卵黄形成前期,卵母细胞的细胞器有所增加,大量核仁外排物进入胞质中;出现卵黄膜和胶质膜,卵黄膜遍布微孔。卵黄形成初期卵母细胞核有较多突起,细胞器大量增加,出现分散分布的卵黄粒。卵黄旺盛合成期卵母细胞迅速增大,卵黄大量积累。成熟期卵母细胞核膜突起回缩,胶质膜易于脱落。(3)卵黄分为2种。Ⅰ型卵黄电子密度高,不发生融合,为内源性卵黄;II型卵黄中等电子密度,可融合为无定形卵黄块,为外源性卵黄。(4)成熟卵母细胞卵黄膜为三层,外层为颗粒层,表面具有粒状突;中层初始为均质结构,不断增厚并纤维化;内层致密,厚度不均。胶质膜电子密度极低。(5)卵子外有滤泡细胞,其核质比很大,细胞器少,在卵母细胞成熟期发生凋亡。文章还探讨了胶质膜、卵黄膜、滤泡细胞的功能,以及细胞器在卵黄形成中的作用。研究结果积累了光裸星虫卵子发生细胞学资料,为光裸星虫生殖调控与人工繁育研究提供理论参考。  相似文献
