Alkylphenolic compounds are present in estuarine and marine environments. They are moderately bioaccumulative, and oestrogenic to fish following three week, in vivo, exposures. The effects of prolonged exposure are, as yet, unclear. Sand goby (Pomatoschistus minutus) were exposed to measured concentrations of 4-tert-octylphenol (OP) in a 28-day, dose-ranging study, and a six month temporal response study. Following 28-days exposure to measured 31+/-6 or 101+/-47 microg l(-1) OP (mean+/-SD), immature male sand goby showed elevated vitellogenin (VTG) mRNA expression. Plasma alkali-labile phosphate concentrations were elevated in both sexes and were higher in females than males. Although measured OP concentrations were variable, they were significantly different for each treatment and a range of concentration-dependent effects were observed. The LC(50) for chronic OP exposure (8 weeks) was 29+/-6 microg l(-1), and both sexes demonstrated concentration and duration dependent increases in VTG mRNA expression. Exposure to 28+/-20 microg l(-1) OP (median+/-95% CI) for 6 months inhibited development of male nuptial colouration and sperm duct glands. These findings are discussed in relation to environmental concentrations of alkylphenols and ecological concerns.  相似文献

Tributyltin (TBT) is toxic to aquatic organisms and occurs widely in sediments and surface waters of American and European rivers and lakes. This study investigated TBT effects on development and population growth rate (r) of the common, hermaphroditic European freshwater snail Physa fontinalis. Egg ropes of similar age (1-3 days old) were exposed to a control (solvent only) and nominal concentrations of 0.01, 1.0 and 10 microg TBT l(-1) in triplicate. Hatching and mortality were recorded during 0-40 days of exposure. At day 40, 18 juveniles were randomly selected from each concentration (i.e., six from each test vessel) and individually exposed to the same concentration of TBT in 50 ml beakers. A cohort of 20 juveniles was allowed to continue developing in the original test vessels, so that individual and grouped results could be compared. Mortality and reproduction were recorded at 48-h intervals throughout the study period (110 days). Abnormal embryonic development was observed at 1 and 10 microg TBT l(-1). Although 50% of eggs hatched at 10 microg TBT l(-1), all these hatchlings failed to survive. Survivorship of hatchlings was significantly reduced by TBT at 1 microgl(-1). In general, there was a delay in egg production in isolated snails when compared with the grouped snails. Survival, fecundity and population growth rate (r) were reduced in both individual and grouped P. fontinalis at 1.0 microg TBT l(-1). Only a decline in r was observed in snails exposed individually to 0.01 microg TBT l(-1).  相似文献

Neomysis integer (Peracarida: Mysidacea) occupies the upper, low-salinity regions of estuaries in Europe, where it can experience periods of reduced oxygen concentration associated with the maximum turbidity zone. The present study reports the distribution of N. integer in response to gradients of dissolved oxygen in combination with environmentally realistic cadmium concentrations (0.1, 0.5 and 1.0 microg Cd2+ l(-1). Control and 0.1 microg Cd2+ l(-1) mysids were distributed evenly along a flume containing an oxygen gradient, and demonstrated no avoidance to hypoxia (down to 30% air saturation). In contrast, mysids exposed to 1.0 microg Cd2+ l(-1) migrated away from areas of low dissolved oxygen and, after 90 min, no mysids were found in regions of <75% air saturation. These results indicate that cadmium poses an increased risk to estuarine organisms inhabiting upper estuarine areas subjected to reduced dissolved oxygen concentrations.  相似文献

Naphthalene (NAP) and phenanthrene (PHE) effects on Salminus brasiliensis, a carnivorous freshwater fish, were investigated using behavioral tests. Larval stages of S. brasiliensis were exposed to water concentrations of 0, 0.04 mg/l, 0.20mg/l and 0.50mg/l for naphthalene and 0, 0.01 mg/l, 0.05 mg/l and 0.1mg/l for phenanthrene during two developmental phases. The prey fish Prochilodus lineatus were not exposed. Visual acuity was measured at the end of phase 2 in individual S. brasiliensis, which were also tested at the end of each phase for number of attacks on prey, number of prey captured, prey capture efficiency, and distance swam. Vision was impaired by PHE exposure, as acuity angles increased in exposed fish. At Stage I 2.3+/-1.2 prey were captured with 46% efficiency in controls compared to 0.4+/-0.3 prey captured with 13.4% efficiency in fish exposed to 0.05 mg PHEl(-1), the LOEL for both endpoints. At Stage II 4.0+/-1.1 preys were captured in controls compared to 2.5+/-0.9 preys captured in fish exposed to 0.01 mg PHEl(-1), the LOEL. Stage II control fish captured prey with 70% efficiency compared to 30% efficiency at Stage II fish exposed to 0.05 mg PHEl(-1), the LOEL. Distance swam was not affected by either NAP or PHE exposure. The exposure of larval stages of S. brasiliensis to realistic water concentrations of PHE impairs foraging skills and could affect recruitment of the species.  相似文献

Bioaccumulation of Cd, Co, Cu, Ni, Pb and Zn in Antarctic gammaridean amphipod collectives, Orchomene plebs (Hurley, 1965), was investigated during a cruise of RV "Polarstern" to the Wedell Sea. With the sole exception of Cd the organisms accumulated metals during exposure and depurated them in uncontaminated seawater. Four independent toxicokinetic experiments and one field study were modelled simultaneously to calculate the following size-dependent bioconcentration factors for organisms with body length 10 mm (BCF(10 mm)): 130 (Co), 4030 (Cu), 190 (Ni), 2900 (Pb), and 5210 (Zn). On the time scale of our experiments the data suggest an increased metal uptake by previously exposed test organisms. The collectives investigated may be regarded as potentially suitable biomonitors for Co, Cu, Ni, Pb and Zn but not for Cd. An approach to evaluate the sensitivity of Orchomene plebs as a biomonitor of waterborne metals in the field indicates minimal increments of the ambient exposure concentrations of 0.01 microg Co l(-1), 0.2 microg Cu l(-1), 0.4 microg Ni l(-1), 0.6 microg Pb l(-1) and 0.3 microg Zn l(-1).  相似文献

To determine the estrogenicity of effluents from sewage treatment plants (STPs) to larval fish, 2-day-old sunshine bass were exposed to effluents from three STPs serving New York City (NYC), varying in size and treatment level. Estrogenic response was evaluated by measuring vitellogenin (VTG) and estrogen receptor (ER) expression in cytosolic fractions of whole body homogenates. Concentrations of the presumptive endocrine disruptors in the effluents were also measured. VTG and ER levels in sewage-exposed fish were 3-5 times that observed in controls. Combined concentrations of estradiol and estrone ranged from 5 to 13 ng/l and nonylphenol-ethoxylate metabolites (NPEOs: 4-nonylphenol, and 1-, 2-, and 3-nonylphenol-ethoxylates) ranged from 180 to 470 microg/l in chlorinated effluent. Results indicate that both ER and VTG can be used as biomarkers for endocrine disruption in larval fish, and that 4-day exposure to sewage effluent is sufficient to elicit significant expression of these markers in sunshine bass larvae. The extremely higher concentrations of NPEOs found in effluent relative to hormones (approximately 40,000-fold) indicates that surfactant metabolites may be contributing significantly to the estrogenic effects observed.  相似文献

Following the in vivo exposure of dab (Limanda limanda L.) to cadmium chloride, kidney phagocytes were collected and their respiratory burst measured in vitro using chemiluminescence. Fish were exposed to mean measured concentrations of 1.3, 2.7 and 5.5 mg Cd litre−1 (as total cadmium ion) for a total of nine weeks, followed by a three week depuration period in clean sea water. Compared with control fish, the respiratory burst of kidney phagocytes from dab sampled after six weeks was significantly reduced in the 2.7 and 5.5 mg Cd litre−1 treatments (Steel's test, p < 0.05). Significant reductions were observed in the respiratory burst of phagocytes from all cadmium exposed fish compared with control fish after nine weeks (Steel's test, p < 0.05). After a further three week depuration period in clean sea water, the respiratory burst of phagocytes from fish previously exposed to 1.3 and 2.7 mg Cd litre−1 were still significantly less than in the control group (Steel's test, p < 0.05). Muscle tissue cadmium concentrations were also analysed, although there was no clear relationship between the muscle total cadmium levels and kidney phagocyte chemiluminescence. The results are discussed with respect to the possible mechanism(s) of cadmium immunotoxicity in dab and recommendations made for future work.  相似文献

The present study examines the impact of exposure to oil-derived products on the behaviour and physiology of the Australian 11-armed asteroid Coscinasterias muricata. Asteroids were exposed to dilutions of water-accommodated fraction (WAF) of Bass Strait stabilised crude oil, dispersed oil or burnt oil (n = 8) for 4 days whereby, prey-localisation behaviour was examined immediately after exposure, and following 2, 7, and 14 days depuration in clean seawater. The prey-localisation behaviour of asteroids exposed to WAF and dispersed oil was significantly affected though recovery was apparent following 7 and 14 days depuration, respectively. In contrast, there was no significant change in the prey-localisation behaviour of asteroids exposed to burnt oil. Behavioural impacts were correlated with the total petroleum hydrocarbon concentrations (C6-C36) in each exposure solution, WAF (1.8 mg l(-1)), dispersed oil (3.5 mg l(-1)) and burnt oil (1.14 mg l(-1), respectively. The total microsomal cytochrome P450 content was significantly lower (P(Dunnett test) < 0.01) in asteroids exposed to dispersed oil than in any other asteroids, whilst asteroid alkaline phosphatase activity was not significantly affected (P(ANOVA) = 0.11). This study further documents the deleterious impact of dispersed oil to marine organisms and supports further research in the area of in situ burning as a less damaging oil spill response measure towards benthic macro-invertebrates.  相似文献

Cypermethrin is a synthetic pyrethroid that is particularly toxic to crustacea. It is therefore applied as a chemotherapeutant in the salmonid aquaculture industry for the treatment of sea lice infestations. After use, cypermethrin is released directly into the marine environment, to be diluted by fresh seawater. The shore crab, Carcinus maenas is found in the vicinity of fish farms, and may come into contact with released cypermethrin. The detoxification enzyme, glutathione S-transferase (GST) has been implicated in cypermethrin metabolism in terrestrial arthropods, but this has not yet been demonstrated in crustacea. In this paper we investigate the response of GST activity in Carcinus to cypermethrin exposure, and also the time course of the induction process. GST activity is significantly increased in Carcinus exposed to nominal concentrations of 50 and 500 ng/l of water-borne cypermethrin. Carcinus demonstrate a significant elevation in GST activity following intra-cephalothoracic injection with 10 ng of cypermethrin. GST activity returns to basal levels after 36 h. The potential application of GST activity in Carcinus as a biomarker of cypermethrin exposure is discussed.  相似文献

Metallothionein has been assayed in a range of aquatic animal tissues as an indicator of metal exposure. We sequenced chub (Leuciscus cephalus) metallothionein cDNA which showed over 90% homology to common carp, goldfish and stone loach and 77% homology to rainbow trout sequences for metallothionein. We then used the extended primer method to develop an accurate quantitative competitive RT-PCR assay for metallothionein mRNA. RT-PCR was used to measure metallothionein mRNA in feral chub from a range of field sites, with different levels of heavy metal pollution, in the West Midlands, UK. Measurements were complemented by analysis of liver and gill metallothionein protein by capillary electrophoresis. There was no significant difference in the metallothionein protein levels between fish of different rivers and there was no evidence of elevation of mRNA at the sites of highest metal exposure. The level of metal exposure (e.g. zinc, nickel and cadmium each ranging between 15 and 28 microg/l ) at the pH (7.5-8.5) of these rivers appears insufficient to elevate hepatic or gill metallothionein in chub. A lack of elevation of hepatic metallothionein mRNA in chub exposed to zinc, copper and manganese for 24 h and 10 days in the laboratory also suggests a non-responsiveness of this species.  相似文献

In order to compare the acute toxicity of tributyltin (TBT) between sexes, five species of caprellid amphipods were exposed to seven levels (0, 0.001, 0.01, 0.1, 1, 10 and 100 microg l(-1)) of TBT for 48 h at 20 degrees C. The 48-h LC50 values in male and female were 1.3-6.4 microg TBTCl l(-1) and 1.2-8.6 microg TBTCl l(-1), respectively. No sex-specific differences of the acute toxicity of TBT were observed in the caprellids. These findings suggest that the risk of survival in response to exposure to TBT between sexes is similar in the caprellids.  相似文献

Experimental studies were performed on healthy, 80-100 g carp (Cyprinus carpio). Fish were exposed by emersion in Roundup (205 mg of glyphosate/l or 410 mg of glyphosate/l) in concentrations of 40- to 20-fold lower than those used in practice. Electron microscopy revealed that the herbicide caused appearance of myelin-like structures in carp hepatocytes, swelling of mitochondria and disappearance of internal membrane of mitochondria in carp at both exposure concentrations. It means that Roundup was harmful to carp when used in applied concentrations. Results of these studies enhance our knowledge of ultrastructural pathomorphology of fish organs following exposure to Roundup.  相似文献

Rainbow trout were exposed to 200 ppb cadmium in the water during four months at 5°C. The liver, kidney and gills were analyzed for cadmium, copper, zinc and metallothionein. Cadmium accumulated in all three organs and reached the highest concentration in the kidney. The zinc and copper concentrations were not altered during the experiment. The metallothionein concentration was significantly higher in liver of exposed fish than in control fish after three months. The kidneys reached significantly elevated levels of metallothionein in the exposed group after four months. These results demonstrate that metallothionein is induced by cadmium after exposure to the metal via water.  相似文献

In coastal marshes, fish larvae may be exposed simultaneously to extreme salinities and to atrazine, a widely used herbicide. To assess the effects of salinity on the toxicity of atrazine, newly-hatched mummichog (Fundulus heteroclitus) were exposed to atrazine (0, 5, 50 and 500 microg/L) at three salinities (3, 15 and 35PSU). Whole body cortisol was measured after 24 h. Body length, condition factor, whole body proteins, lipids, residual masses and water contents were assessed after 96 h. Significants effects were found for both condition factor and water content. Condition factors were lower at salinity 3 and 35 compared to near isoosmotic salinity, 15PSU. In addition, atrazine decreased condition factor at 500 microg/L. Reduction in condition was likely due to retardation in axial growth since body length, percentages of proteins or lipids were not affected. In the absence of atrazine, salinity had no effect on the prevalence of dehydrated (81% water) or hyperhydrated (85% water) larvae. In larvae exposed to atrazine, the prevalence of hyperhydrated larvae increased at 3PSU and 5 microg/L atrazine and that of dehydrated larvae increased at 15 and 35PSU and 5 microg/L atrazine. Severity of dehydratation increased with atrazine concentration at salinity 35PSU. Thus, a short-term exposure to environmentally realistic concentrations of atrazine affects osmotic control in mummichog larvae with possible effects on buoyancy, survival and recruitment.  相似文献

Juveniles of the dendrobranchiate decapod Penaeus indicus take up radiolabelled cadmium from solution over the exposure concentration range of 1.8-31.5 microg L(-1), with an uptake rate constant of 0.090 L g(-1)d(-1) at 15 salinity and 25 degrees C. New cadmium taken up is added to the existing cadmium content of the prawn with no significant excretion, and the rate of accumulation of radiolabelled cadmium is a measure of the absolute cadmium uptake rate from solution. Moulting had no significant effect on the accumulation of cadmium. Newly accumulated cadmium is distributed to all organs with the highest proportions of body content being found in the hepatopancreas, exoskeleton, gills and remaining soft tissues, the hepatopancreas and gills containing the highest labelled cadmium concentrations. Like other crustaceans, penaeid prawns inhabiting anthropogenically contaminated coastal waters with raised cadmium bioavailabilities can be expected to contain raised body concentrations of cadmium. Cadmium concentrations of most field-collected adult penaeids are relatively low, as a probable consequence of the growth dilution of their cadmium contents as a result of the rapid growth rates of penaeid prawns.  相似文献

Certain environmentally persistent compounds can adversely affect reproduction by acting as steroid hormone agonists or antagonists. The goal of the present study was to determine the developmental stage most susceptible to exogenous hormone (estradiol and testosterone) exposure using a small teleost model. In the first (pilot study) of two experiments, medaka (Oryzias latipes), at varying developmental stages, were bath-exposed to 5 micrograms/l 17 beta-estradiol for 24 h. At 5 months of age, fecundity, fertility and embryo and larval viability (reproductive success) were investigated in control and exposed groups. Fish at 1, 1.5, 2 and 5.5 months of age were also sampled, processed and examined histologically for gonadal alteration. No significant differences in mortality, gonadal morphology, body weight, sex-ratio or time to maturity were seen between control and exposed fish. At 5 months, however, when exposure groups were compared to controls, significant differences were seen in reproductive success and viability of offspring. A second experiment exposed embryo stage 10, and 1-, 7- and 21-day-old larvae for 6 days to 15 micrograms/l 17 beta-estradiol or 100 micrograms/l testosterone. No significant differences were seen at 5 months in mortality, body weight, or time to sexual maturity. However, sex-ratios were significantly biased toward female in the stage 10, 1- and 7-day post-hatch estradiol exposure groups. No significant changes in sex-ratio were associated with testosterone exposure at any developmental stage. Further, intersex gonads were observed in fish from all groups exposed to 15 micrograms/l estradiol. Only those fish exposed as newly hatched fry or at 1 week post-hatch displayed intersex gonads following 100 micrograms/l testosterone exposure. Data from these experiments show that newly hatched fry are that life stage most sensitive to hormone exposure and the most appropriate to use in determining effects of known endocrine-disrupting compounds.  相似文献

In the present study the induction of metallothioneins (MTs) and its relation to cytosolic metal concentrations (Zn, Cu and Cd) in the euryhaline crustacean Neomysis integer exposed to Cd at different salinities was studied. N. integer was exposed to the same free cadmium ion activity of 5.74 x 10(-9) mol l(-1) (i.e. 1/5 of the 96 h LC(50) value expressed as cadmium activity) in hypo-osmotic (5 psu), isosmotic (16 psu) and hyper-osmotic media (25 psu) for 7 days. In this way, the effect of salinity on cadmium speciation was eliminated and therefore the physiological effect of salinity on Cd accumulation and MT induction could be studied. The accumulation of cytosolic Cd in N. integer changed with salinity from 1.11+/-0.05 micromol l(-1) at 5 psu up to 1.43+/-0.17 micromol l(-1) at 25 psu. This could indicate that the physiological response of euryhaline estuarine invertebrates like N. integer to salinity changes can influence the rate of trace metal uptake from solution. While the salinity changes did not cause significant differences in cytosolic Zn concentrations (mean value of all tested salinities: 34.4+/-2.8 micromol l(-1)), an inverse relationship between salinity and cytosolic Cu concentration was observed. The highest concentration of 15.7+/-2.3 micromol Cul(-1) was determined at 5 psu and the lowest 10.9+/-1.4 micromol Cul(-1) at 25 psu. This could point to a possible relationship between the copper concentration and the hemocyanin metabolism in N. integer. This is the first time that differential pulse voltammetry method was applied to MT assays with N. integer. Although the exposure to Cd resulted in a higher Cd cytosolic concentration, no subsequent MT increase was detected. The significant positive correlation between MT levels and cytosolic Cu concentrations (Spearman correlation coefficient r(s)=0.356, p=0.009) implies a strong relationship between MT and Cu in N. integer.  相似文献

Copper (Cu) is an essential element for biological systems, however, when present in excess, is toxic. Metallothioneins can play an important role in Cu homeostasis and detoxification. Moreover, Cu can catalyse the production of toxic hydroxyl radicals that cause lipid peroxidation but defence systems in the cells can limit the oxidative damage. The present study was performed to investigate the effect of three Cu concentrations (0.5, 2.5 and 25 microg l(-1)) on the response of antioxidant enzyme activities (superoxide dismutase (SOD), catalase (CAT), selenium-dependent glutathion peroxidase and total glutathion peroxidase), total proteins, metallothioneins (MT), malondialdehyde (MDA) concentrations in the gills of the clam, Ruditapes decussatus. The activity of antioxidant enzymes and total proteins, MT and MDA concentrations were measured in the gills of the clams after 1, 3, 7, 14, 21 and 28 days of Cu exposure. Results indicate that Cu only induces an imbalance in the oxygen metabolism during the first week of Cu exposure due to a decrease in mitochondrial SOD and CAT, selenium-dependent and total glutathion peroxidase activities. Cu also causes lipid peroxidation, measured by the MDA formation, that was Cu dependent. In the gills of clams exposed to 25 microg Cu/l, the excess of Cu triggers the induction of MT synthesis after 3 days of exposure.  相似文献

There is a body of literature indicating that pre-exposure to low levels of metals may increase tolerance during subsequent exposure.1 We have previously demonstrated that exposure to cadmium at 100 to 500 μg/liter will retard the rate of fin regeneration in the killifish Fundulus heteroclitus.2 These experiments were designed to investigate the effects of pre-exposure to cadmium, using fin regeneration as the parameter of effect. The results indicate that pretreatment allows this species to regenerate faster during cadmium treatment as compared to non-pretreated fish, and that the relationship between the pretreatment and treatment concentrations is relevant to pretreatment effects.  相似文献

Arsenic compounds in the marine polychaete Arenicola marina collected from Odense Fjord, Denmark were determined by HPLC-ICPMS. In contrast to most other marine animals, A. marina contained most of its water soluble arsenic as inorganic forms, arsenite (58%) and arsenate (16%), and arsenobetaine was present as a minor constituent (6%) only. Other arsenic compounds detected in A. marina were dimethylarsinate (4%), two arsenosugars (1 and 3%), tetramethylarsonium ion (1.5%), and arsenocholine (<1%). A new arsenobetaine -trimethylarsoniopropionate-previously only reported in fish, was also present at trace levels (<1%), and an unknown anionic arsenical (approximately 10%) remains unidentified. When A. marina was exposed in laboratory experiments to different concentrations of arsenate in seawater (10, 50. 100, 500 and 1000 microg As 1(-1)) the polychaetes accumulated arsenic in a dose dependent. non-linear manner. Most of the accumulated arsenic was biotransformed to arsenite and dimethylarsinate. with the remainder being accumulated as unchanged arsenate. None of the other arsenic compounds naturally present in A. marina increased in concentration following arsenate exposure.  相似文献
