Histopathological changes in various cellular constituents of gill, liver and kidney were studied in Heteropneustes fossilis (Bloch) after treatment with cadmium. The changes included fusion, vacuolization, necrosis, hypertrophy and formation of cellular debris in the gill; swellings in nuclei, vacuolization, necrosis and coagulation of cytoplasm in the liver; expansion of renal tubules and formation of cellular debris in the kidney. The changes in the tissue architecture have been discussed in reference to fish mortality.  相似文献

The effects of exposure to LC50 (10.5 mg/l) for 96 h and to a sublethal concentration (1.05 mg/l) of Sevin for 120 days have been observed on some haematological, biochemical and enzymatic parameters of Channa punctatus. The fish were hyperglycemic and hyperlactemic and the glycogen content of the liver and muscles decreased, but the lactic acid content of two tissues increased. The activity of hexokinase increased in the kidney, intestine, liver and muscles but decreased in the gills and brain. The activity of lactate dehydrogenase decreased in the kidney and intestine, and increased in the liver, muscles, brain and gills. The pyruvate dehydrogenase activity decreased in all the six tissues. Both the exposures produced a decrease in the succinate dehydrogenase activity of the liver. The enzyme activity decreased in the brain, gills and muscles after chronic exposure, whereas the kidney and intestine showed an elevation in enzyme activity.  相似文献

In the present work, histological changes in the gill, liver, kidney and intestine of Notopterus notopterus, induced by pulp waste, paper waste and mixed pulp and paper waste, have been studied both at sublethal and lethal concentrations. The pulp waste produces much more histological changes both at sublethal and lethal concentrations in the gill, liver, kidney and intestine as compared to paper waste and mixed pulp and paper waste. The changes are much more remarkable in the case of sublethal concentration, as compared to a lethal concentration. In a sublethal concentration, the localized necrosis, atrophy and partial and total liquefaction of epithelial cells, pilaster cells, mucous cells of the gill, hepatic cells of the liver, mesenchymatous cells, renal tubule cells and Bowman's capsule of the kidney, mucosal and submucosal cells and blood vessels of intestine have been observed. In lethal concentration, the changes are much more marked in the gill, as compared to the liver, kidney and intestine. The histological changes, produced in the different types of cells and in the tissue as a whole, can be correlated with the different chemical components present in the pulp waste, paper waste and mixed pulp and paper waste.  相似文献

The effects of short-term exposure to tetravalent platinum on survival, opercular movement and post-treatment growth of coho salmon fry (Oncorhynchus kisutch) was investigated. Employing a static water acute toxicity bioassay with platinum as PtCl42HCl·6 H2O, at 8.5±0.2°C, and a water hardness of 55.9±3.5 mg l.?1 (as CaCo3), the 24, 48, and 96-h LC50 values were 15.5, 5.2, and 2.5 mg Pt4+ l.?1 respectively.Rates of opercular movement for fish exposed to platinum increased with increasing concentrations to a level of 1.0 mg l.?1. No further significant increases were evident above this level. Hypoactivity of fish exposed to 0.3 mg l.?1 and higher was evident during the acute toxicity bioassay and much of the post-treatment study. Post-treatment rate of growth for fish exposed to sublethal concentration of platinum for 96 h was less than that of the controls.All organs examined histopathologically were within normal ranges with the exception of the gills and olfactory organ. Lesions in fish exposed to concentration of 0.3 mg l.?1 and higher were characterized by branchial epithelial hypertrophy and hyperplasia, and necrosis of olfactory epithelial cells.  相似文献

Three experiments were conducted with Atlantic salmon (Salmo salar) to assess the effects of dietary exposure to copper and cadmium. The results presented here provide an overview, details of each experiment will be published in full elsewhere. In the first experiment, salmon parr exposed for four weeks to 35 and 700 mg Cu kg−1 diet had significantly elevated intestinal copper concentrations, cell proliferation (PCNA) and apoptosis rates compared to control fish. No differences were observed in gill or plasma copper concentrations among the groups. In contrast to the controls, the Cu exposed groups did not grow significantly during the exposure period. The second experiment (three months exposure) was conducted to assess the effects of dietary copper (control, 35, 500, 700, 900 or 1750 mg Cu kg−1 diet) on growth and feed utilization in salmon fingerlings. Growth was significantly reduced after three months exposure to dietary Cu concentrations above 500 mg kg−1. Similarly, copper body burdens were significantly higher in fish exposed to elevated dietary copper concentrations (above 35 mg Cu kg−1 diet). In the third experiment, salmon parr were exposed to one of six dietary cadmium concentrations (0, 0.5, 5, 25, 125 or 250 mg Cd kg−1 diet) for four months. Cadmium accumulated in the liver>intestine>gills of exposed fish. Rates of apoptosis and cell proliferation in the intestine increased following exposure to dietary cadmium. Exposure to elevated concentrations of dietary cadmium had no effect on growth in salmon parr. Results from these studies indicate that cellular biomarkers have potential as early warning signs of negative effects on the overall fitness of an organism.  相似文献

Genetic transformation is gaining importance for developing plant types suitable to metal accumulate and/or hyperaccumulate. In this study, the transgenic tobacco plant which transferred the ScMTII gene from Saccharomyces cerevisiae to wild type tobacco cultivar Petite Havana (SR1) was grown on soils with low and high cadmium (Cd) and zinc (Zn) concentrations in a growth chamber for 6 weeks and compared to wild type tobacco for Cd and Zn accumulation. Cadmium and Zn accumulations in the transgenic and wild type tobacco plants were increased with the increasing Cd and Zn concentrations. Unlike Zn, the transgenic plant accumulated significantly higher amount of Cd compared to the wild type control plants. Shoot Cd concentrations of transgenic tobacco in higher Cd dosages reached the above the hyperaccumulation threshold value of 100 mg Cd kg?1 in the dry weight (DW). Transgenic tobacco accumulated 354, 400, 372, and 457 mg Cd kg?1 DW, for 10, 20, 40, and 80 mg Cd kg?1 soil treatments, respectively. These values are 3.5–4.5‐fold higher than that of Cd hyperaccumulation threshold value. With 10 mg kg?1 Cd treatment, the bioconcentration factor (BCF) of transgenic tobacco plants for Cd reached up to 35 in which the threshold value for BCF should be at least 10. Our results showed that the transgenic tobacco may be used as a good Cd hyperaccumulator plant and for phytoextraction of Cd contaminated soils, but not for Zn.  相似文献

This study was undertaken to determine the cadmium removal efficiency of Lemna minor when it was used for treatment of wastewater having different characteristics, i. e., pH, temperature and cadmium concentration. Plants were cultivated in different pH solutions (4.5–8.0) and temperatures (15–35°C) in the presence of cadmium (0.1–10.0 mg/L) for 168 h. The amount of biomass obtained in the study period, the concentrations of cadmium in the tissues and in the media and net uptake of cadmium by Lemna have been determined for each condition. The percentages of cadmium uptake (PMU) and bioconcentration factors (BCF) were also calculated. The highest accumulation was obtained for the highest cadmium concentration of 10.0 mg Cd/L as 11.668 mg Cd/g at pH 6.0, and as 38.650 mg Cd/g at 35°C and pH 5.0. The cadmium accumulation gradually increased with initial concentration of the medium, but the opposite trend was observed for the PMU. However, the maximum PMU was obtained as 52.2% in the solution with the lowest concentration of 0.1 mg Cd/L. A mathematical model was used to describe the cadmium uptake and the equation obtained was seen to fit the experimental data very well.  相似文献

Static bioassay tests were conducted to determine the acute toxic effects of copper, cadmium and their mixtures on Heteropneustes fossilis in two different seasons. Median lethal concentrations (24, 48, 72 and 96 h LC50) revealed that copper is more toxic to fish even at very low concentrations in comparison to cadmium, tested separately or in combination with cadmium. The data indicate that the level of tolerance of the fish to metallic ions tested was temperature specific. Generally, fishes were found to be more susceptible to metallic ions at higher temperatures as revealed by the threshold concentration, MATC and LC50 values. The behavioural changes of the test fishes were also observed in reference to different concentrations of the metallic ions and temperatures.  相似文献

In the present investigation the effect of water-borne copper on the gill architecture of a freshwater teleost, Heteropneustes fossilis (BLOCH ) was studied through scanning electron microscopy. The changes produced on the gill surface in a sublethal measured copper concentration (8.17 mg/1) in static bioassay after 96 h of exposure were fusion of secondary lamellae, rupture of various cellular components and deposition of excessive mucus and blood on the gill surface. The epithelial surface of treated gill thus lost the organized structures like microvilli, arborizing ridges and channels. The cause of fish mortality due to action of copper has been discussed in reference to reduced relative diffusing capacity along with other wide ranging effects on the metabolism of fish.  相似文献

The haematological, biochemical, and enzymological alterations produced on exposure of Channa punctatus to LC50 (11.2 mg/L) for 96 h and to a sublethal concentration (1.12 mg/L) of cadmium (Cd2+) for 15, 30, 60, and 120 days have been studied. The fish were hypoglycemic and hypolactemic. The pyruvate content of blood and liver decreased in acute and all stages of chronic exposure except for 30 days where significant increase was recorded. Depletion was noted in the total protein and glycogen content of liver and muscle, and the level of lactate in liver in the two types of exposure. The levels of muscle lactate and pyruvate increased in both exposures. The activities of hexokinase, glucose-6-phosphatase, and lactate dehydrogenase decreased in liver after 96 h and all stages of chronic exposure except 30 days. The activities of hexokinase and lactate dehydrogenase in muscle and glutamate-oxalacetate transaminase and glutamate-pyruvate transaminase in serum, liver, and muscles increased in all exposures. The activities of succinate and malate dehydrogenases in liver and muscles decreased after acute and all stages of chronic exposure. The above results suggest that glycolysis and gluconeogenesis were impaired in liver but glycolysis prevailed in muscle. Aerobic oxidation is adversely affected in metal exposed fish, while proteolysis was increased.  相似文献

The tolerance level of the fertilizer NPK for the freshwater siluroid Mystus M. vittatus (BL .) and histopathological changes in the gill of this fish against two concentrations of NPK were investigated. It was found that at 400 ppm (24 h) mortality in the fish started, and at 700 ppm (96 h) all the fish died. Hence the concentration of NPK at 400 ppm was taken as an experimental concentration for this fish. Curling towards the upperside in the secondary lamellae resulting in the joining with each other and profuse mucus secretion and subsequent death of 40% of the fish (96 h) were observed. At 700 ppm (96 h) NPK, which is far higher than the highest tolerance limit of the fish, all the fish died. Profuse mucus secretion, formation of subepithelial spaces, sloughing of epithelial layer, leaving only the rows of pilaster cells, resulting in the death of the fish, were observed. The mortality of the fish was due to asphyxiation and damage in the gill tissues resulting in the alteration in the respiratory and osmoregulatory metabolism.  相似文献

Puntius sophore (HAMILTON ) were exposed to several concentrations of mercury up to 10 d under static bioassay conditions. Abnormal behaviour in fish were increase in swimming activity, absence of shoaling behaviour, surfacing, loss of equilibrium and death. The body and gills of dead and living fish in the higher mercury concentrations are covered by coagulated mucus. The 96 h and 240 h LC50 values and their 95% confidence limits were 0.145 (0.133 … 0.165); 0.065 (0.057 … 0.072) mg/l of Hg, respectively. The F-ratios are significant at the 1% level of significance and thus indicate that the per cent mortality caused by mercury is distributed in a heterogeneous manner while there is no significant overall difference between the replications. The histopathological assessment of the gill was also carried out. In the control fish, gills are characterized by thin secondary gill lamellae and epithelial layer closely associated with the basement membrane. Several histopathological changes were observed in the gill, including the swelling in secondary gill lamellae, nuclei increases in size and number, and degeneration of epithelial layer.  相似文献

Sewage effluent is a powerful agent of ecological change in estuaries. While the effects of sewage pollution on water quality are usually well documented, biological responses of exposed organisms are not. We quantified health impacts in the form of pathological tissue changes across multiple organs in estuarine fish exposed to elevated levels of treated wastewater. Structural pathologies were compared in wild populations of four fish species from two subtropical estuaries on the east coast of Australia that differ substantially in the amount of direct wastewater loadings. Uptake of sewage-derived nitrogen by fish was traced with stable nitrogen isotopes. Pathologies were common in the liver, spleen, gill, kidney and muscle tissues, and included granulomas, melanomacrophage aggregates, and multiple deformities of the gill epithelia. Tissue deformities were more frequent in fish exposed directly to wastewater discharges. Mullet (Valamugil georgii) were most affected, with only a single specimen free of pathologies in the sewage-impacted estuary. Similarly, in those fish that had structural abnormalities, more deformities were generally found in individuals from sites receiving sewage. These spatial contrasts in impaired fish health correspond to significantly enriched delta15N values in fish muscle as a consequence of fish assimilating sewage-N. Overall, the pattern of lower health and enriched delta15N values in fish from sewage-impacted areas suggests that organism health is lowered by sewage inputs to estuaries. Measurements of organism health are required to understand the effects of sewage on estuarine ecosystems, and histopathology of fishes is a powerful tool to achieve this.  相似文献

Oreochromis niloticus is an endemic species in the River Nile and represents the main object of fishery in the Nile Delta Lakes of which Lake Maryût is the smallest and most polluted. Due to current pollution problems, fish production in the lake greatly declined. Besides fish biology, this study considered a wide array of water databases in selected sites along the lake. Hydrologic and biologic data were compared to that of a reference fish hatchery. Water concentrations of ammonia, manganese, nickel, cadmium, lead and mercury proved hazardous to fish, whereas chromium, copper, iron, zinc, pH, alkalinity, hardness, phosphate, nitrate, and nitrite always fell within acceptable levels. Physiologic evaluation of O. niloticus pointed out improper growth, protein inadequacy, and functional impairment in fish inhabiting polluted sites, in particular the lake main basin. These were reflected by data of specific formulae as RNA/DNA and the relative RNA content (r) in cells of the liver and gill arches as well as by the relative mobilization of serum protein fractions. It seems, however, that growth competence is additionally affected by the stress impact exerted upon fish by massive population numbers in some cramped commercial ponds.  相似文献

The lead, cadmium and mercury content of soft tissues and bone were determined for the common seal (Phoca vitulina) collected from off the coast of East Anglia and the West of Scotland. Lead levels were low in all tissues, cadmium accumulated with age in the kidney of West Scotland seals and mercury accumulated to high levels in the liver of seals from both areas. Comparison of our results with those of other workers, suggests that the rate of mercury accumulation in the liver of seals increases in the order: Canadian Arctic and Atlantic coast < West Scotland < East Anglia < Netherlands coast. Accumulation of mercury in the kidney and spleen of older seals, lends tentative support to the hypothesis that a protective demethylation and retention process in the liver may begin to leak mercury to other tissues at the high concentrations which have been recorded in the liver of older seals, and at the high dose rates which have been used in toxicological experiments.  相似文献

The present paper deals with the determination of LTm values of zolone, rogor, sumithion and abate for Channa gachua by conducting static bioassay experiments. The TLm values for zolone came out to be 0.08275 mg/l, 0.08225 mg/l, 0.0815 mg/l and 0.0810 mg/l; for rogor 5.155 mg/l; 4.955 mg/l, 4.65 mg/l and 4.475 mg/l; for sumithion 12.60 mg/l, 12.50 mg/l, 12.30 mg/l and 12.20 mg/l and for abate 217.25 mg/l, 225.575 mg/l, 221.575 mg/l and 217.25 mg/l for 24, 48, 72 and 96 hours, respectively. Relative susceptibility indicates that this fish is not susceptible to zolone and less to rogor, sumithion and abate, respectively. Present studies also show that increased temperature and pH increase the toxicity of all the biocides, but increased hardness of water decreases the toxicity of biocide solutions. The safe concentrations for zolone, rogor, sumithion and abate came out to be 0.02437 mg/l, 1.3733 mg/l, 3.6907 mg/l and 66.678 mg/l, respectively.  相似文献

The accumulation of crimidin in total samples and in some organs of five fish species was studied under experimental conditions. The species were Cyprinus carpio, Carassius carassius, Tinca tinca, Scardinus erythrophthalmus and Leucaspius delineatus. Fish were exposed to concentrations of 10 mg · l?l and 50 mg · l?1. During short-term experiments water and fish samples were taken at intervals of t = 0, 6, 12, 24, 48 and 72 hours, during the long-term experiment sampling was performed weekly for six weeks. Total samples were analysed for C. carassius, T. tinca, S. erythrophthalmus and L. delineatus. Samples of individual organs and tissues were taken as follows: C. carpio – gills, digestive tract, muscular tissue, kidneys, gonads; C. carassius – gills, digestive tract, muscular tissue, ovaries, testes. Crimidin was determined by gas chromatography. In samples from the short-term experiments at a concentration of 10 mg · l?1 in the water the amount was roughly 101–10 mg · kg?1, at a concentration of 50 mg · l?1 in the water roughly 101 mg · kg?1. In most cases the accumulation coefficient was lower than 1.0. The accumulation capacity of individual fish species did not differ greatly. Of the internal organs only the kidneys had a high accumulation capacity, otherwise the highest values were found in muscular tissue and the gills. After one – three weeks the amount of crimidin in most organs falls, and after transfer to clean water there is a general sharp decline. Thus crimidin is not firmly bound in the body.  相似文献

Monocrotophos (0,0-dimethyl-0-(1-methyl-2-N-methyl-carbamoyl)-cis-vinylphosphate) as an active component of some insecticides is highly soluble in water at a low hydrolysis rate: half-time at 2 mg/l, pH = 7.0 and 38 °C 23 d. Individuals of the following fish species (individual weight in g) were kept during the static bioassay for 6… 72 h at 40 mg/l monocrotophos in the medium: Cyprinus carpio (70), Carassius carassius (45) and Leucaspius delineatus (1.5), and then the concentration of monocrotophos was gas-chromatographically determined in different tissues. Initially, till the 24th hour a high accumulation in the gills took place (in mg/kg fresh weight: Cyprinus 0.49, Carassius 3.88), then the concentration remained uniform or slightly falling here, whereas in the other organs (digestive organs, muscle, gonads) a further accumulation up to 1 mg/kg occurred. Leucaspius was investigated only as whole individuals, and after 72 h their fresh weight had increased to 6.96 mg/kg. The rates of accumulation decrease in the following order: Cyprinus: digestive organs, gills, muscle; Carassius: gills, digestive organs, gonads, muscle.  相似文献

In recent years, there has been a decline in the wild populations of river pufferfish, Takifugu obscurus. Besides overexploitation for commercial purposes, environmental pollution is believed to have contributed to its decline. However, almost no information exists about genes involved in metabolism of xenobiotics by this species. Nevertheless, there is interest in fugu fishes, since they possess the smallest genome among vertebrates. We cloned and characterized the full-length cDNA sequence of a cytochrome P450 1A (CYP1A) gene from T. obscurus. Phylogenic relationship of T. obscurus CYP1A was also compared to other fish species. The tissue distribution and time-dependant induction of CYP1A mRNA were studied by real-time PCR in T. obscurus exposed to an aryl hydrocarbon receptor (Ahr) agonist, β-naphthoflavone (BNF). The greatest basal expression in livers of control as well as BNF-treated individuals. However, brain, gill, gonad, intestine, and kidney also expressed CYP1A. Muscles expressed the least CYP1A. The results of the time-course study revealed induction in brain and gills after 6 h and at 12 h in most tissues. Except for gills, all other organs retained induced expression of CYP1A mRNA up to 96 h.  相似文献

Cadmium, copper and zinc have been analysed in some tissues and organs of Loggerhead, Kemp’s Ridley (only muscle for this species) and Leatherback turtles stranded along the Atlantic coasts of France. The pancreas analysed only in Leatherback turtles exhibited the highest metal concentrations, which is very surprising for an organ which does not play a role in the detoxification processes. The distribution of these elements in kidney, liver and muscle were quite similar to that found in marine mammals or seabirds. Nevertheless, mean cadmium concentrations in the kidney were as high as 13.3 μg g−1 wet weight in the Loggerhead turtles and 30.3 μg g−1 wet weight in the Leatherback turtles. Such high concentrations in the Leatherback turtles have never been recorded before. The main source of cadmium for marine turtles is probably the food. The Leatherback turtles are known to feed mainly on jellyfish in this area. Ten times higher cadmium concentrations have been determined in jellyfish compared to fish. This would imply a greater exposure to cadmium for Leatherback turtles, which probably need to eat great quantities of jellyfish to cover their needs.  相似文献
