Biological markers of produced water (PW) exposure were studied in Atlantic cod (Gadus morhua) in both laboratory and field experiments, using authentic PW from a North Sea oil field. In the laboratory study, the PW exposure yielded significantly elevated levels of metabolites of polycyclic aromatic hydrocarbons (PAHs) and alkylphenols (APs) in bile even at the lowest exposure dose (0.125% PW). Other biomarkers (hepatic CYP1A induction and DNA adduct formation) responded at 0.25% and 0.5% PW concentrations. In the field study, bile metabolite markers and hepatic CYP1A were clearly increased in fish caged close to the PW outfall. Induction of plasma vitellogenin was not found in laboratory or field exposures, suggesting that the levels of oestrogen agonists (such as APs) might not have been sufficient to elicit induction, under the present conditions. The applicability of the biomarkers for use in water column biomonitoring programs is discussed.  相似文献

The biological impact of a treated produced water (PW) was investigated under controlled laboratory conditions in the blue mussel, Mytilus edulis. Mussel health status was assessed using an integrated biomarker approach in combination with chemical analysis of both water (with SPMDs), and mussel tissues. Acyl-CoA oxidase activity, neutral lipid accumulation, catalase activity, micronuclei formation, lysosomal membrane stability in digestive cells and haemocytes, cell-type composition in digestive gland epithelium, and the integrity of the digestive gland tissue were measured after 5 week exposure to 0%, 0.01%, 0.1%, 0.5% and 1% PW. The suite of biomarkers employed were sensitive to treated PW exposure with significant sublethal responses found at 0.01-0.5% PW, even though individual chemical compounds of PW were at extremely low concentrations in both water and mussel tissues. The study highlights the benefits of an integrated biomarker approach for determining the potential effects of exposure to complex mixtures at low concentrations. Biomarkers were integrated in the Integrative Biological Response (IBR/n) index.  相似文献

A biotest system for environmentally realistic exposure of fish to produced water (PW) was developed and tested. Authentic PW was collected at an oil production platform in the North Sea and preserved by freezing in multiple aliquots a 25 L. After transport to the test laboratory onshore, daily PW aliquots were thawed, homogenised and administered to the test fish, Atlantic cod (Gadus morhua), in two diluted exposure concentrations, 0.1% and 0.5%, during a 15 d period, using a continuous flow-through exposure setup. Positive control groups were exposed to two crude oil treatments for comparison. Chemical analyses showed that alkylphenol (AP) and PAH concentrations in PW exposure waters were very low. Observations of significantly increased AP and PAH metabolite levels in PW exposed fish demonstrated the suitability of the biotest system for its use in biological exposure/effect studies of PW, and it also demonstrated the sensitivity of bile metabolites as PW exposure markers in fish. The relevance of the biotest system for PW effect studies and for validating modelled environmental risk estimates of PW dischargers from offshore oil production is discussed.  相似文献

Lysosomal integrity in mussels has been applied as a biomarker to detect the pollution of trace organics and metals in the natural environments. However, few studies have examined the effects of water quality on the response of lysosomal integrity, in particular total ammonia-nitrogen (TAN) and dissolved oxygen (DO). This study demonstrated that high level of TAN (2.0 mg/l) and low DO (2.5 mg O2/l) could significantly reduce the lysosomal integrity in green-lipped mussel Perna viridis, respectively by 33% and 38%, whereas the mussel lysosomal integrity decreased by 70% in the combined treatment of TAN and low DO under laboratory conditions after one week. The mussel lysosomal integrity of all treatment groups could return to the control level after a three week recovery period. In the field validation in Victoria Harbour, Hong Kong during an one-year study period, lysosomal integrity in P. viridis identified the cleanest site east to the harbour, where the lowest TAN and highest DO concentrations were found. While lysosomal integrity in mussels seemed not affected by seasonal changes, approximately 40% of the variation of this biomarker could be attributable to the changes in TAN and DO in seawater. In conclusion, the response of the mussel lysosomal integrity can be confounded by both TAN and DO prevailing in the natural environments and thus caution must be exercised in relating the observed changes in lysosomal integrity to any specific pollutant in coastal water quality monitoring studies.  相似文献

The effects of the new antifouling compound zinc pyrithione (Zpt) on the embryonic development of sea urchin (Paracentrotus lividus) and mussel (Mytilus edulis) were investigated in laboratory toxicity tests. The median effective concentrations (EC50) were 7.7 nM for sea urchin embryos and 8 nM for mussel embryos. Toxic effects of Zpt on the larval growth of the sea urchin were detected at 0.5 nM. Predicted environmental concentrations of Zpt in pleasure craft harbours are higher than the predicted no effect concentrations for sea urchin and mussel embryos, indicating that Zpt may pose a threat to those species from exposure in the field.  相似文献

The effect of chronic exposure to Escherichia coli on morphometrical parameters, different antioxidant defenses, lipid peroxidation and protein oxidation has been studied in digestive gland of the freshwater mussel Diplodon chilensis in the laboratory during 6 weeks. In, a parallel field study, we evaluated morphometrical and oxidative stress parameters in digestive glands of mussels collected from a sewage polluted area (SMA) and from a pristine area (control). Both sites were characterized by analyzing bacteriological and physic-chemical parameters. In the laboratory D. chilensis was able to filter bacteria at high concentrations with a clearing rate of 0.510 ± 0.036 L/h per gram of dry soft tissue mass. The digestive gland mass to shell length ratio (DGM/SL), reduced glutathione (GSH), lipid peroxidation, as concentration of thiobartituric acid reactive substances (TBARS), and glutathione-S-transferase (GST) activity of mussels fed with bacteria were significantly higher than those of control mussels after the fourth week. Fecal bacteria in lake water samples were undetectable in the control, and higher than 24,000 MPN (most probable number)/100 mL in SMA. DGM/SL was higher in SMA. No differences between sites were observed in total lipid and protein content, neither in superoxide dismutase activity. GSH content was higher in SMA, with no difference in the oxidized form. GST activity and MDA were significantly higher in SMA but protein oxidation was not affected.  相似文献

Polycyclic aromatic hydrocarbons (PAHs) and organochlorine pesticides (OCs) are generally present in the marine environment in complex mixtures. The ecotoxicological nature of contaminant interactions, however, is poorly understood, with most scientific observations derived from single contaminant exposure experiments. The objective of this experiment was to examine dose-response relationships between antioxidant parameters and body contaminant levels in mussels exposed to different exposure regimes under laboratory conditions. Accordingly, the green-lipped mussel, Perna viridis, was challenged with a mixture of PAHs (anthracene, fluoranthene, pyrene, benzo[a]pyrene) and OC pesticides (α-HCH, aldrin, dieldrin, p,p′-DDT) over a 4 week period. Contaminants were delivered under four different dosing regimes, with all treatments receiving the same total contaminant load by the end of the exposure period. Antioxidant biomarkers were measured after 1, 2, 3 and 4 weeks, including glutathione (GSH), gluathione-S-transferase (GST), superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx), glutathione reductase (GR) and lipid peroxidase (LPO). GST and CAT were induced in hepatic tissues in most of the exposure regimes, with the majority of significant induction occurring in a constant exposure regime and a two-step alternate exposure regime. Significant differences among exposure regimes were detected in the body burden of contaminants after 28 days. Hepatic CAT and GSH are proposed as potentially useful biomarkers as they showed good correlation with target contaminants and were not readily affected by different dosing patterns.  相似文献

Biofilms on submerged surfaces are important in determining larval settlement of most marine benthic invertebrates. We investigated if exposure of biofilms to hypoxia would alter the larval settlement pattern and result in a shift in benthic invertebrate community structure in the field. Biofilms were first exposed to hypoxia or normoxia in laboratory microcosms for 7 days, and then deployed in the field for another 7 days to allow for larval settlement and recruitment to occur. Using terminal-restriction fragment length polymorphism of the 16S rRNA gene, this study showed that hypoxia altered the biofilm bacterial community composition, and the difference between the hypoxic and normoxic treatments increased with the time of exposure period. This study also demonstrated significantly different benthic invertebrate community structures as a result of biofilm exposure to hypoxia and that the hypoxic and normoxic treatments were dominated by Hydroides sp. and Folliculina sp., respectively.  相似文献

The freshwater pearl mussel (Margaritifera margaritifera) is a highly specialized and sensitive freshwater bivalve, whose survival in the juvenile phase is indicative of high quality habitats. This contribution investigates the use of juvenile freshwater pearl mussels as bioindicators, considering the influence of mussel stock and study stream conditions on juvenile performance, as described by survival and growth rates. A standardized cross experiment was carried out investigating juvenile performance in four different pearl mussel stocks originating from the Rhine, Danube and Elbe drainages, representing distinct genetic conservation units. The juveniles were exposed in five study streams which were selected to integrate pearl mussel streams with different water qualities and recruitment status of the mussel population. Per study stream, five standard mesh cages containing an equal number of 20 (10 × 2) juvenile pearl mussels per stock in separate chambers were installed. Survival and growth rates of juveniles were checked after three months (i.e. before their first winter) and after nine months (i.e. after their first winter). Mussel stock and study stream conditions significantly influenced juvenile performance. Growth rates were determined by study stream conditions and increased with stream water temperature, organic carbon and C/N ratios. Survival rates varied stock-specifically, indicating different levels of local adaptation to their native streams. Due to the detection of stream-specific differences in juvenile performance, freshwater pearl mussels appear suitable as bioindicators. However, a careful consideration of stock-specificity is necessary to avoid false interpretation of bioindication results. The comparison of stock-specific survival in native versus non-native streams implicates that exposure of juveniles outside their native habitats is able to increase breeding success or else serve for risk spreading in breeding programs.  相似文献

The artificial mussel (AM), a novel chemical sampling device, has been developed for monitoring dissolved trace metals in marine environments. The AM consists of Chelex-100 suspended in artificial seawater within Perspex tubing and enclosed with semi-permeable polyacrylamide gel at both ends. To validate the field performance of the AM in temperate waters, we deployed AMs alongside transplanted blue mussels Mytilus edulis in coastal environments in Scotland (Holy Loch, Loch Fyne, Loch Striven and Millport) and Iceland (Reykjavikurhöfn, Gufunes, South of þerney, Hofsvik, Hvalfjörður and Sandgerði) for monitoring trace metals. While uptake patterns of Cd between the AM and M. edulis were highly comparable, discrepancies were found in the accumulation profiles of the other metals (Cu, Cr, Pb and Zn), in particular Zn. Nonetheless, the AMs gave a better resolution to accurately reveal the spatial difference in dissolved metal contamination when compared with M. edulis. AMs complement the use of mussels since AMs indicate dissolved metals in seawater, whereas uptake by mussels indicates a mixture of dissolved and particulate metals. Our results also indicated that historical metal exposure of the transplanted M. edulis could significantly confound their metal concentrations especially when the deployment period was short (i.e. <34 d). This study suggested that the AM can overcome problems associated with variable biological attributes and pre-exposure history in the mussel, and provides a standardized and representative time-integrated estimate of dissolved metal concentrations in different marine environments.  相似文献

Identification of organisms able to act as biological indicators of hypoxia exposure is important given the increasing frequency of hypoxic events worldwide. Grass shrimp, Palaemonetes pugio, are ubiquitous estuarine residents in the southeastern US, where they commonly experience cyclic hypoxia. We hypothesized that reproductive and gene expression patterns of grass shrimp in cyclic hypoxic field sites relative to normoxic sites would be similar to previous results from cyclic hypoxia laboratory experiments showing differential up- and down-regulation of hypoxia-responsive genes. There were few differences in gene expression of indigenous shrimp collected during summer from two Gulf of Mexico estuarine systems [East Bay, FL (EB) and Weeks Bay, AL (WB)], although down-regulation of the gene coding for vitellogenin-1 corresponded with a decreased relative fecundity and fewer ovigerous females at cyclic hypoxic field sites, suggesting an overall impact on reproduction. The gene expression profiles of grass shrimp caged for 7 days in field sites differed by estuary, but few hypoxia-responsive genes identified in laboratory studies were differentially expressed in field shrimp. Overall, genes coding for protein synthesis, protein degradation, carbohydrate and lipid metabolism and electron transport were mostly up-regulated in EB caged shrimp but generally down-regulated in WB caged shrimp and laboratory shrimp. Thus, caged grass shrimp from different bay systems exhibited profoundly different gene expression profiles. Such profiles may serve as sensitive bioindicators of differences in water quality, habitat quality or food resources among estuaries but are not effective as indicators of hypoxia exposure.  相似文献

The sinking of the tanker Prestige in November 2002 off the coast of Galicia resulted in the release of about 60,000 tons of heavy oil. The oil-spill provoked a serious environmental impact in Spanish and French coasts, which biological consequences are still being assessed. In this study we address the temporal dynamics of genetic diversity in some mussel populations impacted by the oil-spill. Changes in genetic diversity can be measured in natural populations provided that serial samples are available from before (year 2000) and after (years 2003, 2005) the oil-spill. Analyses of seven microsatellites indicate a weak but significant increase of genetic variation after the spill. This phenomenon is interpreted herein in terms of a balance between a enhanced genome mutability on microsatellite variation and a low genetic drift due to toxicants and asphyxia although other stochastic phenomena cannot be ruled out. Per locus annotation showed that in spite of the allelic changes observed in the period 2000-2005, the final size of most allelic series remained quite alike to those of year 2000. Present genetic data suggest that the genotoxic impact of the Prestige spill did not compromise the genetic diversity of studied mussel populations, at least regarding the genetic markers analysed.  相似文献

The Western Scheldt river and estuary is known to be highly polluted as it receives industrial, agricultural and domestic effluents from one of the most populated and industrialised zones in Europe. Aquatic organisms are exposed to pollutants, specifically metals that are present in different environmental phases, e.g. dissolved, suspended material or sediment phases. The objective of this study was to study the relationship that exists between environmental metal levels, the degree of metal uptake by aquatic organisms with the concomitant biological responses. For this purpose the bivalve mollusk, Mytilus edulis, was selected as bioaccumulation indicator organism. Environmental (water and sediment) and mussel samples were collected during the late winter (March 2000) from sampling sites in the Scheldt estuary. Sites were selected to represent a salinity gradient from the mouth of the estuary to the furthest distribution area of mussels upstream in the system. Together with standard water quality parameters (e.g. salinity, dissolved oxygen, dissolved organic carbon, etc.) concentrations of twelve metals were analysed in the water (dissolved and suspended matter) and sediments. Levels of these metals were also measured in the soft tissue of M. edulis, together with concomitant biomarker responses in resident mussel populations at each site. The biomarkers that were included in this study were condition index, scope for growth, survival in air, cell membrane stability, DNA damage, and metallothioneins. Data were subjected to multivariate statistical analysis. The physico-chemical parameters and metals in the environmental samples clustered the sites to reflect the distribution based on the salinity gradient. Bioaccumulation results revealed increased metal uptake along a pollution gradient with the highest metal bioaccumulation occurring at the upstream most sites and therefore closest to the in the industrial activities. However, the biomarker responses clustered the sites in a manner that reflected the influence of combination of internal exposure (bioaccumulation) and external exposure (physico-chemical conditions). These differences in biomarker responses clearly demonstrated were attributed to abiotic factors other than metal pollution alone e.g. localized short-term increases in increased suspended sediment concentrations and decreased dissolved oxygen concentrations.  相似文献

Responses of the green-lipped mussel Perna viridis (L.) to suspended solids   总被引:1,自引:0,他引:1  
Laboratory experiments were conducted to investigate the lethal and sublethal effects of suspended solids on the survival and physiological, behavourial and morphological changes of the green-lipped mussel Perna viridis collected from Tolo Harbour, Hong Kong. Results showed that P. viridis survived in all test conditions of suspended solids from 0 to 1200 mg/l over a period of 96 h. Physiological responses of the green-lipped mussel under 14-d exposure of suspended solids from 0 to 600 mg/l, followed by 14-d recovery in natural seawater, revealed no significant changes (p>0.05) in oxygen consumption and dry gonosomatic index for treatments in different concentrations of suspended solids and exposure time. Changes in clearance rate were only found to be significant (p<0.001) with exposure time. Responses in behavourial and morphological changes of the green-lipped mussel were also studied under similar experimental treatments and exposure time. Byssus production was significantly (p<0.001) related to exposure time. Gill damage, however, was significantly greater in treatments (p<0.001). Present findings suggested that P. viridis could tolerate a high level of suspended solids in the laboratory. There were dose-dependent effects of suspended solids on morphology of gill filaments. Implications of survival and responses of the green-lipped mussel to suspended solids in the marine environment are discussed.  相似文献

The occurrence of three endocrine disrupting compounds, 4-n-nonylphenol, triclosan and bisphenol A, was investigated in different bivalves originating from the Aegean Sea (Greece). The bioconcentration potential of these compounds was studied using the Mediterranean mussel (Mytilus galloprovincialis). Tissue samples were extracted by sonication. Analysis was performed by gas chromatography-mass spectrometry. According to the field survey results, the average concentrations of 4-n-nonylphenol, triclosan and bisphenol A, were 158, 461 and 404 ng g−1 (dry weight), respectively. During 28 days of exposure at 300 ng L−1, the tissue concentrations of compounds were constantly increased. Steady state was not observed up to the end of the experiment. Kinetic bioconcentration factors varied from 1.7 (4-n-nonylphenol and triclosan) to 4.5 L g−1 (bisphenol A). Following exposure, mussels were relocated to clean water for 28 days. This experiment revealed that depuration rates for all of the target compounds were lower than uptake rates. The biological half-lives of each compound ranged between 12 days (triclosan) and 26 days (bisphenol A).  相似文献

Biological monitoring of metals in marine waters often employs tissue residue analyses from bivalves. Chemical residues in tissues do not elucidate biological injury, however, whereas biomarkers, do afford sensitive stress indices. Physiological and cytological responses were evaluated in laboratory experiments from the mussel Perna viridis in order to assess the species’ suitability for pollution monitoring in the tropics. Periodic copper exposure increased tissue residues but failed to perturb cardiac activity. Copper destabilized haemocyte lysosomal membranes in a exposure-dependent relationship showing a sensitive, rapid, indication of stress. Cytological biomarkers from P. viridis have potential as indices of pollutant exposure and could facilitate effective biomonitoring of water quality throughout South-east Asia.  相似文献

Polycyclic aromatic hydrocarbons (PAHs) are important environmental pollutants due to their persistence and bioaccumulation potential both in organisms and in sediments. In this study, bioaccumulation and depuration experiments were performed employing local Mediterranean mussel species, Mytilus galloprovincialis, with two biomarkers: filtration rate and lysosomal stability (neutral red retention) assay. Benzo(a)anthracene (BaA) was chosen as the model PAH compound due to its common presence in several matrices in the marine environment. Bioconcentration Factors (BCFs) for the mussels exposed to different BaA concentrations were calculated from both kinetic rate constants and from the experimental data and found between 27-3184 and 16-2745, respectively. Experimental QSAR values were found comparable to the previous QSAR models created for BaA. The effect of BaA exposure on the mussels showed a more clear dose-response relationship according to the results of lysosomal stability compared to the filtration rate.  相似文献

The lysosomal membrane destabilization and the metallothionein content in the digestive gland cells of mussels (Mytilus galloprovincialis Lam.), collected along the east coast of the North Adriatic (Istrian and Kvarnerine coast, Croatia), were examined over a period of four years (1996–1999). The lysosomal membrane stability, as a biomarker of general stress, showed that the membrane labilization period in mussels from polluted, urban- and industrial-related areas was significantly decreased (p<0.05) when compared to mussels from control, clean sea water sites. In the harbour of Rijeka, the most contaminated site, the lysosomal membrane stability was reduced by more than 70% compared to the control. This method also proved to be a useful biomarker for detection of additional stress caused by short-term hypoxia that occurred once during this study inside the polluted and periodically quite eutrophic Pula Harbour. The concentration of metallothioneins in the mussel digestive gland, as a specific biomarker of exposure to heavy metals, did not reveal significant differences (p<0.05) between sites covered by this study.  相似文献
