Many aquatic organisms are negatively affected by exposure to high copper concentrations.We investigated the biochemical response of the mussel Mytilus coruscus(Mytiloida:Mytilidae) to copper exposure.In vivo bioassays using M.coruscus and different copper concentrations were conducted.The activity of six biomarkers,namely superoxide dismutase(SOD),catalase(CAT),acid phosphatase(ACP),alkaline phosphatase(AKP),glutamic-oxaloacetic transaminase(GOT) and glutamic-pyruvic transaminase(GPT) were measured.Survival rates decreased with increased copper concentrations and exposure times.The LC50 values at 48,72,and 96 h exposure were 0.48,0.37,and 0.32 mg/L,respectively.Within digestive glands,CAT activity increased with increasing Cu concentrations.The activity of AKP showed no significant change,while the remaining four enzymes showed decreasing activity with increasing Cu concentrations.Within the gills,AKP activity increased when the Cu concentration was 0.05 mg/L,but showed no significant changes at higher concentrations.Activity of CAT and ACP within gills tended to decrease with increasing Cu concentration.The activity of SOD and GPT decreased at an exposure concentration of 0.2 mg/L.GOT activity within gills decreased at 0.1 mg/L and increased at an exposure concentration of 0.2 mg/L.Within the adductor muscle,AKP activity increased at 0.05 mg/L but did not change at higher exposure concentrations.ACP activity within adductor muscle tissue showed no change,while activities of CAT,GOT and GPT decreased with increasing Cu concentrations.SOD activity within the adductor muscle tissue significantly decreased at the 0.02,0.05 and 0.2 mg/L exposure concentrations.Our results show tissue specific differences for the six biomarkers in for M.coruscus.Our findings provide the basis for the establishment of reference activity levels against which biomarker changes can be estimated,and are essential preliminary steps in development of in vivo bioassays.  相似文献

The pharmacokinetic profiles and sulfamethoxazole (SMX) acetylation process in turbot reared at 18°C were investigated. Either SMX (parent drug) or its acetylized metabolite, N4-acetylsulfamethoxazole (AcSMX), was administered intravascularly to turbot at a dosage of 50 mg/kg BW. Serum concentrations of the parent drug and its metabolite were both measured by HPLC, and the changes in concentration over time were analyzed in two- and non-compartment models because SMX treatment produced multiple peaks. The results demonstrated that the elimination half-life of the parent drugs, SMX and AcSMX, were 159.2 and 5.9 h, respectively. The apparent volume of distribution was 0.2 and 0.8 L/kg, and the clearance was 0.038 and 0.222 L/(h·kg), for SMX and AcSMX, respectively. SMX acetylation in turbot was 2.8%, and the deacetylation of AcSMX was 0.2%. These findings may be useful in optimizing SMX dosage regimens in turbot aquaculture.  相似文献

Scallop Chlamys farreri was exposed to different concentrations of benzo(a)pyrene (BaP) (0.5 μg/L, 1.0 μg/L, 10.0 μg/L and 50.0 μg/L) for 30 days in seawater. Specimens of Clarias gariepinus were treated with lethal(70,75,80,85,90,and 95 mg/L) and sub-lethal concentrations(8,12 and 16 mg/L) of uranyl acetate,a low-radiotoxicity uranium salt. The LC 50 value was registered as 81.45 mg/L. The protein and glycogen concentrations in liver and muscles were decreased in the fish exposed to sub-lethal concentrations. The red blood cell(RBC) and white blood cell(WBC) counts,haemoglobin(Hb) concentration and haematocrit(Hct) values were decreased. Different blood indices like mean corpuscular volume(MCV),mean corpuscular haemoglobin(MCH) and mean corpuscular haemoglobin concentration(MCHC) were negatively affected. Level of plasma glucose was elevated whereas protein was decreased. The level of calcium concentration(Ca) was declined in the blood of exposed fish whereas magnesium(Mg) remains unchanged. The activity level of glutamic-oxaloacetic transaminase(GOT) and glutamic-pyruvic transaminase(GPT) was elevated in exposed fish. These effects were more pronounced in the last period of exposure and in higher concentrations. Results of the present study indicate that uranyl acetate has adverse effects on C larias gariepinus and causes changes in the biochemical and hematological parameters of the fish.  相似文献

Turbot(Scophthalmus maximus L.) reddish body iridovirus(TRBIV) was propagated in turbot fin cells(TF cells) and inactivated as the TRBIV vaccine with its protection efficiency evaluated in this study.TF cells were cultured in 10% bovine calf serum(BCS)-containing MEM medium(pH7.0) at 22℃,in which TRBIV propagated to a titer as high as 105.6 TCID50 mL-1.The TRBIV was inactivated with 0.1% formalin and formulated with 0.5% aluminum hydroxide.The inactivated vaccine caused neither cytopathogenic effect(CPE) on TF cells nor pathogenic effect on turbots.After being administered with the vaccine twice via muscle injection,the turbot developed high-tittered TRBIV neutralizing antibodies in a dose-dependent manner.The vaccine protected the turbot from dying with an immunoprotection rate of 83.3% as was determined via subcutaneous vaccination in the laboratory and 90.5% via bath vaccination in turbot farms,respectively.The inactivated vaccine was very immunogenic,efficiently preventing tur-bot from death.It holds the potential of being applied in aquaculture.  相似文献

Using modified clay is one of the most promising methods for the mitigation of harmful algal blooms(HABs). However, the environmental impact of modified clay has become a subject of global concern. In this study, turbot(Scophthalmus maximus L.) embryos were used as a model to assess the effect of modified clay on this sensitive stage of fish development. Results show that the 24 and 48 h LC_(50)(median lethal concentrations) of the modified clay were 1.70 and 1.65 g/L, and the safe concentration was 0.47 g/L, which is much higher than the concentration used to treat HAB. The modified clay did not affect significantly the hatchability of turbot embryos but when the concentration exceeded 0.50 g/L, the deformity rate of newly hatched larvae increased significantly. The total length, specific growth rate(SGR)and yolk sac absorption rate of larvae reached their peaks at 0.50 g/L and then gradually decreased as the concentration of modified clay increased. Therefore, a moderate amount of modified clay does not harm the survival and hatching of turbot embryos, or the growth and development of newly hatched larvae.  相似文献

The polychaete Perinereis aibuhitensis, a key species in estuarine ecosystems, can improve the culture condition of sediment. Endosulfan is an organochlorine pesticide used globally to control insects and mites; however, it is a source of pollution in aquaculture as a result of runoff or accidental release. In this study, we evaluated the toxicity of endosulfan to polychaeta and its ability to improve polluted sediment. Specifically, the effects of a series of endosulfan concentrations (0, 1.25, 2.5, 5, 10, 15, and 20 mg/L) were investigated, and the results indicated that the 24-h median lethal concentration (24-h LC50) was 55.57 mg/L, while the 48-h median lethal concentration (48-h LC50) was 15.56 mg/L, and the safe concentration was about 1.556 mg/L. In a 30-d exposure experiment, the animal specimen could decompose endosulfan effectively while improving endosulfan-polluted aquatic sediment.  相似文献

Effects of iron and silicon nutrients and of orthogonal combinations of nitrogen, phosphorus, iron and silicon on the growth rate of three benthic diatoms were studied by using an in situ OD measurement method. The results of monofactor tests of iron and silicon showed that Fe-EDTA was better than ferric citrate in the culture of these diatoms; that the favourable concentration of Fe-EDTA was about 0.5 mg/L of Fe and that of silicate was 2.5 mg/L of Si or more. The orthogonal test showed that the optimal concentration ratios (mg/L) of nitrogen, phosphorus, iron and silicon were: 5.0: 0.25: 0.5: 2.0 forCocconeis scutellum var.parva; 2.5: 1.0: 1.0: 2.0 forAmphora coffeaeformis; and 5.0: 0.15: 0.5: 2.0 forN. mollis. The biomarkers determined in this experiment had important roles in detoxification, and showed great potential as biomarkers for oxidative stress. Controlled laboratory experiments designed to simulate field exposure scenarios are particularly useful in ascertaining biomarkers suitable for use with complex contaminant mixtures in the marine environment. 长江中游平原区面临着一系列严重的地质环境问题,其中地下水铵氮和磷的问题十分突出,但目前对于二者共存规律的认识还十分薄弱。以长江中游沿岸故道区为典型研究区,对采集的地下水样品进行了水文地球化学分析,并综合运用因子分析和随机森林模型探讨了铵氮和磷的赋存规律。结果表明:地下水整体处于还原环境中,NH4-N的质量浓度为0.03~71.0 mg/L(均值9.92 mg/L),P的质量浓度为0.02~3.38 mg/L(均值0.51 mg/L)。地下水中高浓度的铵氮与磷均主要为天然成因,但铵氮与磷的迁移富集过程存在差异:铵氮的迁移富集与含氮有机质矿化过程密切相关;磷的迁移富集与铁氧化物或氢氧化物的还原性溶解密切相关,而含磷有机质矿化是磷富集的次要过程。Eh很低的地下水环境易产生高浓度铵氮的地下水,相对中等的还原环境会产生高浓度的磷但通常不会伴生高浓度的铵氮;当地下水中S2-,DOC,I均处于相对较高的浓度水平时会伴生大量的铵氮,而磷的浓度在很大程度上受控于Fe2+浓度;当DOC,I和Fe2+浓度都高时,通常会出现铵氮和磷浓度都较高的现象。   相似文献

Two modes of regulating the water quality of experimental ponds in indoor raceway culture of Litopenaeus vannamei were evaluated using simple water treatment facilities. A self-made water purifying net, aeration stone, composite microbe preparation, and Ceratophyllum demersum were placed in the experimental ponds and the culture water was circulated along the raceway inside the pond using a paddle wheel aerator. In addition, the water quality in the experimental pond was improved by draining effluent from the pipeline at the bottom of ponds 7 and 8 (mode I) and exchanging the circulating water in pond 10 (mode II) with the reservoir water in pond 9 using a pump and pipeline. The water quality in the experimental ponds was similar in response to regulation using mode I or mode II. Water quality parameters in the experimental ponds were controlled within a suitable range by simple facilities during culture period without using any chemical treatments. The rich content of dissolved oxygen was maintained by the circular flow and continuous aeration of the pond water. The respective average values of the main water parameters in experimental ponds 7 and 10 in response to regulation of the water quality using modes I and II were as follows: pH 8.17 and 7.99; DO 5.16 mg/L and 5.97 mg/L; CODMn 18.45 and 12.61 mg/L; TAN (NH3-N) 0.854 mg/L (0.087 mg/L) and 0.427 mg/L (0.012 mg/L); NO2-N 0.489 mg/L and 0.337 mg/L. Moreover, the average body length and body weight of harvested shrimp of pond 7 and pond 10 were 7.56 cm and 8.99 cm, 5.10 g and 8.33 g, respectively. Furthermore, the survival rate, average biomass yield and average condition factor of the shrimp harvested were 70% and 60%, 2.54 kg/m2 and 2.14 kg/m2, and 0.675 g/cm and 0.927 g/cm, respectively. Linear equations describing the relationship between body length and culture time and cubic or power functions describing the relationship between body weight and body length were obtained based on evaluation of the growth data of shrimps throughout the culture period.  相似文献

We studied the effects of mercury (Hg2+) on antioxidant and digestive enzyme activities in terms of LC50 value and on hepatopancreas histostructures of juvenile Chinese mitten crabs Eriocheir sinensis in 40-day exposure to various concentrations of Hg2+ (0, 0.01, 0.05, 0.10, 0.20, and 0.30 mg/L). The results show that the activities of superoxide dismutase (SOD), glutathione peroxidase (GPX) and catalase (CAT) significantly increased in the concentrations of 0.01 and 0.05 mg/L, while that of enzyme decreased in 0.10, 0.20 and 0.30 mg/L treatments. Meanwhile, Hg2+ disrupted the histostructures of the hepatopancreas, causing decreases in activities of pepsin, tryptase, amylase, and cellulose, which are synthesized in the hepatopancreas. Moreover, as the Hg2+ concentration increased, the survival rate of the crabs decreased, worst at 56.57% in 0.30 mg/L. Therefore, although crabs are able to tolerate low levels of mercury pollution, high levels lead to cellular injury and tissue damage in hepatopancreas, which then loses some of its vital physiological functions such as absorption, storage, and secretion.  相似文献

用单因子对比实验和正交实验的方法研究了氮、磷、铁和硅对简单双眉藻生长的影响。结果表明:简单双眉藻对于硝酸钠和尿素均能很好地利用,其相对生长常数无显著差异,对柠檬酸铁和三氯化铁的利用亦无显著差异,FeC6H5O7-Fe的适宜浓度为0.1~0.5mg/L;Na2SiO3-Si的适宜浓度为0.8~1.6mg/L。正交实验表明,简单双眉藻氮、磷、铁、硅的最佳浓度(mg/L)配比为6.6∶0.66∶0.1∶0.4。  相似文献

A 3-month feeding experiment was conducted in an in-door seawater system to investigate the effect of dietary vitamin E (Ve) on the sperm quality of turbot (Scophthalmus maximus). D-α-tocopherol acetate was supplemented to the basal (control) diet (65.14 mg kg?1 Ve) to obtain low and high levels of dietary Ve (244.60 mg kg?1, LVe; 721.60 mg kg?1, HVe). Compared with the control, sperm concentration was significantly increased in Ve-supplemented groups (LVe and HVe); while relative sperm volume and testis-somatic index were significantly increased in group HVe only. Sperm motility duration was significantly longer in group HVe than in the control, but no significant difference was observed in percent motility among groups. Sperm size, the uniformity of mitochondrial size, and the integrity of mitochondria cristae and plasma membrane were improved by dietary Ve, especially in HVe. The content of Ve in testis and liver as well as polyunsaturated fatty acids in sperm increased with dietary Ve. These results suggested that dietary Ve, especially at the high level (721.60 mg kg?1), significantly improved sperm concentration and motility duration and maintained normal sperm morphology of turbot.  相似文献

To increase the cell concentration and the accumulation of astaxanthin in the cultivation of Haematococcus pluvialis, effects of different iron eleetrovalencies (Fe2+-EDTA and Fe3+-EDTA) and species (Fe-EDTA, Fe(OH)32x and FeC6H5O7) addition on cell growth and accumulation of astaxanthin were studied. Results show that different iron electrovalencies have various effects on cell growth and astaxanthin accumulation of H. pluvialis. Compared with Fe3+-EDTA, Fe2+-EDTA stimulate more effectively the formation of astaxanthin. The maximum astaxanthin content (30.70 mg/g biomass cell)was obtained under conditions of 18 μmol/L Fe2+-EDTA, despite the lower cell density (2.3x105 cell/ml)in such condition. Fe3+-EDTA is more effective than Fe2+-EDTA in improving the cell growth. Especially,the maximal steady-state cell density, 2.9x105 cell/ml was obtained at 18 μmol/L Fe3+-EDTA addition. On the other hand, all the various species of iron (EDTA-Fe, Fe(OH)32x, FeC6H5O7) are capable to improve the growth of the algae and astaxanthin production. Among the three iron species, FeC6H5O7 performed the best. Under the condition of a higher concentration (36 μmol/L) of FEC6H5O7, the cell density and astaxanthin production is 2 and 7 times higher than those of iron-limited group, respectively. The present study demonstrates that the effects of the stimulation with different iron species increased in the order of FeC6H5O7, Fe(OH)32x/x and EDTA-Fe.  相似文献
