中国对虾细胞色素P450基因CYP4原核表达条件优化   总被引:1,自引:0,他引:1

根据克隆得到的中国对虾(Fenneropenaeus chinensis)CYP4基因开放阅读框设计引物,构建了中国对虾CYP4基因原核表达载体p28a-CYP4,并对重组菌株Rosetta/28a-CYP4的原核表达条件进行了优化。结果表明:p28a—CYP4转化Rosetta后可实现CYP4基因的原核表达,SDS—PAGE分析显示其在56.0kDa处有显著诱导条带;通过对诱导温度、IPTG浓度、诱导时机(OD600)及诱导时间的优化表明,重组菌株Rosetta/p28a-CYP4的最佳诱导温度为37℃,最佳IPTG浓度为1.2mmol/L,最佳诱导时机及诱导时间分别为0.59和6h。  相似文献

以褐菖鲉(Sebastiscus marmoratus)细胞色素P4501A四种生物标志物,即鳃丝EROD活性、肝EROD活性、肝CYP1A mRNA和肝CYP1A蛋白为指标,在泉州湾海域选择6个站位(包括1个对照站位),开展了为期两周的野外监测实验,研究了上述生物标志物的变化特征以及它们同表层海水中石油类含量和沉积物物中16种优先监控的PAHs含量之间的相关性,并利用新一代综合生物标志物响应指数法(IBRv2)对监测站位石油类和PAHs污染效应进行了评价.结果显示,不同站位的上述四种生物标志物变化趋势基本一致;生物标志物的变化与相应站位表层水中石油类含量及沉积物中PAHs总含量相吻合;IBRv2分析结果表明,各监测站位IBRv2指数介于1.5~4.2之间,其中2号站环境污染压力最大,4号站最小.在复杂的海洋污染条件下,采用CYP1A多种生物标志物监测技术能够更加全面反映石油类等污染及其生物效应状况.  相似文献

Abstract. The supply of nitrogen (N) often limits the productivity of marine macrophytes. In vitro and in vivo assays of glutamine synthetase (GS) activity were employed to investigate patterns of N assimilation by the Mediterranean seagrass Posidonia oceanica (L.) DELILE. Biomass-specific GS activity wa measured in root tissue, in leaves within a shoot, in shoots collected at two sites during two season and over a depth range of 5–33 m. Root tissue was less important than shoot tissue in assimilating inorganic N in P. oceanica , due both to the small roots' biomass (ca. 3% of total plant biomass and greatly lower (10- to 50-fold) GS activities. While the GS activity and N assimilatory potentia (biomass × GS activity; μmol N-h--1) were greatest in leaf 2, leaves 1 and 3–5 assimilated N a significant rates. Shoots from a site characterized by elevated N availability in the winter water column and no significant sediment N reservoir exhibited GS activities that were 9-times higher than shoot from a more oligotrophic site. Shoot GS activities in July increased linearly from 5 to 33 m and wer correlated with light availability as defined by Hsat, (daily period during which photosynthetic reaction are light-saturated). This may represent metabolic compensation by P. oceanica to maintain N influx. Factors contributing to the ecological success of P. oceanica include the ability to assimilate N under conditions of severe light limitation (< 35 μmol photons. m-2. s-1), and metabolic plasticity to ensur the de novo generation of N-containing organic compounds.  相似文献

条纹斑竹鲨线粒体控制区和细胞色素b基因序列分析   总被引:1,自引:0,他引:1  
对厦门和湛江海域的条纹斑竹鲨Chiloscyllium plagiosum(Bennett,1830)]的线粒体DNA控制区和细胞色素6(Cytb)基因进行PCR扩增和测序,获得线粒体DNA控制区、Cytb基因和tRNA^Pro。基因全序列(GenBank序列号:EU363740~EU363752)。结果表明,tRNA^Pro。基因长度为69bp,控制区长度为1094~1096bp,Cytb基因全序列长度为1146bp。tRNA^Pro。基因非常保守,未发现碱基变异。所测30个个体中,控制区只有4个多态位点,5种不同的单倍型;Cytb基因全序列共有10个多态性位点,7种单倍型。Cytb基因的1146个碱基编码381个氨基酸,氨基酸链共有3个变异位点,4种单倍型,厦门群体具有4种单倍型,湛江群体只有1种单倍型。综合Cytb基因和控制区序列,2个条纹斑竹鲨群体共有10种单倍型,其中厦门群体有9种单倍型,湛江群体仅4种单倍型。研究结果表明条纹斑竹鲨在控制区和Cytb基因序列上的遗传多样性均较低,厦门群体的遗传多样性水平高于湛江群体。  相似文献

Mitochondrial DNA sequences were used to identify an invasive swimming crab found in Waitemata Harbour, New Zealand. A 457 base sequence of the cytochrome oxidase 1 gene was compared in New Zealand specimens and nine species of Charybdis from Australia and Asia. The New Zealand specimens aligned with C. japonica. The diagnostic morphological characters of C. japonica were also checked in 54 specimens of the species collected in Waitemata Harbour, and concur with the mtDNA result. This is the first record of C. japonica establishing populations outside its native range. C. japonica, along with C. hellerii and the Lessepsian migrant C. longicollis, are the only known invasive species of Charybdis. C. japonica and C. hellerii are two of the few Charybdis species that inhabit the intertidal zone, and it is likely that the intertidal characteristics of these species contribute to their success as invasive species.  相似文献

参考鳗鲡等鱼类线粒体 DNA序列进行了中国花鲈线粒体 DNA细胞色素 b基因片断的引物设计、PCR扩增及其序列测定。得到中国花鲈的碱基序列为 4 10 bp,其 A、T、G、C含量分别为 10 1bp(2 4 .6 3% )、112 bp(2 7.32 % )、72 bp(17.56 % )、12 5bp(30 .4 9% ) ,与鳗鲡等其他鱼类相同基因片断序列碱基含量相似。  相似文献

The preferential inorganic nitrogen source for the seagrass Zostera noltii was investigated in plants from Ria Formosa, South Portugal. Rates of ammonium and nitrate uptake were determined at different concentrations of these nutrients (5, 25 and 50 μm ), supplied simultaneously (NH4NO3) or separately (KNO3 and NH4Cl). The activity of the enzymes nitrate reductase (NR) and glutamine synthetase (GS) was also assessed. The results showed that ammonium is the preferential inorganic nitrogen source for Z. noltii, but, in the absence of ammonium, the species also has a high nitrate uptake capacity. The simultaneous availability of both inorganic nitrogen forms enhanced the uptake rate of ammonium and decreased the uptake rate of nitrate compared to when only one of the nitrogen forms was supplied. The activity of both enzymes was much higher in the leaves than in the roots, highlighting the importance of the leaves as primary reducing sites in the nitrogen assimilation process.  相似文献

测定了石鲈科的花尾胡椒鲷Plectorhynchus cinctus、少棘胡椒鲷Diagramma pictum、斜带髭鲷Hapaloge-nys nitens、大斑石鲈Pomadasys maculates和点石鲈Pomadasys kaakan5种鱼类的线粒体细胞色素b基因的部分序列402bp,选用鲤Cyprinus carpio作外群,生成代表石鲈科4个属鱼类的数据集,采用邻接法和最大简约法构建了分子系统树,结果显示髭鲷属Hapalogenys和胡椒鲷属Plectorhynchus之间的亲缘关系较近,这2个属与石鲈属Pomadasys亲缘关系相近;少棘胡椒鲷属Diagramma与其它3个属的亲缘关系较远,支持把少棘胡椒鲷从胡椒鲷属归入到少棘胡椒鲷属的分类。  相似文献

本研究旨在建立用超低水平液闪谱仪测定雨水、海水中的宇生核素32P3、3P的方法.通过对液闪谱仪的主要性能与技术参数进行优化设置,对液闪谱仪的效率及能量进行刻度,建立淬灭校正曲线,探讨影响测量本底的因素,实现液闪谱仪对样品的优化测量.并通过液闪谱仪对厦门沿岸地区雨水及厦门湾海水进行测量,得到2004年3月至2005年2月雨水中32P3、3P的月平均比活度分别为0.30×10-2~3.18×10-2、0.28×10-2~3.05×10-2Bq/dm3,33P/32P)A.R.月平均值介于0.74~0.96之间,平均为0.87±0.03;厦门湾海水中32P、33P的比活度分别为1.52×10-2~4.62×10-2、1.28×10-2~3.72×10-2Bq/m3,33P/32P)A.R.为0.81~0.91.  相似文献

黄河三角洲典型潮汐湿地碳、氮、磷生物地球化学特征   总被引:4,自引:0,他引:4  
选择位于黄河三角洲的广饶潮汐湿地为研究对象,对比了在不同的植被覆盖和水文条件下潮汐湿地土壤碳、氮、磷元素的垂直分布及生态化学计量学特征。结果表明,在芦苇(Phragmites australis)覆盖下的GRA2区域沉积物总有机碳(TOC)和总氮(TN)含量明显高于碱蓬(Suaeda salsa)覆盖下的GRA1区域。GRA1柱样TOC含量均值为1.798 mg/g,而GRA2柱样则为3.109 mg/g。GRA1柱样的TN含量范围在0.108~0.213 mg/g,均值为0.165 mg/g,而GRA2柱样TN含量范围则为0.307~0.473 mg/g,均值为0.353 mg/g。总体上,总磷(TP)含量的变化水平相对于TOC和TN来说较大,其垂直变异系数较高。而与TOC和TN相反的是,GRA2柱样中TP含量均值为0.298 mg/g,低于GRA1的0.388 mg/g,这可能跟GRA2区域芦苇的生长比碱蓬需要吸收更多的P元素有一定关系。研究区域沉积物C、N、P元素生态化学计量学比值普遍较低。相对较高的C/N值,较低的C/P值和N/P则显示黄河三角洲潮汐湿地的生物地球化学过程可能更多地受营养元素N和P等的限制,而前者可能是主要限制因素。本研究可为滨海湿地对全球碳和氮的储存提供基础数据,为滨海湿地可持续发展的管理和保护提供科学依据。  相似文献

2001年7月测定大鹏澳表层叶绿素a含量为2.61—19.87mg·m-3,初级生产力为404.08—1097.25mgC·(m2·d)-1,N∶P值远高于Redfield比值16。回归分析表明,该水域浮游植物的现存量或生长量并不受某个单一因子调控。在营养盐加富试验中,加入含有P系列的浮游植物增殖速度最快,揭示P是此时大鹏澳浮游植物的潜在限制因子。将大鹏澳浮游植物水样置于不同的N∶P值环境中进行实验,结果表明,浮游植物在N∶P值为30的梯度组中生物量最高,浮游植物生物多样性程度也最高,其次是N∶P值为16和8的梯度组。N∶P值为30的梯度组的浮游植物优势种的时间变化最稳定。  相似文献

Populations of continuously distributed marine organisms that disperse via pelagic larvae are often assumed to exist in a state of genetic panmixia because of their potential ability for long‐distance dispersal. However, obligate symbionts may have more restricted gene flow due to recruitment limitations, making host specificity an important potential driver of biodiversity. To explore the tension between broad dispersal ability and limited recruitment potential, we used sequences of mtDNA cytochrome c oxidase subunit I (COI) to assess the fine‐scale phylogeography of the cleaner shrimp, Ancylomenes pedersoni, an obligate symbiont of sea anemones, along 300 km of the Florida Reef Tract (FRT). The results indicate high genetic diversity within A. pedersoni populations along the FRT. Pair‐wise ?ST values indicate significant genetic structuring between northern (Fort Lauderdale and Upper Keys) and southwestern (Lower Keys) populations, with significant isolation by distance. Two divergent COI haplotype lineages were detected through statistical parsimony analysis: one evenly distributed across the entire FRT (Clade 1) and one found primarily in two sample localities in the Lower Keys (Clade 2). Phylogenetic analyses using 16S‐rDNA indicates that this genetic diversity is of paraphyletic origin, and possibly the result of multiple colonization events. These results reveal a complex demographic and evolutionary history for A. pedersoni populations and provide the first evidence of highly divergent intra‐specific lineages independently colonizing the FRT. Because the FRT is a highly impacted coral reef system, understanding phylogeographic patterns along it has value beyond documentation of the factors that generate genetic diversity in tropical reef systems: these data are critical for creating scientifically based management strategies.  相似文献

为探讨石斑鱼亚科鱼类的系统发育关系,测定了此亚科中具有代表性的石斑鱼属、鳃棘鲈属、九棘鲈属、驼背鲈属和白线光腭鲈属等5个属10种石斑鱼的细胞色素b基因全序列,分析了这些序列的特性,运用Kimura 2-parameter模型,以邻接法构建了分子系统树。结果表明,九棘鲈属与鳃棘鲈属位于系统进化树的基部,与石斑鱼类其它类群亲缘关系较远;驼背鲈属与白线光腭鲈属网结于石斑鱼属构成的分支中。  相似文献

随着对虾养殖业的发展,对虾疾病越来越受到人们的关注,尤其对虾病毒性疾病已成为各国学者研究的焦点之一。由于没有有效的抗病毒药物,早期预防就显得更为重要,而这依赖于快速、敏感的诊断方法。酶联免疫吸附试验(enzyme-linked immunosorbent assay,ELISA)是1972年由Engvall等在酶免疫试验(enzyme immunoassay,EIA)的基础上发展起来的。目前,在病毒等疾病有关的抗原抗体系统的定性或定量测定领域内已被广泛应用。本文通过病毒纯化,免疫新西兰兔,获得多克隆抗血清,并对抗体进行纯化、酶标记,建立了双抗体夹心直接ELISA的检测方法。  相似文献

随着P2P日益被人们重视,对P2P流特征的研究也越来越多。传统的研究都是以在网络出口截获网络流的分析为主,但是这些分析方法都是被动方式的,不仅无法了解到P2P应用的内部特征,有时还无法对某些流特征给出解释。通过运行典型的P2P应用——1个加入到实际网络中的BitTorrent客户端,以主动的方式统计了客户端对消息的收发情况,并观察和收集到客户端内部状态的变化特征。根据这些统计数据,总结BT协议在实际应用时的特点,分析了协议所使用的策略的性能,另外还针对消息在被动测量时出现的特征作了分析。  相似文献

古湖泊砂砾堤研究表明,24kaB.P.,兹格塘错、错鄂分别存在10m、30m(高出现代湖面)的高湖面,湖泊面积分别可达272km2、220km2,是现代湖泊的1.42、3.12倍.与此相比,9.0-6.0kaB.P.湖面略低;在兹格塘错,古湖泊高出现代湖面8m,面积约为246km2,比现代大56km2;在错鄂,古湖面高出现代12m,面积约为138km2.基于Kutzbach水能联合方程,利用逐次逼近法取得有关参数,重建的各流域上述各时期的降水量分别可达(400±20)mm/a (兹格塘错 24kaB.P.)、(535±20)mm/a (错鄂 24kaB.P.)、(370±20)mm/a (兹格塘错 9.0-6.0kaB.P.)、(470±20)mm/a (错鄂 9.0-6.0kaB.P.).  相似文献
