The health benefits of regular consumption of fish and seafood have been espoused for many years. However, fish are also a potential source of environmental contaminants that have well known adverse effects on human health. We investigated the consumption risks for largemouth bass (Micropterus salmoides; n = 104) and striped mullet (Mugil cephalus; n = 170), two commonly harvested and consumed fish species inhabiting fresh and estuarine waters in northwest Florida. Skinless fillets were analyzed for total mercury, inorganic arsenic, polychlorinated dibenzo-p-dioxins and polychlorinated dibenzofurans (PCDD/F), polychlorinated biphenyls (PCBs), and organochlorine pesticides. Contaminant levels were compared to screening values (SV) calculated using U.S. Environmental Protection Agency (EPA) recommendations for establishing consumption advisories. Largemouth bass were found to contain high levels of total mercury at all sampling locations (0.37–0.89 ug/g) and one location exhibited elevated total PCBs (39.4 ng/g). All of the samples exceeded Florida fish consumption advisory trigger levels for total mercury and one location exceeded the U.S. EPA SV for total PCBs. As a result of the high mercury levels, the non-cancer health risks (hazard index–HI) for bass were above 1 for all locations. Striped mullet from several locations with known point sources contained elevated levels of PCBs (overall range 3.4–59.3 ng/g). However, total mercury levels in mullet were low. Eight of the 16 mullet sampling locations exceeded the U.S. EPA SV for total PCBs and two locations exceeded an HI of 1 due to elevated PCBs. Despite the elevated levels of total PCBs in some samples, only two locations exceeded the acceptable cancer risk range and therefore cancer health risks from consumption of bass and mullet were determined to be low at most sampling locations.  相似文献

New cellular and molecular tools are enabling researchers to study the basic mechanisms of action of natural reproductive hormones, and of endocrine disrupters, including xenoestrogens. Zonagenesis is defined by the hepatic synthesis of the eggshell Zona radiata proteins (Zrp), their secretion and transport in the blood and their uptake by maturing oocytes. Zrps are comprised of three-to-four protein monomers in the 50–60 kDa MW range and show structural similarity to proteins of the Zona pellucida; an extracellular envelope that lies immediately outside the plasma membrane in the eggs of placental mammals. We studied the xenoestrogen-induced elevation in hepatic mRNA and plasma levels of Zrp and vitellogenin (Vtg) in several fish species. Eggshell Zrp protein and mRNA was induced in juvenile salmon by exposure to nonylphenol (NP: given singly or in combination with other xenoestrogens/xenobiotics). In English sole (Pleuronectes vetulus), Zrp was induced in the plasma of mature female, E2- and NP-treated male fish. We are currently using salmon cDNA probes to study estrogen-receptor, Zrp and Vtg mRNA expression in English sole treated with E2- and NP. Validation in a semi-field experiment showed that effluent from an oil refinery treatment plant caused a dose-dependent induction of Zrp and Vtg in juvenile salmon. However, a component corresponding to Zrp-β was more responsive than Zrp-α and Zrp-γ and also more than Vtg. Zrp seem to be a sensitive complement to Vtg analysis, and appear to be a more ecologically significant biomarker of estrogenicity.  相似文献

In this research the hypothesis that Mediterranean top predator species (such as large pelagic fish) are potentially at risk due to EDCs is investigated. These marine organisms tend to accumulate high concentrations of EDCs such as polyhalogenated aromatic hydrocarbons (PHAHs). The potential effects of EDCs on a fish species of commercial interest, the top predator Xiphias gladius (swordfish), were investigated using vitellogenin (Vtg) and Zona radiata proteins (Zrp) as diagnostic and prognostic biomarkers. Dramatic induction of typically female proteins (Vtg and Zrp) was detected by ELISA and Western Blot in adult males of the species. These results are the first warning of the potential risk for reproductive function of Mediterranean top predators, and suggest the need for continuous monitoring of this fragile marine environment.  相似文献

Multixenobiotic resistance mechanism (MXR) in aquatic organisms is mediated by the activity of the P-glycoprotein (Pgp) transporter that binds and actively effluxes different chemicals out of cell. In addition to the Pgp, several other, non-Pgp transport proteins have been recently identified in different human and animal tissues. Given their characteristics and tissue distribution we hypothesized that members of the so-called multidrug resistance-associated protein (MRP) family may be expressed in aquatic organisms. This study attempted to identify MRP related genes in different tissues of several marine and freshwater bivalves (Mytilus galloprovincialis, Dreissena polymorpha, Anodonta cygnea) and fish species (Mullus barbatus, Cyprinus carpio, Salmo trutta). Following an alignment of known MRP1 and MRP2 human sequences, as well as the GenBank available mrp2 sequences from different animals, we determined highly conserved regions and used them to design three pairs of consensus primers. Total RNA was isolated, reverse transcribed to cDNA and the obtained cDNAs were PCR amplified with the corresponding primers. The amplified PCR products were sequenced and their homology compared with Pgp and MRP protein sequences from different species. The expression of MRP related mRNA was clearly identified only in liver tissue isolated from red mullet, with homologies at the protein level ranging from 75% to 76%. Described results clearly pointed at the possibility that at least in the red mullet MXR as a general defense mechanism may be mediated by the activities of at least two different types of transport proteins.  相似文献

Previous studies have shown that aryl hydrocarbon receptor (AhR) agonists possess anti-estrogenic activities and several mechanisms have been proposed to explain the interactions between AhR and estrogen receptor (ER) signalling pathways. In the present study, we show that 3,3'4,4'5-pentachlorobiphenyl (PCB126 - a dioxin-like AhR agonist) produced estrogenic responses in the absence of ER agonist, in fish in vitro system. We exposed salmon primary hepatocytes to PCB126 (1, 10 and 50 pM) and the ER agonist nonylphenol (NP; 5 and 10 microM) singly and also in combination. Vitellogenin (Vtg) and zona radiata proteins (Zr-proteins) levels were analysed by semi-quantitative ELISA. We observed that the protein levels of Vtg and Zr-proteins were significantly induced in a concentration-specific manner in cells treated with PCB126 and NP, singly or in combination. In general, these results show a novel aspect of dioxin-like PCB effect not previously demonstrated in fish system.  相似文献

Tilapia is a common fish species inhabiting inland waters and estuarine regions in Hong Kong and Southeast Asia, and useful for bio-monitoring of metal pollution. Metallothionein (MT) gene expression in fish tissues has been useful to sub-lethal risk assessment as biomarker of exposure to metal ions in fishes inhabiting metal contaminated area. To investigate metal inductions of Tilapia MT gene expression in vivo, Tilapias were injected with different concentrations of heavy metals and tissues were then removed for quantitative PCR assay using mimic PCR methods. All of the metal ions tested (Cu(2+), Cd(2+), Hg(2+), Ni(2+), Pb(2+) and Zn(2+)) were able to induce hepatic MT mRNA levels. Renal MT mRNA levels of Cd(2+) and Zn(2+) treated fish was not induced with significant fold induction, however MT mRNA levels in gills were sensitive to the administrations of these metal ions. These data indicated that Tilapia MT mRNA levels in gills and liver are sensitive biomarker of exposure to various metal ions.  相似文献

Metallothionein (MT) is a protein found in most eukaryotes where its primary role is the regulation of homeostasis of the essential metals copper and zinc. MTs have an additional protective role through their binding of toxic metals such as cadmium and mercury. Increased expression of MT in response to harmful levels of these metals has been demonstrated for several aquatic species. In this study, the expression of MT in the liver tissue of the New Zealand common bully Gobiomorphus cotidianus was measured by quantifying MT mRNA. A statistically significant increase in MT expression was observed in fish exposed to copper for 48 h, but not those exposed to zinc. Field sampling of common bullies showed a wide variation in individual hepatic MT mRNA levels which did not correlate with fish age, sex, or sampling location. A comparison of two populations of common bullies from a polluted and a control site showed a two‐fold higher mean MT mRNA level in fish from the polluted site. Implications for using MT expression in the common bully as a bio‐monitoring tool are discussed.  相似文献

World-wide many cetaceans drown incidentally in fishing nets. To reduce the unwanted bycatch in gillnets, pingers (acoustic alarms) have been developed that are attached to the nets. In the European Union, pingers will be made compulsory in some areas in 2005 and in others in 2007. However, pingers may effect non-target marine fauna such as fish. Therefore in this study, the effects of seven commercially-available pingers on the behaviour of five North Sea fish species in a large tank were quantified. The species tested were: sea bass (Dicentrarchus labrax), pout (Trisopterus luscus), thicklip mullet (Chelon labrosus), herring (Clupea harengus), and cod (Gadus morhua). The fish were housed as single-species schools of 9-13 individuals in a tank. The behaviour of fish in quiet periods was compared with their behaviour during periods with active pingers. The results varied both between pingers and between fish species. Sea bass decreased their speed in response to one pinger and swam closer to the surface in response to another. Thicklip mullet swam closer to the bottom in response to two pingers and increased their swimming speed in response to one pinger. Herring swam faster in response to one pinger, and pout and cod (close relatives) showed no behavioural responses to any of the pingers. Of the seven pingers tested, four elicited responses in at least one fish species, and three elicited no responses. Whether similar responses would be elicited in these fish species in the wild, and if so, whether such responses would influence the catch rate of fisheries, cannot be derived from the results of this study. However, the results indicate the need for field studies with pingers and fish. Based on the small number of fish species tested, the present study suggests that the higher the frequency of a pinger, the less likely it is to affect the behaviour of marine fish.  相似文献

Lipid peroxidation, which is promoted in animal tissues by a variety of toxic agents, is believed to be associated with disruption of cell membranes and loss of activity of membrane-bound enzymes. While the process of lipid peroxidation has been well studied in mammals, only a few non-mammalian species have been examined.1 In the present study, in vitro assays for lipid peroxidation were performed using microsomes prepared from the livers of rats (Rattus rattus) and a marine fish, mullet (Mugil cephalus). Although lipid peroxidation was observed in microsomes prepared from both mullet and rat tissues, and was stimulated by xenobiotics, the mechanisms of generation appeared to be different. In contrast to rats, NADH was much more effective than NADPH in stimulating lipid peroxidation in mullet liver microsomes. The results suggest that NADH-dependent, as well as NADPH-dependent, lipid peroxidation may be important in vertebrate tissues.  相似文献

Coastal urban environments have high plastic pollution levels, and hence interactions between plastic debris and marine life are frequent. We report on plastic ingestion by mullet Mugil cephalus in Durban Harbour, KwaZulu-Natal, South Africa. Of 70 mullet (13.0–19.5?cm total length), 73% had plastic particles in their guts, with a mean of 3.8 particles per fish (SD 4.7). Plastic ingestion showed no relation to digestive tract content or fish length. White and clear plastic fibres were ingested most commonly. This urban population of M. cephalus had a higher incidence of plastic ingestion than has been reported in studies on fish from other coastal areas or the oceanic environment.  相似文献

The hypothesis that Mediterranean top predator species, such as large pelagic fish, are potentially at risk due to endocrine disrupting chemicals (EDCs), is investigated. The potential estrogenic effects of PHAHs in three fish species of commercial interest, the top predators bluefin tuna (Thunnus thynnus thynnus), swordfish (Xiphias gladius), and Mediterranean spearfish (Tetrapturus belone), were investigated using vitellogenin (Vtg), zona radiata proteins (Zrp) and mixed function oxidases (EROD, BPMO) as diagnostic tools. High induction of Vtg and Zrp was detected by western blot and ELISA techniques in adult males of X. gladius and T. thynnus thynnus, suggesting that these species are at high toxicological risk in the Mediterranean sea. Comparison of BPMO and EROD activities in the three species indicated, both in male and female, much higher MFO activity in bluefin tuna. This data suggests high exposure of this species to lipophilic xenobiotic contaminants in the Mediterranean environment.  相似文献

The short-term effects of the commercial PBDE flame retardant mixtures Penta-BDE and cta-BDE on the expression of cytochrome P450 1A (CYP1A), vitellogenin (Vtg) and zona radiata proteins (Zrp) were investigated in juvenile salmon (Salmo salar). For this purpose, groups of fish were dosed twice (oral intake at days I and 4) with 10 and 50 mg/kg body weight of both commercial mixtures. The fishes were sacrificed at day 7 (n = 5 for each group) and 14 (n = 6 for each group), and blood, liver, fillet, and brain were collected. Blanks and positive controls were also part of the experiment. The expressions of Vtg, Zrp, and CYPIA were measured with several techniques (EROD, ELISA, Western, Northern and Slot Blot). The values in the groups of fish treated with Penta-BDE or Octa-BDE did not significantly differ from the reference group for any of the parameters tested. In contrast, the positive control groups treated with estradiol-17beta for Vtg and Zrp expression, and beta-naphthoflavone for CYP1A expression did show a significant response, indicating the potential sensitivity of the fishes for the parameters measured. Since the results of the chemical analyses showed concentrations of a number of PBDE congeners in liver, fillet, and brain that were about three orders of magnitude above those of fish from the North Sea, it is concluded that the short-term toxicity of both commercial PBDE mixtures for these endpoints was low.  相似文献

Vitellogenin (Vtg) is a yolk protein precursor that has been identified as a sensitive biomarker for exposure to estrogenic compounds. We evaluated specific monoclonal and polyclonal antibodies for reactivity with plasma Vtg from two Australian Perciformes, the tropical barramundi (Lates calcarifer) and the temperate black bream (Acanthopagrus butcheri). Blood plasma from 17beta-estradiol exposed (E2) male barramundi (20 mg kg(-1)) and male black bream (2.5-5.0 mg kg(-1)) were sent to Biosense Laboratories (Norway) for cross-reactivity testing using their extensive anti-Vtg antibody selection. Indirect ELISA results determined barramundi plasma displayed the highest binding affinities to ND-3G2 (monoclonal-Mab) and PO-1 (polyclonal-Pab). Black bream was most cross-reactive with ND-1C8 (Mab) and PO-2 (Pab). Next, plasma was assessed for Vtg induction in E2-dosed (5 mg kg(-1)), hatchery-reared barramundi and black bream versus a non-injected control group. Vtg production was assessed by Western blot and indirect ELISA using ND-3G2 and ND-1C8 Mabs, respectively. A prominent band was identified in the range of 100-200 kDa for all female black bream and for all E2-treated barramundi and black bream males, which was confirmed as Vtg by Western blot. Indirect ELISA results for barramundi demonstrated highly significant differences in E2-dosed fish as compared to control fish (Student t, P<0.001). E2 male black bream were significantly different than control males (Student t, P<0.001) and control and E2 females displayed highly significant differences (Student t, P<0.001). These results indicate that exposure to 17beta-estradiol induces significant Vtg production in males of the two Australian Perciformes, with potential use as a biomarker for exposure to estrogenic compounds.  相似文献

World-wide, underwater background noise levels are increasing due to anthropogenic activities. Little is known about the effects of anthropogenic noise on marine fish, and information is needed to predict any negative effects. Behavioural startle response thresholds were determined for eight marine fish species, held in a large tank, to tones of 0.1-64 kHz. Response threshold levels varied per frequency within and between species. For sea bass, the 50% reaction threshold occurred for signals of 0.1-0.7 kHz, for thicklip mullet 0.4-0.7 kHz, for pout 0.1-0.25 kHz, for horse mackerel 0.1-2 kHz and for Atlantic herring 4 kHz. For cod, pollack and eel, no 50% reaction thresholds were reached. Reaction threshold levels increased from approximately 100 dB (re 1 microPa, rms) at 0.1 kHz to approximately 160 dB at 0.7 kHz. The 50% reaction thresholds did not run parallel to the hearing curves. This shows that fish species react very differently to sound, and that generalisations about the effects of sound on fish should be made with care. As well as on the spectrum and level of anthropogenic sounds, the reactions of fish probably depend on the context (e.g. location, temperature, physiological state, age, body size, and school size).  相似文献

双锯鱼(Amphiprion)亦称为海葵鱼,又称为小丑鱼,是一类经济价值较高的海水观赏鱼。目前国内在双锯鱼的胚胎发育、人工饲养以及形态学观察等方面已经取得了有效成果,但在分子生物学水平上对其基因表达的研究较少。为了筛选出适用于双锯鱼胚胎不同发育阶段以及成鱼组织的内参基因,分析酪氨酸酶(TYR)基因的表达情况,以眼斑双锯鱼(A.ocellaris)和白条双锯鱼(A.frenatus)为材料,利用实时定量聚合酶链式反应(qPCR)对18SrRNA、GAPDH(甘油醛-3-磷酸脱氢酶基因)、Ef-1α(转录延伸因子基因)和β-actin(β-肌动蛋白基因)这4个候选内参基因的表达水平进行检测,同时通过geNorm和Norm Finder软件对其稳定性进行评估,最后以合适的内参基因作为参考,研究TYR mRNA的表达水平。结果表明,在双锯鱼胚胎的不同发育阶段,18S和β-actin的表达量相对于其他基因较为稳定;在双锯鱼的不同组织中,稳定性依次为18S>β-actin>Ef-1α>GAPDH。以18S作为内参基因时分析发现TYR基因的表达在双锯鱼胚胎发育过程中呈先上升后下降的趋势,在体节期表达量最高;在双锯鱼各组织中均有表达,且在眼、尾和红皮中表达量最高。  相似文献

In this project we investigated the ecotoxicological effects of endocrine disrupters in a four-year survey of the Mediterranean population of swordfish (Xiphias gladius). In the Mediterranean environment, top predators, such as swordfish, accumulate high concentrations of polyhalogenated aromatic hydrocarbons (PHAHs) and toxic metals, potentially incurring high toxicological risk. The effects of organochlorines and trace elements (Hg, Cd and Pb) in 192 swordfish specimens, caught in the Strait of Messina, Sicily, Italy, were investigated using vitellogenin (Vtg), zona radiata proteins (Zrp) and CYP1A (BPMO, EROD) activities. Vtg and Zrp were found to be dramatically induced in some adult male specimens, suggesting that this species is highly exposed to estrogens in the Mediterranean Sea. A role of organochlorines in this induction phenomenon is suggested by the statistically significant correlations between Zrp in plasma and PCB concentrations in muscle (p<0.032) and Vtg in plasma and PCB concentrations in liver (p<0.034) of male specimens. Levels of trace elements in liver were in the following ranges: Hg 1-22, Cd 1-28 and Pb 0-1.6 ppm d.w. These data indicate potential reproductive alterations in large pelagic fish and suggest the need for continuous monitoring to avoid reductions in the population of this fish species of high commercial and ecological interest.  相似文献

Concentrations of ten polybrominated diphenyl ethers (PBDEs) and eight methoxylated polybrominated diphenyl ethers (MeO-PBDEs) in mullet (Mugil cephalus) and sea bass (Dicentrarchus labrax) collected from the Bizerte Lagoon and the Mediterranean Sea were investigated. To the best of our knowledge, this is the first report of these compounds in marine fishes from Tunisia. The PBDE mean concentrations in fish from Bizerte Lagoon were 45.3 and 96.2 ng g(-1) lw respectively in mullet and sea bass, while the concentrations of these compounds in mullet and sea bass from Mediterranean Sea were 7.80 and 27.9 ng g(-1) lw respectively. MeO-PBDE concentrations in mullet and sea bass from Bizerte Lagoon ranged from 6.46 to 286 ng g(-1) lw and from 49.4 to 798 ng g(-1) lw respectively, while the concentrations of these compounds in mullet and sea bass from Mediterranean Sea ranged from 190 to 401 ng g(-1) lw and from 353 to 578 ng g(-1) lw respectively. The total PBDEs and total MeO-PBDEs concentration in fish from Bizerte Lagoon were similar or slightly lower than those reported for other species from other locations around the world.  相似文献
