In this project we investigated the ecotoxicological effects of endocrine disrupters in a four-year survey of the Mediterranean population of swordfish (Xiphias gladius). In the Mediterranean environment, top predators, such as swordfish, accumulate high concentrations of polyhalogenated aromatic hydrocarbons (PHAHs) and toxic metals, potentially incurring high toxicological risk. The effects of organochlorines and trace elements (Hg, Cd and Pb) in 192 swordfish specimens, caught in the Strait of Messina, Sicily, Italy, were investigated using vitellogenin (Vtg), zona radiata proteins (Zrp) and CYP1A (BPMO, EROD) activities. Vtg and Zrp were found to be dramatically induced in some adult male specimens, suggesting that this species is highly exposed to estrogens in the Mediterranean Sea. A role of organochlorines in this induction phenomenon is suggested by the statistically significant correlations between Zrp in plasma and PCB concentrations in muscle (p<0.032) and Vtg in plasma and PCB concentrations in liver (p<0.034) of male specimens. Levels of trace elements in liver were in the following ranges: Hg 1-22, Cd 1-28 and Pb 0-1.6 ppm d.w. These data indicate potential reproductive alterations in large pelagic fish and suggest the need for continuous monitoring to avoid reductions in the population of this fish species of high commercial and ecological interest.  相似文献

This paper describes the development of a RT-PCR method for assaying Vtg gene expression in different marine fish as a potentially valuable and sensitive biomarker of exposure to estrogenic chemicals. The levels of Vtg mRNA have been analyzed using primers specifically designed for the various species and the procedures have been standardized relative to actine mRNA expression levels. Different species were analyzed including organisms with a great potential as bioindicators in the Mediterranean (i.e. the red mullet Mullus barbatus, the striped mullet Mugil cephalus, the European eel Anguilla anguilla) or exposed to biomagnification of halogenated hydrocarbons and with elevated commercial value (the bluefin tuna Thunnus thynnus). The analysis of vitellogenin mRNA levels has been standardized in feral fish providing suitable indications for a future development of this approach.  相似文献

The short-term effects of the commercial PBDE flame retardant mixtures Penta-BDE and Octa-BDE on the expression of cytochrome P450 1A (CYP1A), vitellogenin (Vtg) and zona radiata proteins (Zrp) were investigated in juvenile salmon (Salmo salar). For this purpose, groups of fish were dosed twice (oral intake at days 1 and 4) with 10 and 50 mg/kg body weight of both commercial mixtures. The fishes were sacrificed at day 7 (n=5 for each group) and 14 (n=6 for each group), and blood, liver, fillet, and brain were collected. Blanks and positive controls were also part of the experiment. The expressions of Vtg, Zrp, and CYP1A were measured with several techniques (EROD, ELISA, Western, Northern and Slot Blot). The values in the groups of fish treated with Penta-BDE or Octa-BDE did not significantly differ from the reference group for any of the parameters tested. In contrast, the positive control groups treated with estradiol-17β for Vtg and Zrp expression, and β-naphthoflavone for CYP1A expression did show a significant response, indicating the potential sensitivity of the fishes for the parameters measured. Since the results of the chemical analyses showed concentrations of a number of PBDE congeners in liver, fillet, and brain that were about three orders of magnitude above those of fish from the North Sea, it is concluded that the short-term toxicity of both commercial PBDE mixtures for these endpoints was low.  相似文献

The feeding and spawning habitats of the overfished Atlantic bluefin tuna (BFT) are mapped in the Mediterranean Sea and used in the present proposal for selecting restricted fishing grounds. The feeding habitat is mainly traced by oceanic fronts of satellite-derived temperature and chlorophyll while the spawning habitat is mostly characterized by an important heating of surface waters. The proposal recommends opening the fishery in feeding areas in case the BFT stock is low (current situation). Only spawning areas at its latest stage could be opened once the stock has recovered to its optimum yield. Due to the possible concentration of fishing vessels if fishing areas are restricted (e.g. four-fold increase with a 1/16th restriction of the Mediterranean Sea) the inspection activities could be better targeted. Identified spawning grounds, opened or closed to fishing, could also be particularly monitored by control operations. Within the authorized areas, the habitat maps would guide fishermen to the favourable habitat reducing their costs. The habitat guided management could be able to adapt the spatial and temporal distribution of the effort to the requirements of both the fisheries’ control and the resource. Its implementation is likely to protect the stock (a) by apparently decreasing illegal fishing which accounts in the recent years for more than 1/3rd of total catches, (b) by protecting the spawners to ensure a suitable recruitment and (c) by distributing the effort to respect the population structure. The first species studied is the emblematic bluefin tuna which is at high risk of collapse due to overfishing. The approach is a priori transposable to other epipelagic species of commercial importance.  相似文献

研究了实验室条件下原油水溶性组分(WSF)暴露对黑鲷、黄鳍鲷和褐菖鲉肝微粒体EROD活性的剂量-效应,时间-效应和恢复过程。实验结果表明,在剂量诱导实验中,褐菖鲉肝EROD活性在原油WSF浓度为50μg/dm3时呈现生物统计学上的显著差异,而黑鲷和黄鳍鲷肝EROD活性在75μg/dm3时才呈现生物统计学上的显著差异;褐菖鲉肝EROD活性诱导倍数最高,但黑鲷的诱导浓度范围较广。在时间诱导实验中,在40μg/dm3原油WSF暴露下黄鳍鲷肝EROD活性在2 d时首先呈现显著差异;三种鱼肝EROD活性均在第4天达到最高,并呈现显著性变化,此后随着暴露时间的延长而逐渐下降并接近对照组水平。在恢复实验中三种鱼肝EROD活性下降并恢复到对照组水平。研究结果表明:对于石油污染物,黑鲷、黄鳍鲷和褐菖鲉肝EROD活性都可以作为污染生化效应监测指标,然而就三种鱼类比较而言,褐菖鲉最敏感,更适合于作为石油类污染及其生化效应,尤其是低剂量效应的监测生物。  相似文献

Various studies have revealed high concentrations of contaminants such as organochlorines (OCs) and heavy metals in Mediterranean cetaceans. A geographical trend of contamination (PCBs and DDTs) has been found for striped dolphin (Stenella coeruleoalba). In this study we used a non-lethal approach (skin biopsy) to investigate bioaccumulation of OCs, including polychlorobiphenyls (PCBs), DDTs, polychlorodibenzo-p-dioxins (PCDDs), polychlorodibenzofurans (PCDFs), trace elements (Hg, Cd, Pb) and CYP1A activity (BPMO) in nine striped dolphins sampled in the Aeolian area (Sicily – Italy) in summer 2002. The arithmetic mean value of BPMO activity in this group was 43.46 AUF/g tissue/h. This value is approximately 3 times and 5 times lower, respectively, than the value found in the Ionian and in the Ligurian groups. Skin biopsies of striped dolphins emerged as a suitable material for assessing the toxicological status of the various Mediterranean groups.  相似文献

Resource partioning among the planktivorous stages of eight fish species occurring in a Mediterranean coastal lagoon was studied. Five species were migratory mullets that spawned in the sea (Liza ramada, L. aurata, L. saliens, Chelon labrosusandMugil cephalus), while the other three species were resident and spawned in the lagoon (Atherina boyeri, Cyprinus carpioandGambusia holbrooki).Mullet fry exhibited similar diets, based on the consumption of zooplanktonic Crustacea such as copepods and cladocerans, although adult chironomids were also important.Gambusia holbrookiandA. boyerifed on small prey such as copepod nauplii, copepodites and rotifers, whileC. carpioconsumed larger prey preferentially (cladocerans and copepods).According to the trophic overlap and niche width results, seven species pairs could be competing in the estuary, although further experimental evidence is still required.  相似文献

The recent overexploitation of East Atlantic and Mediterranean bluefin tuna stock has been well documented in the media where it has become the archetype of overfishing and general mis-management. Beyond the public debate, the crisis also highlighted how the interactions between science and management can change through time according to the awareness of the public opinion. To reflect these issues, the history of Atlantic bluefin tuna overfishing is first described. Then, the major uncertainties that undermine the current scientific advice are summarized and the importance of reducing their impacts by improving knowledge and developing robust scientific framework is considered. The study also discusses how uncertainty was used by different lobbies to discredit science-based management. The recent improvement in bluefin tuna stock status following the implementation of the rebuilding plan shows that, despite uncertainty in the scientific advice, the management of a heavily exploited fish stock can be positive when there is a political will. However, optimizing long-term yields of modern fisheries implies good science. The study concludes by advocating for the implementation of a scientific quota that should be part of the management framework to support the scientific advice.  相似文献

Conjugation of phenolic xenobiotics and metabolites through sulfation and glucuronidation is an important biotransformation pathway. Sulfotransferases (SULT) are generally considered non-inducible, while some UDP-glucuronosyltransferase (UGT) isoenzymes are co-induced with cytochrome P450-1A by Ah-receptor ligands. To test these assumptions for two fish species, we measured sulfation and glucuronidation of 9-hydroxybenzo[a]pyrene in 3-methylcholanthrene (3-MC) treated channel catfish (Ictalurus punctatus) and in mummichog (Fundulus heteroclitus) from the creosote contaminated Atlantic Wood site in the Elizabeth River, VA. The results show a significant induction of both UGT and SULT activity in 3-MC treated catfish, linked to the expected induction of EROD activity. In mummichog, significant induction of UGT was measured at the contaminated site over the reference site (King's Creek, VA), as well as extremely low SULT activities at both sites. Western blots, using a polyclonal antibody for catfish phenol-type SULT, confirmed the absence of phenol-type SULT in mummichog. Residual, though slightly inducible, SULT activity may be attributed to other SULT isoforms.  相似文献

Early life stages from a marine fish species, Fundulus heteroclitus, were exposed to sublethal doses of 3,3',4,4',5 pentachlorobiphenyl (PCB126) to evaluate its effects on ecologically relevant responses: growth and behavior. A few hours after fertilisation, eggs were treated topically with PCB126 (2.5-50 pg egg?1). Four days post-hatching (dph), morphological changes (body length and malformations), spontaneous locomotor activity (active swimming speed, rate of travel, % inactivity), prey capture ability (Artemia franciscana nauplii) and whole body EROD activity were evaluated in larvae. Untreated larvae collected at 0.5, 1, 2, 3, 4 dph were also examined. PCB126 did not increase the mortality or malformation rates. Body length and spontaneous locomotor activity were altered only in larvae treated with the highest dose. Treatment with PCB126 caused a dose-responsive reduction in prey capture ability (rate of decline in the number of Artemia) and induction of EROD activity. The lowest observed effective dose for both of these responses was 5.0 pg PCB126 egg?1 or 5.0 TCDD-toxic equivalents pg g?1 egg, using a TCDD-toxic equivalent factor of 0.005 and an egg mass of 5 mg. Prey capture efficiency (number of Artemia captured per feeding strike) was reduced at ≥ 10.0 pg egg?1. In untreated developing larvae, prey capture ability and efficiency increased as post-hatching development progressed and EROD activity remained low. The pattern of behavioral responses observed in PCB126-exposed Fundulus larvae differed from that observed in less-developed larvae indicating that other mechanisms than retarded development were involved. Behavioral dysfunction was a more sensitive response to PCB126 than morphological alterations and it occurred at environmentally relevant concentrations.  相似文献

This work aims to compile knowledge on ecologically rich habitats that deserve special protection in the Mediterranean high seas. Many marine ecosystems are currently threatened by fishing activities, such as bottom trawling, gillnets and demersal and pelagic long-lines. Especially vulnerable are areas described as either sensitive habitats or essential fish habitats. Among these we highlight the associations of sessile organisms, such as cold coral reefs, mostly detected on continental slopes, seamounts and on the walls of submarine canyons, and those areas considered as hot spots of diversity and habitat of vulnerable fauna, like cold seeps, hydrothermal vents and submarine canyons. Essential habitats for pelagic species are defined by oceanographic features like productive areas associated with upwelling, and in the Mediterranean the spawning areas and migratory routes of bluefin tuna, swordfish, and albacore are of high conservation interest. We propose that in a context of difficult fisheries management in Mediterranean international waters, or high seas, these vulnerable habitats should be protected through the establishment of a web of Marine Protected Areas coordinated by all the involved countries and controlled by strict surveillance.  相似文献

Aquaculture of bluefin tuna in Mediterranean coastal waters has generated growing concern about the negative environmental effects. In the present isotopic study we examined the dispersal and fate of organic matter derived from a Mediterranean tuna farm in the surrounding environment. An overall enrichment in the heavy nitrogen isotope was found in the feed and in farmed tunas, indicating the input of isotopically traceable organic matter in the system. Waste was clearly traceable in the water column up to 1000 m from the cages, while only slight accumulation occurred in the sediment just below the cages. Waste was isotopically shown also to contribute to the diet of demersal and benthopelagic wild fish collected around the cages. As a result, waste undertook multiple pathways. In the water column its was diluted and dispersed due to hydrodynamism, which prevented great accumulation of aquaculture-derived organic matter in sediments. In addition, biological constraints such as benthopelagic and demersal fish further prevented organic matter accumulation through the benthic trophic route.  相似文献

Various studies on Mediterranean cetaceans have revealed bioaccumulation of contaminants such as organochlorines (OCs) and heavy metals. The susceptibility of these animals to organic pollutants and the relationship between bioaccumulation and population decline (as in the case of Delphinus delphis) are unexplored fields. In this study, we used a non-destructive approach (skin biopsy) to explore OC bioaccumulation processes and mixed-function oxidase activity (BPMO) in four species of cetaceans: striped dolphin (Stenella coeruleoalba), bottlenose dolphin (Tursiops truncatus), common dolphin (D. delphis) and fin whale (Balaenoptera physalus). Significant differences in BPMO induction and OC levels were found between odontocetes and mysticetes, the former having mixed-function oxidase activities four times higher than the latter, binding with levels of OCs one order of magnitude higher in odontocetes. A significant correlation (P < 0.05) between BPMO activities and OC levels was found in B. physalus. In an ongoing project, fibroblast cultures have been used as an alternative in vitro method of evaluating interspecies susceptibility to contaminants such as OCs and polycyclic aromatic hydrocarbons (PAHs). These results suggest that cetacean skin biopsies are a powerful non-invasive tool for assessing ecotoxicological risk to Mediterranean marine mammals species.  相似文献

Epibenthic fishes were collected with daytime beam trawl tows (n = 1713) in three shallow (<10 m) habitats of submerged aquatic vegetation (SAV), Zostera marina (eelgrass), Laminaria longicruris (kelp), Phyllophora sp. (algae), and unvegetated sandy/mud areas. We divided the Maine coast into three broad zones based upon geological features and sampled over five consecutive years; during April–November 2000 in the mid coast, in 2001 and 2002 along the south coast and in 2003 and 2004 along the eastern Maine coast. We quantified habitat use by eight economically important fish species (Gadus morhua, Microgadus tomcod, Pollachius virens, Urophycis chuss, Urophycis tenuis, Osmerus mordax, Tautogolabrus adspersus, and Pseudopleuronectes americanus) and 10 other common epibenthic species (n = 18 571). We identified the physical and biological variables most important in discriminating between habitats with and without individual fish species. Logistic regression models based on nearshore habitat characteristics were developed to predict the distribution of these species along the three zones representing broad geological regions of the Maine coast. Logistic regression models correctly classified individual fish species 58.7–97.1% of the time based on the temporal and physical habitat variables (month, temperature, salinity, and depth) and the presence–absence of submerged aquatic vegetation (Zostera, Laminaria, or Phyllophora). Overall fish presence and economically important fish presence were correctly classified 61.1–79.8% and 66.0–73.6% of the time, respectively. The Maine shallow water fish community was composed primarily of young-of-the-year and juvenile fishes with all habitats functioning as facultative nursery areas. Presence of most fish species was positively associated with Zostera, Laminaria, and to a lesser extent, Phyllophora. This study provides direct evidence of shallow waters of the Gulf of Maine as critical facultative nursery habitat for juvenile G. morhua, M. tomcod, P. virens, U. tenuis, U. chuss, T. adspersus, O. mordax and P. americanus, and many ecologically important species.  相似文献

An enhanced version of the spatial ecosystem and population dynamics model SEAPODYM is presented to describe spatial dynamics of tuna and tuna-like species in the Pacific Ocean at monthly resolution over 1° grid-boxes. The simulations are driven by a bio-physical environment predicted from a coupled ocean physical–biogeochemical model. This new version of SEAPODYM includes expanded definitions of habitat indices, movements, and natural mortality based on empirical evidences. A thermal habitat of tuna species is derived from an individual heat budget model. The feeding habitat is computed according to the accessibility of tuna predator cohorts to different vertically migrating and non-migrating micronekton (mid-trophic) functional groups. The spawning habitat is based on temperature and the coincidence of spawning fish with presence or absence of predators and food for larvae. The successful larval recruitment is linked to spawning stock biomass. Larvae drift with currents, while immature and adult tuna can move of their own volition, in addition to being advected by currents. A food requirement index is computed to adjust locally the natural mortality of cohorts based on food demand and accessibility to available forage components. Together these mechanisms induce bottom-up and top-down effects, and intra- (i.e. between cohorts) and inter-species interactions. The model is now fully operational for running multi-species, multi-fisheries simulations, and the structure of the model allows a validation from multiple data sources. An application with two tuna species showing different biological characteristics, skipjack (Katsuwonus pelamis) and bigeye (Thunnus obesus), is presented to illustrate the capacity of the model to capture many important features of spatial dynamics of these two different tuna species in the Pacific Ocean. The actual validation is presented in a companion paper describing the approach to have a rigorous mathematical parameter optimization [Senina, I., Sibert, J., Lehodey, P., 2008. Parameter estimation for basin-scale ecosystem-linked population models of large pelagic predators: application to skipjack tuna. Progress in Oceanography]. Once this evaluation and parameterization is complete, it may be possible to use the model for management of tuna stocks in the context of climate and ecosystem variability, and to investigate potential changes due to anthropogenic activities including global warming and fisheries pressures and management scenarios.  相似文献
