Over the past few decades, an increasing number of marine activities have been conducted in the East China Sea, including the construction of various marine structures and the passage of large ships. Marine safety issues are paramount and are becoming more important with respect to the likely increase in size of ocean waves in relation to global climate change and associated typhoons. In addition, swells also can be very dangerous because they induce the resonance of floating structures, including ships. This study focuses on an investigation of swells in the East China Sea and uses hindcast data for waves over the past 5 years in a numerical model, WAVEWATCH III (WW3), together with historical climate data. The numerical calculation domain covers the entire North West Pacific. Next, swells are separated and analyzed using simulated wave fields, and both the characteristics and generation mechanisms of swells are investigated.  相似文献

Karenia is one of the most harmful and representative red tide genus in a temperate zone. Blooms caused by this genus have resulted in massive fish death in the South China Sea and the East China Sea. However, the potential effects of this dinoflagellate on human health through the transfer of toxins via marine food webs, and the mechanisms of toxicity, are still unknown. Therefore, we examined the toxic effects of a strain of K. mikimotoi (isolated from the South China Sea) on the proliferation and morphology of four mammalian cell lines (two normal cell lines and two cancer cell lines). In addition, we carried out a preliminary investigation on the mechanism of toxicity of the alga. The results show that the polar lipid-soluble component of K. mikimotoi significantly inhibited proliferation of the four cell lines, and resulted in the cells becoming spherical, swollen and damaged. The result of Annexin V and PI double-staining confirmed that cell membranes were disrupted. The malonaldehyde (MDA) contents in the medium of the four cell lines treated with the polar-lipid extracts all increased significantly, which indicates that the polar-lipid toxins produced by K. mikimotoi could adversely affect mammalian cells by inducing lipid peroxidation. We conclude that K. mikimotoi is a potential threat to human health, and the comprehensive effect of this dinoflagellate and its mechanisms should be investigated further.  相似文献

Marine microalga Nannochloropsis oceanica LAMB0001 were domesticated(-73 0 generations,-two days each)to adapt freshwater BGll medium.A number of freshwater medium adapted colonyderived strains were obtained.The strains were verified phylogenetically to be N.oceanica LAMB0001 based on the 18 S ribosomal RNA gene.Freshwater-medium adapted strain(FA1)grew faster in the BG11 medium prepared with freshwater than wild-type N.oceanica grew in f/2 medium prepared with seawater.We assumed that(1)the expression patterns of the genes that expressed differentially between FA1 and the wild-type N.oceanica exposing to the BG11 medium(WT-F)have been reprogrammed;(2)the physiological processes in which these genes involved have been modified;and(3)a Gene Ontology(GO)term or a KEGG pathway enriched by DEGs between FA1 and WT-F has been up-or down-regulated if it was enriched simultaneously by up-or down-regulated DEGs between FA1 and WT-F,respectively.Under these assumptions,we found that FA1 reprogrammed the expression patterns of a set of genes that involved in cell adhesion,membrane and membrane integrity,material transportation,cell movement,and cellular signaling network.These changes in cellular functions and metabolic pathways indicate that the microalga modified its gene expression pattern in a wide function range and at a high regulation rank in order to adapt to the freshwater medium.It is feasible to domesticate marine microalgae to a freshwater habitat,which may aid to modify their cultivation performances.  相似文献

Fibroblast growth factor 19(FGF19) functions as a hormone by affecting glucose metabolism. FGF19 improves glucose tolerance when overexpressed in mice with impaired glucose tolerance or diabetes. A functional cellular FGF19 receptor consists of FGF receptor(FGFR) and glycosaminoglycan complexed with either α Klotho or β Klotho. Interestingly, in mice with diet-induced diabetes, a single injection of FGF1 is enough to restore blood sugar levels to a healthy range. FGF1 binds heparin with high affinity whereas FGF19 does not, indicating that polysaccharides other than heparin might enhance FGF19/FGFR signaling. Using a FGFs/FGFR1 c signaling-dependent Ba F3 cell proliferation assay, we discovered that polyguluronate sulfate(PGS) and its oligosaccharides, PGS12 and PGS25, but not polyguluronate(PG), a natural marine polysaccharide, enhanced FGF19/FGFR1 c signaling better than that of heparin based on ~3H-thymidine incorporation. Interestingly, PGS6, PGS8, PGS10, PGS12, PGS25, and PGS, but not PG, had comparable FGF1/FGFR1 c signal-stimulating activity compared to that of heparin. These results indicated that PGS and its oligosaccharides were excellent FGF1/FGFR1 c and FGF19/FGFR1 c signaling enhancers at cellular level. Since the inexpensive PGS and PGS oligosaccharides can be absorbed through oral route, these seaweed-derived compounds merit further investigation as novel agents for the treatment of type 2 diabetes through enhancing FGF1/FGFR1 c and FGF19/FGFR1 c signaling in future.  相似文献

Gongga Mountain, locates on the eastern edge of Tibetan Plateau of China, is the highest mountain in China except summits in Tibet. Only limited meteorological data on Gongga Mountain have been published so far. Here we present the meteorological records from two stations, Moxi Station (at 1,621.7 m above sea level (a.s.l.), 1992–2010) and Hailuogou Station (at 2,947.8 m a.s.l., 1988–2010), on the eastern slope of Gongga Mountain. In the past two decades, the annual precipitation decreased while the annual mean temperature increased at Hailuogou Station. Both precipitation and temperature increased at Moxi Station. The precipitation variation on the eastern slope of Gongga Mountain is influenced by both East Asian Monsoon and Indian Monsoon, so that the precipitation concentrated between May and October. The temperature variation on the eastern slope of Gongga Mountain in the past two decades showed similar trends as that of the northern hemispheric and global. In the past two decades, the temperature increased 0.35°C and 0.3°C/decade at Hailuogou Station and Moxi Station respectively, which was higher than the increase extents of northern hemispheric and global temperature. The most intense warming occurred at the first decade of 21st century. The winter temperature increased more at Hailuogou Station than at Moxi Station. A remarkable increase of temperature in March was observed with only a little precipitation at both high and low altitude stations.  相似文献

Volatile organic compounds in marine algae have been reported to comprise characteristic flavor of algae and play an important role in their growth, development and defensive response. Yet their biogeneration remain largely unknown. Here we studied the composition of volatile compouds in Pyropia yezoensis and their variations in response to methyl jasmonate(MeJA) and diethyldithiocarbamic acid(DIECA) treatment using gas chromatography-mass spectrometry(GC-MS). A total of 44 compounds belonging to the following chemical classes(n) were identified, including aldehydes(11), alcohols(8), acids and esters(6), alkanes(5), ketones(5), alkenes(3), and S-or N-containing miscellaneous compounds(6). External treatment with plant hormone MeJA increased the content of 1-dodecanol, 4-heptenal, and 2-propenoic acid-2-methyl dodecylester, but decreased the content of phytol, 3-heptadecene, 2-pentadecanone, and isophytol. When pretreated with DIECA, an inhibitor of the octadecanoid pathway leading to the biosynthesis of endogeneous jasmonates and some secondary metabolites, phytol and isophytol were increased, while 4-heptenal, 1-dodecanol, and 2-propenoic acid-2-methyl dodecylester were decreased, both of which were negatively correlated with their variations under MeJA treatment. Collectively, these results suggest that MeJA does affect the volatile composition of P. yezoensis, and the octadecanoid pathway together with endogenous jasmonate pathway may be involved in the biosynthesis of volatile compounds, thereby providing some preliminary envision on the composition and biogeneration of volatile compounds in P. yezoensis.  相似文献

Experiments were designed to expose the filter-feeding bivalve Perna viridis to different Cd-contaminated water environments in order to compare the different pathways through which Cd is accumulated. Results show that mussels can accumulate Cd through seawater, food, sediment and suspended particle pathways in a short period of time. Mussels' uptake of Cd through the seawater pathway reaches the highest concentration approximately 3 and 9 times larger than through the algae and sediment pathways respectively after 7 d. This indicates that the Cd-accumulation through seawater is most efficient. Results also indicate that the uptake directly through contaminated algae, particles or sediments ingested by mussels is less important when compared with the uptake of Cd by mussels through the seawater pathway. Metal uptake pathways and mechanisms of bioaccumulation by marine bivalve are also discussed in this paper.  相似文献

Serotonin (5-HT) has been found to stimulate meiotic maturation of oocytes in many molluscs. During maturation, several signaling pathways are involved, especially the phosphatidylinositol and cAMP pathways. In order to examine the possible role of the phosphatidylinositol signaling pathway in regulating oocyte maturation in Mercenaria mercenaria, the effects of the activator/inhibitor of phospholipase (PLC) and protein kinase C (PKC) on serotonin-induced maturation were studied. Results show that high-concentrations of neomycin (inhibitor of PLC) blocked oocyte maturation, while 9, 10-dimethyl-1, 2-benzanthracene (DMBA, activator of PLC) promoted oocyte maturation in the presence of serotonin. It was also found that in the presence of serotonin, phorbol 12-myristate 13-acetate (PMA, activator of PKC) inhibited oocyte maturation, while sphingosine (inhibitor of PKC) stimulated oocyte maturation. These results indicate that serotonin-induced oocyte maturation requires the activation of the phosphatidylinositol pathway. Decrease of PLC inhibited serotonin-induced oocyte maturation, whereas a decrease of PKC stimulated the maturation. Thus, our study indicates that serotonin promotes maturation of M. mercenaria oocytes through PLC stimulated increase in calcium ion concentration via inositol-1, 4, 5-trisphosphate (IP3) but not PKC.  相似文献

通过对2 131km高分辨率浅地层剖面资料分析和研究,揭示了威海市近海海域浅地层结构特征,分析了海洋灾害地质因素类型及分布特征,并绘制了主要灾害地质因素分布图。威海市近海海域65m以浅地层共分为5层,第一层为全新世泥质楔形体,地层厚度8~15m;第二层为末次盛冰期以来的海侵层,厚度一般小于3m;第三层为MIS2期的陆相沉积层,地层厚度3~8m;第四层为MIS3期的海相沉积层,厚度3~6m;第五层未完全揭示。近海海域灾害地质因素主要包括埋藏下切谷、浅部断层、浅层气、海底沙波沙脊、浅埋起伏基岩等5种类型;埋藏下切谷主要分布在南部海域U3地层单元中;浅部断层主要分布在南部海域水深大于20m的海底堆积平原;浅层气主要分布在北部海域,东部和南部海域面积较小;海底沙波沙脊主要分布在荣成成山头外海海域;浅埋起伏基岩主要分布在基岩海岸和岛屿较近的海域。  相似文献

【目的】综述重金属元素镉Cd和铜Cu对海洋浮游植物的毒性效应研究,总结重金属对海洋浮游植物的毒性作用机制,为开展重金属对海洋浮游植物毒害机理的深入研究提供借鉴。【方法】以毒理实验中常用的重金属镉和铜为代表,总结了重金属对海洋浮游植物毒理研究的常用测试指标,归纳了重金属对海洋浮游植物光合作用、抗氧化系统等方面的毒性作用机制。【结果】重金属对海洋浮游植物毒性作用机制概括为:1)重金属替换与其结构相似的作为酶辅助因子的金属元素,使浮游植物体内某些酶失活;2)重金属直接或间接诱导活性氧的产生,使浮游植物受到氧化胁迫;3)重金属与生物大分子中的某些基团亲和性高而结合,阻断相关的生理生化过程。【结论】重金属的毒性效应因浮游植物种类不同而有所差异,今后相关研究中需更加关注重金属在多种海洋浮游植物共存环境下的毒性效应及机制,将有助于全面系统地理解重金属对海洋浮游植物的致毒机理。  相似文献

Recently, the widespread use of TiO_2 nanoparticles(nano-TiO_2) has attracted the global concern regarding their adverse effects to aquatic organisms. Nanoparticles may get to the aquatic environments and harm the growth of algae. This paper presents the studies of nano-TiO_2 concentrations influence on the photosynthetic characteristics, antioxidant system and antitumor activity of marine macroalgae Gracilaria lemaneiformis after exposed to 0, 5, 10, 20 or 40 mg L-1 of nano-TiO_2. Results showed that the photosynthetic pigment content of G. lemaneiformis obviously decreased and the antioxidant defense system was significantly disturbed after exposure to nano-TiO_2. Compared with control, the maximum level of malondialdehyde showed 3.3-fold increase for 15 days exposure to nano-TiO_2. The activity of superoxide dismutase and catalase enhanced obviously by 1.10-2.24-fold. The aqueous extract of G. lemaneiformis(AEGL) possesses anticancer activity against HepG2 liver cancer cells proliferation and the 48 h-cell inhibition exceeds 35%. But the toxicity of nano-TiO_2 would inhibit the anticancer activity of AEGL. When G. lemaneiformis was exposed to nano-TiO_2, the effect of the aqueous extract of G. lemaneiformis on the cell inhibition, reactive oxygen species production, apoptosis, cycle arrest of HepG2 cells significantly lessened, and which were linearly associated with nano-TiO_2 exposure concentration. Those results further confirmed that nano-TiO_2 can inhibit the anticancer activity of G. lemaneiformis. The results in this work imply that macroalgae can be an effective biomarker of nano-TiO_2 contaminant, and can be useful indicator to evaluate ecological effects of increasing pollutants in marine ecosystem.  相似文献

This study investigated the molecular mechanism of anti-tumor effect of fascaplysin, a nitrogenous red pigment firstly isolated from a marine sponge. Microarray analysis show that the TNF and TNF receptor superfamily in human umbilical vein endothelial cells (HUVEC) and human hepatocarcinoma cells (BEL-7402) were significantly regulated by fascaplysin. Western Blot results reveal that fascaplysin increased the expression of cleaved caspase-9, active caspase-3, and decreased the level of procaspase-8 and Bid. Flow cytometry and cytotoxicity tests indicate that fascaplysin sensitized cells to tumor necrosis-related apoptosis-inducing ligand-(TRAIL) induced apoptosis, which was markedly blocked by TRAIL R2/Fc chimera, a dominant negative form of TRAIL receptor DR5. Therefore, our results demonstrate that fascaplysin promotes apoptosis through the activation of TRAIL signaling pathway by upregulating DR5 expression.  相似文献

We studied the phagotrophic ability of dinoflagellate strain Karenia mikimotoi KM-Lü (isolated from the South China Sea), using fluorescent microspheres, bacteria isolated from the culture of K. mikimotoi and a marine microalgae Isochrysis galbana. We found that K. mikimotoi cultured under conditions of high light intensity could ingest fluorescent microspheres (diameters 0.5 and 2.0 μm) and fluorescence-labeled bacteria and microalgae. Under a low light intensity, however, only fluorescent microspheres (diameter 0.5 μm) and fluorescence-labeled microalgae were ingested. K. mikimotoi showed better growth by ingesting living marine bacteria or microalgae I. galbana than the controls, either in nutrient-depleted or nutrient-replete conditions. In nutrient-depleted conditions, the growth of K. mikimotoi was more significant with I. galbana as the prey item. In conclusion, the harmful dinoflagellate K. mikimotoi from the South China Sea has apparent phagotrophic ability, and some marine bacteria and microalgae may promote the growth of K. mikimotoi.  相似文献
