The effects of temperature, light intensity, salinity, and initial pH on the growth and fatty acid composition of Pinguiococcus pyrenoidosus 2078 were studied for eicosapentaenoic acid (EPA) production potential. The fatty acid composition was assayed by gas chromatography-mass spectrometry, which indicated that the main fatty acids were C14:0, C16:0 and EPA. The highest EPA percentage 20.83% of total fatty acids was obtained at 20℃ with the temperature being set at 20, 24, and 28℃. Under different salinities and light intensities, the highest percentages of total polyunsaturated fatty acids (PUFAs) and EPA were 17.82% and 31.37% of total fatty acids, respectively, which were achieved at salinity 30 and 100μmol photon m-2s-1 illumination. The highest percentages of total PUFAs and EPA were 38.75% and 23.13% of total fatty acids, respectively, which were reached at an initial pH of 6 with the test range being from 5.0 to 9.0.  相似文献

From March 2004 to February 2005,seasonal variations in lipid content and fatty acid composition of gonad of the cockle Fulvia mutica(Reeve) were studied on the eastern coast of China in relation to the reproductive cycle and environment conditions(e.g.,temperature and food availability).Histological analysis as well as lipid and fatty acid analyses were performed on neutral and polar lipids of the gonad.Results showed that gametogenesis occurred in winter and spring at the expense of lipids previously accumulated in summer and autumn,whereas spawning occurred in summer(20.4-24.6℃).The seasonal variation in lipid content was similar to that of the mean oocyte diameter.In both neutral and polar lipids,the 20:5n-3 and 22:6n-3 levels were relatively higher than saturated fatty acids,and polyunsaturated fatty acids were abundant,with series n-3 as the predominant component.Seasonal variations in the 20:5n-3 and 22:6n-3 levels and the principal n-3 fatty acids were clearly related to the reproductive cycle.The ∑(n-3) and ∑(n-6) values were relatively high during January-May,and the associated unsaturation index was significantly higher than that in other months.The results suggest that fatty acids play an important role in the gametogenesis of F.mutica.  相似文献

This study was conducted to compare lipid and fatty acid composition of cod, haddock and halibut.Three groups of cod (276 g±61 g), haddock (538 g±83 g) and halibut (3704 g±221 g) were maintained with commercial feeds mainly based on fish meal and marine fish oil for 12 weeks prior to sampling.The fatty acid compositions of muscle and liver were determined by GC/FID after derivatization of extracted lipids into fatty acid methyl esters (FAME).Lipids were also fractionated into neutral and polar lipids using Waters silica Sep-Pak.The phospholipid fraction was further separated by high-performance thin-layer chromatography (HPTLC) and the FAME profile was obtained.Results of the present study showed that cod and haddock were lean fish and their total muscle lipid contents were 0.8% and 0.7%, respectively, with phospholipid constituting 83.6% and 87.5% of the total muscle lipid, respectively.Halibut was a medium-fat fish and its muscle lipid content was 8%, with 84% of the total muscle lipid being neutral lipid.Total liver lipid contents of cod, haddock and halibut were 36.9%, 67.2% and 30.7%, respectively, of which the neutral lipids accounted for the major fraction (88.1%-97.1%).Polyunsaturated fatty acids were the most abundant in cod and haddock muscle neutral lipid.Monounsaturated fatty acid level was the highest in halibut muscle neutral lipid.Fatty acid compositions of phospholipid were relatively constant.In summary, the liver of cod and haddock as lean fish was the main lipid reserve organ, and structural phospholipid is the major lipid form in flesh.However, as a medium-fat fish, halibut stored lipid in both their liver and muscle.  相似文献

This study investigated the effects of long-term low-salinity on the growth performance,proximate composition,amino acid and fatty acid profiles of Scylla paramamosain.The salinity of the low-salinity-culture group(LC)and maintained-salinity-cul-ture group(MC)were set at 5 and 25 respectively.After rearing for 30 d,the survival rate(SR)of the LC group was significantly lower,whereas the average daily molting frequency(ADMF)was significantly higher than those in MC group(P<0.05).There were no significant difference for weight gain rate(WGR)and specific growth rate(SGR)between the two groups.The moisture content of the LC group was slightly higher than the MC group without significant difference.The contents of ash,crude lipid and crude pro-tein in the LC group were lower than those in the MC group,but no significant differences were found except for ash content.A total of 17 free amino acids and 23 fatty acids were identified from the muscle of S.paramamosain.The total amino acids(TAA),essen-tial amino acids(EAA)and essential amino acid index(EAAI)had no significant difference between the two groups.The crabs from LC group had a significantly higher HUFA,including EPA,DHA,ARA.The percentage of n-3 PUFA and the ratio of n-3/n-6 PUFA were significantly higher in the LC group.These results indicated that S.paramamosain rearing at low salinity had significantly de-creased SR;however,the low-salinity environment significantly promoted the accumulation of unsaturated fatty acids(UFAs),espe-cially the n-3 PUFA,which might be related to the osmoregulation mechanism.  相似文献

The effect of light intensity ( 1500 Ix and 5000 Ix) on the total lipid and fatty acid composition of six strains of marine diatoms Cylindrotheca fusiformis (B211 ), Phaeodactylum tricornutum (Bl14, Bl18 and B221) Nitzschia closterium (B222) and Chaetoceros gracilis (B13) was investigated.The total lipids of B13, Bl14, and B211 grown at 5000 Ix were lower than those grown at 1500 Ix. No evident changes were observed in Bl18, B221 and B222. Fatty acid composition changed considerably at different light intensity although no consistent correlation between the relative proportion of a single FA and light intensity. The major fatty acids of the 6 strains were 14:0, 16:0, 16:l(n- 7) and 20:5(n-3) . Cylindrothecafusiformis had high percentage of 20:4n - 6 (9.2 - 10.9% ) . The total polyunsaturated fatty acid /rl all 6 strains decreased with increasing light intensity. The percentage of the highly unsaturated fatty acid eicosapentaenoic acid (EPA) decreased with increasing light intensity in all strains except Chaetoceros gracilis.  相似文献

In this study, the proximate composition and the amino and fatty acid profi les of shrimp Litopenaeus vannamei(prey) and wild and cultured cuttlefi sh Sepia pharaonis(the latter fed the prey) were determined and compared with FAO/WHO recommendations. The resulting scores for isoleucine, phenylalanine+tyrosine, histidine, lysine, threonine, and tryptophan in cultured cuttlefi sh were ≥150. The ratio of EAA(essential amino acids)/nonessential amino acids in cultured cuttlefi sh(0.82) was higher than in the wild form(0.80). All EAA amino acid scores for cultured cuttlefi sh were higher than their wild counterparts, except for histidine and tryptophan. Both groups of cuttlefi sh possessed similar saturated fatty acid content, with the cultured containing much more total(Σ) monounsaturated fatty acids, Σ n-6 polyunsaturated fatty acids(PUFA), and eicosapentaenoic acid(C20:5 n-3) but less Σ n-3 PUFA, arachidonic acid(C20:4 n-6), and docosahexaenoic acid(C22:6 n-3) than their wild counterparts. Therefore, the present results suggest that these cultured cuttlefi sh were better than the wild form for human health. Notably, these results also indicate that the nutritional composition of these cuttlefi sh might have been signifi cantly affected by diet.  相似文献

Sediment samples obtained from the South Mid-Atlantic Ridge were studies by gas chromatography-mass spectrometer(GC-MS) for the abundance and distributions of total fatty acids(TFAs). Approximately 34 fatty acids were identified, with the chain-lengths ranging from C12 to C30. The total concentrations of TFAs(∑TFA) ranged from 7.15 to 30.09 μg g-1 dry sediment, and ∑TFA was weakly correlated with bitumen content(R2 = 0.69). The ∑TFA of samples around hydrothermal areas were significantly higher than that of samples away from hydrothermal areas, indicating intense primary production and large biomass in the hydrothermal areas, and suggesting a close relationship between hydrothermal activity and ∑TFA of samples. The characteristics of the TFA composition in the present study are rich in monounsaturated fatty acids and lacking in polyunsaturated fatty acids, and the ratios between the concentrations of monounsaturated fatty acids and ΣTFAs in samples close to the hydrothermal areas, are about 0.8, but for samples far from the hydrothermal areas, they are only about 0.5. Several fatty acids(e.g., a/i C15:0 and C16:1ω7), which are signature biomarkers for sulfur-metabolizing bacteria, show the same distribution trend as ∑TFA of samples, further highlighting the close relationship between fatty acid content and hydrothermal activity and/or hydrothermal communities. The metabolic activities of hydrothermal communities, especially those of microorganisms, are likely the main source of fatty acids in samples.  相似文献

In order to explore the standing stock and preservation of organic carbon, different forms of compound specific lipids were analyzed, based on a 1.8 m sediment core collected in the northern continental shelf of South China Sea in May, 2016. In the core sediments, 21 species of fatty acids, 6 species of alcohols, 8 species of sterols and phytol were detected. Fatty acids were dominated components with the percent of 67%–91% in the total lipids, in which saturated fatty acid C16:0 accounted for about 50%. The contents of lipid species generally decreased from the sediments at surface to those deep, except alcohols waving in a certain range at the same time. Fatty acids primarily existed in free form, followed by base hydrolytic form, and then acid hydrolytic form. Free alcohols were obviously dominated, while base and acid hydrolytic forms contributed small comparably. Phytol gave a significant priority to base hydrolytic form, while acid hydrolytic form could be neglected. Similar to phytol, sterols mostly presented in base hydrolytic form, and then in free form. The compositions of lipids and their forms suggested that organic carbon basically originated from primary production in upper waters in studying area, and were well preserved in the sediments.  相似文献

Genetic modifi cation is useful for improving the nutritional qualities of cyanobacteria. To increase the total unsaturated fatty acid content, along with the ratio of ω-3/ω-6 fatty acids, genetic engineering can be used to modify fatty acid metabolism. S ynechococcus sp. PCC7002, a fast-growing cyanobacterium, does not contain a Δ6 desaturase gene and is therefore unable to synthesize γ-linolenic acid(GLA) and stearidonic acid(SDA), which are important in human health. In this work, we constructed recombinant vectors Syd6 D, Syd15 D and Syd6Dd15 D to express the Δ15 desaturase and Δ6 desaturase genes from Synechocystis PCC6803 in Synechococcus sp. PCC7002, with the aim of expressing polyunsaturated fatty acids. Overexpression of the Δ15 desaturase gene in S ynechococcus resulted in 5.4 times greater accumulation of α-linolenic acid compared with the wild-type while Δ6 desaturase gene expression produced both GLA and SDA. Co-expression of the two genes resulted in low-level accumulation of GLA but much larger amounts of SDA, accounting for as much to 11.64% of the total fatty acid content.  相似文献

This study was conducted to evaluate the effects of dietary lipid sources on the growth performance and fatty acid composition of the swimming crab, P ortunus trituberculatus. Four isonitrogenous and isoenergetic experimental diets were formulated to contain four separate lipid sources, including fish, soybean, rapeseed, and linseed oils(FO, SO, RO, and LO, respectively). With three replicates of 18 crabs each for each diet, crabs(initial body weight, 17.00 ±0.09 g) were fed twice daily for 8 weeks. There were no significant differences among these groups in terms of weight gain, specific growth rate, and hepatosomatic index. However, the RO groups' survival rate was significantly lower than FO groups. The feed conversion and protein efficiency ratios of RO groups were poorer than other groups. The proximate compositions of whole body and hepatopancreas were significantly affected by these dietary treatments. Tissue fatty acid composition mainly reflected dietary fatty acid compositions. Crabs fed FO diets exhibited significantly higher arachidonic, eicosapentaenoic, and docosahexaenoic acid contents in muscle and hepatopancreas compared with VO crabs. Linoleic, oleic, and linolenic acids in muscle and hepatopancreas were the highest in the SO, RO, and LO groups, respectively. The present study suggested that SO and LO could substitute for FO in fishmeal-based diets for swimming crabs, without affecting growth performance and survival.  相似文献

Fish maw(the dried swimbladders of fish) is ranked in the list of the four sea treasures in Chinese cuisine. Fish maw is mainly produced from croaker, which is the most highly priced. However, some of the fish maw being sold as croaker maw are in fact not from croaker, but from the Nile perch Lates niloticus. The present work determined and compared the proximate composition, amino acid and fatty acid composition of croaker Protonibea diacanthus maw and perch L. niloticus maw. The results indicated that both maws were high protein sources and low in fat content. The dominant amino acids in both maws were glycine, proline, glutamic acid, alanine and arginine. These amino acids constituted 66.2% and 66.4% of the total amino acids in P. diacanthus and L. niloticus, respectively. The ratio of FAA: TAA(functional amino acids: total amino acids) in both maws were 0.69. This is a good explanation for why fish maws have been widely utilized as a traditional tonic and remedy in Asia. Except valine and histidine, all the essential amino acid contents in P. diacanthus were higher than in L. niloticus. Moreover, croaker P. diacanthus maw contained more AA and DHA than perch L. niloticus maw, showing a higher ratio of n-3 / n-6, which is more desirable.  相似文献

Phaeodactylum tricornutum is a potential livestock for the combined production of eicosapentaenoic acid(EPA) and fucoxanthin. In this study, six marine diatom strains identified as P. tricornutum were cultured and their total lipid, fatty acid composition and major photosynthetic pigments determined. It was found that the cell dry weight concentration and mean growth rate ranged between 0.24–0.36 g/L and 0.31–0.33/d, respectively. Among the strains, SCSIO771 presented the highest total lipid content, followed by SCSIO828, and the prominent fatty acids in all strains were C16:0, C16:1, C18:1, and C20:5(EPA). Polyunsaturated fatty acids, including C16:2, C18:2, and EPA, comprised a significant proportion of the total fatty acids. EPA was markedly high in all strains, with the highest in SCSIO828 at 25.65% of total fatty acids. Fucoxanthin was the most abundant pigment in all strains, with the highest in SCSIO828 as well, at 5.50 mg/g. The collective results suggested that strain SCSIO828 could be considered a good candidate for the concurrent production of EPA and fucoxanthin.  相似文献

Lipids were extracted from organs of the starfish Asterias amurensis associated with different treatments(raw-control,boiling and heating),and then analyzed for lipid content,lipid oxidation index,lipid classes and fatty acid composition.Results showed that boiling softened the hard starfish shells,thus facilitating the collection of starfish organs.As compared with raw organs,the boiled organs had lower water content and higher lipid content,possibly due to the loss of water-holding capacity caused by protein denaturation.Both boiling and heating increased the peroxide value(PV),thiobarbituric acid(TBA) value and carbon value(CV) of lipids.Despite slight increases in the content of complex lipids,associated lipid composition had no substantial variations upon boiling and heating.For simple lipids,the content of 1,2-diglyceride decreased in boiled and heated organs,with free fatty acids observed on thin layer chromatography(TLC).However,neither boiling nor heating significantly changed the fatty acid compositions of simple or complex lipids in starfish organs,suggesting that these two treatments had no significant effects on complex lipids in starfish organs.Together,our results indicated that boiling of starfish soon after capture facilitated the handling and extraction of useful complex lipids consisting of abundant glucosylceramide and eicosapentaenoic acid(EPA)-bounded phospholipids.  相似文献

The fatty acid compositions of seston and Calanus sinicus were investigated to study trophic relationships in Jiaozhou Bay. Principal component analysis was carried out to ordinate the fatty acid patterns of seston in stations and months. The results showed that diatoms were most abundant in the phytoplankton at station A5 (located in the northwest of the bay:36°9′N, 120°20′E) and least abundant at station D7 (located outside of the bay: 35°59′N, 120°26′E). By contrast, dinoflagellates were most abundant at station D7 and least abundant at station A5. According to the annual variations of 16:1ω7 and 18:4ω3/ 16:1ω7, diatoms flourished mainly in spring and summer, while dinoflagellates bloomed exclusively in summer. A distinctive feature of the fatty acid composition of C. sinicus was the prevalence of 20:5ω3 and 22:6ω3. The higher content of 16:1ω7 over 18:4ω3 in females indicated that diatoms contributed more than dinoflagellates to the diet of C. sinicus. The feeding intensity of C. sinicus on diatoms was higher in spring and autumn than in other seasons. The herbivorous indicators 20:1 and 22:1 were comparatively low, suggesting that besides phytoplankton, C. sinicus might feed on a wider range of particles including organic detritus, bacteria and small copepods.  相似文献

To compare the flesh quality of large yellow croaker cultured with three different modes, enclosure culture(EC), cage culture with feeding trash fish(CCF) and cage culture with feeding formula diet(CCD), thirty six fish individuals of EC(484.6 ± 79.8 g), CCF(432.7 ± 87.9 g) and CCD(416.9 ± 49.5 g) were sampled to measure body color, flesh water holding capacity, flesh proximate composition, amino acids and fatty acids composition, and collagen and inosinic acid contents. The analysis showed that there was no significant difference in the yellowness of skin and the contents of crude protein and inosinic acid between CCD and EC(P 0.05), and the two modes showed higher values than CCF(P 0.05). Most of the amino acids(AA) in flesh were higher in EC than in CCF(P 0.05), but CCD showed no difference in AA composition from the other two modes(P 0.05). EC and CCD had higher contents of collagen, free AA and lower drip loss than CC(P 0.05). In polyunsaturated fatty acids, EC showed significantly higher levels of C20:5 n-3 and C22:6 n-3 than CCF and CCD(P 0.05), but CCD had significantly higher C18:2 n-6 than CCF and EC(P 0.05). In conclusion, the flesh quality of CCD fish is similar to EC fish, and these two modes have better flesh quality than CCF.  相似文献

Monthly difference of the chemical composition of oyster cultured along the eastern coast of Shandong Province was analyzed. The components analyzed included glycogen, fatty acid and free amino acid (FAA). The content of glycogen was high in January and March (2.89 and 2.82 g(100 g) ^-1 on average, respectively) and low in October (2.07g(100g)^- 1 on average). The low content of neutral lipids in October reflected a relatively poor nutritional value of oyster (1.42 g( 100 g)^- 1 on average). The main fatty acids of oyster were palmitic acid (16:0), oleic acid (18: 1), eicosapentaenoic acid (EPA, 20: 5(0 3) and docosahexaenoic acid (DHA, 22:6(0 3). The major FAAs of oyster were Taurine, Glutamicacid, Glycin, Alanine, Arginine and Proline. Taurine was the most abundant FAA with its content ranging from 603 mg(100 g) ^-1 to 1 139 mg( 100 g) ^-1. The high contents of glycogen, polyunsaturated fatty acid and FAA showed that oyster cultured along the eastern coast of Shandong Province was nutritionally good in January and March.  相似文献

The objective of this study was to evaluate the effect of anthropogenic pollution on DNA damage and the fatty acid profile of the bottom dweller fish( Cirrhinus mrigala), collected from the River Chenab, in order to assess the effect of the toxicants on the quality of the fish meat. The levels of Cd, Hg, Cu, Mn, Zn, Pb, Cr and Sn and of phenols from this river were significantly higher than the permissible limits set by the USEPA. Comet assays showed DNA damage in Cirrhinus mrigala collected from three different sampling sites in the polluted area of the river. Significant differences were observed for DNA damage through comet assay in fish collected from polluted compared to control sites. No significant differences were observed for DNA damage between farmed and fish collected from upstream. The micronucleus assay showed similar trends. Fish from the highly polluted sites showed less number of fatty acids and more saturated fatty acids in their meat compared to fish from less polluted areas. Several fatty acids were missing in fish with higher levels of DNA in comet tail and micronucleus induction. Long-chain polyunsaturated fatty acids, eicosapentaenoic acid(20:5n-3) was found missing in the fish from polluted environment while it was found in considerable amount in farmed fish 7.8±0.4%. Docosahexaenoic acid(22:6n-3) also showed significant differences as 0.1±0.0 and 7.0±0.1% respectively, in wild polluted and farmed fishes.  相似文献

Considering the economic importance in traditional Chinese medicine(TCM) and lack of baseline information, we evaluated the proximate composition, fatty acid and amino acid profiles, trace element content and C:N ratio in the alligator pipefish, Syngnathoides biaculeatus. Amongst proximate principals, a crude protein formed the major biochemical component((58.9±2.2)% dry weight). Mean percent concentrations(dry weight) of other components such as a total lipid(TL), ash and nitrogen-free extract measured were,(1.8±0.2)%,(19.2±2.2)% and(20.1±0.45)%, respectively. The fatty acid profile revealed the presence of 27 saturated fatty acids(SFA) with 13 straight-chained and 14 branched-chained, 28 unsaturated fatty acids(UFA) with 14 monounsaturated and 14 polyunsaturated and nine other minor fatty acids. Mean percent contributions of total SFAs and UFAs to TL were found to be(55.41±0.24)% and(44.05±0.25)%, respectively. Altogether, 16 dif ferent amino acids with an equal number of essential(EAA) and non-essential(NAA) ones were identified. Percent contributions by EAA and NAA to the total amino acid content were 38.11% and 61.89%, respectively. Trace metal concentrations in S. biaculeatus were generally low and their distribution followed the order, MgFeZnMnCuCrNiHgCo. The C:N ratio was(4.37±0.04)%. The profile of major biochemical constituents in alligator pipefish, S. biaculeatus revealed its potential use in TCM as well as a nutritional diet for human consumption. The results of the study would also form the basis for formulation and optimization of diets for the culture of S. biaculeatus.  相似文献

This study investigated the effect of dietary lipid on the growth, fatty acid composition and Δ5 fatty acyl desaturase genes(Fads) expression of juvenile abalone(Haliotis discus hannai Ino) hepatopancreas. Six purified diets were formulated to contain tripalmitin(TP), olive oil(OO, 72.87% 18:1n-9), grape seed oil(GO, 68.67% 18:2n-6), linseed oil(LO, 70.48% 18:3n-3), ARA oil(AO, 41.81% ARA) or EPA oil(EO, 44.09% EPA and 23.67% DAH). No significant difference in survival rate was observed among abalone fed with different diets. Weight gain rate(WGR) and daily growth rate of shell length(DGRSL) were significantly increased in abalone fed with diets containing OO, AO and EO, but decreased in abalone fed with LO diet(P 0.05) in comparison with those fed with TP. High level of dietary 18:2n-6 resulted in higher content of n-6 polyunsaturated fatty acids(PUFAs) in abalone fed with GO than those fed with TP, OO, LO and EO(P 0.05). n-3 PUFAs in abalone fed with LO was significantly higher than those in abalone fed with TP, OO, GO and AO(P 0.05). The highest contents of 20:1n-9 and 22:1n-9 were observed in abalone fed with OO. The expression of Δ5 Fads in hepatopancreas of abalone was enhanced by high concentration of 18:3n-3 and suppressed by dietary LC-PUFAs; however it was not affected by dietary high concentration of 18:1n-9 or 18:2n-6. These results provided valuable information for understanding the synthesis of LC-PUFAs and nutritional regulation of Δ5 Fads expression in abalone.  相似文献
