Dinoflagellates are unicellular eukaryotic protists that dominate in all coastal waters, and are also present in oceanic waters. Despite the central importance of dinoflagellates in global primary production, the relationship between dinoflagellates and bacteria are still poorly understood. In order to understand the ecological interaction between bacterial and dinoflageUates communities, denaturing gradient gel electrophoresis (DGGE) and SSU rRNA sequencing were applied to monitoring the population dynamics of bacteria and dinoflagellates from the onset to disappearance of a dinoflagellates bloom occurred in Baltimore Inner Harbor, from April 15 to 24, 2002. Although Prorocentrum minimum was the major bloom forming species under the light microscopy, DGGE method with dinoflagellate specific primers demonstrated that Prorocentrum micans, Gymnodinium galatheanum and Gyrodinium uncatenum were also present during the bloom. Population shifts among the minor dinoflagellate groups were observed. DGGE of PCR-amplified 16S rRNA gene fragments indicated that cyanobacteria, α, β, γ-proteobacteria, FlavobacteriumBacteroides-Cytophaga (FBC), and Planctomcetes were the major components of bacterial assemblages during the bloom. DGGE analysis showed that Cytophagales and α-proteobacteria played important roles at different stages of dinoflagellates bloom. DGGE can be used as a rapid tool to simultaneously monitor population dynamics of both bacterial and dinoflagellates communities in aquatic environments, which is demonstrated here.  相似文献

Distribution of dinoflagellate cysts in Yellow Sea sediments   总被引:1,自引:1,他引:0  
To investigate the distribution,abundance,and species composition of dinoflagellate cysts in the Yellow Sea,surface sediment samples were collected at 37 sites,including the Korean dump site.Twenty-one dinoflagellate cyst taxa were identified,with the assemblages dominated mainly by Spiniferites bulloideus,Operculodinium centrocarpum,and cyst of Alexandrium catenella/tamarense type.A high frequency of O.centrocarpum in the Yellow Sea was observed for the first time,and it is likely that this can be attributed to the dynamics of the Yellow Sea Cold Water Mass and the Changjiang(Yangtze) River runoff.Total cyst concentrations ranged from 23 to 48 442 cysts/g dry weight,and high cyst concentrations were recorded adjacent to the dumping site.This result suggests that anthropogenic activities such as ocean dumping stimulate the growth of dinoflagellates in the Yellow Sea,which in turn leads to high levels of dinoflagellate cyst production.  相似文献

Differential expression of genes is crucial to growth and development of fish. To select the appropriate genes for gene normalization during Cynoglossus semilaevis early developmental process, eight candidate reference genes (ACTB, B2M, EF1A, GADPH, RPL7, TUBA, UBCE and 18S) were tested for their adequacy by using quantitative real-time PCR. The results showed that the expression of all the examined genes exhibited tissue dependent variations in the mature C. semilaevis. EF1A was listed as the most stable reference among the 14 tissues by RefFinder. Furthermore, the recommended comprehensive ranking of the stability determined by RefFinder showed that 18S was the most stable gene during the early developmental stages (from oosphere to 90 days old) in this study. However, when divided the Ct value data of the above mentioned early developmental stages into two separate periods (embryo and post-hatching periods), TUBA and 18S represented the most stable references of these two developmental periods, respectively. Consequently, the reference gene should be carefully and accurately chosen even for studies of the same species at various developmental processes. The relevant data may help in selecting appropriate reference genes for mRNA expression analysis, and is of great value in the studies of fish growth and development.  相似文献

In a grid investigation, dinoflagellate cysts were collected from sediments in Xiamen Western Harbor in May of 2000, from which five species of cysts were identified: Alexandrium tamarensis, A. minutum, Lingulodinium polyedra, Gonyaulax scrippsae and Gymnodinium catenatum, account for about 21% in the species composition. The quanti-tative analysis of the sediments shows that the number of dinoflagellate cysts varies from 51 to 256 cysts/g of sediment, the highest value (>200 cysts/g) being recorded at the stations of the central part of the bay, while the lowest (<100 cysts/g) at the bay mouth. A good linear relationship is found between cyst amount and fine-grained sediments. Complex physiognomies on the seabed, topographty in the bay and weak water exchange are the main factors not only in cyst accumulation but also in their distribution pattern, and have resulted in the difference in cyst densities between the inner bay and the outer bay in the harbor.  相似文献

Most of reported harmful algal blooms(HABs)of microalgae(75%)have been caused by dinoflagellates.Studies on the negative effects of HABs have generally focused on animals,valuable organisms in particular,and environmental factors such as dissolved oxygen and nutrients,but relatively fewer on community level,particularly that using metagenomic approach.In this study,we reported an investigation on the effects of a HAB caused by the dinoflagellate Prorocentrum donghaiense on the species diversity and community structure of the dinoflagellate sub-community via a pyrosequencing approach for the samples taken before,during,and after the bloom season of P.donghaiense in the East China Sea.We sequenced partial 28S rRNA gene of dinoflagellates for the field samples and evaluated the species richness and diversity indices of the dinoflagellate community,as a sub-community of the total phytoplankton.We obtained 800185 valid sequences(categorized into 560 operational taxonomic units,OTUs)of dinoflagellates from 50 samples and found that the biodiversity of dinoflagellate community was significantly reduced during the blooming period in comparison to that in pre-and after-blooming periods,as reflected in the four diversity indices:the species richness expressed as the number of OTUs,Chao1 index,Shannon index(evenness),and Gini-Simpson index.These four indices were all found to be negatively correlated to the cell density of the bloom species P.donghaiense.Correlation analyses also revealed that the P.donghaiense cell abundance was correlated negatively with NO3--N,and NO2--N,but positively with total nitrogen(TN)and total phosphorus(TP).Principal coordinates analysis(PCoA)showed that the community structure of dinoflagellates was markedly different among the different sampling periods,while the redundancy analysis(RDA)revealed P.donghaiense abundance,salinity,NO3--N,and SiO32-were the most four significant factors shaping the dinoflagellate community structure.Our results together demonstrated that HABs caused by the dinoflagellate P.donghaiense could strongly impact the aquatic ecosystem on the sub-community level which the blooming species belongs to.  相似文献

Growth and biochemical responses of the coenobial green alga Scenedesmus obliquus to salinity stress were monitored across different phases of growth.The alga was cultured on BG11 growth medium and subjected to 0,30,100,and 200 mmol/L NaCl for a period of 20 d,during which algal cultures were harvested at 4-d intervals.The salinity-induced inhibition of algal growth was accompanied with prolongation of timing of the different growth phases.The sharp and progressive salinity-induced inhibition of algal growth rate during the early phase of growth points to salt shock but moderation of inhibition at the subsequent stages of growth means algal acclimation to salinity.The concentrations of chlorophylls a and b,soluble sugars,proteins as well as those of K+and Na+in the alga exhibited peaks at the initiation of the exponential phase of growth,with increasing magnitude in proportion to the increase in the level of salinity.Nevertheless,whereas soluble sugars of the alga peaked at initiation of the exponential phase,starch concentration progressively increased with culture age,reaching saturation towards the stationary phase.Whereas the salinity-induced increase in soluble sugars was most evident at the early stages of growth the reverse was true for starch.The present results point to fast acclimation of S.obliquus to salt stress post a brief salt shock,utilizing soluble sugars,K+and Na+for osmotic adjustment.Increasing salinity from 0 to 200 mmol/L NaCl led to progressive increase in soluble sugars,proteins,K+and Na+concentrations of the algal cells,particularly at the early stages of growth.However,the salinity-induced increase in chlorophyll concentration approached a limit at 100 mmol/L NaCl whereas that in starch concentration was more evident at the later stages of growth.  相似文献

The abundance and temporal patterns of viable diatom resting stage cells in sediments of the East China Sea in 2006 have been investigated. The abundance of viable resting stages was enumerated with the most probable number (MPN) technique. Overall, 25 diatom species (including varieties) belonging to 16 genera were detected. Viable resting stage cells were common, on the order of 103 to 106 cells/g dry mass. The abundant taxa included Skeletonema marina, S. dohrnii, Chaetoceros curvisetus and Thalassiosira spp., with Skeletonema accounting for 83.3% of the cell. The effects of temperature (10, 15, 20, 25℃), salinity (20, 25, 30, 35 psu) and light intensity (0, 300, 4 000, 8 000 lux) on the germination of resting stages in sediment samples were also determined. The results showed that light intensity had a significant positive effect on the germination of diatom resting stage cells (p <0.01) and that diatoms could not germinate in darkness. Temperature and salinity had no noticeable effects on the germination. The present study suggests that there are abundant diatom resting cells in sediments of the East China Sea, which could serve as a "seed bank" in the phytoplankton population succession and contribute to the initiation of algal blooms.  相似文献

Spatial distribution patterns of the different life stages of Euphausia superba in the region of the South Shet- land Islands and southern Scotia Sea (Antarctica) were assessed based on scientific survey data collected in lanuary and February of 2010. Adults, eggs, nauplii, metanauplii, calyptopis I-Ill, and furcilia I-II were found in the investigation. The abundance of larvae averaged 1 172.8 ind./mz, with calyptopis I and II as the dominant stages. Habitat occupancy patterns varied among Euphausia superba at different stages, and three sub-regions were identified by cluster analysis. The degree of larval development increased from west to east. Larvae were not observed north of the South Shetland Islands. Calyptopis I was predominant in the water between Elephant Island and the South Orkney Islands, which featured no thermocline. Older stages, including calyptopis II and III and furcilia I and II, were common in north and northeast of the South Orkney Islands, which were characterized by high temperature and high chlorophyll concentration. Distri- bution and abundance of the early life stages of E. superba were associated with specific environmental con- ditions. According to Biota-Environment matching (BIOENV), the distributions of E. superba larvae were correlated with a combination of temperature at the surface and 200 m, and 0-100 m integrated chlorophyll a concentration.  相似文献

To explore the effects of temperature changes on dinoflagellate bloom succession in the coastal waters of the East China Sea, changes in the growth, photophysiology, and Rubisco gene expression of Prorocentrum donghaiense and Karenia mikimotoi, two harmful algal species, were investigated at different temperatures (16 to 28°C). The maximal specific growth rate and the maximal mRNA expression of Rubisco gene in P. donghaiense and K. mikimotoi occurred at 20 and 24°C, respectively. The photosynthetic activity of P. donghaiense was generally stable, but K. mikimotoi photosynthesis increased when temperatures rose from 16 to 28°C. The effective photochemical efficiency (F q /F m ) and the maximal relative electron transfer rate (rETRmax) of K. mikimotoi increased significantly with increasing temperature, and the lowest and highest values occurred at 16 and 28°C, respectively. It seems that P. donghaiense has higher photosynthetic capacity than K. mikimotoi due to its higher F q /F m , rETRmax, and photosynthetic efficiency (α). However, K. mikimotoi has a higher growth rate than P. donghaiense. These results suggest that the photosynthetic activity and genetic responses of dinoflagellates are species-dependent. It is likely that temperature changes affect species composition during blooms, leading to the observed patterns of bloom succession.  相似文献

The feeding of abundant herbivore plankton species has been investigated during different stages of coccolithophorid development in the northeastern Black Sea, namely, at the initial stage of development (March 2009) and at the massive bloom stage (June 2011 and May 2013). The role of coccolithophorids as a food source for Black Sea copepods Calanus euxinus, Acartia clausi, and Pseudocalanus elongatus; the heterotrophic dinoflagellate Noctiluca scintillans; and the larvacean Oikopleura dioica has been characterized for the first time. The contribution of coccolithophorids to copepod ration was low (0.2 to 13% of total carbon intake) during all study periods. The content of these organisms in the diet of N. scintillans ranged from 17 to 100%. The coccolithophorid bloom apparently had a stronger positive effect on the nutrition of fine filter feeders, such as N. scintillans and O. dioica, than on copepod nutrition. Daily coccolithophorid consumption by zooplankton ranged from 0.7 to 39.4% of the biomass in different study periods. The grazing of N. scintillans and O. dioica populations made the greatest contribution to coccolithophorid consumption (up to 26 and 23% of the coccolithophorid biomass, respectively).  相似文献

The food sources of aquacultured Apostichopus japonicus and the trophic levels of organisms in a sea cucumber(A. japonicus) and prawn(Penaeus japonica) polyculture system in a saltwater pond in Zhuanghe, Liaoning Province were examined using carbon and nitrogen stable isotopes. Across organisms, δ13C ranged from(–25.47±0.20)‰ to(–16.48±0.17)‰(mean±SD), and δ15N ranged from(4.23±0.49)‰ to(12.44±0.09)‰. The δ13C and δ15N contents of A. japonicus, P. japonica and Fenneropenaeus chinensis were comparatively higher than those of other organisms. Values of δ13C and δ15N revealed that P. japonica, Hemigrapsus sanguineus and Neomysis japonica comprised the largest component of the diet of A. japonicus. The mean trophic level of the organisms in this saltwater pond polyculture system was(2.75±0.08). P. japonica, A. japonicus, F. chinensis,Synechogobius hasta and Neomysis japonica were in the 3rd trophic level(2–3); jellyfish, H. sanguineus and zooplankton were in the 2nd trophic level(1–2); and Enteromorpha prolifera, benthic microalgae, periphyton and suspended matter primarily consisting of phytoplankton, bacteria and humus were in the primary trophic level(0–1).  相似文献

In the East China Sea(ECS), the succession of causative species responsible for blooms is a recurrent phenomenon during the spring, which changes from diatoms to dinoflagellates. Observations from space and in situ cruises captured this pattern of succession during spring of 2005. In this study, we coupled two biological models, which were developed previously for Skeletonema costatum and Prorocentrum donghaiense,into a circulation model tailored for the ECS. The coupled biophysical model was used to hindcast the blooms and to test the hypothesis proposed in earlier studies that phosphate(PO4 3–) is the first-order decider of the succession. The coupled model successfully reproduced the hydrodynamics(as described in a companion paper by Sun et al.(1), the spatiotemporal distribution of the chlorophyll a(Chl a) concentration, and the species succession reasonably well. By analyzing the effects of different factors on the surface Chl a distribution, we confirmed that the offshore boundaries of the blooms were confined by PO4 3–. In addition, we suggest that surface wind fields may modulate the horizontal distribution of blooms. Thus, during the dispersal of blooms, surface winds coupled with PO4 3– may control the succession of blooms in the ECS. The proposed coupled model provides a benchmark to facilitate future improvements by including more size classes for organisms, multiple nutrient schemes, and additional processes.  相似文献

The species composition, phytoplankton abundance, and relative yield of the variable fluorescence (F v /F m ) were determined in the mesotrophic Nhatrang Bay in October–November of 2004. The species diversity (250 taxonomic units) and heterogeneity of the phytoplankton structure were high. With respect to the number of species and their abundance, diatoms prevailed. In selected parts of the bay, dinoflagellates dominated. The mean biomass in the water column under 1 m2 (B t ) varied from 2.3 to 64.4 mg C/m3 being 31.0 mg C/m3 on average. The values of B t were the lowest at the stations nearest to the river mouth. Seaward, B t increased. The values of B t increased with depth at some stations and decreased at others. In the surface sea layers, the biomass was lower than that in the underlying waters. The values of F v /F m ranged from 0.10 to 0.64 (at a mean value of 0.49). The lowest values of F v /F m were observed in the area close to the seaport. Over the greater part of the bay, the values of F v /F m were higher than 0.47. Such values are indicative of the relatively high potential photosynthetic activity of the phytoplankton. The abundance and species diversity were higher than those in the dry season (March–April).  相似文献

Green macroalgae Chaetomorpha aerea and C. linum are taxonomically confused. In this paper, we tried morphological and molecular analyses to separate these two species. C. aerea and C. linum can be distinguished from morphological characteritics, such as frond dimension, cells size and shape, their mean length/width ratios(LWR), and cell walls constriction. Thalli of C. aerea attenuate basipetally, with diameter 270–500 μm at upper portion, 160–360 μm at middle portion, 100–160 μm at basal portion. For the upper part, the length of cells is less than their diameter. Cell walls usually constrict at the dissepiments, which are pellucid or colorless and give the filament beaded appearance. In contrast, thalli of C. linum often have a constant diameter of 90–300 μm within the same individual, cell walls usually do not constrict and cells are cylindrical or barrel shaped. The LWR is larger than that of C. aerea. Results show that the pairwise distance between two species is 3.6%–3.7% for 18 S r RNA gene and 53.5%–54.3% for ITS region. In phylogeny, they distribute at distant clades, which confirms a genetic divergence at molecular level. In addition, morphological data indicates that filament diameter of C. linum samples is highly variable, ranging from 90 μm to 300 μm. In order to clarify the phylogeny and relationships of the most confused hypotrichous ciliates, Holosticha-complex, four closely related holostichids (five populations), Holosticha bradburyae, H. diademata, Anteholosticha sp., and A. manca, were compared and analyzed using ITS2 secondary structures, ITS1-5.8S-ITS2 region and SSrRNA gene sequences. The ITS1-5.8S-ITS2 region sequences of these four species were first sequenced, and they shared sequence identities ranging from 68.0% to 90.1%, while two populations of Anteholosticha sp. differed in three nucleotides (sequence identity 99.8%). There were several minor differences among ITS2 secondary structures of these species, while two populations of Anteholosticha sp. had the identical secondary structure. Phylogenetic trees inferred from the ITS1-5.8S-ITS2 region sequences of stichotrichs using multiple algorithms (Neighbor-Joining, Maximum Parsimony and Bayesian) revealed similar topologies. The results show that:(1) Holosticha bradburyae and H. diademata firmly clustered together with strong bootstrap supports, forming a sister clade with Anteholosticha sp., (2) Anteholosticha appeared to be a paraphyletic assemblage, in which the morphotype A. manca was more closely related to Diaxonella trimarginata than to its congener Anteholosticha sp. Phylogenetic analyses based on the SSrRNA gene and the combined sequences of SSrRNA gene and ITS1-5.8S-ITS2 region revealed the similar relationships between Holosticha and Anteholosticha, nevertheless their positions within the subclass Stichotrichia differed from each other inferred from different genes.  相似文献

Bacterial abundance, phytoplankton community structure and environmental parameters were investigated to study the relationships between bacteria and phytoplankton during giant jellyfish Nemopilema nomurai blooms in the central Yellow Sea during 2013. N. nomurai appeared in June, increased in August, reached a peak and began to degrade in September 2013. Results showed that phosphate was possible a key nutrient for both phytoplankton and bacteria in June, but it changed to nitrate in August and September. Phytoplankton composition significantly changed that pico-phytoplankton relative biomass significantly increased, whereas other size phytoplankton significantly decreased during jellyfish bloom. In June, a significantly positive correlation was observed between chlorophyll a concentration and bacterial abundance(r=0.67, P0.001, n=34).During jellyfish outbreak in August, there was no significant correlation between phytoplankton and bacteria(r=0.11, P0.05, n=25), but the relationship(r=0.71, P0.001, n=31) was rebuilt with jellyfish degradation in September. In August, small size phytoplankton occupied the mixed layer in offshore stations, while bacteria almost distributed evenly in vertical. Chlorophyll a concentration significantly increased from(0.42±0.056) μg/L in June to(0.74±0.174) μg/L in August, while bacterial abundance just slightly increased. Additionally, the negative net community production indicated that community respiration was not entirely determined by the local primary productivity in August. These results indicated that jellyfish blooms potentially affect coupling of phytoplankton and bacteria in marine ecosystems.  相似文献

Outbreaks of paralytic shellfish poisoning (PSP) and dense blooms caused by Alexandrium species in Jinhae-Masan Bay, Korea have been nearly annual events for many years. However, excluding some Alexandrium species responsible for PSP, there are no critical reports on the morphology of Alexandrium species in this bay. To identify the Alexandrium species based on detailed morphological features, vegetative cells collected water samples and established by the incubation of resting cysts isolated from sediment trap samples were analyzed. Four species of Alexandrium were identified: Alexandrium affine, A. fundyense, A. catenella, and A. insuetum. Morphological features of these species were basically consistent with those outlined in previous studies. However, the ventral pore and the connecting pore on the sulcal plate, which have been accepted as diagnostic characteristics for the identification of A. fundyense and A. catenella, need to be reevaluated, indicating that useful morphological features for identifying these two species should be recommended to avoid confusion in the classification of species in genus Alexandrium.  相似文献

This study investigated alkaline phosphatase (APase) activity and dissolved organic and inorganic phosphorus utilization by the harmful dinoflagellate Gymnodinium impudicum (Fraga et Bravo) Hansen et Moestrup isolated from the South Sea of Korea. Under conditions of limited phosphorus, observation of growth kinetics in batch culture yielded a maximum growth rate (μmax) of 0.41 /day and a half saturation constant (Ks) of 0.71 μM. In time-course experiments, APase was induced as dissolved inorganic phosphorus (DIP) concentrations fell below 0.83 μM, a threshold near the estimated Ks; APase activity increased with further DIP depletion to a maximum of 0.70 pmol/cell/h in the senescent phase. Thus, Ks may be an important index of the threshold DIP concentration for APase induction. G. impudicum utilizes a wide variety of dissolved organic phosphorus compounds in addition to DIP. These results suggest that DIP limitation in the Southern Sea of Korea may have led to the spread of G. impudicum along with the harmful dinoflagellate Cochlodinium polykrikoides in recent years.  相似文献
