以大弹涂鱼为研究对象,应用单细胞凝胶电泳技术并结合外周血基因组DNA琼脂糖凝胶电泳,研究了镉胁迫对大弹涂鱼外周血细胞的遗传损伤。结果表明:(1)镉对大弹涂鱼外周血细胞是遗传毒性而非细胞毒性,并且产生的遗传损伤存在显著(P0.01或P0.05)的剂量效应,染毒1h的大弹涂鱼的全长和尾长与浓度的相关方程分别是y=29.592x-10.576(r2=0.9786),y=28.417x-14.859(r2=0.9857),染毒2h的大弹涂鱼的全长和尾长与浓度的相关方程分别是y=32.044x-5.0235(r2=0.941),y=29.911x-9.539(r2=0.9635)。(2)DNA的损伤程度和污染胁迫之间存在着时间效应。以上结果表明,DNA损伤可作为大弹涂鱼受镉胁迫时的生物指标。  相似文献

苯并(a)芘对大弹涂鱼肝脏超氧化物歧化酶活性的影响   总被引:18,自引:1,他引:18  
在实验生态条件下,研究了在苯并(a)芘(BaP)胁迫下大弹涂鱼肝脏超氧化物歧化酶(SOD)活性的变化,结果显示:暴露3d时,不同BaP含量组大弹涂鱼肝脏SOD活性无显著差异(P>0.05),而暴露7d时,随着B aP含量的升高,0.5mg/dm^3 BaP含量组SOD活性被显著诱导(P<0.05),为对照组的1.87倍;随着暴露时间的延长,各含量组肝脏SOD的活性均表现出不同程度的下降趋势,其中对照组肝脏SOD活性显著降低,表明SOD活性易受污染以外的因素,如水体容量、铒料、光照等的影响。污染解除后,0.5mg/dm^3含量组SOD活性显著升高,表明大弹涂鱼肝脏仍具有较强的生理调节机能,也可能表明SOD活性的变化相对于环境因子改变的“迟滞性”。以上这些结果表明SOD有可能作为大弹涂鱼受BaP胁迫的生物指标。  相似文献

采用急性毒性实验方法,研究了不同浓度氯化镉(0、7.25、14.50、29.00、58.00和116.00mg/L)处理不同时间(1d、3d、5d和7d)对长江华溪蟹心肌细胞凋亡的影响。结果表明,低浓度组及染毒时间较短时镉对心肌的影响不明显,但随着染毒浓度的增加和时间的延长,心肌出现空泡变性、玻璃样变性、炎细胞浸润等病理现象,在染毒7d虽然观察到疑似的凋亡细胞,但凋亡率较低;荧光显微镜未观察到心肌细胞典型的凋亡细胞核形态学变化;琼脂糖凝胶电泳图谱未出现凋亡细胞所特有的梯状条带;流式细胞术检测结果显示镉处理组细胞凋亡率增加,但与对照组相比,无统计学意义(P>0.05);Caspase-3活性也没有显著性变化(P>0.05)。因此,在本实验所选取的浓度和时间范围内,镉未诱导长江华溪蟹心肌细胞发生细胞凋亡,这与本课题组前期研究中观察到的肝胰腺、精巢组织在染毒2d、7d发生细胞凋亡的情况明显不同,这可能与心脏组织的特殊性、不同组织对镉的积累量不同有关。  相似文献

采用静态毒性实验方法,研究重金属镉胁迫条件下大弹涂鱼外周血微核率和肝脏过氧化物酶活性的变化。结果表明,在0.05mg/L Cd2 浓度组,10d时检测到微核率极显著升高;0.5mg/L和5mg/L组5d时就可检测到微核率极显著升高,到10d时微核率达到最高值;10d时转入清洁海水后,3个组在15d再次出现一个微核率的最高值。大弹涂鱼肝脏过氧化物酶标记包括谷胱甘肽过氧化物酶(GSH-Px)、谷胱甘肽转硫酶(GST)。3个组的GSH-Px酶活性和5mg/L浓度组GST酶活性在12h后显示出极显著变化,3个组GST酶活性在24h检测到极显著差异;在转入清洁海水5d时,3个组GSH-Px酶活性和对照组相比可检测到极显著差异,而3个组的GST酶活性均无显著差异。研究结果表明,大弹涂鱼外周血微核标记和肝脏过氧化物酶标记能够灵敏地指示水环境中的镉污染;大弹涂鱼外周血微核标记和肝脏内过氧化物酶标记具有一定的互补作用。  相似文献

应用透射电镜的方法,研究了镉的三种浓度对鲫鱼肾细胞超微结构的影响。结果表明:镉离子染毒4天后,肾细胞超微结构即出现变化,且随着处理浓度的升高,变化渐趋明显,主要表现在细胞核和线粒体。细胞核核膜肿胀、弥散、最后解体。线粒体双层膜解体,形成空白区,嵴断裂、膨大和扭曲。此外,内质网膨胀、空泡化、扭曲打转,溶酶体数量和类型随镉处理浓度的升高而增多。  相似文献

作者提出镉对中华绒螯蟹肝胰脏R—细胞亚显微结构和血清谷丙转氨酶活力影响。在亚显微结构水平上,R—细胞中,受影响变化最明显的是线粒体;线粒体体积和其内膜嵴的破坏程度与介质镉浓度成显著的正相关关系;随镉浓度的升高,细胞器减少,内质网膨胀,细胞质和线粒体基质变稀薄。SGPT活力也随镉浓度升高而显著增大,并与R—细胞亚显微结构变化程度一致。本文还就处于不同镉浓度下的标本,与生理机能相联系的R—细胞形态变化的意义、R-细胞线粒体亚显微结构变化与SGPT活力变化的关系及SGPT活性变化的特点等进行了初步探讨。  相似文献

通过暴露染毒方法研究了栉孔扇贝对镉离子(Cd2+)的35d富集与30d释放规律。结果表明:栉孔扇贝对镉有很强的富集性,体内的镉含量与暴露溶液里的Cd2+浓度呈正相关;栉孔扇贝对镉的释放速率随着暴露水体中镉浓度的增加而减慢。应用双箱动力学模型,通过非线性拟合得到了栉孔扇贝对Cd2+的吸收速率常数k1随着外部水体中镉浓度的增大而减小,生物富集系数BCF则随着外部水体中镉浓度的增大而减少。当镉浓度为0.05mg/L时BCF仅为3 730,而当镉浓度为0.000 5mg/L时BCF达到1.79×1011。平衡状态下栉孔扇贝体内镉含量(CAmax)随着水体中镉浓度增加急剧下降,与水体中镉浓度呈负相关;排出速率常数k2、生物学半衰期B1/2与低浓度镉无明显相关性,随着镉浓度的增大k2显著降低而B1/2显著增大。验证了双箱动力学模型适用于栉孔扇贝对Cd2+的生物富集与释放规律研究。  相似文献

生物体内金属硫蛋白含量变化,可作为环境中重金属污染的生物标志物.通过暴露实验研究了扁额细首纽虫Cephalothrix simula暴露在不同浓度的铜,锌和镉溶液96 h,其体内金属硫蛋白含量的变化规律.结果表明3种重金属离子的胁迫作用均导致金属硫蛋白含量增加,在实验初期(6~24 h),金属硫蛋白含量达到高峰;金属硫蛋白含量与暴露溶液金属浓度具有明显的剂量~效应关系,金属离子浓度越高,金属硫蛋白含量越高;锌离子诱导金属硫蛋白合成的作用大于铜、镉离子.  相似文献

本文研究了浙江温岭池塘养殖的2龄大弹涂鱼的生长特性、各基本生长指标间的关系以及肥满度等.结果表明:大弹涂鱼雌雄生长差异不显著;体长与体高、体宽的相关关系分别为Ls=5.685 7Hb 12.219(r=0.991 1)、Ls=5.397 3D 23.084(r=0.989 5),体长与体重、纯体重的相关关系分别为W=2×10-6Ls3.502 5(r=0.996 6)、Wn=1×10-6Ls3.519(r=0.997 2);大弹涂鱼6-7月生长速度较快,8月生长减慢,9-11月又较快生长,12月至翌年2月生长最慢,翌年3-6月生长又相对加快.大弹涂鱼体长和体重相对增长率的年变化规律与池塘水温年变化一致.大弹涂鱼的肥满度随着鱼的生长而增大.与已报道的野生大弹涂鱼及虾池养殖大弹涂鱼的肥满度进行了比较,提出了提高池塘养殖大弹鱼肥满度的措施.  相似文献

以大黄鱼(Larimichthys crocea)为研究对象,通过研究急性低盐度胁迫对大黄鱼血清生化组分的影响,评估盐度骤降引起鱼体的应激反应。急性低盐度(15、8)胁迫下,在72 h的实验周期中,大黄鱼血清葡萄糖(GLU)含量整体显著下降(P<0.05);血清总蛋白(TP)、白蛋白(ALB)及球蛋白(GLB)含量在15盐度胁迫下呈波动变化,与对照组相比无显著性差异(P>0.05),8盐度胁迫下,TP含量呈显著下降后逐渐升高趋势;血清甘油三酯(TG)和总胆固醇(CHOL)含量在15盐度胁迫下均在初始阶段显著变化后逐渐恢复至对照组水平,而8低盐胁迫后二者含量均在实验后期(48 h和72 h)大幅度显著升高;血清中的丙氨酸氨基转移酶(ALT)和天门冬氨酸氨基转移酶(AST)含量在低盐胁迫下均显著高于对照组水平,且呈现出盐度相关性。研究结果表明,急性低盐胁迫会引起鱼体应激反应,实际生产中应避免养殖环境盐度的剧烈变化。  相似文献

采用半静态染毒实验研究了菲和Cd2+单一及复合污染下胁迫9 d和清水释放2 d过程中毛蚶体内活性氧(ROS)、谷胱甘肽(GSH)含量和谷胱甘肽硫转移酶(GST)活性的变化。结果表明,菲和Cd2+单独胁迫能诱导毛蚶体内ROS含量的增加及GSH含量、GST活性的降低,且随染毒浓度的增加,对各指标影响愈明显,存在剂量-效应关系。毛蚶体内GSH和GST的变化趋势具有一致性。菲和Cd2+复合污染对毛蚶体内的氧化胁迫效应整体大于二者单独污染。在清水释放阶段,单一及复合污染胁迫组各指标均能恢复至对照组水平。表明毛蚶体内抗氧化系统适合作为监测海洋多环芳烃和重金属复合污染的生物标志物。  相似文献

基于取自辽东湾北部海域表层沉积物和岩心样品,利用XRF等方法对沉积物中的重金属含量进行系统的测定,结合137 Cs、210Pb年代测定,分析了研究区表层沉积物中重金属分布特征、环境质量状态以及重金属污染历史。结果表明:辽东湾表层沉积物重金属As、Cd、Cr、Cu、Pb、Zn、Hg含量高值区主要分布在葫芦岛附近海域,并且呈现由近岸向远岸递减的趋势;辽东湾海域部分站位表层沉积物已经受到重金属污染,轻度和重度污染率为34.5%,清洁率为65.5%,重度污染站位集中在葫芦岛附近海域;辽东湾沉积物中重金属污染大致出现在1975年,于1997年达到最大,随后,重金属污染开始明显减少。辽东湾沉积物中重金属分布及演化特征与沿海社会经济发展和国家环境保护政策有着密切的联系。  相似文献

Dogwhelks Nucella lapillus feed mainly on mussels and barnacles, and may experience periods of starvation. We report effects of nutritional state and prey type on the survival, growth, cadmium (Cd) accumulation, metallothionein (MT) induction and glycogen stores in N. lapillus exposed to Cd in water. Adult dogwhelks, with similar shell length (30.0+/-1.5 mm), were either starved or fed to satiation with barnacles Semibalanus balanoides, mussels Mytilus edulis or Cd-dosed M. edulis, and kept in filtered natural seawater (< 0.01 microg Cd 1(-1)) or Cd-contaminated (400 microg Cd 1(-1)) seawater for 80 days. Mortality and individual growth rate were determined. Cd, MT and glycogen were measured in different tissues. Prolonged starvation and exposure to Cd significantly reduced the survivorship of N. lapillus, but feeding could help dogwhelks to combat Cd toxicity and minimise mortality. Extended starvation also caused tissue wastage, leading to higher concentrations of Cd and MT in tissues, whereas fed animals increased in weight and had lower Cd and MT concentrations because of the tissue dilution effect. Prey type significantly affected growth rate of dogwhelks and indirectly influenced Cd accumulation, MT induction and glycogen stores. Eating mussels promoted better growth and higher glycogen reserves than eating barnacles. Individual growth rate decreased with increasing Cd accumulation. Cd-exposed survivors grew faster and consumed more than control animals, implying that these survivors may have better fitness and greater tolerance to Cd toxicity. The use of growth, condition index, MT and glycogen as biomarkers of environmental pollution are discussed. These results indicate a need to incorporate biological data including growth (or at least condition index) and prey type into biomonitoring programmes to allow sound interpretation.  相似文献

The Taiwan Strait region has many miles of coastline, and the Taiwan Straits Tunnel (TST) project faces many potential pollution problems as construction proceeds through sensitive areas. Conventional approaches for pollution mitigation require further examination. The recent development of nanoscale particle technology has shown distinct advantages for contaminant attenuation and ground improvement. This paper is focused on trace metals and is part of the overall effort to develop the nanoscale particle technology. Trace metals in ground and surface waters represent a continued threat to human and ecological health. One of the difficulties in removing toxic concentrations of trace metals from solution is the variable oxidation state and amphoteric nature of multiple constituents. In particular, while cationic metals (e.g., Pb2+, Cd2+, Ni2+) may be rendered less mobile under high pH conditions, anionic metals (e.g., AsO43-, CrO42-, SeO42-)may become more mobile. The objective of this research was to evaluate the sorption of both cationic and anionic trace elements, including arsenic (As), cadmium (Cd), chromium (Cr) and selenium (Se) under batch conditions. Mixtures of a local residual soil were tested alone and in combination with lime and zero valent iron. It was hypothesized that lime would raise the pH and precipitate positively charged metals while zero valent iron would create reducing conditions favorable to the immobilization of negatively charged metals. Results indicate that the use of lime and/or zero valent iron can increase the sorption capacity of soil. Compared to the baseline soil, sorption capacity increased with addition of lime for arsenic and cadmium while it decreased for chromium and selenium. In the case of zero valent iron addition, sorption capacity increased for cadmium, chromium and selenium, while showing no change for arsenic. When both lime and zero valent iron were used, the sorption capacity increased for all metals tested. These results suggest that the combined use of lime and zero valent iron may serve as an alternative treatment technology for removing trace metals from contaminated water systems.  相似文献

The clam Meretrix meretrix was used as a biomonitor to implement an environmental monitoring program along the coast of Beibu Gulf in October 2011. This program not only analyzed biomarkers including acetylcholinesterase, glutathione peroxidase, glutathione S-transferase, catalase and superoxide dismutase activities, total glutathione content and lipid peroxidation level in M. meretrix but also adopted a multi-biomarker approach – integrated biomarker response (IBR) to assess the environmental quality in this ecosystem. In addition, the metal (Hg, As, Cu, Pb, Zn, Cd and Cr) and polychlorinated biphenyls (PCBs) content in the surface sediment at the study area were also measured. The results showed that IBR index was able to distinguish a space trend between sampling sites with different degrees of anthropogenic environmental stress. Integrated contamination degree were displayed in the form of star plots and compared to IBR plots. There was a visual consistency between the pollution level and IBR variation. Based on the results, it was proved that the IBR method coupled with chemical analysis was quite useful for the assessment of environmental pollution in the coastal system.  相似文献

在2010年6月对连云港市对虾养殖区的沉积物环境状况进行这站调查,分析了对虾池养殖区表层沉积物中重金属的含量及分布,并采用海洋沉积物质量国家标准和地积累指数法对该养殖区沉积物中重金属污染进行了评价。结果表明: (1)Cu、Zn、Pb、Cd、Hg、Cr、As的平均浓度分别为22.31 mg/kg、283.9 mg/kg、64.86 mg/kg、0.58 mg/kg、0.0056 mg/kg、89.23 mg/kg、35.58 mg/kg,通过计算单项污染指数表明所有元素的污染指数均在 0~1 之间,属于非污染,表明虾池沉积物属于清洁土壤。(2)利用地积累指数 (Igeo)法对所得到的调查结果进行了评价,发现各样点沉积物呈现出轻洁至中等的污染程度,参与评价的6种重金属的污染程度依次为As> Zn> Pb、Cd> Cr> Cu。进一步的生态风险评价结果表明,所有站位沉积物Cu和Cd(除站位5)的含量皆低于ERL值,说明Cu和Cd很少产生不利的生态效应;全部站位的重金属Zn、Pb和8个站位的Cr的含量均在ERL和ERM之间,表明这三种金属偶尔产生不利的生态效应。  相似文献

The use of biomarkers such as cytochrome P4501A (CYP1A) or metallothionein (MT) for pollution monitoring is based on the assumption that an increased activity or concentration is primarily caused by exposure to contaminants. Previous studies have shown that the response of biomarkers may be affected by factors such as season, temperature, gender, nutritional status or size. The objective of the present study was to identify natural factors that affect the hepatic activity of CYP1A and hepatic concentration of MT in flounder (Platichthys flesus L.). Thirty flounder were sampled at each of two sites in the Hvaler archipelago, southern Norway, monthly through one year. A set of variables were recorded for each sampling and individual including water temperature, salinity, external and internal lesions, intestinal content, sex, size and organ weights. Hepatic CYP1A activity (EROD), MT and metal concentrations were determined for each individual flounder. The influence of environmental and endogenous factors on the response of these two biomarkers was assessed in multiple regression models. For both biomarkers, 50–60% of the total variability could be explained from factors directly related to season, gender and maturation. Season contributed significantly in the model for EROD, as did external lesions. Size, condition and diet did not contribute greatly when the above factors were included. The results confirm previous findings that season, gender and maturation must be taken into account in biomarker monitoring, but also indicate that other factors such as external lesions should be considered.  相似文献

生物标志物是指能够指示或表征暴露于环境化学污染物而引起生物机体发生响应变化的标志物或指标。采用生物标志物指示海洋环境的水质变化及其生态预警作用的研究是当今海洋环境科学研究的热点和前沿,在海洋污染评价领域中应用广泛。本文介绍了生物标志物的定义及分类,综述了国内外基于不同种类的生物标志物的评价方法在海洋溢油污染监测与评价中的应用特点和研究现状,以及多种生物标志物综合应用的典型方法及重要意义,例如生物标志物响应指数(biomarker response index,BRI)和综合生物标志物响应指数法(Integrated biomarker responses index,IBR)。最后,本文总结并指出了今后开展相关溢油污染评价的研究方向,包括针对海洋溢油污染特点筛选特异性强、敏感性高的生物标志物以及建立基于生物标志物的海洋溢油污染预警系统等,从而提供更加精准有效的技术方法和科学保障。  相似文献
