利用近源物种马氏珠母贝的EST-SSR引物对企鹅珍珠贝DNA进行扩增,获得7对可扩增出目标条带的近缘分子标记,分析野生群体(YS)和养殖群体(YZ)企鹅珍珠贝的遗传多样性。结果显示,7个位点共获得26个等位基因;野生和养殖群体的等位基因数分别为3~5和3~4,平均期望杂合度(He)分别为0.623 5、0.496 8;平均观测杂合度(Ho)分别为0.431 8、0.302 8;HNUPM047位点的多态信息含量较低,为0.446 7,其他位点均在0.5以上,说明该遗传标记可提供比较丰富的遗传信息。表明养殖群体遗传多样性水平低于野生群体。  相似文献

【目的】研究JAK3抑制剂对珍珠囊发育及移植免疫反应的影响。【方法】以马氏珠母贝(Pinctada fucata martensii)为材料,植核前用JAK3抑制剂处理细胞小片和珠核,植核后18 d和30 d取珍珠囊,用石蜡切片技术和H.E.染色法(Hematoxylin-eosin Staining)观察JAK3抑制剂对珍珠囊发育的影响,利用荧光定量技术检测植核后不同时间(6、12、24 h和3、6、12、18、30 d)免疫相关基因NF-kappaB(Nuclear factor kappa B)、IkappaK(I-kappaB kinase)和炎性诱导因子IL-17(Interleukin 17)的表达变化。【结果】植核后18 d和30 d,与对照组相比,JAK3抑制剂对珍珠囊的发育没有显著影响(P> 0.05)。荧光定量结果显示,植核后6 h和12 h,JAK3抑制剂可以显著性抑制IL-17的表达,植核后6、3、6、12 d,JAK3抑制剂可以显著性抑制NF-kappaB的表达,植核后3、6 d,JAK3抑制剂可以显著性抑制IkappaK的表达(P <0.05)。【结论】JAK3抑制剂对马氏珠母贝免疫基因表达有明显的抑制作用,可以有效调控珍珠贝植核后的移植免疫,而对珍珠囊发育的影响不明显。  相似文献

3种珍珠贝精子发生及其超微结构的比较研究   总被引:1,自引:0,他引:1  
在透射电子显微镜下观察合浦珠母贝(Pinctadafucata)、大珠母贝(Pinctadamaxima)和企鹅珍珠贝(Pteriapenguin)的精子发生及精子形态.结果表明.大珠母贝从精原细胞到精子,核内都有数量不等的板层小体(nuclearlamellarbody);企鹦珍珠贝从精原细胞的晚期直到精子细胞早期,大多数细胞出现核内包含体(nuclearinclusion);而合浦珠母贝的生精细胞核无特殊结构.合浦珠母贝在初级精母细胞时由高尔基体分泌形成前顶体粒;企鹅珍珠贝的前顶体粒主要在次级精母细胞和早期精子细胞阶段形成。虽然大珠母贝的各级生精细胞中都没有出现典型的高尔基体,但前顶体粒仍在精母细胞时期形成。三种贝的精子结构均为典型的原生型,由头部、中段和尾部构成.合浦珠母贝精子的顶体最长,中段的线粒体数有变异;大珠母贝的精子核最高,其中含有板层小体;而企鹅珍珠贝精子中段的体积是最大的.  相似文献

设定温度范围16~40°C,盐度范围10~50,采用复合设计和响应曲面分析法在实验室条件下研究不同温度、盐度对马氏珠母贝(P.fucata)外套膜Hsp70基因表达量的综合效应并建立模型。采用方差分析(ANOVA)、决定系数等对实验所得回归方程模型进行显著性及拟合度检验。结果表明,温度的一次、二次效应对马氏珠母贝外套膜Hsp70基因表达量影响显著(p值<0.05);盐度的一次效应对马氏珠母贝外套膜Hsp70基因表达量无显著影响(p值>0.05),二次效应对马氏珠母贝外套膜Hsp70基因表达量影响极显著(p值<0.05);温度、盐度之间的互作效应对马氏珠母贝外套膜Hsp70基因表达量无显著影响(p值>0.05)且温度的效应大于盐度的效应。经响应曲面分析法和优化,得到马氏珠母贝外套膜Hsp70基因表达量在适宜的温度、盐度范围内呈峰值变化,在温度26.85°C、盐度29.39时,马氏珠母贝外套膜Hsp70基因表达量达到最小值0.95,满意度达到99.36%。建立了马氏珠母贝外套膜Hsp70基因表达量模型方程,其决定系数98.69%,矫正决定系数97.38%,预测决定系数91.42%,模型的拟合度极高,可用于预测马氏珠母贝外套膜Hsp70基因表达量。  相似文献

以马氏珠母贝糖胺聚糖粗品为主要功能成分,以β-环糊精为辅料,制成珠母贝糖胺聚糖胶囊,并进行检测。结果表明:该胶囊中水分、总糖、糖胺聚糖(GAG)和粗蛋白质量分数分别为8.85%、85.23%、3.00%和3.60%;胶囊的装量差异与崩解时限均符合质量标准;胶囊制剂中细菌总数小于1000 g-1,大肠杆菌MPN小于30 g-1,霉菌和酵母菌总数小于25 g-1,符合《保健食品通用卫生要求》规定;致病菌均未检出。  相似文献

The industrially important organic compound 1,3-propanediol(1,3-PDO) is mainly used as a building block for the production of various polymers. In the present study, response surface methodology protocol was followed to determine and optimize fermentation conditions for the maximum production of 1,3-PDO using marine-derived Klebsiella pneumoniae HSL4. Four nutritional supplements together with three independent culture conditions were optimized as follows: 29.3 g/L glycerol, 8.0 g/L K 2 HPO 4, 7.6 g/L(NH 4) 2 SO 4, 3.0 g/L KH 2 PO 4, pH 7.1, cultivation at 35°C for 12 h. Under the optimal conditions, a maximum 1,3-PDO concentration of 14.5 g/L, a productivity of 1.21 g/(L·h) and a conversion of glycerol of 0.49 g/g were obtained. In comparison with the control conditions, fermentation under the optimized conditions achieved an increase of 38.8% in 1,3-PDO concentration, 39.0% in productivity and 25.7% in glycerol conversion in flask. This enhancement trend was further confirmed when the fermentation was conducted in a 5-L fermentor. The optimized fermentation conditions could be an important basis for developing lowcost, large-scale methods for industrial production of 1,3-PDO in the future.  相似文献

翡翠贻贝形态性状对软体部质量的影响   总被引:2,自引:0,他引:2  
随机选择200个湛江南三岛翡翠贻贝野生个体,测量壳长(SL)、壳宽(SW)、壳高(SH)、韧带长(LL)和软体重(EW)等5个性状,计算性状间的相关系数,采用通径分析方法计算以壳性状和韧带长为自变量对软体重作依变量的通径系数、决定系数。结果表明:壳长、壳宽、壳高、韧带长与软体部质量相关极显著(P<0.01)。壳长对软体部质量的直接影响最大,壳高对软体部质量的直接影响次之;与壳长和壳高相比,壳宽、韧带长对软体部质量的直接影响较小。采用逐步回归分析方法建立了以壳长和壳高为自变量估计软体部质量的多元回归方程:YEW=-39.287+0.466XSL+0.994XSH,为翡翠贻贝选种提供了理论依据和测度指标。  相似文献

The main taste components of triploid and diploid oyster (Crassostrea gigas) were compared. Free amino acids, inosine monophosphate, succinate, trimethylamine oxide and betaine in fresh and boiled extractives were analyzed. The protein, lipids, glycogen, moisture and ash, which may affect the flavour, were evaluated. In boiled extractives, the amino acids were 394.1 mg(100 g)−1 in diploid and 183.5 mg(100 g)−1 in triploid. However, in fresh oyster extractives, they were 320.0 mg(100g)−1 and 147.3 mg(100g)−1 respectively. The inosine monophosphate in triploid was 44% more than that in diploid, and a little difference existed in the content of trimethylamine oxide between them. The contents of these taste components were the basis for taste flavour pattern determination.  相似文献

The shell color of Pacific oyster (Crassostrea gigas) is a desirable trait; but the nutritional studies on C. gigas with different shell colors have not been conducted. Through successive selective breeding, five shell color strains of black (B), purple (P), orange (O), golden (G) and white (W) C. gigas have been developed. The aim of this study was to evaluate the chemical composition and nutritional value of five shell color strains and one commercial population with a common color. The biochemical composition including moisture, total protein, glycogen, ash, total fat, fatty acids (FA), amino acids and minerals was detected. The results indicated that the protein (50.76%–56.57%) was the major component. The content of glycogen showed a significant difference between orange shell and golden shell strains, as well as between commercial population and golden shell strain. In addition, all shell color strains contained a large amount of essential amino acids (12.20–14.15 g (100 g)?1), of them leucine (2.81–3.29 g (100 g)?1) and lysine (2.79–3.28 g (100 g)?1) were predominant. The oysters were rich in polyunsaturated fatty acids (42.26%–45.24% of total fatty acid) with high levels of DHA (18.53%–21.16% of total fatty acid) and EPA (17.23%–18.68% of total fatty acid). Significant differences of mineral contents (Mg, Zn, Fe and Cu) were identified among the six populations. These results indicated that C. gigas with different shell colors presented rich nutritional value with high protein, glycogen, essential amino acids and polyunsaturated fatty acids. The biochemical composition obtained in this study is useful for selective breeding of C. gigas with different shell colors.  相似文献

Oyster peptides were produced from Crassostrea hongkongensis and used as a new protein source for the preparation of an oyster peptide-based enteral nutrition formula (OPENF). Reserpine-induced malabsorption mice and cyclophosphamide-induced immunosuppression mice were used in this study. OPENF powder is light yellow green and has a protein-fat-carbohydrate ratio of 16:9:75 with good solubility in water. A pilot study investigating immune functional impacts of the OPENF on mice show that the OPENF enhanced spleen lymphocyte proliferation and the activity of natural killer (NK) cells in BALB/c mice. Furthermore, OPENF can improve intestinal absorption, increase food utilization ratio, and maintain the normal physiological function of mice. These results suggest that oyster peptides could serve as a new protein source for use in enteral nutrition formula, but more importantly, also indicate that OPENF has an immunostimulating effect in mice.  相似文献

An active peptide against herpes virus was isolated from the enzymic hydrolysate of oyster (Crassostrea gigas) and purified with the definite direction hydrolysis technique in the order of alcalase and bromelin. The hydrolysate was fractioned into four ranges of molecular weight (〉10 kDa, 10-5 kDa, 5-1 kDa and 〈1 kDa) using ultrafiltration membranes and dialysis. The fraction of 10-5 kDa was purified using consecutive chromatographic methods including DEAE Sephadex A-25 column, Sephadex G-25 column, and high performance liquid chromatogram (HPLC) by activity-guided isolation. The antiviral effect of the obtained peptide on herpetic virus was investigated in Vero cells by observing cytopathic effect (CPE). The result shows that the peptide has high inhibitory activity on herpetic virus.  相似文献
