Algal biotechnology has advanced greatly in the past three decades. Many microalgae are now cultivated to produce bioactive substances. Odontella aurita is a marine diatom industrially cultured in outdoor open ponds and used for human nutrition. For the first time, we have systematically investigated the effects of culture conditions in cylindrical glass columns and fiat-plate photobioreactors, including nutrients (nitrogen, phosphorus, silicon, and sulfur), light intensity and light path, on O. aurita cell growth and biochemical composition (protein, carbohydrate, β-1,3-glucan, lipids, and ash). The optimal medium for photoautotrophic cultivation of O. aurita contained 17.65 mmol/L nitrogen, 1.09 mmol/L phosphorus, 0.42 mmol/L silicon, and 24.51 mmol/L sulfur, yielding a maximum biomass production of 6.1-6.8 g/L and 6.7-7.8 g/L under low and high light, respectively. Scale-up experiments were conducted with fiat-plate photobioreactors using different light-paths, indicating that a short light path was more suitable for biomass production of O. aurita. Analyses of biochemical composition showed that protein content decreased while carbohydrate (mainly composed of 15-1,3-glucan) increased remarkably to about 50% of dry weight during the entire culture period. The highest lipid content (19.7% of dry weight) was obtained under 0.11 mmol/L silicon and high light conditions at harvest time. Fatty acid profiles revealed that 80% were Cx4, C~6, and C20, while arachidonic acid and eicosapentaenoic acid (EPA) accounted for 1.6%-5.6% and 9%-20% of total fatty acids, respectively. High biomass production and characteristic biochemical composition profiles make O. aurita a promising microalga for the production ofbioactive components, such as EPA and D-1,3-glucan.  相似文献

Anti-lipopolysaccharide factors (ALFs) are important antimicrobial peptides that are isolated from some aquatic species. In a previous study, we isolated ALF genes from Chinese mitten crab, Eriocheir sinensis. In this study, we optimized the production of a recombinant ALF by expressing E. sinensis ALF genes in Escherichia coli maintained in shake-flasks. In particular, we focused on optimization of both the medium composition and the culture condition. Various medium components were analyzed by the Plackett-Burman design, and two significant screened factors, (NH4)2SO4 and KH2PO4, were further optimized via the central composite design (CCD). Based on the CCD analysis, we investigated the induction start-up time, the isopropylthio-D-galactoside (IPTG) concentration, the post-induction time, and the temperature by response surface methodology. We found that the highest level of ALF fusion protein was achieved in the medium containing 1.89 g/L (NH4)2SO4 and 3.18 g/L KH2PO4, with a cell optical density of 0.8 at 600 nm before induction, an IPTG concentration of 0.5 mmol/L, a post-induction temperature of 32.7°C, and a post-induction time of 4 h. Applying the whole optimization strategy using all optimal factors improved the target protein content from 6.1% (without optimization) to 13.2%. We further applied the optimized medium and conditions in high cell density cultivation, and determined that the soluble target protein constituted 10.5% of the total protein. Our identification of the economic medium composition, optimal culture conditions, and details of the fermentation process should facilitate the potential application of ALF for further research.  相似文献

Vitreoscilla hemoglobin is an oxygen-binding protein that promotes oxygen delivery and reduces oxygen consumption under low oxygen conditions to increase the efficiency of cell respiration and metabolism. In this study, we introduced a Vitreoscilla hemoglobin gene (vgb) into Chlorella vulgaris by Agrobacterium tumefaciens-mediated transformation (ATMT). PCR analysis confirmed that the vgb gene was successfully integrated into the Chlorella vulgaris genome. Analysis of biomass obtained in shake flasks revealed transformant biomass concentrations as high as 3.28 g/L, which was 38.81% higher than that of the wild-type strain. Lutein content of transformants also increased slightly. Further experiments recovered a maximum lutein yield of 2.91 mg/L from the transformants, which was 36.77% higher than that of the wild-type strain. The above results suggest that integrated expression of the vgb gene may improve cell growth and lutein yield in Chlorella vulgaris, with applications to lutein production from Chlorella during fermentation.  相似文献

Endosulfan, an organochlorine pesticide, is highly toxic and effective at controlling pests in agriculture, horticulture, and public health programs. In this study, static bioassays were used to evaluate the toxicity of endosulfan to freshwater prawns( Macrobrachium rosenbergii) of various lengths(1.5±0.03,4±0.08, and 7±0.06 cm). Additionally, the activities of peroxidase(POD), acid phosphatase(ACP),alkaline phosphatase, acetylcholinesterase(AChE), and Na + /K +-ATPase were analyzed to refl ect the effects of endosulfan exposure. The 96 h LC 50 of endosulfan for prawns 1.5, 4, and 7 cm long were 1.86, 4.53,and 6.09μg/L, respectively, improved tolerance to endosulfan with growth. The POD activities of test organisms exposed to low concentrations of endosulfan were inhibited, indicating the presence of oxygen damaged tissue. Moreover, a notable decrease in AChE activity was observed due to overstimulation of neurotransmission, which might result in abnormal behavior. The effect caused by endosulfan on phosphatase production in the hepatopancreas of prawns 1.5, 4, and 7 cm long was different because the ability of nonspecifi c immune regulation increased with growth. The 96 h LC 50 values obtained in this study could be used in the formulation of water-quality criteria in China. Moreover, the changes in enzymes activities of M. rosenbergii under stress of endosulfan could be applied in the establishment of early warning indicators for bio-safety.  相似文献

We adopted the response surface methodology using single factor and orthogonal experiments to optimize four types of antimicrobial agents that could inhibit biofilm formation by Streptococcus mutans, which is commonly found in the human oral cavity and causes tooth decay. The objective was to improve the function of marine Arthrobacter oxydans KQ11 dextranase mouthwash (designed and developed by our laboratory). The experiment was conducted in a three-level, four-variable central composite design to determine the best combination of ZnSO4, lysozyme, citric acid and chitosan. The optimized antibacterial agents were 2.16 g/L ZnSO4, 14 g/L lysozyme, 4.5 g/L citric acid and 5 g/L chitosan. The biofilm formation inhibition reached 84.49%. In addition, microscopic observation of the biofilm was performed using scanning electron microscopy and confocal laser scanning microscopy. The optimized formula was tested in marine dextranase Arthrobacter oxydans KQ11 mouthwash and enhanced the inhibition of S. mutans. This work may be promoted for the design and development of future marine dextranase oral care products.  相似文献

Yeast strain 10 with high yield of protease was isolated from sediments of saltern near Qingdao, China. The protease had the highest activity at pH 9.0 and 45℃. The optimal medium for the maximum alkaline protease production of strain 10 was 2.5 g soluble starch and 2.0 g NaNO3 in 100 mL seawater with initial pH6.0. The optimal cultivation conditions for the maximum protease production were temperature 24.5 ℃, aeration rate 8.0 L min^- 1 and agitation speed 150 r min^-1 . Under the optimal conditions, 623.1 Umg^-1 protein of alkaline protease was reached in the culture within 30 h of fermentation.  相似文献

Small heat shock proteins encompass a widespread but diverse class of proteins,which play key roles in protecting organisms from various stressors.In the present study,the full-length cDNAs of two small heat shock proteins(MgsHSP22 and MgsHSP24.1)were cloned from Mytilus galloprovincialis,which encoded peptides of 181 and 247 amino acids,respectively.Both MgsHSP22 and MgsHSP24.1 were detected in all tissues examined by real-time PCR,with the highest expression being observed in muscle and gonad tissues.The real-time PCR results revealed that Cd signifi cantly inhibited MgsHSP22 expression at 24 h and MgsHSP24.1 at 24 and 48 h under 5 μg/L Cd 2 + exposure.MgsHSP24.1 expression was also signifi cantly inhibited after 50 μg/L Cd 2+ exposure for 48 h.With regard to antioxidant enzymes,increased GPx and CAT activity were detected under Cd 2+ stress(5 and 50 μg/L),while no signifi cant difference in SOD activity was observed throughout the experiment.Overall,both MgsHsps and antioxidant enzymes revealed their potential as Cd stress biomarkers in M.galloprovincialis.  相似文献

Acid hydrolysis is a simple and direct way to hydrolyze polysaccharides in biomass into fermentable sugars. To produce fermentable sugars effectively and economically for fuel ethanol, we have investigated the hydrolysis of Enteromorpha using acids that are typically used to hydrolyze biomass:H2SO4, HCl, H3PO4 and C4H4O4 (maleic acid). 5%(w/w) Enteromorpha biomass was treated for different times (30, 60, and 90 min) and with different acid concentrations (0.6, 1.0, 1.4, 1.8, and 2.2%, w/w) at 121°C. H2SO4 was the most effective acid in this experiment. We then analyzed the hydrolysis process in H2SO4 in detail using high performance liquid chromatography. At a sulfuric acid concentration of 1.8% and treatment time of 60 min, the yield of ethanol fermentable sugars (glucose and xylose) was high, (230.5 mg/g dry biomass, comprising 175.2 mg/g glucose and 55.3 mg/g xylose), with 48.6% of total reducing sugars being ethanol fermentable. Therefore, Enteromorpha could be a good candidate for production of fuel ethanol. In future work, the effects of temperature and biomass concentration on hydrolysis, and also the fermentation of the hydrolysates to ethanol fuel should be focused on.  相似文献

In karst regions,the spatial heterogeneity of soil mineral oxides and environmental variables is still not clear.We investigated the spatial heterogeneity of SiO2,Al2O3,Fe2O3,CaO,MgO,P2O5,K2O,and MnO contents in the soils of slope land,plantation forest,secondary forest,and primary forest,as well as their relationships with environmental variables in a karst region of Southwest China.Geostatistics,principal component analysis(PCA),and canonical correlation analysis(CCA)were applied to analyze the field data.The results show that SiO2was the predominant mineral in the soils(45.02%–67.33%),followed by Al2O3and Fe2O3.Most soil mineral oxide components had a strong spatial dependence,except for CaO,MgO,and P2O5in the plantation forest,MgO and P2O5in the secondary forest,and CaO in the slope land.Dimensionality reduction in PCA was not appropriate due to the strong spatial heterogeneity in the ecosystems.Soil mineral oxide components,the main factors in all ecosystems,had greater influences on vegetation than those of conventional soil properties.There were close relationships between soil mineral oxide components and vegetation,topography,and conventional soil properties.Mineral oxide components affected species diversity,organic matter and nitrogen levels.  相似文献

The effects of different NaCl concentrations, nitrogen sources, carbon sources, and carbon to nitrogen molar ratios on biomass accumulation and polyhydroxybutyrate (PHB) production were studied in batch cultures of the marine photosynthetic bacterium Rhodovulum sulfidophilum P5 under aerobic-dark conditions. The results show that the accumulation of PHB in strain P5 is a growth-associated process. Strain P5 had maximum biomass and PHB accumulation at 2%-3% NaCl, suggesting that the bacterium can maintain growth and potentially produce PHB at natural seawater salinity. In the nitrogen source test, the maximum biomass accumulation (8.10±0.09 g/L) and PHB production (1.11±0.13 g/L and 14.62%±2.25% of the cell dry weight) were observed when peptone and ammonium chloride were used as the sole nitrogen source. NH 4 + -N was better for PHB production than other nitrogen sources. In the carbon source test, the maximum biomass concentration (7.65±0.05 g/L) was obtained with malic acid as the sole carbon source, whereas the maximum yield of PHB (5.03±0.18 g/L and 66.93%±1.69% of the cell dry weight) was obtained with sodium pyruvate as the sole carbon source. In the carbon to nitrogen ratios test, sodium pyruvate and ammonium chloride were selected as the carbon and nitrogen sources, respectively. The best carbon to nitrogen molar ratio for biomass accumulation (8.77±0.58 g/L) and PHB production (6.07±0.25 g/L and 69.25%±2.05% of the cell dry weight) was 25. The results provide valuable data on the production of PHB by R. sulfidophilum P5 and further studies are on-going for best cell growth and PHB yield.  相似文献

We evaluated the dietary protein requirements of juvenile turbot (Scophthalmus maximus L.) and their effects on aquatic quality. Five experimental diets were formulated containing 450, 480, 500, 520, and 540 g/kg. Each diet was randomly assigned to triplicate groups of juvenile turbot (mean initial body weight 34.5 ± 5.5 g) for 88 d. Both the weight gain ratio and feed efficiency increased with increasing dietary protein up to 500 g/kg, but no further improvement was detected when dietary protein levels were >500 g/kg. Protein intake and digestion increased with protein levels, while fecal nitrogen and nitrogen content in seawater increased only when dietary protein exceeded 500 g/kg. Protein digestibility was highest at intermediate dietary protein levels. Chemical oxygen demand, nitrite-nitrogen (NO2--N) and phosphatic-phosphor (PO43--P) levels increased in the rearing water as dietary protein levels increased. The optimum eco-nutrition level of dietary protein for juvenile turbot was 500 g/kg under the current experimental conditions. The diets containing 540 and 500 g/kg protein had similar growth rates and feed conversion ratios, but levels of ammonia (NH4+) and nitrogen were considerably higher in the water and feces, respectively, at the higher level of dietary protein. The difference in the pattern of change between body weight gain and ammonia concentration in water with increasing dietary protein is described by rhomb characteristics.  相似文献

Cyanothece sp. 113 is a unicellular, aerobic, diazotrophic and photosynthetic marine cyanobacterium. The optimal medium for exopolysaccharide yield by the strain was 70.0 g/L of NaCl, and 0.9 g/L of MgSO4 based on the modified F/2 medium for cultivation of marine algae. The optimal cultivation condition for exopolysaccharide yield by this cyanobacterial strain was 29°C, aeration, and continuous illumination at 86.0 μE/M2/S. Under the optimal conditions, over 18.4 g/L of exopolysaccharide was produced within 12 days. This was so far the highest exopolysaccharide yield produced with strains of Cyanothece sp. obtained.  相似文献

Changes in sea surface temperature(SST), seawater oxygen isotope(δ 18 O sw), and local salinity proxy(δ 18 O sw-ss) in the past 155 ka were studied using a sediment core(MD06-3052) from the northern edge of the western Pacifi c Warm Pool(WPWP), within the fl ow path of the bifurcation of the North Equatorial Current. Our records reveal a lead-lag relationship between paired Mg/Ca-SST and δ 18 O during Termination II and the last interglacial period. Similarity in SST between our site and the Antarctic temperature proxy and in CO 2 profi le showed a close connection between the WPWP and the Antarctic. Values of δ 18 O sw exhibited very similar variations to those of mean ocean δ 18 O sw, owing to the past sea-level changes on glacial-interglacial timescale. Calculated values of δ 18 O sw-ss refl ect a more saline condition during high local summer insolation(SI) periods. Such correspondence between δ 18 O sw-ss and local SI in the WPWP may refl ect complex interaction between ENSO and monsoon, which was stimulated by changes in solar irradiance and their infl uence on the local hydrologic cycle. This then caused a striking reorganization of atmospheric circulation over the WPWP.  相似文献

为研究生物酶采油解堵剂中产蛋白酶菌株的初、复筛选及培养条件优化,从大庆原油样品中筛选菌种,通过水解酪素的透明圈实验及福林酚测蛋白酶酶活的方法进行菌株的初、复筛选;以蛋白酶酶活为优化指标,采用单因素实验对筛选的产蛋白酶菌株的培养基及培养条件进行优化,优化最适培养基:可溶性淀粉为15g/L,蛋白胨为20g/L,酵母膏为20g/L,NaCl为1.0g/L,CaCl2为0.02g/L,Na2HPO4为0.2g/L,NaH2PO4为0.1g/L;在初始pH为6.0、接种量为5%(体积分数)、温度为31℃、摇床转速为160r/min的条件下,培养72h后,菌株的蛋白酶酶活为551.0U/mL,为复筛选菌株的蛋白酶酶活的22.92倍,即为菌株生长繁殖及代谢的最佳条件,能够获得更高的蛋白酶酶活,有利于后续实验的进行.结果表明:菌株产蛋白酶对原油作用效果为发酵液表面张力从作用前的56.2mN/m降低到作用后的30.5mN/m,表面张力显著降低,还有降解降黏原油等效果,具有一定的研究价值.  相似文献

Sixteen surface sediment samples were collected and analysed to evaluate the residues of organochlorine pesticides(OCPs) from intertidal flat in Jiangsu Province. Overall, 22 OCPs were detected with total concentrations of OCPs ranging widely from 0.96 to 12.14 ng/g(dry wt). Total hexachlorocyclohexane(HCH) and total dichlorodiphenyltrichloroethane(DDT) levels varied from 0.01 to 0.67 ng/g and from 0.23 to 4.85 ng/g, respectively. DDTs were the predominant compounds. The dominance of β-HCH indicated a history of HCH pollution. According to the ratios of( p, p'-DDD+ p, p'-DDE)/ p, p '-DDT and o, p ′-DDT/ p, p ′-DDT, new input of DDTs did not occur in most sites, and the main sources were historical usage of technical DDTs. OCPs such as dieldrin, endrin, p, p ′-DDD, and p, p ′-DDT exceeded the effects range low, showing adverse biological effects that would occasionally occur at some sites of the study area.  相似文献

Harmful algal blooms (HABs) have led to extensive ecological and environmental issues and huge economic losses. Various HAB control techniques have been developed, and biological methods have been paid more attention. Algicidal bacteria is a general designation for bacteria which inhibit algal growth in a direct or indirect manner, and kill or damage the algal cells. A metabolite which is strongly toxic to the dinoflagellate Alexandrium tamarense was produced by strain DH46 of the alga-lysing bacterium Alteromonas sp. The culture conditions were optimized using a single-factor test method. Factors including carbon source, nitrogen source, temperature, initial pH value, rotational speed and salinity were studied. The results showed that the cultivation of the bacteria at 28°C and 180 r min?1 with initial pH 7 and 30 salt contcentration favored both the cell growth and the lysing effect of strain DH46. The optimal medium composition for strain DH46 was determined by means of uniform design experimentation, and the most important components influencing the cell density were tryptone, yeast extract, soluble starch, NaNO3 and MgSO4. When the following culture medium was used (tryptone 14.0g, yeast extract 1.63g, soluble starch 5.0 g, NaNO3 1.6 g, MgSO4 2.3 g in 1L), the largest bacterial dry weight (7.36 g L?1) was obtained, which was an enhancement of 107% compared to the initial medium; and the algal lysis rate was as high as 98.4% which increased nearly 10% after optimization.  相似文献

Land use changes are known to alter soil organic carbon (SOC) and microbial properties, however, information about how conversion of natural forest to agricultural land use as well as plantations affects SOC and microbial properties in the Changbai Moun- tains of Northeast China is meager. Soil carbon content, microbial biomass carbon (MBC), basal respiration and soil carbon mineraliza- tion were studied in five selected types of land use: natural old-growth broad-leaved Korean pine mixed forest (NF); spruce plantation (SP) established following clear-cutting of NF; cropland (CL); ginseng farmland (GF) previously under NF; and a five-year Mongolian oak young forest (YF) reforested on an abandoned GF, in the Changbai Mountains of Northeast China in 2011. Results showed that SOC content was significantly lower in SP, CL, GF, and YF than in NF. MBC ranged from 304.4 mg/kg in CL to 1350.3 mg/kg in NF, which was significantly higher in the soil of NF than any soil of the other four land use types. The SOC and MBC contents were higher in SP soil than in CL, GF, and YF soils, yielding a significant difference between SP and CL. The value of basal respiration was also higher in NF than in SP, CL, GF, and YF. Simultaneously, higher values of the metabolic quotient were detected in CL, GF, and YF soils, indicat- ing low substrate utilization of the soil microbial community compared with that in NF and SP soil. The values of cumulative mineral- ized carbon and potentially mineralized carbon (Co) in NF were significantly higher than those in CL and GF, while no significant dif- ference was observed between NF and SP. In addition, YF had higher values of Co and C mineralization rate compared with GF. The results indicate that conversion from NF into agricultural land (CL and GF) uses and plantation may lead to a reduction in soil nutrients (SOC and MBC) and substrate utilization efficiency of the microbial community. By contrast, soils below SP were more conducive to the preservation of soil organic matter, which was reflected in the comparison of microbial indicators among CL, GF, and YF land uses. This study can provide data for evaluating soils nutrients under different land use types, and serve as references for the rational land use of natural forest in the study area.  相似文献

Wave fi elds of the South China Sea(SCS) from 1976 to 2005 were simulated using WAVEWATCH III by inputting high-resolution reanalysis wind fi eld datasets assimilated from several meteorological data sources. Comparisons of wave heights between WAVEWATCH III and TOPEX/Poseidon altimeter and buoy data show a good agreement. Our results show seasonal variation of wave direction as follows: 1. During the summer monsoon(April–September), waves from south occur from April through September in the southern SCS region, which prevail taking about 40% of the time; 2. During the winter monsoon(December–March), waves from northeast prevail throughout the SCS for 56% of the period; 3. The dominant wave direction in SCS is NE. The seasonal variation of wave height H s in SCS shows that in spring, H s ≥1 m in the central SCS region and is less than 1 m in other areas. In summer, H s is higher than in spring. During September–November, infl uenced by tropical cyclones, H s is mostly higher than 1 m. East of Hainan Island, H s 2 m. In winter, H s reaches its maximum value infl uenced by the north-east monsoon, and heights over 2 m are found over a large part of SCS. Finally, we calculated the extreme wave parameters in SCS and found that the extreme wind speed and wave height for the 100-year return period for SCS peaked at 45 m/s and 19 m, respectively, SE of Hainan Island and decreased from north to south.  相似文献
