雾化进样石墨炉原子吸收光谱法的研究 Ⅰ.直接雾化进样装置及其性能

介绍一种自行设计和加工的直接雾化石墨炉进样装置的结构、工作原理和操作方法。该装置由微机控制与石墨炉加热程序同步工作,可以实现自动控制和自动测量。对雾化进样石墨炉分析的仪器参数和实验条件进行了研究和选择。结果表明,该装置与石墨炉结合具有自动化程度高、重现性好、样品利用率高和分析速度快等特点。  相似文献

Generation of reactive oxygen species in different fractions of the coelomocytes of holothurian Eupentacta fraudatrix in response to the thermostable toxin of Yersinia pseudotuberculosis in vitro

Pure fraction (92%–95%) of phagocytes (FP) and a mixture of amoebocytes (62%) and morula cells (38%)-FPMC- of the holothurianEupentacta fraudatrix' (Holothuroidea, Dendrochirota) were obtained by using ficoll-verographine step gradient. Basal production of reactive oxygen species (ROS) in FP quantified by using reduction of nitroblue tetrazolium (NBT) was more than twice that in FPMC. Thermostable toxin ofYersinia pseudotuberculosis (TST) at different concentrations (0.2; 0.5; 2.5 μg/ml, but not 0.1 μg/ml) stimulated NBT reduction in FPMC after 24 h incubation. In FP, TST at concentrations of 0.1 and 0.2 μg/ml inhibited and at concentrations of 0.5 and 2.5 μg/ml stimulated NBT reduction after 24 h incubation. Maximal effect was observed in FP and FPMC at TST concentrations of 0.5 and 0.2 μg/ml, respectively. Addition of catalase (0.7 μg/ml) to the cells treated with TST (2.5 μg/ml) was followed by a decrease in NBT reduction compared to that under toxin treatment alone. TST stimulated superoxide dismutase activity in concentration-dependent manner (maximum at 0.5 μg/ml concentration in FP) after 24 h treatment, and this stimulation was prevented by a commercial catalase. Plant lectin concanavalin A stimulated NBT reduction more than 5-fold in FPMC compared to the control. With addition of TST, lectin stimulated ROS to lesser extent than that with lectin alone. When catalase, TST, and lectin were added into the FPMC simultaneously, ROS increase was similar to that under lectin treatment alone. On the whole, data obtained indicated that ROS generation in holothurian coelomocytes especially occurs in both stimulated and not stimulated phagocytes, and that changes in ROS production by these cells may be one of the mechanisms of antibacterial protection of holothurians. Study on Application of Polyenzyme Method to Offal of Harengula zunasi

The new polyenzyme method for making gravy from Harengula zunasi offal involves protein enzymolysis with flavorase after proper alkaline and neutral protease levels were established by orthogonal trials to select the best hydrolytic conditions for processing offal with alkaline and neutral protease. The conditions for the polyenzyme method were pH of 7.0, temperature of 50℃ , alkaline and neutral protease concentration of 1.5% respectively, hydrolysis time of 120 min, andflavorase concentration of 2.0% , for 60 min. The new gravy-making technology yields a nutritious and delicious gravy containing 40.3 % of total essential amino acids, with delicious amino acids Glu, Asp, Gly, Ala, Pro and Ser comprising 49.5% , total and amino nitrogen being respectively1.9 and 1.1 g/100 g (amino acid nitrogen being 61.0% of total nitrogen), The polyenzyme method was used to make 14.8% protein gravy from Harengula zunasi offal. Starting with analysis on the evolving course of oasis and the characteristics and evolution of transitional zone between oasis and desert, in consideration of ecological elements including plant stomata resistance, area covered by vegetation, and physical elements including albedo of vegetation and bare soil, atmosphere temperature, and humidity, under the condition of the balance among net radiation flux, latent heat flux, and sensible heat flux, the following are calculated: temperatures of vegetation and bare soil in different conditions, as well as the evapotranspiration rate of ecosystem. Analysis on evapotranspiration rate indicates that it depends on both the climate of environment and the physiological and ecological conditions of plants. On certain conditions, the evapotranspiration rate of transitional zone between oasis and desert (i.e. area covered by vegetation less than 20%), in some parameter domains, appears in bifurcation or multiequilibrium state. Meanwhile, in such area, ecosystem is extremely unstable. Any minor change to the balance will cause either increase or reduction of area covered by vegetation in ecosystem, on the basis of discussion on the emergency of these phenomena. This paper is attempting to propose an effective way of destruction and rebuilt ecosystem in transitional zone. The way is to control the evaporation of plant through selecting anti-drought country plant with big stomata resistance, and modify the roughness of the underlying surface in ecosystem by establishing rational interspace structure of plant community, so as to put the degenerative ecosystem into the natural succession track. This primary theory is being verified through observation and analysis on historical data.  相似文献   

Modelling the April 15 spectrum of Supernova 1993 J

We present the modeling of the ultraviolet and optical spectra obtained simultaneously on 1993 April 15 with the HST and at Lick Observatory. A Monte Carlo code is employed in the modeling and a comparison is made between models reported by different groups. With an atmosphere similar to the Sun in chemical composition, the observed spectral lines are well reproduced by a power law density structure of index around 20 except the strong H and HeI λ5876 lines which have peculiar absorption profiles. The photospheric velocity is found to be 9500 km/s and the blackbody temperature of the spectrum is 7990 K. For H and HeI λ5876, we suggest a two-component density structure which has a smoother layer located immediately outside the steeply decreasing inner envelope. The power law indices are most probably 20 and 3, respectively, with the transition point at about 13 000 km/s. In addition, this outer smooth layer serves to flatten the far UV spectrum as observed.  相似文献
