Numerous studies have utilized surface complexation theory to model proton adsorption behaviour onto mesophilic bacteria. However, few experiments, to date, have investigated the effects of pH and ionic strength on proton interactions with thermophilic bacteria. In this study, we characterize proton adsorption by the thermophile Anoxybacillus flavithermus by performing acid-base titrations and electrophoretic mobility measurements in NaNO3 (0.001-0.1 M). Equilibrium thermodynamics (Donnan model) were applied to describe the specific chemical reactions that occur at the water-bacteria interface. Acid-base titrations were used to determine deprotonation constants and site concentrations for the important cell wall functional groups, while electrophoretic mobility data were used to further constrain the model. We observe that with increasing pH and ionic strength, the buffering capacity increases and the electrophoretic mobility decreases. We develop a single surface complexation model to describe proton interactions with the cells, both as a function of pH and ionic strength. Based on the model, the acid-base properties of the cell wall of A. flavithermus can best be characterized by invoking three distinct types of cell wall functional groups, with pKa values of 4.94, 6.85, and 7.85, and site concentrations of 5.33, 1.79, and 1.42 × 10−4 moles per gram of dry bacteria, respectively. A. flavithermus imparts less buffering capacity than pure mesophilic bacteria studied to date because the thermophile possesses a lower total site density (8.54 × 10−4 moles per dry gram bacteria).  相似文献

Several recent studies have applied surface complexation theory to model metal adsorption behaviour onto mesophilic bacteria. However, no investigations have used this approach to characterise metal adsorption by thermophilic bacteria. In this study, we perform batch adsorption experiments to quantify cadmium adsorption onto the thermophile Anoxybacillus flavithermus. Surface complexation models (incorporating the Donnan electrostatic model) are developed to determine stability constants corresponding to specific adsorption reactions. Adsorption reactions and stoichiometries are constrained using spectroscopic techniques (XANES, EXAFS, and ATR-FTIR). The results indicate that the Cd adsorption behaviour of A. flavithermus is similar to that of other mesophilic bacteria. At high bacteria-to-Cd ratios, Cd adsorption occurs by formation of a 1:1 complex with deprotonated cell wall carboxyl functional groups. At lower bacteria-to-Cd ratios, a second adsorption mechanism occurs at pH > 7, which may correspond to the formation of a Cd-phosphoryl, CdOH-carboxyl, or CdOH-phosphoryl surface complex. X-ray absorption spectroscopic investigations confirm the formation of the 1:1 Cd-carboxyl surface complex, but due to the bacteria-to-Cd ratio used in these experiments, other complexation mechanism(s) could not be unequivocally resolved by the spectroscopic data.  相似文献

Declining sea ice area in the Canadian Arctic has gained significant attention with respect to the prospect of increased shipping activities. To investigate relationships between recent declines in sea ice area with Arctic maritime activity, trend and correlation analysis was performed on sea ice area data for total, first-year ice (FYI), and multi-year ice (MYI), and on a comprehensive shipping dataset of observed vessel transits through the Vessel Traffic Reporting Arctic Canada Traffic Zone (NORDREG zone) from 1990 to 2012. Links to surface air temperature (SAT) and the satellite derived melt season length were also investigated. Between 1990 and 2012, statistically significant increases in vessel traffic were observed within the NORDREG zone on monthly and annual time-scales coincident with declines in sea ice area (FYI, MYI, and total ice) during the shipping season and on a monthly basis. Similarly, the NORDREG zone is experiencing increased shoulder season shipping activity, alongside an increasing melt season length and warming surface air temperatures (SAT). Despite these trends, only weak correlations between the variables were identified, although a step increase in shipping activity is apparent following the former summer sea ice extent minimum in 2007. Other non-environmental factors have also likely contributed to the observed increase in Arctic shipping activity within the Canadian Arctic, such as tourism demand, community re-supply needs, and resource exploration trends.  相似文献

Patterns of social interaction influence how knowledge is generated, communicated, and applied. Theories of social capital and organizational learning suggest that interactions within disciplinary or functional groups foster communication of knowledge, whereas interactions across groups foster generation of new knowledge. We used social network analysis to examine patterns of social interaction reported in survey data from scientists and managers who work on fish and fire issues. We found that few fish and fire scientists and managers interact with one another, suggesting low bridging social capital and thus, limited opportunity for generation of new knowledge. We also found that although interaction occurs among scientists—suggesting modest bonding social capital—few managers interact with other managers, indicating limited opportunity for communication of scientific knowledge for the purposes of application. We discuss constraints and opportunities for organizational learning evident in these patterns of social interaction among fish and fire scientists and managers.  相似文献

We used naturally occurring radium isotopes as tracers of water exchange in Apalachicola Bay, a shallow coastal-plain estuary in northwestern Florida. The bay receives fresh water and radium from the Apalachicola River, and mixes with Gulf of Mexico waters through four inlets. We deployed moored buoys with attached Mn-fibers at several stations throughout the estuary during two summer and two winter periods. After deployment for at least one tidal cycle we measured the ratio of the two short-lived radium isotopes 223Ra (half-life = 11 d) and 224Ra (3.6 d) to estimate “radium ages” of the water in the bay.During our four seasonal deployments the river discharge ranged from 338 to 1016 m3 s 1. According to our calculations the water turnover time in the bay during these samplings ranged from 6 to 12 days. Age contours in the bay showed that winds and tides as well as river discharge influence the water movement and the residence time of freshwater in the bay. We also calculated the mean age of river water in the bay which was between 5 to 9 days during the studied periods. We suggest that this approach can be used to quantify transport processes of dissolved substances in the bay. For example, soluble nutrient or pollutant transport rates from a point source could be examined. We conclude that the radium age technique is well suited for flushing rate calculations in river dominated shallow estuaries.  相似文献

Stratovolcanoes and lava domes are particularly susceptible to sector collapse resulting from wholesale rock failure as a consequence of decreasing rock strength. Here, we provide insights into the influence of thermal and cyclic stressing on the strength and mechanical properties of volcanic rocks. Specifically, this laboratory study examines the properties of samples from Mount St. Helens; chosen because its strength and stability have played a key role in its history, influencing the character of the infamous 1980 eruption. We find that thermal stressing exerts different effects on the strengths of different volcanic units; increasing the heterogeneity of rocks in situ. Increasing the uniaxial compressive stress generates cracking, the timing and magnitude of which was monitored via acoustic emission (AE) output during our experiments. AEs accelerated in the approach to failure, sometimes following the pattern predicted by the failure forecast method (Kilburn 2003). Crack damage during the experiments was tracked using the evolving static Young’s modulus and Poisson’s ratio, which represent the quasi-static deformation in volcanic edifices more accurately than dynamic elastic moduli which are usually implemented in volcanic models. Cyclic loading of these rocks resulted in a lower failure strength, confirming that volcanic rocks may be weakened by repeated inflation and deflation of the volcanic edifice. Additionally, volcanic rocks in this study undergo significant elastic hysteresis; in some instances, a material may fail at a stress lower than the peak stress which has previously been endured. Thus, a volcanic dome repeatedly inflated and deflated may progressively weaken, possibly inducing failure without necessarily exceeding earlier conditions.  相似文献

Activity ratios (AR) of radium isotopes have been used with success to constrain estimates of water ages and to approximate residence times in coastal waters. We compared two common radium sampling methods (grab sampling and stationary moorings) to estimate water ages and the residence time of St. Andrew Bay waters in northwest Florida, USA. Both sampling methods utilize manganese dioxide fibers (“Mn fibers”) to adsorb dissolved radium from the water column. Grab samples capture radium activities at a discrete time while moorings integrate radium activities over longer deployments. The two methods yielded similar results in this study and thus both approaches are useful for water age comparisons and residence time approximations. However, since radium often varies as a function of tidal stage, deploying moorings over a complete tidal cycle is the preferred approach. An estimated residence time for North Bay and West Bay of 8–11 days was approximated using ARs for both ex224Ra/223Ra and ex224Ra/228Ra. Some complications were introduced as St. Andrew Bay is a tidally dominated, rather than a river-dominated bay system where this method has previously been applied. The largest freshwater source to this bay system is from a man-made reservoir, with an average freshwater flow of only 20 m3 s?1. The activity concentrations and ARs measured by both sampling methods suggest that while the reservoir is the prominent radium source, it is not the only radium source. Nonetheless, a tidal mixing model applied to the western half of the system yielded an approximate flushing time of 10–12 days, similar to that derived from our radium-based water age approach.  相似文献

Ti valence measurements in MgAl2O4 spinel from calcium‐aluminum‐rich inclusions (CAIs) by X‐ray absorption near‐edge structure (XANES) spectroscopy show that many spinels have predominantly tetravalent Ti, regardless of host phases. The average spinel in Allende type B1 inclusion TS34 has 87% Ti+4. Most spinels in fluffy type A (FTA) inclusions also have high Ti valence. In contrast, the rims of some spinels in TS34 and spinel grain cores in two Vigarano type B inclusions have larger amounts of trivalent titanium. Spinels from TS34 have approximately equal amounts of divalent and trivalent vanadium. Based on experiments conducted on CAI‐like compositions over a range of redox conditions, both clinopyroxene and spinel should be Ti+3‐rich if they equilibrated with CAI liquids under near‐solar oxygen fugacities. In igneous inclusions, the seeming paradox of high‐valence spinels coexisting with low‐valence clinopyroxene can be explained either by transient oxidizing conditions accompanying low‐pressure evaporation or by equilibration of spinel with relict Ti+4‐rich phases (e.g., perovskite) prior to or during melting. Ion probe analyses of large spinel grains in TS34 show that they are enriched in heavy Mg, with an average Δ25Mg of 4.25 ± 0.028‰, consistent with formation of the spinel from an evaporating liquid. Δ25Mg shows small, but significant, variation, both within individual spinels and between spinel and adjacent melilite hosts. The Δ25Mg data are most simply explained by the low‐pressure evaporation model, but this model has difficulty explaining the high Ti+4 concentrations in spinel.  相似文献

Much work in recent years has reported on the role of submarine groundwater discharge (SGD) on coastal biogeochemistry, but most of those studies have focused on temperate or tropical climates where year-round rainfall recharges surficial aquifers. The aim of this work, however, was to examine SGD behaviors in an arid setting??Marina Lagoon on the Egyptian Mediterranean coast. SGD was estimated via radon surveys and time-series measurements in lagoon waters during two campaigns (wet season in March 2009 and dry season in July 2010). Relatively higher values of radon were detected in March (maximum >30?dpm/L) compared to July (up to 16?dpm/L), which would indicate either enhanced input rates, or lower mixing/atmospheric losses during the wet season. Lower salinity waters within Marina Lagoon were characterized by higher radon and higher concentrations of dissolved inorganic nitrogen (DIN) and silica (DISi), characteristic of groundwater inputs. Based on lagoon and groundwater radon measurements and an advection-diffusion model, SGD average rates between 0.83 to 2.4?×?108?L/day were estimated for both surveys. Since no seasonal pattern was detected, we suspect that either (1) SGD is derived from a regional aquifer not influenced by recharge from local rainfall or (2) local water use for irrigation and domestic purposes artificially recharges the surficial aquifer in the dry summer (tourist) months, which compensates for the lack of rainfall occurring at that time.  相似文献

A novel manganese dioxide (MnO2) resin is suitable for the determination and decontamination of lead isotopes (208Pb, 207Pb, 206Pb) in aqueous solutions at trace concentrations. This is desirable due to the toxic nature and high abundance of lead in the environment. Current techniques are both time consuming, expensive and not suitable for low-level lead decontamination. The MnO2 resin has been demonstrated to pre-concentrate with extraction efficiency above 90% for a range of pH values, and with a mean extraction of 92.5% from fresh waters at a flow rate of 100?ml?min?1. The lead distribution coefficient is 1.3?×?104 (pH 7) with tolerance to calcium and sodium. Adsorption isotherms have been investigated and the resin shown to follow the Langmuir adsorption isotherm with a saturation point of 41.5?mg per g of MnO2 resin.  相似文献
