Generation of reactive oxygen species in different fractions of the coelomocytes of holothurian Eupentacta fraudatrix in response to the thermostable toxin of Yersinia pseudotuberculosis in vitro

Pure fraction (92%–95%) of phagocytes (FP) and a mixture of amoebocytes (62%) and morula cells (38%)-FPMC- of the holothurianEupentacta fraudatrix' (Holothuroidea, Dendrochirota) were obtained by using ficoll-verographine step gradient. Basal production of reactive oxygen species (ROS) in FP quantified by using reduction of nitroblue tetrazolium (NBT) was more than twice that in FPMC. Thermostable toxin ofYersinia pseudotuberculosis (TST) at different concentrations (0.2; 0.5; 2.5 μg/ml, but not 0.1 μg/ml) stimulated NBT reduction in FPMC after 24 h incubation. In FP, TST at concentrations of 0.1 and 0.2 μg/ml inhibited and at concentrations of 0.5 and 2.5 μg/ml stimulated NBT reduction after 24 h incubation. Maximal effect was observed in FP and FPMC at TST concentrations of 0.5 and 0.2 μg/ml, respectively. Addition of catalase (0.7 μg/ml) to the cells treated with TST (2.5 μg/ml) was followed by a decrease in NBT reduction compared to that under toxin treatment alone. TST stimulated superoxide dismutase activity in concentration-dependent manner (maximum at 0.5 μg/ml concentration in FP) after 24 h treatment, and this stimulation was prevented by a commercial catalase. Plant lectin concanavalin A stimulated NBT reduction more than 5-fold in FPMC compared to the control. With addition of TST, lectin stimulated ROS to lesser extent than that with lectin alone. When catalase, TST, and lectin were added into the FPMC simultaneously, ROS increase was similar to that under lectin treatment alone. On the whole, data obtained indicated that ROS generation in holothurian coelomocytes especially occurs in both stimulated and not stimulated phagocytes, and that changes in ROS production by these cells may be one of the mechanisms of antibacterial protection of holothurians. Carbon isotope fractionation between graphite and diamond during shock experiments

Abstract— Carbon isotopic compositions were measured for shock‐produced diamond and shocked graphite formed at peak pressures ranging from 37 to 52 GPa. The δ¹³C values of diamonds produced in a sealed container were generally lower than that of the initial graphite. The differences in the carbon isotopic composition between initial graphite and shocked graphite/diamond may reflect kinetic isotopic fractionation during the oxidation of the graphite/diamond and/or analytical artifacts possibly induced by impurities in the samples. The pressure effect on the isotopic fractionations between graphite and diamond can be estimated from the δ¹³C values of impurity‐free diamonds produced using a vented container from which gases, including oxygen, in pore spaces escaped during or after the diamond formation (e.g., 0.039 ± 0.085‰ at a peak pressure of 52 GPa). Any isotopic fractionation induced by shock conversion of graphite to diamond is too small to be detected in natural shock‐induced diamond‐graphite systems related to terrestrial impact cratering processes.  相似文献

Study on Application of Polyenzyme Method to Offal of Harengula zunasi

The new polyenzyme method for making gravy from Harengula zunasi offal involves protein enzymolysis with flavorase after proper alkaline and neutral protease levels were established by orthogonal trials to select the best hydrolytic conditions for processing offal with alkaline and neutral protease. The conditions for the polyenzyme method were pH of 7.0, temperature of 50℃ , alkaline and neutral protease concentration of 1.5% respectively, hydrolysis time of 120 min, andflavorase concentration of 2.0% , for 60 min. The new gravy-making technology yields a nutritious and delicious gravy containing 40.3 % of total essential amino acids, with delicious amino acids Glu, Asp, Gly, Ala, Pro and Ser comprising 49.5% , total and amino nitrogen being respectively1.9 and 1.1 g/100 g (amino acid nitrogen being 61.0% of total nitrogen), The polyenzyme method was used to make 14.8% protein gravy from Harengula zunasi offal. 讨论运用PCI 9054(美国PLX公司生产的接口芯片)作为接口芯片的PCI(Peripheral Component Interconnect)板卡的软硬件设计,以实现天线跟踪的两个时间同步中断。利用标准秒信号中断作为系统时钟同步信号,并同步产生时间间隔为20ms(或40、50ms,可选)的中断信号,来处理天线跟踪指令输出。中断信号通过PCI中断口INTA#接入计算机,在驱动中识别不同的中断信号,并在应用程序响应中断处理后,命令ACU(Antenna Control Unit)机,实现射电天文望远镜的同步跟踪。其控制过程分3部分阐述:硬件设计、驱动程序设计、安装及应用;着重讨论了前两者的设计方法及思路。  相似文献   

Modelling the April 15 spectrum of Supernova 1993 J

We present the modeling of the ultraviolet and optical spectra obtained simultaneously on 1993 April 15 with the HST and at Lick Observatory. A Monte Carlo code is employed in the modeling and a comparison is made between models reported by different groups. With an atmosphere similar to the Sun in chemical composition, the observed spectral lines are well reproduced by a power law density structure of index around 20 except the strong H and HeI λ5876 lines which have peculiar absorption profiles. The photospheric velocity is found to be 9500 km/s and the blackbody temperature of the spectrum is 7990 K. For H and HeI λ5876, we suggest a two-component density structure which has a smoother layer located immediately outside the steeply decreasing inner envelope. The power law indices are most probably 20 and 3, respectively, with the transition point at about 13 000 km/s. In addition, this outer smooth layer serves to flatten the far UV spectrum as observed.  相似文献   

Isolation and pharmacological activities of bromophenols from Rhodomela confervoides

Four bromophenols were isolated from the extract of marine red alga,Rhodomela confervoides by column chromatography and HPLC methods. By means of spectroscopic methods inclding IR, MS, 1D, and 2D-NMR techniques, their structures were elucidated as (1) 3-bromo-4,5-dihydroxy benzoic acid methyl ester; (2) bis (2,3-dibromo-4,5-dihydroxybenzyl) ether; (3) 3,4-dibromo-5-(methoxymethyl)-1,2-benzenediol and (4) 3-bromo-4,5-dihydroxy-benzaldehyde. Compound 1 was first isolated from the algae in nature, and 1, 4 were found to have selective cytotoxic activities against KB, Bel 7402 and A549 cells, 2 showed powerful antibacterial activities againstStaphylococcus aurens andPseudomonas aeruginosa.
