Bulk Cd adsorption isotherm experiments, thermodynamic equilibrium modeling, and Cd K edge EXAFS were used to constrain the mechanisms of proton and Cd adsorption to bacterial cells of the commonly occurring Gram-positive and Gram-negative bacteria, Bacillus subtilis and Shewanella oneidensis, respectively. Potentiometric titrations were used to characterize the functional group reactivity of the S. oneidensis cells, and we model the titration data using the same type of non-electrostatic surface complexation approach as was applied to titrations of B. subtilis suspensions by Fein et al. (2005). Similar to the results for B. subtilis, the S. oneidensis cells exhibit buffering behavior from approximately pH 3-9 that requires the presence of four distinct sites, with pKa values of 3.3 ± 0.2, 4.8 ± 0.2, 6.7 ± 0.4, and 9.4 ± 0.5, and site concentrations of 8.9(±2.6) × 10−5, 1.3(±0.2) × 10−4, 5.9(±3.3) × 10−5, and 1.1(±0.6) × 10−4 moles/g bacteria (wet mass), respectively. The bulk Cd isotherm adsorption data for both species, conducted at pH 5.9 as a function of Cd concentration at a fixed biomass concentration, were best modeled by reactions with a Cd:site stoichiometry of 1:1. EXAFS data were collected for both bacterial species as a function of Cd concentration at pH 5.9 and 10 g/L bacteria. The EXAFS results show that the same types of binding sites are responsible for Cd sorption to both bacterial species at all Cd loadings tested (1-200 ppm). Carboxyl sites are responsible for the binding at intermediate Cd loadings. Phosphoryl ligands are more important than carboxyl ligands for Cd binding at high Cd loadings. For the lowest Cd loadings studied here, a sulfhydryl site was found to dominate the bound Cd budgets for both species, in addition to the carboxyl and phosphoryl sites that dominate the higher loadings. The EXAFS results suggest that both Gram-positive and Gram-negative bacterial cell walls have a low concentration of very high-affinity sulfhydryl sites which become masked by the more abundant carboxyl and phosphoryl sites at higher metal:bacteria ratios. This study demonstrates that metal loading plays a vital role in determining the important metal-binding reactions that occur on bacterial cell walls, and that high affinity, low-density sites can be revealed by spectroscopy of biomass samples. Such sites may control the fate and transport of metals in realistic geologic settings, where metal concentrations are low.  相似文献

The variation in the precipitation phase in polar regions represents an important indicator of climate change and variability.We studied the precipitation phase at the Great Wall Station and Antarctic Peninsula(AP)region,based on daily precipitation,synoptic records and ERA-Interim data during the austral summers of 1985?2014.Overall,there was no trend in the total precipitation amount or days,but the phase of summer precipitation(rainfall days versus snowfall days)showed opposite trends before and after 2001 at the AP.The total summer rain days/snow days increased/decreased during 1985?2001 and significantly decreased at a rate of?14.13 d(10 yr)?1/increased at a rate of 14.31 d(10 yr)?1 during 2001?2014,agreeing well with corresponding variations in the surface air temperature.Further,we found that the longitudinal location of the Amundsen Sea low(ASL)should account for the change in the precipitation phase since 2001,as it has shown a westward drift after 2001[?41.1°(10 yr)?1],leading to stronger cold southerly winds,colder water vapor flux,and more snow over the AP region during summertime.This study points out a supplementary factor for the climate variation on the AP.  相似文献

To achieve a high-quality simulation of the surface wind field in the Chukchi/Beaufort Sea region, quick scatterometer (QuikSCAT) ocean surface winds were assimilated into the mesoscale Weather Research and Forecasting model by using its three-dimensional variational data assimilation system. The SeaWinds instrument on board the polar-orbiting QuikSCAT satellite is a specialized radar that measures ice-free ocean surface wind speed and direction at a horizontal resolution of 12.5 km. A total of eight assimilation case studies over two five-day periods, 1–5 October 2002 and 20–24 September 2004, were performed. The simulation results with and without the assimilation of QuikSCAT winds were then compared with QuikSCAT data available during the subsequent free-forecast period, coastal station observations, and North American Regional Reanalysis data. It was found that QuikSCAT winds are a potentially valuable resource for improving the simulation of ocean near-surface winds in the Chukchi/Beaufort Seas region. Specifically, the assimilation of QuikSCAT winds improved, (1) offshore surface winds as compared to unassimilated QuikSCAT winds, (2) sea-level pressure, planetary boundary-layer height, as well as surface heat fluxes, and (3) low-level wind fields and geopotential height. Verification against QuikSCAT data also demonstrated the temporal consistency and good quality of QuikSCAT observations.  相似文献

Microfossils in isolation basin sediments are frequently used to reconstruct sea‐level change, but preservation problems and non‐analogue situations can limit their usefulness. Here we investigate the potential of stable carbon isotopes (δ13C) and C/N ratios from bulk organic matter, as an alternative proxy of salinity within isolation basin sediments from a basin in northwest Scotland. Within the Holocene sediment δ13C and C/N are determined largely by the mean weighted values of the predominant source of the organic material. Analysis of modern materials and comparison with the diatom record shows that the marine parts of the sequence are dominated by high δ13C and variable C/N. In the fresh water sequences the organic material is a mixture of both freshwater aquatic and terrestrial plant input that have relatively low δ13C and high C/N. The application of δ13C and C/N ratios in the studied basin in general follow the environmental change recorded by the diatoms and shows the potential of bulk organic matter in the investigation of salinity change in isolation basins. We study the evolution of a rigidly rotating protoneutron star (PNS) with hyperons and nucleons or solely nucleons in its core due to the escape of trapped neutrinos. As the neutrinos escape, the core nucleonic neutron star (NS) expands and the stellar rotation slows. After the neutrinos escape, the range of the spin periods is narrower than the initial one, but the distribution is still nearly uniform. A PNS with hyperons, at the late stage of its evolution, keeps shrinking and spinning up until all the trapped neutrinos escape. Consequently, the distribution of the stellar initial spin periods is skewed towards shorter periods. If the hyperonic star is metastable, its rotational frequency accelerates distinguishedly before it collapses to a black hole.  相似文献

We report infrared photometry of the extrasolar planet HD 209458b during the time of secondary eclipse (planet passing behind the star). Observations were acquired during two secondary eclipses at the NASA Infrared Telescope Facility (IRTF) in 2003 September. We used a circular variable filter (1.5 per cent bandpass) centred at 3.8 μm to isolate the predicted flux peak of the planet at this wavelength. Residual telluric absorption and instrument variations were removed by offsetting the telescope to nearby bright comparison stars at a high temporal cadence. Our results give a secondary eclipse depth of 0.0013 ± 0.0011, not yet sufficient precision to detect the eclipse, whose expected depth is  ∼0.002 –0.003  . We here elucidate the current observational limitations to this technique, and discuss the approach needed to achieve detections of hot Jupiter secondary eclipses at 3.8 μm from the ground.  相似文献

Owing to effects arising from quantum electrodynamics (QED), magnetohydrodynamical fast modes of sufficient strength will break down to form electron-positron pairs while traversing the magnetospheres of strongly magnetised neutron stars. The bulk of the energy of the fast mode fuels the development of an electron-positron fireball. However, a small, but potentially observable, fraction of the energy (~1033 erg) can generate a non-thermal distribution of electrons and positrons far from the star. This paper examines the cooling and radiative output of these particles. Small-scale waves may produce only the non-thermal emission. The properties of this non-thermal emission in the absence of a fireball match those of the quiescent, non-thermal radiation recently observed non-thermal emission from several anomalous X-ray pulsars and soft-gamma repeaters. Initial estimates of the emission as a function of angle indicate that the non-thermal emission should be beamed and therefore one would expect this emission to be pulsed as well. According to this model the pulsation of the non-thermal emission should be between 90 and 180 degrees out of phase from the thermal emission from the stellar surface.  相似文献
