AMP deaminase catalyzes the conversion of AMP into IMP and ammonia. In the present study, a full-length cDNA of AMPD1 from skeletal muscle of Japanese flounder Paralichthys olivaceus was cloned and characterized. The 2 526 bp cDNA contains a 5’-UTR of 78 bp, a 3’-UTR of 237 bp and an open reading frame (ORF) of 2 211 bp, which encodes a protein of 736 amino acids. The predicted protein contains a highly conserved AMP deaminase motif (SLSTDDP) and an ATP-binding site sequence (EPLMEEYAIAAQVFK). Phylogenetic analysis showed that the AMPD1 and AMPD3 genes originate from the same branch, but are evolutionarily distant from the AMPD2 gene. RT-PCR showed that the flounder AMPD1 gene was expressed only in skeletal muscle. QRT-PCR analysis revealed a statistically significant 2.54 fold higher level of AMPD1 mRNA in adult muscle (750±40 g) compared with juvenile muscle (7.5±2 g) (P<0.05). HPLC analysis showed that the IMP content in adult muscle (3.35±0.21 mg/g) was also statistically significantly higher than in juvenile muscle (1.08±0.04 mg/g) (P<0.05). There is a direct relationship between the AMPD1 gene expression level and IMP content in the skeletal muscle of juvenile and adult flounders. These results may provide useful information for quality improvement and molecular breeding of aquatic animals.  相似文献

Soil-structure interaction (SSI) of a building and shear wall above a foundation in an elastic half-space has long been an important research subject for earthquake engineers and strong-motion seismologists. Numerous papers have been published since the early 1970s; however, very few of these papers have analytic closed-form solutions available. The soil-structure interaction problem is one of the most classic problems connecting the two disciplines of earthquake engineering and civil engineering. The interaction effect represents the mechanism of energy transfer and dissipation among the elements of the dynamic system, namely the soil subgrade, foundation, and superstructure. This interaction effect is important across many structure, foundation, and subgrade types but is most pronounced when a rigid superstructure is founded on a relatively soft lower foundation and subgrade. This effect may only be ignored when the subgrade is much harder than a flexible superstructure: for instance a flexible moment frame superstructure founded on a thin compacted soil layer on top of very stiff bedrock below. This paper will study the interaction effect of the subgrade and the superstructure. The analytical solution of the interaction of a shear wall, flexible-rigid foundation, and an elastic half-space is derived for incident SH waves with various angles of incidence. It found that the flexible ring (soft layer) cannot be used as an isolation mechanism to decouple a superstructure from its substructure resting on a shaking half-space.  相似文献

合成地震动一直是地震学研究中一个非常跃的领域。针对2长周期结构物抗震设计缺少宽频带强震记录的现状,利用“反射率法”和“围道积分法”等合成地震动方法对地震动长周期特性进行了探讨。结果表明:合成地震动方法是研究地震动长周特性的一个有效方法;在地震动长周期成份中,面波起着重要作用。因此长周期结构物的抗震设计必须考虑面波的影响。  相似文献

由于套管的厚度小于目前声波测井仪器工作频率下的波长,声波仪器工作时可在套管中激发Lamb波或SH模式波.目前评价水泥胶结质量的声波测井仪器主要是利用套管中的对称Lamb波(拉伸波,S0模式)或反对称Lamb波(泄漏弯曲波,A0模式)的相对幅度或衰减评价套管与水泥之间的胶结程度.Lamb波可看作是纵波和横波在套管边界的耦合形成的,在套管中还存在另一种非耦合的SH横波,偏振方向与套管轴向平行沿着周向传播.本文基于定向声源辐射技术,实现了对称Lamb波、反对称Lamb波的分波模拟技术,数值研究了套管后耦合不同声阻抗介质时,对称Lamb波、反对称Lamb波以及SH横波的测井响应,通过频散衰减曲线以及模式波的波形响应幅度的直观显示表明,由于套管中的SH波仅向固体水泥中辐射剪切波,这一传播特征与套管中的Lamb波相比可以极大提高套管-水泥界面胶结状况的测量灵敏度.Lamb波的固有缺陷是套管与两侧介质的较大声阻抗反差使得在井中测量套管后面水泥界面的灵敏度受到相当大的限制.  相似文献

Products and mechanisms of the reaction between the nitrate radical (NO3) and three of the most abundant reduced organic sulphur compounds in the atmosphere (CH3SCH3, CH3SH and CH3SSCH3), have been studied in a 480 L reaction chamber using in situ FT-IR and ion chromatography as analytical techniques. In the three reactions, methanesulphonic acid was found to be the most abundant sulphur containing product. In addition the stable products SO2, H2SO4, CH2O, and CH3ONO2 were identified and quantified and thionitric acid-S-methyl ester (CH3SNO2) was observed in the i.r. spectrum from all of the three reactions. Deuterated dimethylsulphide (CD3SCD3) showed an isotope effect on the reaction Deuterated dimethylsulphide (CD3SCD3) showed an isotope effect on the reaction rate constant (kH/kD) of 3.8±0.6, indicating that hydrogen abstraction is the first step in the NO3+CH3SCH3 reaction, probably after the formation of an inital adduct.Based on the products and intermediates identified, reaction mechanisms are proposed for the three reactions.  相似文献

In this paper, we have attempted a problem of reflection and refraction of plane harmonic SH‐wave at a corrugated interface between two different perfectly conducting self‐reinforced elastic half‐spaces. Rayleigh's method is employed to find out the expressions of reflection and refraction coefficients for first‐ and second‐order approximation of the corrugation. The expressions of these coefficients show that they depend on the properties of half‐spaces, angle of incidence, frequency of the incident wave and are strongly influenced by the corrugation of the interface. Numerical computations are performed for a particular model having special type of interface and the variation of these coefficients are depicted graphically against the angles of incidence, frequency parameter, corrugation parameter at different values of reinforcement parameters. Seismic ground motion in central Mexico is amplified relative to ground motion observed at the same epicentral distance along the Pacific Coast in a frequency band that includes destructive ground motion at Mexico City. Although several hypothesis have been advanced, at present there is no generally accepted explanation of such amplification. We have analyzed coda-length magnitude data reported by Servicio Sismológico Nacional (SSN) for events recorded during 1993 to increase our understanding of the spatial distribution of this phenomenon. Our results indicate that regional amplification: (a) is detected by magnitude residual computed at each station, relative to the average of SSN network;and (b) is likely related to the crustal structure under the central portion of the Transmexican Volcanic Belt (TMVB). Finally, preliminary wave propagation modelling (using SH wave, finite difference method) suggests that crustal heterogeneity is a possible cause of regional amplification. However, if this is so, it is required that both geometry and velocity distribution vary between the coast and Mexico City.  相似文献

对Wolf理论进行拓展,使之可解决沉积谷地对波的散射问题,进而利用间接边界元法,求解了弹性层状半空间中沉积谷地对入射平面SH波的放大作用问题。通过自由场反应分析,求得假想边界上各点的位移和各单元的应力反应。施加虚拟均布荷载,求得位移和应力的格林函数。根据应力和位移连续边界条件确定虚拟分布荷载,将自由场反应和虚拟分布荷载产生的反应叠加起来,即得到问题的解答。最后,以基岩上单一土层中沉积谷地对入射平面SH波的放大作用为例进行了数值计算分析,结果表明,层状半空间中沉积谷地和均匀半空间中沉积谷地对波的放大作用存在显著差别。文中分析了造成差别的原因,并讨论了覆盖层厚度和刚度对放大作用的影响。  相似文献

A continuum model for the interaction analysis of a fully coupled soil–pile–structure system under seismic excitation is presented in this paper. Only horizontal shaking induced by harmonic SH waves is considered so that the soil–pile–structure system is under anti‐plane deformation. The soil mass, pile and superstructure were all considered as elastic with hysteretic damping, while geometrically both pile and structures were simplified as a beam model. Buildings of various heights in Hong Kong designed to resist wind load were analysed using the present model. It was discovered that the acceleration of the piled‐structures at ground level can, in general, be larger than that of a free‐field shaking of the soil site, depending on the excitation frequency. For typical piled‐structures in Hong Kong, the amplification factor of shaking at the ground level does not show simple trends with the number of storeys of the superstructure, the thickness and the stiffness of soil, and the stiffness of the superstructure if number of storeys is fixed. The effect of pile stiffness on the amplification factor of shaking is, however, insignificant. Thus, simply increasing the pile size or the superstructure stiffness does not necessarily improve the seismic resistance of the soil–pile–structure system; on the contrary, it may lead to excessive amplification of shaking for the whole system.
