采用半连续培养方法研究了温度和营养盐(N和P)限制对中国东海2种重要赤潮生物东海原甲藻和中肋骨条藻生长及种间竞争的影响。结果表明,东海原甲藻在20℃和25℃时生长状态良好,具有明显的指数增长期,15℃时细胞生长明显受到影响;中肋骨条藻具有较广的温度适应性,15~25℃时均具有明显的指数增长期。东海原甲藻可以忍受低营养盐环境并种群增长,而中肋骨条藻细胞增长需要较丰富的营养盐。在营养盐充足的环境里中肋骨条藻具有竞争优势,相反,在营养盐限制的环境中,东海原甲藻是竞争的优胜者。实验结果与东海原甲藻赤潮爆发现场的环境调查结果基本一致,可以作为解释东海原甲藻赤潮形成原因的依据。  相似文献

为了开展无乳链球菌(Streptococcus agalactiae)核糖结合蛋白(Ribose binding protein B,RbsB)结构功能的研究,本实验根据已知无乳链球菌ZQ0910全基因组序列设计相关引物。采用PCR方法扩增其RbsB基因,随后将该基因定向克隆到原核表达载体pGEX-6p-1中,在大肠杆菌BL21(DE3)感受态细胞中进行IPTG诱导表达;采用HRV 3C蛋白酶切除GST标签,分子筛分离获得RbsB蛋白;运用生物信息学软件对RbsB基因序列进行分析,并对RbsB蛋白二级和三级结构进行预测;采用NeXtal Tubes JCSG Core Suite结晶试剂盒筛选蛋白结晶条件。研究结果表明,该基因全长为969碱基,编码322个氨基酸,RbsB蛋白理论分子量33.9ku,等电点为9.41,二级结构中α螺旋结构所占比重最高,建立RbsB蛋白三维结构模式图;经IPTG诱导后表达的融合蛋白分子量为59ku,筛选RbsB蛋白的初始结晶条件为(0.2mol/L di-Potassium hydrogen phosphate,20%(W/V)PEG3350;1.5mol/L ammonium sulfate,25%(V/V)Glycerol),获得RbsB蛋白结晶体。本研究结果可为无乳链球菌核糖结合蛋白(RbsB)的功能解析提供实验及理论基础。  相似文献

房山岩体是由 3次脉动式侵位形成的复式岩体 ,不同期次侵位的岩石及其中的包体来自相同的岩浆源区 ,为下地壳的中、基性火成岩。全岩 矿物Rb Sr等时线年龄为 12 6± 3Ma。第 1期至第 2期 ,主要是由以斜长石为主的矿物平衡结晶造成的 ;第 2期至第 3期 ,则可能是批式熔融的结果。包体的成因机制可能不止一种 :英安玢岩质包体和石英粗安玢岩质包体可能分别代表第 2期和第 3期脉动式侵入体的冷凝壳 ;石英正长岩包体则可能代表该侵入体演化到最后的产物 ;而石英闪长质和石英二长闪长质包体则可能为同源岩浆早期结晶的产物。  相似文献

In this study,the effects of ‘initial’ soil moisture(SM) in arid and semi-arid Northwestern China on subsequent climate were investigated with a regional climate model. Besides the control simulations(denoted as CTL),a series of sensitivity experiments were conducted,including the DRY and WET experiments,in which the simulated ‘initial’ SM over the region 30 –50°N,75 –105°E was only 5% and 50%,and up to 150% and 200% of the simulated value in the CTL,respectively. During several triaxial compression experiments on plastic hardening, softening, and failure properties of dense sand specimens, it was found on various stress paths that the size of the failure surface was not constant. Instead, it changed depending on the current state of hydrostatic pressure. This finding is in contrast to the standard opinion consisting of the fact that the failure surface remains constant, once it has been reached during an experiment or in situ. In general, the behaviour of cohesionless granular‐material‐like sand is somehow characterised in between fluid and solid, where the solid behaviour results from the angle of internal friction and the confining pressure. Although the friction angle is an intrinsic material property, the confining pressure varies with the boundary conditions, thus defining different solid properties like plastic hardening, softening, and also failure. Based on our findings, it was the goal of the present contribution to introduce an improved setting for the plastic strain hardening and softening behaviour including the newly found yield properties at the limit state. For the identification of the material parameters, a complete triaxial experimental analysis of the tested sand is given. The overall elasto‐plasticity concept is validated by numerical computations of several laboratory foundation‐ and slope‐failure experiments. The performance of the proposed approach is compared with the standard concept of a constant failure surface, where the corresponding yield surfaces are understood as contours of equivalent plastic work or plastic strain. 江苏省东海、宁镇和苏州地区分布有若干分属于两类不同成因类型的花岗岩类岩体。在这些岩体的边部和顶部分布着许多形态极不规则、大小各异的暗色包体,它们的矿物组合、岩石化学和地球化学特征与主体岩石相似,为同源产物。包体中矿物的结晶粒度明显细于主体岩石,并含有大量因骤冷而晶出的针状磷灰石,且见不同程度的重熔现象。故它们是由于岩浆脉动上侵、挤碎并重熔的早期形成的岩体边缘相岩石,而不是早期火成岩和围岩捕虏体,也不是岩浆析离体和深源包体。  相似文献

Abstract Data are presented on a garnet population in a specimen of garnet-biotite-plagioclase-quartz schist from the cordierite zone of an Archaean thermal dome in the Southern Slave Province of the Canadian Shield. Garnet crystals are bounded by planar dodecahedral faces and by trapezohedral faces which on the 10-μm scale are corrugated. Crystal distribution, as revealed by dissection of a small cubic volume of rock, is random. The size distribution is normal, with a mean diameter of 0.81 mm and a standard deviation of 0.32 mm. In the largest crystal of the population (mean radius 0.83 mm), [Mn] = 100 Mn/(Fe + Mg + Mn + Ca) decreases from 14.5 at the centre to 7.5 and then increases in the outer margin to 8.5; [Fe] increases continuously from 67 at the centre to 77 at the surface; [Mg] increases from 12.5 to 13.5 and then falls sharply to 11; [Ca] remains unchanged at 4.0 and then drops to 3.3. Progressively smaller crystals have progressively lower [Mn] and higher [Fe] concentrations at their centres, while all crystals have the same margin composition. Growth vectors extending from given concentration contours to crystal surfaces are of equal length regardless of the size of the crystal in which the vector is located. A garnet-forming model is presented in which reaction was initiated by a rise in temperature. Nucleation sites were randomly selected. The nucleation rate increased with time and then declined. Crystal faces advanced at a constant linear rate, which implies an increase in volume proportional to surface area. Initially, the composition of garnet deposited on crystal surfaces was determined by van Laar equations of equilibrium, which demanded the withdrawal of Mn and Fe from within chlorite crystals. This transfer reaction was then accompanied by an ion exchange reaction which moved Mn and Fe to garnet surfaces from biotite, in exchange for Mg. The exchange reaction provides an explanation for the high overall concentration of Mn and Fe in garnet and for the observed Mn and Mg reversals in the margins of crystals. The increase of garnet volume in the garnet population is found to be parabolic, i.e. Vαα5.  相似文献

Bent-over buoyant jets distorted by a crosscurrent develop a vortex pair structure and can bifurcate to produce two distinct lobes which diverge from one another downwind. The region downwind of the source between the lobes has relatively low proportions of discharged fluid. Factors invoked by previous workers to cause or enhance bifurcation include buoyancy, release of latent heat at the plume edge by evaporating water droplets, geometry and orientation of the source, and the encounter with a density interface on the rising path of the plume. We suggest that the pressure distribution around the vortex pair of a rising plume may initially trigger bifurcation. We also report new experimental observations confirming that bifurcation becomes stronger for stronger bent-over plumes, identifying that bifurcation can also occur for straight-edged plumes but gradually disappears for stronger plumes which form a gravity current at their final level and spread for a significant distance against the current. Observations from satellites and the ground are reviewed and confirm that volcanic plumes can show bifurcation and a large range of bifurcation angles. Many of the bifurcating plumes spread out at the tropopause level and suggest the tropopause may act on the plumes as a density interface enhancing bifurcation. Even for quite moderate bifurcation angles, the two plume lobes become rapidly separated downwind by distances of tens of kilometers. Such bifurcating plumes drifting apart can only result in bilobate tephra fall deposits. The tephra fall deposit from the 16 km elevation, SE spreading, bifurcating volcanic plume erupted on 15 May 1981 from Mt Pagan was sampled by previous workers and clearly displayed bilobate characteristics. Examples of bilobate tephra fall deposits are reviewed and their origin briefly discussed. Bilobate deposits are common and may result from many causes. Plume bifurcation should be considered one of the possible mechanisms which can account for come examples of bilobate tephra fall deposits.  相似文献
