Acidic oligosaccharide sugar chain (AOSC), a D-mannuronic acid oligosaccharide, derived from brownalgaepolysaccharide, has been completed Phase I clinical trial in China as an anti-Alzheimer's Disease (AD) drug candidate. The identification of AOSC-binding protein(s) in neurons is very important for understanding its action mechanism. To determine the binding protein(s) of AOSC in neurons mediating its anti-AD activities, confocal microscopy, affinity chromatography, and liquid chromatography-tandem mass spectrometry (LC-MS/MS) analysis were used. Confocal microscopy analysis shows that AOSC binds to SH-SY5Y cells in concentration-, time-, and temperature-dependent fashions. The AOSC binding proteins were purified by affinity chromatography and identified by LC-MS/MS analysis. The results showed that there are 349 proteins binding AOSC, including clathrin, adaptor protein-2 (AP-2) and amyloid precursor protein (APP). These results suggest that the binding/entrance of AOSC to neurons is probably responsible for anti-AD activities. 相似文献

Frontal affinity chromatography was applied to characterizing the mechanism of binding of silver with sediment particulates collected from Lake Ontario, Canada. The results showed that there was one major binding site for Ag+ in the particulates. The binding capacity ranges from 6.06 to 1.01 μ·mol·g-1, and the binding constant (lgK) from 6.23 to 7.43 M-1 in 0.005 M ion strength at pH=3-7. Conditional surface binding constants and complexation capacities for Zn, Pb, Cd, and Cu were determined from surface titration experiments of heterogeneous natural aquatic particulate matter of different origin and composition. Metals and particles were evaluated in naturally occurring concentration ranges in river water.The adsorption of trace metals can be adequately described with a single conditional binding constant over a wide range of metal : particle ratios. Ca = 2.5 (pH 8.0). Maximum surface binding capacities for all metal ions were found for particles containing high fractions of Mn-oxides which are associated with large specific surface areas. Generally, we found sorption capacities to decrease in the sequence Cu Pb, Zn > Cd.The experiments suggest that the conditional surface binding constants and complexation capacities are applicable to model trace metal adsorption in the concentration ranges of natural waters under conditions similar to the experiments. 增长是数量的增加,是量变;发展,是质量的提升,是质变。囿于煤炭开发过程中的收益递减规律,煤炭资源有效供给不足和对煤烟型污染控制的法律约束导致煤炭需求受限,故而煤炭工业增长具有不可持续之特性。我国煤炭工业的持续增长已经付出了高昂的资源成本、环境成本和安全健康成本。煤炭产量的无序增长,开采技术低水平粗放式的外延扩张,将进一步破坏资源、环境。我国西部将会再现新的楼兰化景象,我们对此深感忧虑。煤炭工业的可持续发展之路为:以煤炭资源的充分合理利用为基础,以环境容量为前提,以废弃物资源化为指导思想,以适度规模的循环经济为目标模式。煤炭工业的可持续发展应以其不可持续增长——有限增长为约束条件。 相似文献

A thin-walled steel tube/bamboo plywood composite hollow column with binding bars (SBCCB) was developed using transverse binding bars to reinforce a thin-walled steel tube and bamboo plywood composite hollow column. Low reversed cyclic loading tests were performed on 9 SBCCB specimens to observe the damage process and morphology in the SBCCBs. The effects of the slenderness ratio, the net cross-sectional area of the bamboo plywood, the assembly patterns of the cross sections, and the binding bars on the stress and seismic performance were investigated. The results show that the main failure modes of the SBCCBs were cracking of different adhesion interfaces at the base of the column and fracturing of the bamboo plywood. The cross-sectional assembly pattern has a significant effect on the failure modes; increasing the cross-sectional area and the slenderness ratio of the composite column will dramatically improve the seismic performance of the SBCCBs. SBCCBs have excellent elastic deformation and seismic energy consumption characteristics. The binding bars ensure the integrity of the specimen and inhibit the failure of the adhesion interface at the base of the column. Finally, based on the measurement of the axial pressure and the stress-strain curve within elastic range of the composite columns, a simplified mechanical model and a calculation method for bearing capacity was formulated. The calculation values of bearing capacity agree well with the experimental ones, and calculation method can be used to provide guidance for engineering applications of a SBCCB. 相似文献
