A putative tetrasporophyte-specific gene, designated as SSH466 (GenBank accession No. DQ019223), was one of the genes identified
in this work using suppression subtractive hybridization (SSH) method in Gracilaria lemaneiformis. The full length of the gene was obtained using SMART RACE strategy. Sequence analysis revealed that the gene had 1 019 nucleotides,
including an open reading frame of 498 nucleotides encoding 166 amino acid residues, 158 nucleotides of 5′ untranslated region
and 363 nucleotides of 3′ non-coding region. Protein motif and secondary structure prediction showed that there existed a
transmembrane domain with a unique β-sheet. Thus, SSH466 protein might be a cross-membrane protein. Sequence homology search
in the public GenBank databases did not reveal any significant match with SSH466. Virtual Northern blot analysis confirmed
that it was a tetrasporophyte-specific gene. 相似文献

Gametophytes of Laminaria japonica were cryopreserved in liquid nitrogen using encapsulation-dehydration with two-step cooling method. Gametophytes cultured at 10℃ and under continuous irradiance of 30 μmol m-2 s-1 for 3 weeks were encapsulated in calcium alginate beads. The beads were dehydrated in 0.4 molL-1 sucrose prepared with seawater for 6 h, desiccated in an incubator set at 10℃ and 70% relative humidity for 4 h, pre-frozen at either -40℃ or -60℃ for 30 min, and stored in liquid nitrogen for >24 h. As high as 43% of survival rate was observed when gametophytes were thawed by placing the beads in 40℃seawater and re-hydrated in 0.05 molL-1 citrate sodium prepared using 30‰ NaC1 7 d later. More cells of male gametophytes survived the whole procedure in comparison with female gametophytes. The cells of gametophytes surviving the preservation were able to grow asexually and produce morphologically normal sporophytes. 相似文献

克隆了半滑舌鳎脂肪分解关键酶基因即激素敏感脂肪酶(Hormone-sensitive lipase,HSL)和脂肪甘油三酯脂肪酶(Adipose triglyceride lipase,ATGL)cDNA部分序列,并探讨了饲料中脂肪水平对半滑舌鳎稚鱼HSL及ATGL基因表达的影响。克隆所得到的HSL和ATGL片段长度分别为589bp和581bp,序列和系统进化分析表明半滑舌鳎的HSL和ATGL与大部分鱼类具有较高的同源性。采用不同水平的鱼油配制成5种不同脂肪水平(6.68%、9.84%、13.47%、17.89%和21.88%干物质)的等氮饲料,饲养35日龄半滑舌鳎稚鱼30d,养殖过程中每天饱食投喂5次,养殖实验结束后采用实时荧光定量PCR分别测定各处理稚鱼内脏团的HSL及ATGL mRNA相对表达量。结果表明,高脂饲料组(21.88%)显著促进了半滑舌鳎稚鱼内脏团HSL基因的表达(P0.05),但饲料脂肪水平对于半滑舌鳎稚鱼内脏团的ATGL表达量未产生显著影响(P0.05)。 相似文献

采用相关序列扩增多态性(sequence-realted amplified polymorphism,SRAP)技术分析野生草鱼和家养草鱼,并筛选与草鱼种质退化相关的分子遗传标记。共进行88对引物组合的检测,产生标记数目共计905个。依据标记在群体中出现的频率和变化规律,共筛选出21个可能与种质相关的特异性标记,对这些特异性标记进行测序并将测序结果进行BLAST分析,发现测得片段中有8个片段能在GeneBank中找到同源性较高的序列,而其他片段与数据库中序列的相似性较低。 相似文献

When cultured under certain environmental coriditions (25℃, light intensity 80 μmol/m~2 ·s, LD12/12, in enriched seawater medium with 7 × 10~(-4)mol/L NO_3,-N, 1.56× 10~(-4)mol/L, PO_4-P and supplementsof other elements like Mn, Fe, I, etc.), mate and female gametophytes of U pinnatifida keptgrowing vegetatively and propagated fast at average daily fresh weight increase rate of about 20%. Theempirical formula G_m=G_o. 3~m was established to estimate the output of vegeative gametophytes. Vigorousvegetative gametophyte cells began to form reproductive structures (oogonium and spermatangium, whenthe tmperature was lower than 25℃ and other environmental factors wrre kept optimal. The sufficientsupply of gametophyte cells provided enough seeds for raising Undaria sporelings on prodiction scale.Controlled cross-breeding experiments using selcted male and female gametophyte clones which increasetheir cell number by mitosis instead of meiosis were also carried out in vitro.
