There is enormous potential for global transfer of microorganisms, including pathogens, in ships' ballast water. We contend that a major advancement in the study of ballast-water microorganisms in particular, and of aquatic pathogens in general, will be expedited sample analysis, such as provided by the elegant technology of DNA microarrays. In order to use DNA microarrays, however, one must establish the appropriate conditions to bind target sequences in samples to multiple probes on the microarrays. We conducted proof-of-concept experiments to optimize simultaneous detection of multiple microorganisms using polymerase chain reaction (PCR) and Southern hybridization. We chose three target organisms, all potentially found in ballast water: a calicivirus, the bacterium Vibrio cholerae, and the photosynthetic protist Aureococcus anophagefferens. Here, we show simultaneous detection of multiple pathogens is possible, a result supporting the promising future use of microarrays for simultaneous detection of pathogens in ballast water. 相似文献

Appropriate pathogenic bacteria were isolated from diseased (or dead) flounder (Paralichthys olivaceus L) occurring in a mariculture farms of Qinhuangdao, which all diseased fishes expressed bacterial septicaemia. The phenotypic information of the 12 pure cultures was studied extensively, including morphological characteristics, colony characteristics in different media, physiological and biochemical characteristics, and the mole fraction G C ratio of the DNA for representative strain. The results show that the isolates belong to a new species of Vibrio, and are designated as Vibrio qinhuangdaora sp.nov, by the area from which the diseased flounders are collected (Qinhuangdao) based on its biological properties following Rules of International Code of Nomenclature of Bacteria. At the same time,studies on the serum homology ofisolates, and pathogenicity of isolates were conducted by experimental infection. The results show that all the isolates are of serologic similarity, and the isolates have strong pathogenicity to flounder. 本研究采用快速瑞氏-吉姆萨染色法观察鳗弧菌(Vibrio anguillarum)刺激后魁蚶(Scapharca broughtonii)几种血细胞的形态变化,研究不同种类血细胞的免疫功能。魁蚶血淋巴液中红细胞、白细胞和血栓细胞经免疫刺激后均发生形态改变,属于免疫防御的应激反应。经鳗弧菌免疫刺激后,红细胞无丝分裂现象增多,并参与病原菌的吞噬作用;白细胞变形运动、吞噬和融合现象普遍,细胞表面具有粘性,参与血栓的形成;血栓细胞主要表现出粘连和凝血的特性,偶有吞噬病原菌的现象;研究中检测了2种常规抗凝剂的抗凝效果,结果表明,2种抗凝剂分别改变了血栓细胞和白细胞的表面粘连特性,从而阻止它们参与凝血,因而都具有抗凝效果。本研究结果表明,魁蚶血细胞分工明确,能共同防御病原菌的入侵,细胞免疫在魁蚶血淋巴液抗菌免疫中起极其重要的作用,血细胞的形态变化和免疫功能分析可为后期研究魁蚶抗菌机制和筛选抗凝剂奠定基础。 相似文献

The characteristics of a bacterium strain M3, isolated from cultured flounderParalichthys olivaceus with remarkable external sign of skin ulcer during an epizootic outbreak, indicated that the bacterium belonged to the speciesVibrio anguillarum. Challenge by I.M. (intramuscular injection), bath, and oral administration with M3 showed that it was highly pathogenic
forParalichthys olivacues. The LD50 dose was 5.144×103 CFU/ per fish infection by I.M. injection. Recovered inoculated bacteria from the surviving fish revealed that the asymptomatic
carriers could be a latent contagious source. Study of the effect of bacterial culture CFS (cell-free-supernatant) showed
that the exotoxins produced by M3 play an important role in its pathogenicity for flounder. The resistance of M3 to 36 out
of 41 antibiotics indicated that the bacterial disease outbreak was mainly attributable to the frequent and excessive use
of antimicrobial agents; and that vaccination would be an effective precaution against bacterial disease.

Vibrio harveyi cells (dose = > 103 cells mL 1) and extracellular products (ECP; >25μg m L-1 of total protein concentration) destroyed haematopoietic cultures of Nephrops norvegicus within 24 h of exposure. Cytopathic effects (CPE)started after 4 h of exposure to the bacterial cells, with some granularity in the cytoplasm, mostly in cells in the outer periphery of the explant growth. At the end of the infection, a considerable number of nuclei remained attached to the substrate,apparently unaffected. Following exposure to ECP, initial deterioration was observed at 2 h with the presence of granularity in the cytoplasm of< 20% cells, and few cells displayed small vacuoles around the nuclei. Parallel results were obtained using whole animal experiments, with V. harveyi cells being lethal to nephrops within 24 h. 相似文献
