本研究通过对线粒体控制区序列进行分析的技术手段,对中国宁德(ND)、厦门(XM)、漳浦(ZP)、南澳岛(NAD)、湛江(ZJ)、海口(HK)与北海(BH)的7个野生黄鳍棘鲷(Acanthopagrus latus)群体的遗传多样性和遗传分化进行比较分析。结果表明:中国东南沿海的野生黄鳍棘鲷群体呈现出中等以上的遗传多样性特征,48个黄鳍棘鲷样品的控制区序列长度为580 bp,单倍型数目为44个,总体的单倍型多样性指数(Hd)和核苷酸多样性指数(Pi)分别为0.996和0.017 30。7个野生黄鳍棘鲷群体间的遗传分化程度较低,各群体间的遗传距离为0.010 28~0.029 87。群体间的遗传分化系数(FST)为-0.068 41~0.545 86,北海群体和其他群体间存在一定程度的遗传分化。AMOVA分析显示群体内的遗传变异占比为83.34%,大部分遗传变异发生在组群内;但北海群体与其他6个群体分为两个组群后,组群间的遗传变异占比为42.66%,两个组群间存在较大程度的遗传变异。基于Kimrua距离法构建的系统进化树中显示北海群体聚为一个分支,另外6个群体聚为一支。基于TCS network构建的单倍型网络图中44个单倍型混杂在一起,未按照水系格局和地理距离进行分布,表明黄鳍棘鲷群体间未形成多个单系群。本研究可为中国东南沿海野生黄鳍棘鲷群体的种质资源利用和保护提供科学依据。  相似文献

为鉴定鱼肚的正品来源,运用一对特异性引物扩增并测定10 个不同价格(700~1 500 元/kg)鱼肚样本的线粒体DNA 细胞色素c 氧化酶Ⅰ(cox1)约660 bp 的序列.通过BLASTN 比对DNA 序列同源度,判定鱼肚所属种类分别为鲈形目石首鱼科(Sciaenidae)、马鲅科(Eleutheronema)、笛鲷科(Lutjanidae)、带鱼科(Trichiuridae).cox1 条码序列获取便捷,基于此条码序列的物种鉴定技术可用于鱼肚种类鉴别.  相似文献

There have been comparatively few large-scale studies on spatial genetic structure of bathyal seafloor fauna, despite the importance of these data to the successful management of the world's oceans. We use a comparative analysis of mitochondrial DNA from five bathyal (200–3500 m) species of brittle-stars (Ophiuroidea) to assess phylogeographic structure along an extensive (8000 km) longitudinal gradient at temperate latitudes (28–56°S) from south-west Australia (113°E) to seamounts east of New Zealand (175°W). We found no evidence of a genetic discontinuity between Australia and New Zealand, either across the temperate Tasman Sea or across the Southern Ocean between the South Tasman Rise and the Macquarie Ridge. However, there were latitudinal phylogeographical breaks between tropical, temperate and polar regions; longitudinal breaks across the eastern Indian Ocean; and a bathymetric break at approximately 1700 m. Although there was limited regional structure in the frequency of haplotype distributions within the major clades, and no clade appeared to be strictly panmictic, the regional structure in general was not concordant with a simple isolation-by-distance model. Demographic structure varied with three clades having a simplified haplotype network, low effective population sizes and no evidence of significant population expansion, and two clades having a high diversity of haplotypes, relatively high effective population sizes and signs of recent population expansion. These results are discussed with respect to putative dispersal strategies. We hypothesise that the ‘brooding’ species produce both brooded young and pelagic larvae, allowing for both the maintenance of local populations and long-distance dispersal.  相似文献

Humans colonized the Balearic Islands 5–4 ka ago. They arrived in a uniquely adapted ecosystem with the Balearic mountain goat Myotragus balearicus (Bovidae, Antilopinae, Caprini) as the only large mammal. This mammal went extinct rapidly after human arrival. Several hypotheses have been proposed to explain the extinction of M. balearicus. For the present study ancient DNA analysis (Sanger sequencing, Roche-454, Ion Torrent), and pollen and macrofossil analyses were performed on preserved coprolites from M. balearicus, providing information on its diet and paleo-environment. The information retrieved shows that M. balearicus was heavily dependent on the Balearic box species Buxus balearica during at least part of the year, and that it was most probably a browser. Hindcast ecological niche modelling of B. balearica shows that local distribution of this plant species was affected by climate changes. This suggests that the extinction of M. balearicus can be related to the decline and regional extinction of a plant species that formed a major component of its diet. The vegetation change is thought to be caused by increased aridity occurring throughout the Mediterranean. Previous hypotheses relating the extinction of M. balearicus directly to the arrival of humans on the islands must therefore be adjusted.  相似文献

Replicating prebiotic polymers are thought to predate the emergence of true life-forms. The initial mode of replication, a prerequisite for Darwinian selection, is unknown, but demands an explanation based on local physicochemistry. Dual consideration of the conditions of the early terrestrial surface, with the unusual physicochemical properties of nucleic acids like DNA, could explain the emergence of nucleic acids as key biomolecules. The early impact that produced the Moon, and fast terrestrial rotation, subjected coastal areas 3.9 Ga ago to rapid tidal flooding (dilution) and drying (concentration), with a likely periodicity in the range of 2-6 h, and could have provided a driving force for cyclic replication of early biomolecules. Such a mechanism applies only to molecules capable of association/polymerization at high salt concentration, and of dissociation at low salinity. Nucleic acids meet these criteria. It is suggested that tidal cycling, resembling the polymerase chain reaction (PCR) mechanism, could only replicate and amplify DNA-like polymers. This mechanism suggests constraints on the evolution of extra-terrestrial life.  相似文献

In this study, two common pollutants (benzo[a]pyrene and methamidophos) in marine environment were tested by comet assay for their inducement of in vivo genotoxic effect to the blood cells of black porgy ( Acanthopagrus schlegeli). The fish was exposed to 2 μg/L of benzo[a]pyrene (BaP) and methamidophos, and their mixture. The assay was performed on whole blood at 2 h, 5 h, 24 h and 96 h exposure intervals. A significant increase in DNA damage was observed in each treatment with the pollutants. Additive effect of BaP and methamidophos was also found in the experiment. However, the decrease ratios of DNA damage for 5 h and 96 h exposure interals compared with 2 h and 24 h exposure ones, respectively, were noticed. This phenomenon may be explained by the function of repairing process via enzyme cytochrome P450 in the animal. Evidence of the genotoxicity of organophosphorus pesticides (OPs) and polynuclear aromatic hydrocarbons (PAHs) on marine fish are discussed in this paper.  相似文献

生物体可看作是一种特殊的信息处理系统 ,论文的第二章从这种观点出发讨论了生物细胞内最大的信息源基因组DNA序列的信息编码问题。首先探讨了基因组蛋白质编码区的信息编码问题 ;简要回顾转录水平的基因表达调控后 ,讨论了基因组基因表达调控区的信息编码问题。第三章介绍了我们利用internet上可获取的开放的生物信息学资源 ,对基因组DNA调控区的一种可能的编码方式所进行的理论研究。这种可能的编码方式我们称之为双链DNA上的碱基空间模式BSP (BaseSpatialPattern)。首先 ,我们发现了由三联体BSP介导的一种 4与 2 0的关系 ,称为 4 -BSP -2 0关系。并推测 4 -BSP -2 0关系也许是某些DNA—蛋白质相互作用中的一种识别密码 ;第二 ,研究真核细胞转录调控中的DNA—蛋白质相互作用的实验数据表明 :某些DNA结合蛋白质对双螺旋核酸的专一性识别可能与BSP有关。并找到了 7组由其他生物学家公开发表的实验数据支持这一猜测 ;第三 ,研究了BSP的形式化描述问题 ,找到了一种基于等价类的BSP描述方法 ,并开发了相应的计算机处理算法。该描述方法简明且便于用计算机进行处理 ,更重要的是它提供了一种研究途径 ,可把BSP和 4 -BSP -2 0关系推广到单链DNA (ssDNA)和ssDNA -蛋白质相互作用的研究中 ,从而可进一步推广到RN  相似文献

In order to thoroughly investigate the diversity of glacier microorganisms, four DNA extraction methods with different lysis patterns were tested and two screened methods (the Bosshard-Bano method and the Zhou method) were optimized for the most effective form of the filter membrane (cut vs. uncut), the DNA extraction method, and the precipitation method. The two optimized methods were then compared with the commercial Mo-Bio DNA extraction kit, and the results showed that the kit was generally suitable for extraction of microorganism DNA from glacier surface snow. Procedurally, it was found that a modified Bosshard- Bano method (i.e., cutting the filter membrane into pieces, using a specific lysis pattern [lysozyme (5 mg/mL)-protease K (1 mg/mL)-CTAB (1%)-SDS (1%)], performing the extraction only once by chloroform-isoamyl alcohol (24:1), and conducting DNA precipitation by pure ethanol) was also an effective and less expensive method for extraction of microorganism DNA from glacier surface snow.  相似文献

测定了洱海沉积物中有机碳、氮,提取了沉积物中的DNA。有机碳氮呈现相同的变化趋势降解主要发生在表层10cm;7cm前后氧化-还原条件变化,微生物电子受体改变,所以有机质的降解方式不同,出现不同的特征。利用分子生物学和地球化学相结合的方法,能更全面的认识和理解早期成岩过程。  相似文献
