The Australian Institute of Marine Science (AIMS) conducted a pilot study around the Harriet A oil production platform on the Northwest Shelf of Australia. We evaluated hepatic ethoxyresorufin-O-deethylase (EROD) activity, fluorescent aromatic compounds (FACs) in bile and immunodetection of CYP1A-like proteins in two Australian tropical fish species, Gold-Spotted Trevally (Carangoides fulvoguttatus) and Bar-Cheeked Coral Trout (Plectropomus maculatus) to assess exposure to petroleum hydrocarbons associated with produced formation water (PFW). Additionally, the incidence of hydrocarbon-degrading bacteria isolated from the liver and bile of all fish captured was examined. Low EROD activity was found in both species, with EROD activity in C. fulvoguttatus showing significant site differences. FACs and CYP1A protein levels in C. fulvoguttatus showed a clear trend in hydrocarbon exposure consistent with hydrocarbon chemistry data: Harriet A>Harriet C>reference site. P. maculatus showed elevated levels of FACs at Harriet A as compared to the reference site and demonstrated detectable levels of CYP1A-like proteins at these two sites. Hydrocarbon-degrading bacteria were found in the liver and bile of both species, yet there was no correlation by sites. Our results demonstrate that C. fulvoguttatus and P. maculatus have potential as indicator species for assessing the effects from exposure to petroleum hydrocarbons. Both FACs and CYP1A are providing warning signs that there is potential for biological effects on fish populations exposed to PFW around the Harriet A production platform.  相似文献

The incidence of hepatoma, epidermal and other forms of cancerous growths in fish is well documented (Halver, 1967; Matsushima & Sugimura, 1976; Dawe et al., 1964). In fish, as in mammals, cancer may be a result of metabolically activated carcinogens. In mammals the primary enzyme system involved in the activation of environmental carcinogens is the cytochrome P-450 linked mixed-function oxidase (MFO). The hepatic microsomes of the species offish studied contain variable levels of cytochrome P-450. Liver microsomes of the trout Salmo trutta lacustris are surprisingly active in metabolising benzo-[a]pyrene (BP) and other compounds preferentially metabolised by polycyclic aromatic hydrocarbon (PAH)-specific cytochrome P-450. This finding may be significant, because it is apparent that the detrimental effects of PAHs require metabolic activation.We have studied the production of reactive intermediates of BP by following their binding to DNA and by measuring the specific nucleotide-BP metabolite complexes by chromatography. Untreated S. trutta liver microsomes catalyse the production of reactive intermediates of BP which bind to nucleotides of DNA at a rate that is 3–4 times higher than that catalysed by control rat liver microsomes. The most prominent DNA-BP metabolite adducts produced by trout liver microsomes are the nucleoside adduct of BP-7, 8-diol-9,10-epoxide and 9-OH-BP-4,5-oxide and other phenol oxides. In contrast to the trout, another fish species, roach (Rutilus rutilus), has extremely low activity. Although the in vitro binding of BP to DNA is not strictly correlated to in vivo binding or biological effects, our results show that reactive intermediates are formed by trout liver and thus the prerequisite for chemical carcinogenesis or mutagenesis is ful filled. This is further supported by the fact that in Ames's test, trout liver preparations produce mutagenic products from promutagens.  相似文献

Geometric test field calibration of digital photogrammetric sensors   总被引:1,自引:0,他引:1  
Test field system calibration will be a fundamental part of the future photogrammetric production line. Accurate calibration and performance evaluations are necessary for fully assessing the stability and accuracy of digital sensing techniques. In this paper, a method of comprehensive geometric calibration in a test field has been developed and empirically tested using eight image blocks collected with three UltraCamD digital large format photogrammetric cameras. Permanent photogrammetric test fields form the basis of the method. Important components of the method are determination of system parameters, evaluation of systematic errors, and assessment of geometric accuracy. The results showed that UltraCamD images contained systematic deformations that could not be modeled with single lens additional parameter models. Good point determination accuracy was obtained despite the systematic errors; the typical accuracy was 2–3 μm in image space in the horizontal coordinates and 0.05–0.09‰ of the object distance in height. One of the cameras had significantly poorer performance. In the worst cases, the horizontal accuracy was 5 μm in image space and the height accuracy was 0.18‰ of the object distance. The analog cameras gave better results than the UltraCamD, but the development of appropriate mathematical models for UltraCamD as well as improvements in digital sensors may change the situation in the near future.  相似文献

A variety of sources of organic contaminants to the Great Barrier Reef lagoon and near-shore environment exist including boating activity, agriculture and urban run-off. Cytochrome P-450 1A activity as measured by ethoxy-resorufin O-deethylase (EROD) activity has been widely used as an indicator of the exposure of fish to organic contaminants such as polychlorinated biphenyls (PCB), polycyclic aromatic hydrocarbons (PAH) and some organochlorine pesticides. This study demonstrates the successful application of EROD measurements in a common Australian tropical estuarine fish species, Acanthopagrus berda (Pikey Bream), to identify areas under potential stress from organic contaminants. Fish were captured from four creeks draining agricultural land, a creek draining urban land and two creeks with less disturbed catchments. Significant induction of cytochrome P450-1A was observed in fish captured from Ross Creek (urban catchment, 7.4-fold) and Cromarty Creek (agricultural catchment, 6.4-fold). Increased activity was also observed in fish captured from other creeks draining agricultural land (Plantation Creek, Victoria Creek, Seymour River, 1.9–2.6-fold) as compared to those captured from creeks in undisturbed catchments (Baldy Creek, Fisher Creek, 67–114 pmol/min/mg protein).  相似文献

Summary The paper deals, in the light of some examples, with the adoption of petrophysics in the exploration for sulfide ores in Finland. No attempt is made to clarify the practicability of all the known petrophysical properties but to visualize the meaning of the results gained through routine determinations of some properties in practical exploration for sulfide ores. The petrophysical properties examined are the density, the susceptibility, the remanent magnetism and the resistivity. Petrophysical laboratory tests have been carried out on boulders, exposure samples, drill cores and geochemical samples.By routine petrophysical investigations it is possible to obtain additional information which can be utilized in the various stages of the geophysical investigations. The petrophysical investigations b by which the correlation between the geological information and physical properties can be clarified, form an important link between geology and geophysics in the exploration.  相似文献

The aquatic effects of forestry practices and peat extraction continue to cause serious concerns. The effect mechanisms of peat extraction on water quality and aquatic ecology of the receiving surface waters are well known, but the impacts are often difficult to differentiate from those of forest management. A pairwise temporal sediment study was conducted on two adjacent lakes in Central Finland to study whether the unique effects of peat extraction can be detected in an area of intensive forest drainage. Both lakes are affected by forestry, but the reference lake has no history of peat extraction in its watershed. The deepest parts of the lakes were cored through the lacustrine sediments, and the recent carbon and dry matter sedimentation rates were compared to their site-specific reference values. Recent changes in benthic macroinvertebrates (chironomids) and diatom algae were studied to assess the ecological effects of these practices in the lakes. No significant differences in recent increases in carbon accumulation were found between the peat extraction-impacted lake and the reference lake. The pairwise comparison allowed identification of a regional pattern of impacts that is closely related to the history of land use, particularly forestry, in the region. The approach also allowed identification of the transient signs of peat extraction in the chemical and chironomid records of the impacted lake. The recent changes in chironomids and diatoms suggest eutrophication and deterioration in benthic conditions likely caused by drainage ditch network maintenance activities in the catchments.  相似文献

Sandy clinothems are of interest as hydrocarbon reservoirs but there is no proven, economic, clinothem reservoir in the Norwegian Barents Sea. We used high-resolution, 2D and 3D seismic, including proprietary data, to identify a previously untested, Barremian, clinoform wedge in the Fingerdjupet Subbasin (FSB). Data from recent well 7322/7-1 plus seismic have been used to characterize this wedge and older Lower Cretaceous clinoforms in the FSB. In the latest Hauterivian – early Barremian, during post-rift tectonic quiescence, shelf-edge clinoforms (foreset height > 150 m) prograded into an under-filled basin. Increased sediment input was related to regional uplift of the hinterland (northern Barents Shelf). Early Barremian erosion in the north-western FSB and mass wasting towards the SE were followed by deposition of delta-scale (<80 m high), high-angle (c. 8°) clinoform sets seaward of older shelf-edge clinoforms. This may be the local expression of a regional, early Barremian, regressive event. By the close of the Barremian, clinoforms had prograded, within a narrow, elongate basin, across the FSB and towards the uplifted Loppa High. A seismic wedge of high-angle (10–12°), low-relief, delta-scale (25–80 m) clinoform sets occurs between shelf-edge clinoforms to the NW and the uplifted area to the SE. Well 7322/7-1, positioned on a direct hydrocarbon indicator, <1 km NNW of the high-angle, low-relief, delta-scale clinoforms, found upward coarsening siltstone-cycles linked to relative sea-level fluctuations on a marine shelf. Sand may have accumulated, offshore from the well, in high-angle, low-relief foresets of the delta-scale clinothems (which are typical geometries elsewhere interpreted as ‘delta-scale, sand-prone subaqueous clinoforms’). Deposition was controlled by the paleosurface, storms and longshore currents on an otherwise mud-dominated shelf. The study highlights challenges associated with exploration for sandstone reservoirs in seismic wedges on an outer shelf.  相似文献
