Biomarkers of organochlorine exposure, such as the induction of cytochrome P450 1A (CYP1A), can be used to assess the impact of environmental contaminants on the health of free-ranging marine mammal populations. The objective of the present study was to measure CYP1A in skin and liver biopsies obtained from live harbour seals (Phoca vitulina). Twelve harbour seal pups, aged three to five weeks, were captured from the Fraser River estuary, British Columbia, Canada, and temporarily held in captivity. Skin ( approximately 60 mg) and liver ( approximately 40 mg) biopsies, obtained while seals were under general anaesthesia, yielded sufficient tissue for the measurement of CYP1A by immunoblot analysis and ethoxyresorufin O-deethylase activity. A short-term exposure experiment, in which harbour seals (n=3) were treated orally with beta-naphthoflavone (BNF), resulted in increased hepatic and cutaneous CYP1A protein levels, consistent with observations in other mammals. This study is the first to measure CYP1A in skin and liver biopsies from live harbour seals and to report in vivo BNF-associated CYP1A induction in a marine mammal. The results demonstrate that microsamples collected using minimally-invasive techniques can provide toxicologically-relevant information form marine mammals. 相似文献

Blubber of harbour seals (Phoca vitulina) sampled from the Strait of Georgia, BC, in 1991 and 1992 contained higher concentrations of polychlorinated dibenzo-p-dioxins and furans (PCDD/F) than did samples from Quatsino Sound on western Vancouver Island. The higher proportions of 1,2,3,6,7,8-hexachlorodibenzodioxin and 2,3,7,8-tetrachlorodibenzofuran in the Strait of Georgia samples probably reflect contamination arising from the discharge of effluents from bleached kraft mills. Higher concentrations of non-ortho- and mono-ortho substituted polychlorinated biphenyls were also found in the Strait of Georgia samples, probably as a result of general industrial activity in the region. Principal components analysis showed that residues were distributed differently between males and females. Within males, concentrations of PCDD, PCDF and PCBs appeared to increase with age. Toxic equivalents (TEQ) to 2,3,7,8-tetrachlorodibenzodioxin in the Quatsino Sd. samples were close to the working guidelines for edible fish used by Health Canada; TEQ calculated for the Strait of Georgia samples exceeded these guidelines by up to a factor of five. 相似文献

The identification of toxicants affecting aquatic benthic systems is critical to sound assessment and management of our nation's waterways. Identification of toxicants can be useful in designing effective sediment remediation plans and reasonable options for sediment disposal. Knowledge of which contaminants affect benthic systems allows managers to link pollution to specific dischargers and prevent further release of toxicant(s). In addition, identification of major causes of toxicity in sediments may guide programs such as those developing environmental sediment guidelines and registering pesticides, while knowledge of the causes of toxicity which drive ecological changes such as shifts in benthic community structure would be useful in performing ecological risk assessments. To this end, the US Environmental Protection Agency has developed tools (toxicity identification and evaluation (TIE) methods) that allow investigators to characterize and identify chemicals causing acute toxicity in sediments and dredged materials. To date, most sediment TIEs have been performed on interstitial waters. Preliminary evidence from the use of interstitial water TIEs reveals certain patterns in causes of sediment toxicity. First, among all sediments tested, there is no one predominant cause of toxicity; metals, organics, and ammonia play approximately equal roles in causing toxicity. Second, within a single sediment there are multiple causes of toxicity detected; not just one chemical class is active. Third, the role of ammonia is very prominent in these interstitial waters. Finally, if sediments are divided into marine or freshwater, TIEs perforMed on interstitial waters from freshwater sediments indicate a variety of toxicants in fairly equal proportions, while TIEs performed on interstitial waters from marine sediments have identified only ammonia and organics as toxicants, with metals playing a minor role. Preliminary evidence from whole sediment TIEs indicates that organic compounds play a major role in the toxicity of marine sediments, with almost no evidence for either metal or ammonia toxicity. However, interpretation of these results may be skewed because only a small number of interstitial water (n = 13) and whole sediment (n = 5) TIEs have been completed. Using all available major samples of Seyfert galaxies and their corresponding closely matched control samples of nonactive galaxies, we find that the bar ellipticities (or axial ratios) in Seyfert galaxies are systematically different from those in nonactive galaxies. Overall, there is a deficiency of bars with large ellipticities (i.e., "thin" or "strong" bars) in Seyfert galaxies compared to nonactive galaxies. Accompanied with a large dispersion due to small number statistics, this effect is strictly speaking at the 2 sigma level. To obtain this result, the active galaxy samples of near-infrared surface photometry were matched to those of normal galaxies in type, host galaxy ellipticity, absolute magnitude, and, to some extent, redshift. We discuss possible theoretical explanations of this phenomenon within the framework of galactic evolution, and, in particular, of radial gas redistribution in barred galaxies. Our conclusions provide further evidence that Seyfert hosts differ systematically from their nonactive counterparts on scales of a few kiloparsecs. 相似文献

Reconstruction of contaminant trends in a salt wedge estuary with sediment cores dated using a multiple proxy approach 总被引:2,自引:0,他引:2
The Taunton River is a partially mixed tidal estuary in southeastern Massachusetts (USA) which has received significant contaminant inputs, yet little information exists on the history of discharge and the subsequent fate of these contaminants. Three sediment cores taken along a transect were analyzed, reconstructing the spatial and temporal trends of pollution in the estuary. A combination of radiometric dating, contaminant markers, and storm layers from major hurricanes were used to establish age models and sedimentation rates. Age estimates obtained from the different dating methods compared well, establishing an accurate history of contaminant release to the estuary. Polycyclic aromatic hydrocarbons (PAHs) were present in one core at depths corresponding to the early 1860s, earlier than previously established dates of introduction. Temporal and spatial trends of Cr, Cu, Hg and Pb indicated multiple sources of varying input to the river. Polychlorinated biphenyls (PCBs) were present in each of the cores from the 1930s onward, with elevated levels still present in surficial sediments at several sites. A unique organic compound, Topanol, which was produced locally was used as a tracer to track contaminant transport in the river. Tracer data indicates that contaminants are still being transported and deposited to surficial sediments at high concentrations well after their discharge. This reconstruction demonstrates the utility of using multiple dating proxies where often the sole use of radiometric dating techniques is not an option and provides insights into the fate of contaminants discharged decades ago but continue to represent environmental risks. 相似文献

Natural and anthropogenic hydrocarbons in sediments from the Chubut River (Patagonia, Argentina) 总被引:9,自引:0,他引:9
In March of 2001 a study was carried out to evaluate hydrocarbon levels in the lower course of the Chubut River. The study included 12 sample stations along the river from San Cristóbal Bridge to the confluence with the sea, in a 25 km straight-line extension in a urbanized area. In the first 11 stations, resolved aliphatic (RAli) hydrocarbons presented low values, between 0.07 and 0.96 microg/g dry weight (dw); the unresolved complex mixture (UCM) between 0.42 and 2.72 microg/g dw, and the total aliphatic (TAli) hydrocarbons between 0.55 and 3.07 microg/g dw. In the last station, at the mouth of Chubut river, these values increased to 460, 284, and 741 microg/g dw for RAli, UCM and TAli, respectively. The n-alkanes distribution indices and the compositional parameters suggested a predominantly biogenic origin in eleven stations, and a predominantly anthropogenic origin in the last station, with the highest hydrocarbon values. It is possible to conclude that the stations with low hydrocarbon values and biogenic origin predominance would constitute the baseline of aliphatic hydrocarbons for river sediments at this zone. The station with the highest hydrocarbon concentration and predominantly anthropic origin was related to the presence of Rawson city's port, where its activities (harbor and fishing vessels) generate hydrocarbon wastes unrelated to the river base profile in the study zone. Offshore, but within the river influence, there is an important fishing area of Argentine Red and Patagonian shrimps (Pleoticus muelleri and Artemesia longinaris, respectively). 相似文献
