南极硅藻GJ01(Berkeleya rutilans)产谷胱甘肽条件的优化

摘    要:以南极硅藻GJ01(Berkeleya rutilans)为对象,分别采用紫外、可见和荧光分光光度法,研究了营养条件等因子对GJ01生长和胞内谷胱甘肽含量的影响,并对培养的GJ01细胞合成谷胱甘肽的条件进行了优化。结果表明,在单因子影响条件下,1.2g/L的碳酸氢钠对南极硅藻GJ01的生长最好,且最有利于谷胱甘肽的积累;0.8mmol/L氯化钙对GJ01的生长最为有利,而0.2mmol/L氯化钙对谷胱甘肽的产生是适宜的;维生素B_(12)和生物素对GJ01胞内谷胱甘肽的产生影响较小。正交试验表明,GJ01细胞产生谷胱甘肽的最优条件是:光照周期为16h/8h、NaHCO_3为1.6g/L、Ca~(2 )为0.4mmol/L、Cys为10mmol/L。该结果为利用南极硅藻GJ01生产谷胱甘肽提供了参考和依据。

Abstract:GSH,as a key component of Asc-glutathione cycle,plays important roles in protecting tissues from toxicant damage and keeping cells in normal redox state,on which this paper studied.Berkeleya rutilans GJ01,one of so-called Antarctic ice microalgae,an extreme microorganism in the cryosphere,is one of major producers of primary production in marine ecology system of Antarctic polar region.They have become a promising resource of GSH.In this paper,effect of nutrition factor on GSH production level was studied for G J01 with spectrophotometer in searching for the optimization of synthesis condition for GSH.It was showed that 1.2g/L NaHCO_3 and 0.2mmol/L Ca~(2 ) is ideal to the production of GSH. Vitamin B_(12) and biotin impacted slightly on GSH production.Orthogonal test revealed that the optimal condition for GSH production of GJ01 is 16h light/8h dark,1.6g/L NaHCO_3,0.4mmol/L Ca~(2 ),and 10mmol/L Cys.The results should be in- formative to the application of GJ01 for GSH production.
