Cyanobacterial blooms in eutrophic freshwater systems are a worldwide problem, creating adverse effects for many aquatic organisms by producing toxic microcystins and deteriorating water quality. In this study, microcystins (MCs) in Microcystis aeruginosa, and Daphnia magna exposed to M. aeruginosa, were analyzed by HPLC-MS, and the effects of M. aeruginosa on D. magna were investigated. When D. magna was exposed to M. aeruginosa for more than 2 h, Microcystin-LR (MC-LR) was detected. When exposed to 1.5 × 106, 3 × 106, 0.75 × 107, and 1.5 × 107 cell/mL of M. aeruginosa for 96 h, average survival of D. magna for treatments were 23.33%, 33.33%, 13.33%, 16.67%, respectively, which were significantly lower than the average 100% survival in the control group (P < 0.05). The adverse effects of M. aeruginosa on body length, time for the first brood, brood numbers, gross fecundity, lifespan, and population growth of D. magna were density-dependent. These results suggest that the occurrence of M. aeruginosa blooms could strongly inhibit the population growth of D. magna through depression of survival, individual growth and gross fecundity. In the most serious situations, M. aeruginosa blooms could undermine the food web by eliminating filter-feeding zooplankton, which would destroy the ecological balance of aquaculture water bodies.  相似文献

The interspecific interactions between the rotifer Brachionus plicatilis and two harmful algal blooms (HAB) species were investigated experimentally by single culture method. B. plicatilis population and the growth of the two algae were compared at different algal cell densities. The results demonstrated that the B. plicatilis obtained sufficient nutrition from Prorocentrum donghaiense to support net population increase. With exposure to 2.5×104 cells mL−1 of P. donghaiense, the number of B. plicatilis increased faster than it did when exposed to other four algal densities (5, 10, 15 and 20 ×104 cells mL−1), and the increase rate of B. plicatilis population (r) at this algal density was 0.104 ± 0.015 rd−1. Cell densities of P. donghaiense decreased due to the grazing of B. plicatilis. In contrast, Heterosigma akashiwo had an adverse effect on B. plicatilis population and its growth was largely unaffected by rotifer grazing. In this case, B. plicatilis population decreased and H. akashiwo grew at a rate similar to that of the control.  相似文献

From Oct., 1999 to Oct., 2000, the heterotrophic bacterial flora in the aquaculture area around Xuejiadao was investigated. The result shows that the populations of the heterotrophic bacteria are heavier in summer and autumn than those in winter and spring. The average populations in seawater, sediment, the surface of seaweed and the surface of fish are 1.4×104cfu mL−1, 5.4×106cfu g−1, 1.5×106cfu g−1 and 1.8×103cfu cm−2, respectively. A total of 301 strains were isolated, among them 259 were Gram-negative. All the Gram-negative bacteria belong to 13 genera and some genera of Enterobacteriaceae. The communities of bacteria are slightly different among the samples. In the body surface of fish, Genus vibrio is dominant. In the remaining samples, dominant genus is Aeromonas.  相似文献

From Oct. 1999 to Oct. 2000, the heterotrophic bacterial floras in the industrial marine environment around the Qingdao Power Plant (QPP) and in the unpolluted marine environments were investigated. The results showed that the numbers of the heterotrophic bacteria around QPP were much higher than those in unpolluted environments, and the average numbers in QPP Seawater, QPP Sediment, Unpolluted Seawater and Unpolluted Sediment were 5.4×104cfu(mL)−1, 5.0×105cfug−1, 3.0×102cfu(mL)−1 and 1.3×105cfug−1 respectively. Totally, 118 strains were isolated from QPP and 99 of them were Gram-negative. One hundred and twenty one strains were isolated from the unpolluted environments and 104 of them were Gram-negative. All the Gram-negative bacteria belonged to 13 genera. The distribution of the bacteria was varied in different marine environments. The results showed that the unpolluted marine environments contained much more Vibrio than seawater and sediment around QPP.  相似文献

Sodium thiosulphate and neutron activation determination of iodine content in samples of 18Sargassum species collected in Guangdong and Guangxi Province in April 1996 showed large differences (0.47×10−3 inSargassum hemiphyllum, 0.56×10−3 inSargassum assimile, and 4.5×10−3 inSargassum vachellianum). Traditional titration analysis and neutron activation analysis for determination of total iodine yielded similar results showing that certainSargassum species had high capacity to accumulate iodine. A bloom of the dinoflagellate Scrippsiella trochoidea was detected for the first time in inner Tolo Harbor, Hong Kong in 2 000. Water samples were collected at eight stations along a transect passing through a red tide patch for microscopic analysis of phytoplankton composition and high-performance liquid chromatography (HPLC) analysis of phytoplankton pigments. During the bloom, the density of dinoflagellates was 1.1×106 cells L−1 within the patch and 8.6×105 cells L−1 outside the patch where the phytoplankton community was dominated by diatoms. After the bloom the S. trochoidea began to decrease in density and was replaced by diatoms as the dominating bloom-causing organisms at all stations, and the density of dinoflagellates at most stations was less than 1.0 × 106 cells L−1. The status of S. trochoidea as the causative species of the bloom was indicated by the presence of peridinin, the marker pigment for dinoflagellates. The shift from dinoflagellates to diatoms was marked by the decline of peridinin and the prevalence of fucoxanthin. Phytoplankton pigment markers also revealed the presence of other minor phytoplankton assemblages such as cryptomonads and blue-green algal.  相似文献

The SCENTO-System was used to study the carbon dynamics between phytoplankton primary production and heterotrophic bacterial secondary production. Most of the methods used nowadays in situ for limnological synecology studies were applied. Primary production measurement showed an increasing tendency with increasing content of chlorophylla. It provided a true photosynthetic rate lying within the range of eutrophic lakes. Net EOC released from the algae ranged from 8.5 to 27.5 μg C l−1(6h)−1. Accompanying the algal products the number of bacteria increased from 1.475 ×109 to 8.074×109 cells l−1. The bacterial mean cell volume was small, between 0.0315 and 0.0548μm3. Bacterial carbon production from direct growth estimates was compared with independent calculations of bacterial growth from EOC uptake and3H-thymidine incorporation. Direct estimates were 2.97–10.0 μg Cl−1 (24h)−1 with the exception of a zero-growth on the third day. EOC uptake was 123.5–191.0 μg Cl−1 (6h)−1. That calculated from3H-thymidine incorporation was 0.2–0.5 μg Cl−1 (6h)−1.14C-glucose dark uptake ran parallel to the increasing bacterial biomass. The respiration of glucose was 6.5% (avg.) of the gross uptake. Since the system operated without grazing pressure, a real carbon flow from primary production to bacterial secondary production could be observed.  相似文献

The abundance and biomass of benthic heterotrophic bacteria were investigated for the 4 typical sampling stations in the northern muddy part of Jiaozhou Bay, estuary of the Dagu River, raft culturing and nearby areas of Huangdao in March, June, August and December, 2002. The abundance and biomass range from 0.98×107 to 16.87×107 cells g−1 sediment and 0.45 to 7.08 μg C g−1 sediment, respectively. Correlation analysis showed that heterotrophic bacterial abundance and biomass are significantly correlated to water temperature (R=0.79 and 0.83, respectively,P<0.01).  相似文献

When cultured under certain environmental conditions (25°C, light intensity 80 μmol/m2·s, LD 12/12, in enriched seawater medium with 7×10−4 mol/L NO3−N, 1.56×10−4 mol/L, PO4−P and supplements of other elements like Mn, Fe, I, etc.), male and female gametophytes ofU. pinnatifida kept growing vegetatively and propagated fast at average daily fresh weight increase rate of about 20%. The empirical formulaG m=G o·3m was established to estimate the output of vegetative gametophytes. Vigorous vegetative gametophyte cells began to form reproductive structures (oogonium and spermatangium, when the temperature was lower than 25°C and other environmental factors were kept optimal. The sufficient supply of gametophyte cells provided enough seeds for raisingUndaria sporelings on production scale. Controlled cross-breeding experiments using selected male and female gametophyte clones which increase their cell number by mitosis instead of meiosis were also carried out in vitro. Juvenile sporophytes from the cross-breeding had almost the same length and width increase rates as those of the control. The fact that vegetative gametophytes can be purposely selected, propagated quickly, cross-bred, with the resulting sporophytes having almost the same characteristics leads to a new way to select desiredUndaria strains for long time use without losing the desired economic characteristics. Acute toxicities of potassium permanganate, formalin, and Lugol’s iodine solution to a commonly occurred marine ciliate Pleuronema coronatum (Ciliophora, Scuticociliatida) were measured. Linear regression analysis of the results highlighted the close relationships between doses of the medicines and mortalities of the organisms, thus providing a capability to predict toxicity effects from the dose. Toxic effects of the medicines on the ciliates were described in the present paper, and the median lethal concentrations (LC50 values) were given. Results of measurements indicated that 2 h-LC50 and 12 h-LC50 values of formalin on P. coronatum were 59.00×10−6 and 43.57×10−6, while those of Lugol’s solutions were 90.13 and 67.84×10−6 respectively. The tolerance of P. coronatum to formalin is apparently lower than that to Lugol’s iodine solution and potassium permanganate is a suitable medicine to kill ciliates in short time.  相似文献

The ecological processes and interrelationships between protists, either autotrophic or heterotrophic, and environmental factors in mariculture ponds are largely unknown. This study investigated the temporal dynamics of potentially harmful dinoflagellate, Prorocentrum minimum (Pavillard) Schiller, and its relationship to physico-chemical factors and protozoan grazers over a complete cycle in a semi-enclosed shrimp-farming pond near Qingdao, Northern China. P. minimum occurred frequently in low numbers from June to August, followed by a sharp increase from the middle of August, reaching a single maximum peak value of 2.2×105 cells L−1 in October. Temporal variation in abundance was positively correlated with dissolved nitrogen, but showed a significant inverse relationship to abundance of the dominant ciliates, Tintinnopsis lohmanni and Askenasia stellaris. The results provide statistical evidence that the number of P. minimum increased with increasing nitrogen, and the suppression or shortening of algal bloom may be associated with protozoan grazers, such as Tintinnopsis lohmanni, in mariculture ponds.  相似文献

A new kinetic spectrophotometric method has been developed for the determination of vanadium (V). The method is based on the catalytic effect of vanadium (V) on the oxidation of weak acid brilliant blue dye (RAWL) by KBrO3 using the citric acid as activation reagent. The obtained optimum conditions are: c (RAWL) = 1×10−4 molL−1, c (KBrO3) = 3×10−2 molL−1, c (citric acid) = 9×10−3 molL−1, pH = 2.50, the reaction time being 7.0 min and the temperature being 25.0°C. Under the optimum conditions, the proposed method allows the determination of vanadium (V) in the range of 0–70.0 ng mL−1 and the detection limit is down to 0.407 ng mL−1. For standard vanadium (V) solution determination, the recovery efficiency is in the range of 98.5%–102% and the RSD ranges from 0.76%–1.25%. Moreover, it is demonstrated that most cations and anions do not interfere with the determination of vanadium (V) under the analytical condition. The new method was successfully applied in the determination of vanadium (V) in fresh water and seawater samples with satisfactory results. Vanadium (V) in the seawater samples from Qingdao offshore was determined using the method and the distribution of vanadium (V) was mapped. Compared with other instrumental analytical methods, the proposed method shows good selectivity, sensitivity, simplicity, lower cost and rapidity. It can be employed on shipboard easily.  相似文献

A new kinetic spectrophotometric method has been developed for the determination of trace Ni (II) in natural water.The method is based on the catalytic effect of Ni (II) on the oxidation of weak acid brilliant blue dye (RAWL) by KIO4 in acid medium.The concentration of nickel (II) can be determined spectrophotometrically by measuring the decrease of absorbance of RAWL at λ= 626 nm using the fix-time method.The influencing factors are investigated by the orthogonal experimental design.The obtained optimum analytical conditions are:pH=2.00,c RAWL=5.00×10-5 mol L-1,cKIO 4= 2.00×10-5 mol L-1,the reaction time t=10 min and the temperature T=25℃..Under the optimum conditions,the developed method allows the measurement of Ni (II) in a range of 0-40.0 ng mL-1.The standard deviation of eleven independent measurements of blank reaction is S=3.08×10-3 and the limit of detection is 2.20 ng mL-1.The relative standard deviations (RSDs) in six replicate determinations of 5 ng mL-1 and 8 ng mL-1 Ni (II) are 2.87% and 1.11%,respectively.Moreover,the experiments show few cations and anions can interfere with the measurement of Ni (II).The recovery efficiencies of this method are in a range of 97.0%-102.5% in freshwater samples.But there is a decreasing effect,which is about 0.2 times the added Ni (II) in seawater medium.After reasonable calibration this processing method is used for the determination of Ni (II) in seawater samples successfully.The results show this developed method has high accuracy and precision,high sensitivity,large range of linearity and high speed.The method can,therefore,be employed at room temperature.  相似文献

Ten actinomycete strains isolated from the Yellow Sea off China's coasts were identified as belonging to two genera by 16 S r DNA phylogenetic analysis: Streptomyces and Nocardiopsis. Six Streptomyces strains(MA10, 2SHXF01-3, MA35, MA05-2, MA05-2-1 and MA08-1) and one Nocardiopsis strain(MA03) were predicted to have the potential to produce aromatic polyketides based on the analysis of the KSα(ketoacyl-synthase) gene in the type II PKS(polyketides synthase) gene cluster. Four strains(MA03, MA01, MA10 and MA05-2) exhibited significant inhibitory effects on mycelia growth(inhibition rate 50%) and subsequent aflatoxin production(inhibition rate 75%) of the mutant aflatoxigenic Aspergillus parasiticus NFRI-95. The ethyl acetate extracts of the broth of these four strains displayed significant inhibitory effects on mycelia growth, and the IC50 values were calculated(MA03: 0.275 mg m L-1, MA01: 0.106 mg m L-1, MA10: 1.345 mg m L-1 and MA05-2: 1.362 mg m L-1). Five strains(2SHXF01-3, MA03, MA05-2, MA01 and MA08-1) were selected based on their high cytotoxic activities. The ethyl acetate extract of the Nocardiopsis strain MA03 was particularly noted for its high antitumor activity against human carcinomas of the cervix(He La), lung(A549), kidney(Caki-1) and liver(Hep G2)(IC50: 2.890, 1.981, 3.032 and 2.603 μg m L-1, respectively). The extract also remarkably inhibited colony formation of He La cells at an extremely low concentration(0.5 μg m L-1). This study highlights that marine-derived actinomycetes are a huge resource of compounds for the biological control of aflatoxin contamination and the development of novel drugs for human carcinomas.  相似文献

This study deals with contribution of artificial food pellet and natural food to Chinese prawn (Penaeus orientalis) growth in a semiintensive culture pond. The prawn carbon consumption, budget, and the effects of some factors on the budget were investigated. The results showed that 26.2% of P. orientalis growth carbon came from formulated feed at the initial culture stage (when the prawns were 0.06±0.01 g in wet weight), and was 62.5% when the prawns were 9.56±1.04 g. The remaining part of the growth carbon was derived from organic fertilizer and natural food. The highest growth rate occurred at 20×10-3 salinity. Suitable salinity for culturing Chinese prawn was (20-28)×10-3.  相似文献

Hyper-spectrum models for monitoring water quality in Dianshan Lake,China   总被引:1,自引:0,他引:1  
The correlation between water quality parameters and hyper-spectral reflectance is studied with models established for each parameter and applied in Dianshan Lake, in the upstream of the Huangpu River running through Shanghai, China. Models are for dissolved oxygen (DO in mg/L): R720/R680 = 20.362×(R720/R680)2-31.438×(R720/R680)+19.156; for turbidity (NTU): R*714.5 = 206.07× (R*714.5)2-582.5×R*714.5 + 423.24; and for total phosphorus (TP in mg/L): R*509.5 = 16.661× (R*509.5)2-32.646×R*509.5+16.116. The R2 values are 0.770 8, 0.660 4 and 0.738 7, respectively, showing strong positive relationships. The models were then applied to assess water quality of different times. Results are quite satisfactory for some samples.  相似文献

Batch culture experiments were conducted with a red tide dinoflagellateScrippsiella trochoidea (Stein) Loeblch III collected from Jiaozhou Bay, Shangdong, China. Growth rates and oellular Chl—a were measured in media with iron and manganese ion concentrations controlled at different levels using EdTA-trace metal buffer systems. Cell density increased 3.2 times to 6.5 times over the range of lowest (0 mol/L) to highest (10−5 mol/L) iron and manganese ion concentrations. The range of cell density response was much lower than the range of total available iron and manganese, which was >100—fold that of Fe. This nonlinear relationship indicates that the cells adapt to make more efficient use of iron and manganese under limiting conditions. The cellular Chl—a content maximized after 3 days incubation and then decreased gradually under either iron or manganese limitation conditions. It indicated that the algae gained higher photosynthesis ability when transferred to a new environment. Growth responses to iron and manganese limitation can be both modeled according to the equation of Monod. The half—saturation constant for growth,k, is 4.6×10−8 mol/L for Fe and 5.1×10−8 mol/L for Mn. Our results showed that the iron availability in Jiaozhou Bay does not limit the growth ofS. trochoidea.
