A field study was carried out in the Lagoon of Venice (north-east Italy) with the aim of evaluating the potential use of lysosomal destabilisation as a biomarker of anthropogenic stress in the autochthonous mussel Mytilus galloprovincialis. Two different approaches were adopted in biomonitoring six sites in the Lagoon, evaluating indigenous populations of mussels and organisms transplanted from a reference site and checked at several points in time. Lysosomal membrane stability was investigated by means of two tests: neutral red retention assay (NRRA) for evaluating haemocyte lysosomes and lysosomal latency test (LLT) for digestive cell lysosomes. Results indicate that the lysosomal response measured in haemocytes according to NRRA is a more valuable biomarker of anthropogenic stress in the framework both of passive and active biomonitoring in marine coastal environments.  相似文献

The use of lysosomal stability in the mussel, Mytilus galloprovincialis, as a potential biomarker of environmental contamination has been evaluated along the Portuguese coast. To this end, the neutral red retention (NRR) time was measured in mussel haemocytes gathered from nine different locations reflecting different degrees of anthropogenic contamination. Mussels collected in the vicinity of industrial and urban areas showed the lowest lysosomal stability. Additionally, no significant seasonal variability (winter-spring/summer) for NRR time was observed. In order to further support the usefulness of this method as an integrated tool for monitoring marine coastal environments, we compared the levels of xenobiotics in mussel tissues with the obtained NRR values. The results highlighted a consistent pattern, with the lowest lysosomal stability intimately correlated with the higher contaminant concentrations. In summary, this integrated approach further demonstrated that the NRR assay can provide useful and objective indications of the real health status of organisms subjected to different stress agents, being a valid option for environmental monitoring.  相似文献

A high temperature catalytic oxidation (HTCO) technique was used to measure dissolved organic carbon (DOC) during seasonal surveys of the Tamar Estuary, U.K. At the time of the programme, the field of DOC analysis had been plagued by numerous analytical difficulties. However, using thorough calibration of the analytical systems and the systematic analysis of an internal reference material, a valuable estuarine DOC data set was produced. The range of DOC concentrations observed (478–110 μM C) is consistent with the published data for riverine and coastal sea waters respectively. The Tamar Estuary is a freshwater DOC-dominated system, with strong correlation between lateral DOC distribution and salinity. However, mixing behaviour was not strictly conservative. During tidal cycle studies at a fixed station, DOC concentrations appeared to be uncoupled from salinity, and were inversely related to turbidity. It is concluded that tidally-induced resuspension of bottom sediments provided the dominant control mechanism for DOC concentration. The Tamar Estuary shows contrasting behaviour to the larger, more heavily impacted, Severn Estuary. Hence it is likely that the behaviour of DOC in estuaries cannot be classified as typical per se, but is a function of the natural and anthropogenic characteristics of the catchment and hydrology.  相似文献

Many cellular and sub-cellular biomarkers associated with mussel (Mytilus edulis) digestive gland and kidney have been characterised. The lysosomal compartment of these tissues have been recognised as being particularly sensitive, exhibiting pollutant induced responses which could be potentially used as a ‘biomarker’. However, relatively few studies have investigated the lysosomal response within molluscan hemocytes. This study was conducted to test whether lysosomal reactions, in live hemocytes isolated from mussels, can be used as a biomarker of pollutant exposure and deleterious effect. Lysosomal responses to a number of hydrocarbons, including anthracene and phenanthrene, and to the amphiphilic heterocylic chemical, chlorpromazine, were examined. The supravital dye neutral red (NR) was used to examine lysosomal membrane fragility, following xenobiotic exposure. NR was also used to verify the lysosomal compartment as the reported accumulation site of a new molecular probe, BODIPY-FL-verapamil (BFLV). The use of BFLV, with confocal laser microscopy and image analysis enabled visualisation and quantification of lysosomal distribution and perturbation. BFLV showed that exposure of molluscan hemocytes to xenobiotics (20 ppb–10 ppm) induced the formation of pathologically enlarged lysosomes. The internal trafficking of lysosomes was shown to be severely compromised after exposure to chlorpromazine. Exposed molluscan hemocytes exhibited significantly reduced lysosomal retention times, for neutral red. Preliminary data is presented demonstrating the opportunity for these non-destructive biomarker techniques to detect pollution gradients in situ.  相似文献

The potential of eel (Anguilla anguilla) as a monitoring species for the Thames Estuary, UK, was examined. Hepatic cytochrome P4501A [7-ethoxyresorufin O-deethylase (EROD) activity] and blood vitellogenin (Western analysis) were investigated as biomarkers of exposure to, respectively, organic contaminants and to contaminants showing estrogenic activity. Hepatic microsomal EROD activities in A. anguilla from seven sites in the Thames Estuary in May 1998 varied three-fold (111 +/- 24 to 355 +/- 42 pmol min-1 mg protein-1) (mean +/- S.E.M.) and showed correlation with salinity; however, the latter relationship was not maintained at other times of the year. The range of EROD activities was two- to eight-fold higher than the 37 +/- 8 pmol min-1 mg-1 for A. anguilla from the relatively clean Tamar Estuary. beta-Naphthoflavone treatment (5 mg kg-1 wet wt.; 2 days) of Thames A. anguilla produced a two-fold increase in hepatic microsomal EROD activity. Comparing the Thames EROD data with those for A. anguilla from well-characterised contaminated sites in the Netherlands (Van der Oost, R., Goks?yr, A., Celander, M., Heida, H., & Vermeulen, N. P. E. 1996. Aquatic Toxicology, 36, 189-222), the Thames is suggested to be moderately impacted by polycyclic aromatic hydrocarbons and related contaminants. 17-beta-Estradiol treatment produced the appearance of a plasma protein of 211 Kd app. mol. wt. (recognised by antibodies to vitellogenin of Morone saxatilis), but putative vitellogenin could not be detected in A. anguilla from selected sites in the Thames Estuary.  相似文献

This study assessed numerous histological parameters as markers of health status in mussels (Mytilus spp.) collected from several locations along a contamination gradient on the River Tamar and two coastal sites on the north and south coast of Cornwall, UK. Twenty-seven health parameters were assessed including the presence of pathogens, inflammatory lesions, non-specific pathologies and reproductive condition. Logistic regression showed that reproductive condition and inflammatory lesions showed changes indicative of a contamination gradient response. Mussel populations further upstream within the Tamar were relatively less developed compared to mussels situated downstream in the estuary. Compared to coastal locations, mussels sampled within the Tamar also exhibited a higher prevalence of inflammatory lesions that increased at locations situated furthest upstream. Similarly, levels of lipofuscin within the kidney epithelium were highest in mussels sampled within the estuary and Whitsand Bay compared to Trebarwith Strand. The most prevalent pathogens observed during this survey included Rickettsia/Chlamydia-like organisms (R/CLO), gill ciliates including Ancistrum mytili, the copepod Mytilicola intestinalis, Ciliophora-like organisms and Marteilia sp. Marteilia sp. was observed in 31% of the mussels sampled from the Cremyll Ferry site. Generally, the range and prevalence of pathogens was highest from the two estuarine locations at Cremyll Ferry and Wilcove. No significant differences were observed between estuarine sampling sites and Whitsand Bay with respect to median epithelial cell height of digestive tubules, however mussels from Trebarwith Strand did exhibit a reduced median epithelial cell height compared to mussels sampled from other sampling sites. Species genotyping was also undertaken during this study in order to consider any histological differences in relation to species. Results are discussed in relation to histopathology health parameters and biological effects monitoring.  相似文献

Biochemical (aldehyde dehydrogenase and catalase activities), cellular (neutral red retention time) and physiological (survival in air and condition index) biomarkers were determined in mussels collected in four differently impacted lagoon areas. Multivariate analysis showed that samples were distributed mainly according to temperature and four seasonal groups were identified. The combination of spatial and temporal information enabled us to distinguish physiological variations due to natural causes from those due to anthropic stress, and to identify the sampling period when several biomarkers are less influenced by both natural and endogenous factors.  相似文献

To evaluate the impact of land-based contaminants on benthic faunas, as part of the Land-Ocean Contamination Study (LOCS), a study transect was established in Jakarta Bay, Indonesia during the 1996 southeasterly monsoon. The transect extended 72 km between the city's main port, Tanjung Priok, and the Pilau Seribu Island chain in the Java Sea. The dissolved concentrations of Pb, Cu, Zn, Cr and Ni in seawater, in suspended particulate matter (SPM) and surficial sea bed sediments were measured along the transect at nine sites. In addition, metal concentrations were measured in tissues of the corals Goniopora lobata and Lobophyllia hemprichii at five sites, and of the green mussel, Perna viridis at six sites. An assessment of the impact of contaminants on the faunas was made using a biomarker approach, employing coral community analysis and lysosomal stability with Perna viridis. The results of the study showed two main trends. The distributions of metals dissolved in seawater, in SPM, and in the coral and mussel tissues were similar, and failed to show a consistent graded response from inshore to offshore sites. This suggests that the concentration of metals in waters is the primary route for metal uptake by the coral and mussel tissues. By way of contrast, a clear offshore increase in coral generic diversity, coral cover, coral colony numbers and neutral-red retention time (lysosomal stability) was observed, suggesting increasing nearshore stress. Whilst the coral community stucture may reflect the seasonally-averaged metal distributions in the bay (shown by the metal content of sea bed sediments which increase shorewards), it is more likely that the coral community structure and lysosomal biomarker are responding principally to other nearshore stresses, such as sediment and nutrient loading of water (sewage) or, more likely, organic contaminants such as oils and other hydrocarbons.  相似文献

The aim of this study was to investigate the cellular pathological responses of hepatopancreatic digestive cells from the periwinkle Littorina littorea exposed to the polycyclic aromatic hydrocarbon (PAH) fluoranthene and to ascertain whether any injurious effects were reversible within the experimental time scale. A secondary objective was to establish the relationship of the various reactions to animal health status, using lysosomal stability as an index of well-being. Exposure of snails to a concentration of 335 microgl(-1) (1.7 microM) fluoranthene (seawater renewed and spiked daily with fluoranthene) for 5 days resulted in a reduction in lysosomal stability (neutral red retention) and endocytosis; and an increase in smooth endoplasmic reticulum (ER) and 7-ethoxycoumarin-o-deethylase (ECOD; measured as cyano-ECOD) activity measured in isolated live digestive cells. Exposed snails treated with clean seawater for a further 8 days resulted in a return to control levels of lysosomal stability, ECOD and ER; endocytosis showed only a partial recovery. Multi-variate and uni-variate analysis showed that there were strong correlations between the various cellular biomarker responses. These findings are interpretable within the current framework of molluscan biomarker responses to PAHs. Principal component analysis was used to derive the first principal component for endocytosis, ER and ECOD reactions and these were plotted against lysosomal stability as a measure of cellular well-being. The resulting significant regression represents the mapping of the individual biomarkers within health status space for a gradient of fluoranthene toxicity. From this analysis, we concluded that endocytosis is an indicator of healthy snails while proliferation of ER and to a lesser extent induced ECOD are indicative of dysfunction and reduced health. Finally, the results indicate that stress induced by chronic exposure to a PAH is reversible.  相似文献

为了探讨甬江口海洋倾倒区附近海域表层沉积物中多氯联苯的污染水平及生态环境影响,对2014年1月在甬江口倾倒区及附近海域采集的8个沉积物样品用气相色谱法测定PCBs残留量,并且进行环境生态风险评价。结果显示:甬江口海洋倾倒区表层沉积物中PCBs含量为1.310~6.538 μg·kg-1,平均值为3.413 μg·kg-1,其分布表现为西北和东南向PCBs含量高于倾倒区;10种指示性PCBs中,主要以低氯代的PCB28、PCB118和PCB155为主,其中PCB118和PCB155在表层沉积物中的含量相对较高,其含量在各采样站位中占PCBs总量的26.1%~93.1%。甬江口海洋倾倒区沉积物中PCBs污染水平低于长江口、闽江口及珠江口,高于鸭绿江口、辽河口及黄河口。采用潜在生态危害指数法、加拿大环境质量标准ISQG法、生态风险值(EPA)法及毒性当量因子法进行评价,结果基本一致:甬江口海洋倾倒区属于较低生态风险,一般不会引起生物的负效应。  相似文献

Lysosomal destabilization assays have been used as valuable biomarkers of pollutant exposures in a variety of bivalve and fish species. The responses of oysters, Crassostrea virginica, deployed at and native to various reference and degraded sites were evaluated for lysosomal destabilization during both summer and winter seasons. In both native and deployed oysters, lysosomal destabilization rates tended to be higher during the winter at both reference and polluted sites. There are at least two hypothetical explanations. Greater lysosomal destabilization rates may be related to physiological changes associated with mobilization of nutrient reserves during the winter and gametogenesis. However, lysosomal destabilization in deployed oysters was correlated with tissue metal concentrations. These data also support a second hypothesis that seasonal differences in physico-chemical factors (such as reduced levels of acid volatile sulfides) may increase the bioavailability of metals during the winter so that adverse effects are more pronounced.  相似文献

Expanding industrial activities in the Arctic require an urgent assessment of the toxicity of chemicals at low temperatures. Organisms acclimatized to low temperature exhibit specific adaptations. For example, the amount of unsaturated lipids is increased to maintain the fluidity of the cell membranes. It has been hypothesized that such temperature-induced alterations in membrane lipid composition may affect the stability of lysosomal and cell membranes in the common mussel, Mytilus edulis, an organism exposed to seasonal temperature extremes. As mussels may be exposed to petroleum compounds along industrialized coastlines, we tested the combined effects of exposure to low temperature and the petroleum compound, phenanthrene, on haemocyte membrane stability. Test animals, acclimated to either 0 or 10 degrees C, were exposed to phenanthrene (0 = control or 500 micrograms l-1) and haemocytes were examined using the neutral red retention assay (lysosomal stability) and a fluorescence assay (cell membrane stability). At 0 degree C, lysosomal and cell membranes from uncontaminated mussels were destabilized compared with 10 degrees C (P = 0.0005). No significant effects (P > 0.05) of phenanthrene were detected at either temperature. Possible mechanisms underlying membrane destabilization include a weaker physical resistance of the membrane due to a higher amount of unsaturated lipids, a potentially higher level of reactive oxygen radicals at low temperature and the higher susceptibility of unsaturated lipids to oxidative stress. More work is required to better understand the consequences of this membrane destabilization at low temperature on the susceptibility of the organism to pollutants.  相似文献

Sub-cellular perturbations in the lysosomal compartment of molluscan haemocytes were examined in mussels (Perna viridis), collected along a pollution gradient. The neutral red technique was validated using a well defined contamination gradient among indigenous populations from five stations along Tolo Harbour, Hong Kong. Condition indices (shell length:dry tissue wt) and tissue metal concentrations were also measured in an attempt to identify a relationship between contamination level and adverse physiological effects. Correlations were found between lysosomal retention time and condition along the pollution gradient. There were significant differences between mussels collected from stations on offshore islands and those collected from inner harbour sites (p < 0.05). There was, however, little correlation between metal concentrations and retention time or condition (p > 0.05).  相似文献

Coastal zones are among the most productive and vulnerable areas on the planet. An example of impact on these fragile environments was shown in the case of the "Sea Empress" oil tanker, which ran aground in the Bristol Channel in 1996, spilling 72,000 tons of "Forties" crude oil. The objective was to investigate the sub-lethal cellular pathology and tissue hydrocarbon contamination in marine mussel populations, 4 months after the initial spill, using the neutral red retention (NRR) assay for lysosomal stability in blood cells. NRR was reduced in mussels, and indicative of cell injury, from the two sites closest to the spill in comparison with more distant and reference sites. Lysosomal stability was inversely correlated with polycyclic aromatic hydrocarbon concentrations in mussel tissues. Reduced lysosomal stability has previously been shown to contribute to impaired immunocompetence and to autophagic loss of body tissues. The use of this type of technique is discussed in the context of cost-effective, ecotoxicological tools for Integrated Coastal Zone Management.  相似文献

The clam Scrobicularia plana and the polychaete worm Nereis diversicolor were collected in several sites from a littoral enclosure in SW Spain. The aim of our study was to relate various biomarker responses in these species to a pollution gradient caused by untreated domestic discharges and to verify the adequacy of the selected species as sentinels in this habitat. The biomarkers selected were the antioxidant enzymes catalase (CAT), glutathione peroxidase (GPX) and DT-diaphorase (DT-D). In addition, the activities of cytochrome P450-dependent ethoxyresorufin O-deethylase (EROD) activity, the phase II detoxifying enzyme glutathione S-transferase (GST) and the neurotoxicity marker acetylcholinesterase (AChE) were measured. Metallothionein levels were selected as biomarkers of heavy metals exposure in both species. The results suggest a different response in the water filtering organism (clam) and the sediment eater (polychaete), probably as a consequent of different pollution exposure and that samples from the "Ca?o Sancti-Petri" were exposed to biologically active compounds that altered some of their biochemical responses. AChE was the most sensitive biomarker in both species and N. diversicolor proved to be a more robust sentinel in this ecosystem.  相似文献

为了解黄河尾闾及近岸沉积物中重金属的污染特征,采用电感耦合等离子体质谱仪(ICP-MS)测定了黄河河道和河口48个样品中Cr、Cu、Zn、Cd、Pb和As等6种重金属的含量,并对其分布和生态风险进行了评价。结果表明,黄河尾闾及近岸重金属的总含量沿河道至河口方向呈现先增加后降低再增加的趋势,至近岸B断面,As、Pb、Cd和Zn含量呈明显增加趋势,Cu和Cr含量增加幅度较小;黄河河口沉积物中重金属浓度明显高于河道沉积物中重金属的浓度。与我国其他流域相比,研究区域重金属含量处于较低水平。潜在生态风险评价结果表明,黄河尾闾及近岸沉积物6种重金属潜在生态风险系数从高到低依次为:CdAsPbZnCuCr,黄河尾闾及近岸沉积物的潜在生态风险主要由Cd和As引起,两者的贡献率分别为55.90%和29.54%,研究区域生态风险低,污染轻。研究结果将为黄河尾闾及近岸生态保护、环境管理和污染治理提供一定基础数据。  相似文献

Dissolved oxygen (DO) concentrations were surveyed in the Daliao River Estuary in autumn to investigate the temporal and spatial trends of DO distributions, The results indicate that DO is weakly stratified in the Yingkou section of the Daliao River Estuary, and remains constant vertically in the areas near and out of the entrance. Horizontally, both instantaneous and continuous observations show that the Yingkou section of the Daliao River Estuary is affected by hypoxic conditions; while no conditions of low DO have been observed in the areas near and out of the entrance.DO-salinity correlations exhibit different controlling factors of DO distributions in different sections of the Daliao River Estuary. At the upstream part of the Yingkou section of the Daliao River (A1-A4), oxygen demanding materials and the associated biochemical processes are the main factor controlling the DO distribution, but in the lower part of the Yingkou section and the area near and out of the entrance (A5-A15), tidal advections are more influential for DO distributions.  相似文献
