以4个不同生长阶段的球等鞭金藻的胞外滤液为研究对象,研究其对球等鞭金藻藻生长速率和胞内生化成分含量的影响。结果表明,随着胞外滤液老化程度的逐渐增加,藻细胞的生长速率及胞内物质的含量受到越来越强烈的影响,表现为生长速率呈现负值,胞内生化成分的含量降低。即便是补充了足够的营养盐,也无法消除胞外滤液的抑制作用。实验证明,球等鞭金藻在生长过程中产生的抑制物在影响其生长的同时,对胞内物质也有一定的抑制作用。  相似文献

以球等鞭金藻老化培养液的乙酸乙酯粗提物处理微藻细胞,通过对细胞密度、叶绿素a含量、胞内多糖含量及蛋白质含量等生理指标的测定,证实了乙酸乙酯粗提物对球等鞭金藻细胞生长有明显的抑制作用。进一步比较乙酸乙酯粗提物对球等鞭金藻细胞密度、叶绿素a含量及胞内多糖含量96h的半抑制剂量,得出细胞密度、胞内多糖及叶绿素a可以作为反映乙酸乙酯粗提物对球等鞭金藻抑制作用的生物学指标。  相似文献

利用交叉培养的方法,研究了球等鞭金藻与纤细角毛藻、牟氏角毛藻、新月菱形藻和三角褐指藻等4种海洋微藻间的化感作用。并利用高效液相色谱分析了这4种海洋微藻的胞外滤液萃取物。结果表明,(1)球等鞭金藻胞外滤液浓度大于40%时,显著抑制三角褐指藻、新月菱形藻和牟氏角毛藻的生长,而对纤细角毛藻的生长则有明显的促进效果,但当胞外滤液浓度大于80%时则对纤细角毛藻的生长也表现出抑制作用。纤细角毛藻、新月菱形藻、牟氏角毛藻的滤液浓度大于40%时,对球等鞭金藻表现出显著抑制作用,三角褐指藻胞外滤液浓度大于80%时才能抑制球等鞭金藻的生长。(2)4种海洋微藻胞外滤液的萃取物能明显抑制球等鞭金藻的生长。(3)纤细角毛藻、牟氏角毛藻、新月菱形藻和三角褐指藻的胞外滤液分别包含8种、5种、6种和7种物质,同时结果还表明4种海洋微藻产生的抑制球等鞭金藻生长的化感物质并不相同。  相似文献

在含有三个生长阶段(指数生长期、静止期和衰亡期) 的球等鞭金藻细胞再悬浮液、细胞破碎液、上清液、藻壳液和无细胞滤液的培养液中对牟氏角毛藻和三角褐指藻进行了培养,并通过这些培养液对两种微藻生长的影响探讨了球等鞭金藻生长抑制物的产生和释放过程.结果表明,生长抑制物的产生及释放与球等鞭金藻细胞的生长阶段密切相关;生长抑制物在细胞的对数生长初期产生并在细胞内积累,当细胞内的生长抑制物浓度超过一定范围后,细胞开始向培养物中释放生长抑制物,进而导致培养物中生长抑制物的浓度增加.  相似文献

探讨了不同浓度Zn2 对球等鞭金藻细胞生长及细胞内多糖、叶绿素、蛋白质等生化成分含量的影响.研究表明,浓度低于4.0 mg/L的Zn2 对球等鞭金藻的生长无明显抑制作用.当Zn2 浓度高于10.0mg/L时,则对球等鞭金藻的生长产生明显的抑制作用,随着Zn2 浓度的升高,球等鞭金藻胞内叶绿素和多糖含量降低,而蛋白质含量则明显升高.  相似文献

通过一次培养试验,分别研究了培养液中营养盐N、P、Si、Fe浓度变化对球等鞭金藻(Isochrysisgalbana)生长速率的影响。实验结果表明,生长速率与单一营养盐浓度符合Monod方程,而且将Monod方程扩展得到了可以表示多种营养盐对球等鞭金藻生长的复合限制模式。  相似文献

通过分批培养实验研究了氮、磷、铁、硅、碳、维生素B1、B12等营养因子对球等鞭金藻生长的影响,确定了氮、磷、铁、硅、维生素B1、B12对球等鞭金藻生长的必要性,并通过正交实验得出球等鞭金藻生长的最佳配方,即在天然海水中添加NaN03-N 7.5g/m^3,KH2PO4-P 0.5g/m^3,FeCL3-Fe 0.1g/m^3,Na2SiO3-Si0.2g/m^3,B1 100mg/m^3、B12 0.5mg/m^3。  相似文献

于1995年4—12月在中国科学院海洋研究所进行微藻除菌及比较除菌与自然带菌微藻生长特点的研究。经平板培养排除霉菌后,利用组合抗生素(青霉素+卡那霉素+链霉素+庆大霉素)获得除菌球等鞭金藻、三角褐指藻及小球藻,对抗生素处理前后的微藻生长特点进行比较研究。结果表明,与未除菌时相比,除菌后的球等鞭金藻及小球藻不易老化(可保持良好悬浮30d以上);回加细菌于除菌藻,藻细胞下沉附底,说明细菌可促使微藻细胞老化。无维生素时,除菌后的球等鞭金藻生长更差,暗示未除菌的球等鞭金藻培养液内可能存在产(类)维生素细菌。除菌后三角褐指藻细胞形态发生一定变化,回加细菌后藻细胞形态有部分恢复。与未除菌时相比,除菌后三角褐指藻更能耐受高温(如30℃)。另外,某些抗生素能够刺激球等鞭金藻的生长。  相似文献

通过实验室构建不同氮磷比条件下的酸化海水环境,培养麻痹性贝毒(paralytic shellfish toxin,PST)产毒藻微小亚历山大藻(Alexandrium minutum),采用藻细胞计数法、分光光度法、高效液相色谱-质谱法等分析方法,探讨了不同氮磷比条件下的酸化海水对微小亚历山大藻的生长、氮磷营养盐吸收、产毒等参数的影响。结果表明,在一定程度上,酸化环境对A.minutum的生长、PST的胞内累积和胞外释放有促进作用,且胞外培养液中PST含量增加的程度更高;同时,酸化海水促进胞内毒性较低的N-H类膝沟藻毒素(GTX2和GTX3)被氧化成毒性较高的N-OH类膝沟藻毒素(GTX1和GTX4),导致A.minutum的细胞毒性和培养液毒性增加。而海水中氮磷比的进一步升高,在一定程度上促进了微小亚历山大藻细胞毒素含量和毒性的增加,但并未促进其生长,且降低了藻细胞内的磷储量。  相似文献

于1995年8-12月,利用单因子实验法,首次研究了混合稀土对球等鞭金藻(Isochrysisgalbana)生长繁殖的影响,结果表明,海水中稀土最佳浓度范围为1.66-6.60mg/L,细胞浓度和叶绿素含量分别提高11.6%-23.2%和19.9%-45.2%混合稀土加入浓度高于9.9mg/L对球等鞭金藻的生产产生抑制作用,稀土对球等鞭金藻生长的促进作用与其他营养盐类浓度有关,当细胞处于指数生长  相似文献

球等鞭金藻细胞内可富集丰富的二十碳五烯酸(Eicosapentaenoic acid,EPA)和二十二碳六烯酸(Docosahexaenoic acid,DHA),这些多不饱和脂肪酸(Polyunsatured fatty aicd,PUFA)具有重要的生理活性。采用气相色谱法研究了不同浓度的NaNO3和NaH2PO4对球等鞭金藻生长和脂肪酸含量及组成成分的影响。结果表明,NaNO3和NaH2PO4对球等鞭金藻生长和脂肪酸含量(占干重的百分含量)及组成成分均有影响。在初始NaNO3浓度为1~74.8mg/L和初始NaH2PO4浓度为0.47~4.48mg/L范围内,随着NaNO3和NaH2PO4浓度的增加,球等鞭金藻的细胞密度和比生长速率都增大,脂肪酸和多不饱和脂肪酸含量则都呈现下降趋势;C22∶6(DHA)占干重的百分含量随着NaNO3和NaH2PO4浓度的增加而下降,但在总脂肪酸中的百分含量则增加,当NaNO3浓度为74.8mg/L时可达到9.92%,当NaH2PO4浓度为4.48mg/L时可达到7.81%。  相似文献

在f/2全营养培养条件下,采用共培养以及滤液交叉培养的方法研究了中肋骨条藻(Skeletonema costatum)和微小亚历山大藻(Alexandrium minutum)之间的竞争作用,结果表明: 微小亚历山大藻指数生长后期的无藻细胞滤液对中肋骨条藻的生长没有抑制作用,但中肋骨条藻滤液明显抑制了微小亚历山大藻的生长,且抑制作用随着中肋骨条藻滤液比例的增大而增大,他感作用是影响这两种微藻间竞争的重要方式之一;通过向培养基中添加自他感物质标准品15S-hydroxy-5Z,8Z,11Z,13E,17Z-eicosapentaenoic acid (15(S)-HEPE)考察其对中肋骨条藻自身以及微小亚历山大藻生长的影响,发现15(S)-HEPE在中肋骨条藻滤液中并不是能够抑制微小亚历山大藻生长的他感物质,今后应进一步深入分析中肋骨条藻释放的抑制微小亚历山大藻生长的他感物质。  相似文献

--Eight common red tide species including the Dinoflagellates Ale randrium tamarense,Prorocentrum minimum, Prorocentrum mican, Diatoms Skeletonema costatum , Cerataulina pelagica, Leptocylindrus minimus, a Raphidophyte Heterosigma akashiwo and a Chrysophyte Ivohrysis galbana were chosen to study the effect of four nitrogen substrates ( NO3 - N, NH4 - N, Urea - N,Mixed - N) on the growth of these algae. After two transfers, the results showed that at N concentration of 550μrnol, NO3 N was the best nitrogen source among these four nitrogen substrates; Urea - N and Mixed- N were also good for the growth of most algae, but not as good as NO3 - N for some species; NH4 - N inhibited the growth of all these species except H. akashiwo. At concentrations of 280 and 50 μmnol experiments on the growth of I. galbana, P. minimum and L. minimus in NH4 - N and NO3 - N substrates were also performed. The results showed that the growth rates of I. galbana were not significantly different from each other in NO3 - N and NH4 - N substrates both concentrations of 280 and 50 μmol. In concentration of 280μmol NH4 - N substrate, the growth rates of P.minimum and L. minimus were slower than in same concentration of NO3 - N; At concentration of 50μrmol , the growth rate of P. minimum in NH4 - N was not significantly different from that in NO3 - N, while the growth rate of L. minimus was only about 30% of that in NO3 - N substrate.The results indicated that each alga had its own preference in N-substrate and concentration, therefore,different nitrogen substrates may play a role in red tide formation.  相似文献

为研究剧毒卡尔藻(Karlodinium veneficum)与东海原甲藻(Prorocentrum donghaiense)之间的化感作用,在f/2营养盐条件下,分别采用了混合培养及滤液培养的方法。结果表明:剧毒卡尔藻指数生长后期的无藻细胞滤液明显抑制了东海原甲藻的生长,而东海原甲藻滤液对剧毒卡尔藻的生长几乎无影响,化感作用作为重要的影响因素使剧毒卡尔藻获得竞争优势。将剧毒卡尔藻指数生长末期的无藻细胞滤液置于固相萃取(SPE) C18小柱,经吸附、不同比例的甲醇溶液洗脱,考察不同洗脱部分对东海原甲藻生长的影响,发现80%的甲醇洗脱部分明显抑制了东海原甲藻的生长。本研究为了解剧毒卡尔藻的竞争机制提供了重要的科学依据,今后将重点分析剧毒卡尔藻释放的有效抑制东海原甲藻生长的化感物质。  相似文献

本文主要探究了东海原甲藻(Prorocentrum donghaiense)与三角褐指藻(Phaeodactylum tricornutum)之间的他感作用。在实验室模拟条件下,分别研究了添加比例为10%、40%、60%、90%、100%的三角褐指藻无藻细胞滤液的培养基对东海原甲藻生长的影响,以及相同比例的东海原甲藻无藻细胞滤液培养基对三角褐指藻生长的影响。结果表明,低浓度三角褐指藻滤液对东海原甲藻(P.donghaiense)的生长表现为促进作用,高浓度则抑制其生长,因此,不同浓度的三角褐指藻藻液对东海原甲藻生长表现出两种相反的他感作用效果;而东海原甲藻滤液对三角褐指藻(P.tricornutum)的生长没有显著影响,说明东海原甲藻并未对三角褐指藻表现出他感作用。  相似文献

Allelopathic effects of several concentrations of fresh tissue,dry powder and dry tissue of three bloom-forming green macroalgae Ulva pertusa,Ulva linza and Enteromopha intestinalis on the red tide microalga Heterosigma akashiwo were evaluated in microcosms systems.The effects of macroalgae culture medium filtrate were investigated on H.akashiwo using initial or semi-continuous filtrate addition.Preliminary studies on the algicidal effects of one aqueous and four organic solvent extracts from the macroalgae on the microalga were carried out to confirm the existence of allelochemicals in the tissue of these macroalgae.The dry powder of U.pertusa was extracted with methanol,and the methanol extracts were partitioned to petroleum ether phase,ethyl acetate phase,butanol phase and distilled phase by liquid-liquid fractionation.The bioassays of the activity of every fraction were carried out on H.akashiwo.The resultant microcosms assay showed that the growth of H.akashiwo was strongly inhibited by using fresh tissues,dry powder or dry tissue of these three macroalgae,while aqueous and methanol extracts of both macroalgae had strong inhibitory effects on the growth of H.akashiwo,and the EC 50 values for methanol extract of U.pertusa,U.linza or E.intestinalis were 0.016,0.028×10-12 or 0.033×10-12,respectively.While the other three organic solvent extracts(acetone,ether and chloroform) had no apparent effect on its growth,this suggests that the allelochemicals from these three macroalgae had relatively high polarities.The activity of petroleum ether phase,ethyl acetate phase,butanol phase and distilled phase of U.pertusa methanol extract was carried out on H.akashiwo indicating that petroleum ether phase and ethyl acetate phase had stronger algicidal effect on H.akashiwo.The inhibition effect of the ethyl acetate phase was not as strong as that of petroleum ether phase,and effective concentration of petroleum ether phase was 17 mg/L for H.akashiwo.However,no significant algicidal effects were observed on the butanol phase and distilled water phase.These three macroalgae's culture medium filtrate exhibited no apparent growth inhibitory effect on the microalga under initial filtrate addition whereas the growth of H.akashiwo was significantly inhibited under semi-continuous filtrate addition,which suggests that continuous release of small quantities of rapidly degradable allelochemicals from the fresh tissue of both macroalgae was effective in inhibiting the growth of H.akashiwo.  相似文献
