本实验分别针对3株低温藻株:微拟球藻Nannochloropsis sp.ZL-12、四爿藻Tetraselmis chui ZL-33和小球藻Chlorellasp.ZL-45,3株中温藻株:球等鞭金藻Isochrysis galbana CCMM5001、等鞭金藻Isochrysis sp.CCMM5002和微拟球藻Nannochloropsis sp.CCMM7001,3株高温藻株:微拟球藻Nannochloropsis sp.JN1、绿色巴夫藻Pavlova viridis JN2和海洋小球藻Chlorellasp.JN3,研究了在通入0.03%(空气)、5%、10%3个CO2浓度梯度条件下的生长特性,同时考察了其总酯及中性脂的累积情况。结果显示,富碳培养有利于不同温度条件下9株藻株的生长,除微拟球藻Nannochloropsis sp.CCMM7001最适生长的CO2浓度为5%外,其余8株藻株最适生长的CO2浓度均为10%。在低温和高温条件下,6株海洋富油微藻在通入10%CO2时具有最大生物量产率,在中温条件下球等鞭金藻和等鞭金藻在通入10%CO2时获得最大生物量产率,而微拟球藻在通入5%时获得最大生物量产率,随着CO2浓度的增加,9株藻株的总脂含量和中性脂含量有明显提高。低温和中温藻株的总脂含量高于高温藻株的总脂含量,从中性脂的累积规律来看,9株藻株均在平台期的累积达到最大值,GC-MS分析结果表明,9株微藻适合制备生物柴油的C14~C18系脂肪酸相对含量在不同CO2条件下基本保持不变,维持在90%左右。实验结果显示,所研究的藻株作为富油高固碳优良藻株,具备用于海洋生物质能耦合CO2减排开发的潜力。  相似文献

Hydrogen peroxide (H2O2) was measured in marine surface waters of the eastern Atlantic Ocean between 25°N and 25°S. H2O2 concentrations decreased from 80 nM in the north to 20 nM in the south, in agreement with earlier observations. A diel cycle of H2O2 production as a function of sunlight in surface waters was followed twice whilst the ship steamed southward. Around 23°N a distinct diel cycle could be measured which correlated well with irradiance conditions.The wavelength dependency of H2O2 formation was studied near the equator. For 16 hours, water samples were incubated with wavelength bands of the solar spectrum, i.e. visible (VIS: 400–700 nm), VIS and ultraviolet A radiation (UVAR: 320–400 nm) and VIS, UVAR and ultraviolet B radiation (UVBR: 280–320 nm). A significant relationship was found between wavelength band and the production of H2O2. In addition, a clear positive relationship between intensity and production was found. UVAR was 6.5 times more efficient than VIS in producing 1 nM of H2O2, whereas UVBR was 228 times more efficient than VIS. When these data were weighted with respect to the energy of the solar spectrum at zenith hour, 28% of the H2O2 was formed by VIS, 23% was formed by UVAR and 48% was formed by UVBR. Considering the strong attenuation of UVBR in marine waters as compared with UVAR and VIS radiation, the role of UVAR deeper in the water column is recognised. One strain of unicellular greenish algae embedded by mucilage was successfully isolated from equatorial area in the Indian Ocean. Microscopic observation, ultrastructure features and genetic identification confirmed that the strain was closely related to Cyanothece sp., which was a cyanobacteria species with great ecological significance.Cells were solitary with oval or bacilliform shapes. Diameters of this strain were relatively small, ranging from 2.5 to 6.5 μm on average. Ultrastructure of cells was simple. Thylakoids were arranged parietal and keritomized content were observed in the thylakoid region. Various electron-transparent granules with low electron-dense region as well as cyanophycin or glycogen granules-like organelle and carbonxysomes were also observed. For pigment composition, the dominant pigments were chlorophyll a, β-Carotene, Zeaxanthin and an unknown pigment, contributing 23.8%, 26.1%, 14.7% and 15.7% to total pigments respectively. The phylogenetic analysis of16 S rRNA gene and nif H gene confirmed that Strain EIO409 was closely related with Cyanothece sp..  相似文献

过氧化氢(H2O2)等活性氧(ROS)物质因具有绿色高效的特点,在废水处理、杀菌消毒等领域受到研究者广泛关注。利用电催化生成H2O2是一种实时提供活性氧物质的有用方法。然而,目前报道的大多数高性能催化材料都是粉体形式,不便于在实际场景中的应用。因此,制备可直接应用的电极材料显得尤为重要。本研究利用喷涂-热解的制备方法,制备了高活性氧催化活性的氧化炭黑(O−CB)/多孔碳毡电极,通过聚四氟乙烯(PTFE)的修饰优化电极表面的氧气传质,成功制备氧掺杂碳气体扩散电极。研究结果表明,O−CB/PTFE-5 wt%电极具有最高的催化性能,其合成H2O2的速率达27.19 mg·L-1(mg catalyst)-1·cm-1·h-1。海洋典型污损微生物假单胞菌(Pseudomonas sp.)的抗菌实验表明,该电极电催化作用60 min产生的活性氧对Pseudomonas sp.的杀菌率可达到97.69%,作用120 min的杀菌率可达到99.99%。  相似文献

Mangrove forests are known to be inhabited by diverse symbiotic cyanobacterial communities that are capable of N2 fixation. To investigate its biodiversity, root sediments were collected from a mangrove forest in Chuuk State, Federated States of Micronesia (FSM), and an entangled yellow-brown coccoid cyanobacterium was isolated. The isolated cyanobacterium was reproduced by multiple fission and eventually produced baeocytes. Phylogenetic analysis revealed that the isolate was most similar to the genera Myxosarcina and Chroococcidiopsis in the order Pleurocapsales. Compositions of protein, lipid and carbohydrate in the cyanobacterial cells were estimated to be 19.4 ± 0.1%, 18.8 ± 0.4% and 31.5 ± 0.1%, respectively. Interestingly, total fatty acids in the isolate were mainly composed of saturated fatty acids and monounsaturated fatty acids, whereas polyunsaturated fatty acids were not detected. Based on the molecular and biochemical characteristics, the isolate was finally classified in the genus Myxosarcina, and designated as Myxosarcina sp. KIOST-1. These results will contribute to better understanding of cyanobacterial biodiversity in the mangrove forest in FSM as well as the genus Myxosarcina, and also will allow further exploitation of its biotechnological potential on the basis of its cellular characteristics.  相似文献

海洋亚心形扁藻(Platymonas subcordiformis)在光照生长后,经过暗培养诱导其可逆产氢酶,然后转移到光照下可产生氢气。在300 mL产氢反应器中对海洋亚心形扁藻产氢情况的考察表明,不同的光强条件可导致扁藻产生不同的生长曲线,而在不同的培养阶段,扁藻的产氢能力也有很大的差别。其他培养条件相同,高光照强度(E=13000 lx)条件下,扁藻生长较快,而且可以获得更大的产氢能力,300 mL密度为3×106个/mL、叶绿素质量浓度为5.8 mg/L的藻液最大产氢浓度可达17.5%,比低光照强度(E=5000 lx)时提高了32%,叶绿素含量降低了61%。根据产氢前后以及暗诱导后扁藻生理状态的考察结果,可以推测出是不同生长阶段扁藻的生理状态差异导致了其产氢能力的变化。  相似文献

微藻固碳是一种新型节能减排技术,具有长期可持续发展的潜力。本文对两株富油微藻(球等鞭金藻和微拟球藻)进行了富碳培养下生长特性及中性脂积累特性的研究。两株富油微藻的最佳培养条件为10%CO2浓度和f培养基。本研究对两株富油微藻的最大生物量产率、总脂含量、最大油脂产率、微藻的C含量和CO2固定率进行了测定。球等鞭金藻的各参数指标分别为:142.42±4.58g/(m2·d),39.95%±0.77%,84.47±1.56g/(m2·d),45.98%±1.75%和33.74±1.65g/(m2·d)。微拟球藻的各参数指标分别为:149.92±1.80g/(m2·d),37.91%±0.58%,89.90±1.98g/(m2·d),46.88%±2.01%和34.08±1.32g/(m2·d)。实验结果显示,两株海洋微藻均属于高固碳优良藻株,适合应用于微藻烟气减排技术开发,具备用于海洋生物质能耦合CO2减排开发的潜力。  相似文献

Short-lived halocarbons were measured in Arctic sea–ice brine, seawater and air above the Greenland and Norwegian seas (~81°N, 2–5°E) in mid-summer, from a melting ice floe at the edge of the ice pack. In the ice floe, concentrations of C2H5I, 2-C3H7I and CH2Br2 showed significant enhancement in the sea ice brine, of average factors of 1.7, 1.4 and 2.5 times respectively, compared to the water underneath and after normalising to brine volume. Concentrations of mono-iodocarbons in air are the highest ever reported, and our calculations suggest increased fluxes of halocarbons to the atmosphere may result from their sea–ice enhancement. Some halocarbons were also measured in ice of the sub-Arctic in Hudson Bay (~55°N, 77°W) in early spring, ice that was thicker, colder and less porous than the Arctic ice in summer, and in which the halocarbons were concentrated to values over 10 times larger than in the Arctic ice when normalised to brine volume. Concentrations in the Arctic ice were similar to those in Antarctic sea ice that was similarly warm and porous. As climate warms and Arctic sea ice becomes more like that of the Antarctic, our results lead us to expect the production of iodocarbons and so of reactive iodine gases to increase.  相似文献

The effects of low-pH, high-pCO2 conditions on deep-sea organisms were examined during four deep-sea CO2 release experiments simulating deep-ocean C sequestration by the direct injection of CO2 into the deep sea. We examined the survival of common deep-sea, benthic organisms (microbes; macrofauna, dominated by Polychaeta, Nematoda, Crustacea, Mollusca; megafauna, Echinodermata, Mollusca, Pisces) exposed to low-pH waters emanating as a dissolution plume from pools of liquid carbon dioxide released on the seabed during four abyssal CO2-release experiments. Microbial abundance in deep-sea sediments was unchanged in one experiment, but increased under environmental hypercapnia during another, where the microbial assemblage may have benefited indirectly from the negative impact of low-pH conditions on other taxa. Lower abyssal metazoans exhibited low survival rates near CO2 pools. No urchins or holothurians survived during 30–42 days of exposure to episodic, but severe environmental hypercapnia during one experiment (E1; pH reduced by as much as ca. 1.4 units). These large pH reductions also caused 75% mortality for the deep-sea amphipod, Haploops lodo, near CO2 pools. Survival under smaller pH reductions (ΔpH<0.4 units) in other experiments (E2, E3, E5) was higher for all taxa, including echinoderms. Gastropods, cephalopods, and fish were more tolerant than most other taxa. The gastropod Retimohnia sp. and octopus Benthoctopus sp. survived exposure to pH reductions that episodically reached −0.3 pH units. Ninety percent of abyssal zoarcids (Pachycara bulbiceps) survived exposure to pH changes reaching ca. −0.3 pH units during 30–42 day-long experiments.  相似文献

The distribution of the fugacity of CO2 ( $ f_{{{\text{CO}}_{ 2} }} $ ) and air–sea CO2 exchange were comprehensively investigated in the outer estuary to offshore shallow water region (lying adjacent to the Sundarban mangrove forest) covering an area of ~2,000 km2 in the northern Bay of Bengal during the winter. A total of ten sampling surveys were conducted between 1 December, 2011 and 21 February, 2012. Physico-chemical variables like sea surface temperature (SST), salinity, pH, total alkalinity (TAlk), dissolved inorganic carbon (DIC) and in vivo chlorophyll-a along with atmospheric variables were measured in order to study their role in controlling the CO2 flux. Surface water $ f_{{{\text{CO}}_{ 2} }} $ ranged between 111 and 459 μatm which correlated significantly with the SST (r = 0.71, p < 0.001, n = 62). Neither DIC nor TAlk showed any linear relationship with varying salinity in the estuarine mixing zone, demonstrating the significant presence of non-carbonate alkalinity. An overall net biological control on the surface $ f_{{{\text{CO}}_{ 2} }} $ distribution was established during the study, although no significant correlation was found between chlorophyll-a and $ f_{{{\text{CO}}_{ 2} }} $ (water). The shallow water region studied was mostly under-saturated with CO2 and acted as a sink for atmospheric CO2. The difference between surface water and atmospheric $ f_{{{\text{CO}}_{ 2} }} $ ( $ \Updelta f_{{{\text{CO}}_{ 2} }} $ ) ranged from ?274 to 69 μatm, with an average seaward flux of ?10.5 ± 12.6 μmol m?2 h?1. The $ \Updelta f_{{{\text{CO}}_{ 2} }} $ and hence the air–sea CO2 exchange was primarily regulated by the variation in sea surface $ f_{{{\text{CO}}_{ 2} }} $ , since atmospheric $ f_{{{\text{CO}}_{ 2} }} $ varied over a comparatively narrow range of 361.23–399.05 μatm.  相似文献

Two depth inversion algorithms (DIA) applicable to coastal waters are developed, calibrated, and validated based on results of computations of periodic waves shoaling over mild slopes, in a two-dimensional numerical wave tank based on fully nonlinear potential flow (FNPF) theory. In actual field situations, these algorithms would be used to predict the cross-shore depth variation h based on sets of values of wave celerity c and length L, and either wave height H or left–right asymmetry s2/s1, simultaneously measured at a number of locations in the direction of wave propagation, e.g., using video or radar remote sensing techniques. In these DIAs, an empirical relationship, calibrated for a series of computations in the numerical wave tank, is used to express c as a function of relative depth koh and deep water steepness koHo. To carry out depth inversion, wave period is first predicted as the mean of observed L/c values, and Ho is then predicted, either based on observed H or s2/s1 values. The celerity relationship is finally inverted to predict depth h. The algorithms are validated by applying them to results of computations for cases with more complex bottom topography and different incident waves than in the original calibration computations. In all cases, root-mean-square (rms)-errors for the depth predictions are found to be less than a few percent, whereas depth predictions based on the linear dispersion relationship—which is still the basis for many state-of-the-art DIAs—have rms-errors 5 to 10 times larger.  相似文献

The TCO2, O2, TA and δ13C data of the 1969 Geosecs Intercalibration Cruise was analyzed and found to be consistent with a vertical mixing model which assumes that each point along a vertical profile is a mixture of the upper and lower boundaries. Calculated regression coefficients are in agreement with the model of Redfield et al. (1963) and with the assumption that TA variation is due to carbonate reaction. Oxygen consumption and TCO2 production decrease exponentially with depth and approximately 80% of ΔCO2 can be accounted for, on average, by O2 consumption. The remaining 20% are probably due to carbonate solution which seems to take place at depths below 2,500 m. The present study suggests that the isotopic composition (δ13C) of the carbon source, required to account for most of the oxygen consumed, may be heavier than the value of −23%. assigned to dissolved organic carbon and particulate organic carbon.  相似文献

以采自天津海水浴场潮间带的污泥为研究材料,将污泥热休克处理后富集混合菌群进行厌氧发酵产氢,测定混合菌群在发酵过程中累积产氢量和相关产氢指标(吸光度、酸碱度和氧化还原电位)的变化。结果表明,热休克处理后富集的产氢混合菌群产氢量为0.41mol H2/mol葡萄糖。产氢结束后,利用变性梯度凝胶电泳(DGGE)分析混合菌群组成和Fe-氢酶基因的多样性。混合菌群基因组DNA的16S rRNAV3区扩增产物经过DGGE电泳分离,结果得到6条丰度比较高的条带,测序结果表明优势菌为Clostridium sp.。利用梭菌氢酶基因保守性引物克隆混合产氢菌群中梭菌Fe-氢酶基因,扩增产物经DGGE电泳分离,电泳结果表明混合菌群中Fe-氢酶基因分为3个分类单元,NCBI-BLAST比对结果与Clostridium roseum和Clostridium perfringens的Fe-氢酶基因相似度分别为79%和98%。分析产氢混合菌群组成和Fe-氢酶基因多样性,可以为扩大产氢微生物种质资源奠定基础。  相似文献

The thickness of the mixed bottom boundary layer (BBL) has been analyzed based on the CTD data at transoceanic sections in abyssal waters of the Northern Atlantic. The measurements were carried out at two transoceanic sections approximately along 48° N (ASV-99) and 5° N (AI-2000) in 1999 and 2000. These data, and the WOCE data obtained at four zonal sections (AR7E and AR12 along 57° N, AR01 along 24.5° S, and A06 along 7.5° N), were used for the calculation of the statistical characteristics of the BBL??s thickness H B . The probability distribution function F(H B ) was close to lognormal. The mean value ??H B ?? at different latitudes was in the range from 30 to 60 m. The averaged BBL thickness = 46.1 m. The BBL??s thickness was about 1% of the ocean??s depth D; the ratio H B /D was the minimum (0.8%) near the equator and increased up to 1.6% in the polar latitudes.  相似文献

The precision of spectrophotometric measurements of indicator absorbance ratios is sufficient to allow evaluation of small isotopically induced differences in the dissociation constant of boric acid (KB). The quotient of 11KB and 10KB, obtained using isotopically ⩾99% pure borate/boric acid buffers, provides an equilibrium constant for the reaction 10B(OH)3+11B(OH)411B(OH)3+10B(OH)4 which heretofore had not been experimentally determined. Previous theoretical and semi-empirical evaluations of this equilibrium, which is important for assessments of the paleo-pH of seawater and the paleo-pCO2 of the atmosphere, have yielded constants, 11–10KB=10KB/11KB, that have ranged between 1.0194 and approximately 1.033. The experimentally determined value 11–10KB=1.0285±0.0016 (mean±95% confidence interval) obtained at 25 °C and 0.63 molal (mol kg−1 H2O) ionic strength is in much better agreement with recent theoretical assessments of 11–10KB that have ranged between 1.026 and 1.033, than the much-cited original estimate (1.0194) of Kakihana et al. (1977) [Fundamental studies on the ion-exchange separation of boron isotopes. Bulletin of Chemical Society of Japan 50, 158–163]. Since the activity quotient for the fractionation reaction is almost equal to unity, it is expected that the 11–10KB value obtained in this study will be applicable over a wide range of solution compositions and ionic strengths.  相似文献

Vertical profiles of dissolved and particulate 210Po and 210Pb were measured across the redox transition zone at Station F1 in Framvaren Fjord, Norway. In this fjord, a sharp decrease in pH above the O2/H2S interface facilitates the aerobic dissolution of MnO2. In contrast, Fe(II) concentrations begin to increase only at the O2/H2S interface depth. Activity profiles reveal that dissolved 210Po and 210Pb are sequestered efficiently by particulates in surface waters. As polonium-210 and lead-210 activities descend down into the aerobic manganese reduction (AMR) zone, they are remobilized during the reductive dissolution of the carrier phase oxyhydroxides. Both 210Po and 210Pb are highly enriched at the O2/H2S interface where an active community of microbes, such as anoxygenic phototrophs (e.g., Chromatium, Chlorobium sp.), thrives. The coincident peaks in 210Po, 210Pb and microbial biomass suggest a strong biological influence on the behavior of these radionuclides. There is a strong covariance between the vertical distribution of Mn and Pb, indicating that their redox cycling is closely coupled and is likely microbially mediated.  相似文献

The Prévost lagoon (Mediterranean coast, France), was subject to annual dystrophic crises caused by the biodegradation of opportunistic macroalgae (Ulva lactuca) in the past. These crises result in anoxic waters with subsequent blooms of Purple Sulphur Bacteria (red waters) which, by oxidizing sulphide, contribute to the reestablishment of oxic conditions in the water column. Mechanical dredging of the macroalgal biomass has been carried out in the lagoon since 1991 with the aim of preventing the ecological and economic disturbances caused by such crises. Dredging began just before the phototrophic bloom when the water was already hypoxic (O2 = 0.7 mg·L−1) and contained sulphilde (H2S = 7.3 mg·L−1) and purple patches of phototrophic bacteria (Thiocapsa sp.) that were beginning to develop on decaying macroalgae at the sediment surface. The dredging prevented red water formation and drastically modified both phototrophic community structure and activity and biogeochemical sulphur cycling. The dredging permitted the reestablishment of oxic conditions for a short period only (1–13 August). Resuspension of the superficial sediment layers disturbed the phototrophic bacterial community, whose numbers decreased by one order of magnitude (from 2 × 106 to 3.9 × 105 CFU.mL−1). The phototrophic community was no longer effective in reoxidizing the reduced sulphur compounds remaining in the sediments, as shown by a drastic sulphate depletion in the superficial sediment layers. Moreover, the increase in the specific bacteriochlorophyll a concentration of the phototrophic purple bacteria and the rapid development of Green Sulphur Bacteria (Prosthecochloris-like microorganisms) indicated that the phototrophic community was growing under severe light-limiting conditions due to the resuspension of sediment particles in the water. These conditions did not allow the phototrophic bacterial community to efficiently reoxidize the reduced sulphur compounds originating from the sediments. In consequence, hypoxic conditions (O2 = 4.7 to 4.8 mg·L−1) and low sulphide concentrations (H2S = 0.4 to 0.7 mg·L−1) were detected in the water column until September. The ecological balance in the lagoon was reestablished only in October, whereas, in previous years it had been restored in August.  相似文献

大气CO2持续升高,导致溶入海水中的CO2增多,海水表层的H+浓度增加,从而引起海洋酸化。为了探讨近岸定生大型海藻对这种环境变化的响应,本文选择经济海藻坛紫菜为实验材料,研究海洋酸化与紫外辐射对藻体生长以及光合特性的影响。实验分两个CO2处理,分别为正常空气水平(390 ppmv)和高CO2水平(800 ppmv); 三种辐射处理,分别为全波长辐射(PAB)、滤除紫外线B(PA)和仅接受可见光处理(PAR)。研究结果表明,CO2培养下的坛紫菜,在仅有可见光(P)或者同时有紫外线A(PA)存在的情况下,显著促进藻体的生长;但在全波长辐射处理下(PAB),这种作用不明显。高CO2降低了藻体在P和PA处理下的光合作用速率,但对PAB处理作用不显著。高CO2处理下的藻体,UV-B显著降低了全波长辐射下藻体紫外吸收物质的含量,但在正常CO2水平下,紫外辐射的作用不显著。这表明高CO2导致的生长优势被紫外辐射的负面效应所抵消,在全球变化的过程中,紫外辐射的进一步加强在海洋酸化的背景下甚至有可能降低坛紫菜的产量。  相似文献

Microbial plankton metabolism was examined during summer 2010 in sea ice-influenced waters of the Fram Strait, eastern Arctic Ocean. Rates of gross primary production and community respiration were tightly coupled over a wide range of values (33±3–143±6 and 20±3–126±6 mmol O2 m−2  −1, respectively) leading to a prevalence of positive net community production. The high variability in community respiration, similar to that of gross primary production, suggests that heterotrophic metabolism may exhibit a significant response to environmental change. Bacterial respiration was assessed at similar time scales to bacterial production measurements, by determining the in vivo INT reduction capacity without pre-filtering the community. Bacteria seem to play a major role in total community respiration, contributing between 5% and 61% of total community respiration, indicating that a high fraction of the organic carbon in Arctic planktonic food webs could flow through these microbes.  相似文献
