青鳞鱼蛋白风味酶控制水解动力学模型   总被引:1,自引:0,他引:1  
在假设风味酶恒温控制水解动力学遵循内切酶限制水解动力学历程的前提下,通过实验方法求出了风味酶恒温控制水解动力学模型。结果表明,风味酶对青鳞鱼蛋白进行控制水解的动力学模型为:R= ( 54 .782e0 -0 .045s0 )exp( -0 .398DH),DH=2 .513ln[1+(21 .803e0 /s0 -0. 0179)t];其酶失活常数kd=22. 155 6min-1;水解反应能够顺利进行的条件是:e0 /s0 >c0,常数c0 =8 .214×10-4。验证实验证明,根据风味酶恒温控制水解动力学模型得到的理论水解度与实际水解基本吻合。  相似文献

以罗非鱼鱼鳞胶为原料,制备对血管紧张素转换酶(ACE)有抑制活性的降血压肽。以水解度(DH)及ACE抑制率为指标,筛选水解鱼鳞胶的最适酶种,并对其水解工艺条件进行优化;采用超滤技术对所制备的降血压肽进行分离纯化,比较不同组分的ACE抑制率,测定高ACE抑制率组分的氨基酸组成,并运用MALDI-TOF-MS法分析其分子质量分布。结果表明,复合蛋白酶是水解鱼鳞胶最适酶种,在50℃、pH7.5、料液质量体积比1∶5、酶与底物质量比(mE/mS)1.5%条件下,酶解6 h,酶解液的ACE抑制作用最强,其水解度DH 73%,ACE的抑制率76.15%。酶解液经超滤分离所获得的分子质量小于2 000 u的组分对ACE抑制率最高,达83.15%;该组分的氨基酸主要含有Gly和Pro,含量分别为22.04%和11.40%;该组分分子质量范围为1 300~2 000 u。  相似文献

于 1998年 11月至 2 0 0 2年 12月 ,在福建长乐对西施舌 (Coelemactraantiguata)的生物学特征进行测定 ,以人工养殖方法观察其周年生长情况。观测结果表明 ,长乐沿海西施舌的群体组成以体长 6 1~ 110mm占优势 ,其频率 ( fn)为 0 .14 0~ 0 .2 2 2~ 0 .12 5。不同大小的西施舌 ,软体部肥满度指数 (CI)具有明显的季节差异。 1龄西施舌月平均体长增长 4 .0 5mm ;2龄的为 2 .31mm ;3龄的为 1.83mm ;1~ 3龄为成体生长发育阶段 ,3龄之后趋向衰老。不同发育生长阶段的壳高与壳长直线回归关系 :幼虫期H =0 .994L - 13.0 7;稚贝期H =0 .90 5L - 4.76 ;幼贝期H =0 .0 14 +0 .82 4L ;成贝期H =0 .36 7+0 .72 4L。  相似文献

研究了木瓜蛋白酶的酶解温度、酶浓度、酶解时间、加水量、pH值对青鳞鱼可溶性蛋白得率的影响 ,探讨了α 氨态氮含量与青鳞鱼可溶性蛋白得率的关系。结果表明 :温度小于 5 0℃ ,随着酶解温度的提高、酶浓度的增加、酶解时间的延长青鳞鱼可溶性蛋白得率及α 氨态氮的含量均增加 ,α 氨态氮含量增加比可溶性蛋白得率的增加明显 ;加水量的增加 ,可溶性蛋白得率增加 ;pH值对水解效果及可溶性蛋白得率影响不大。综合考虑 ,最佳水解条件为 :温度 5 0℃ ,酶浓度为 1 5 0 0u/g ,酶解时间 80min ,m原料 :m水 =1∶2 ,pH 6 8。酶解所得可溶性蛋白经氨基酸分析仪检测 ,其氨基酸总量为 49 47mg/mL ,必需氨基酸占氨基酸总量的 41 0 % ,游离氨基酸含量为 6 3mg/mL ,肽含量为 87 2 %。  相似文献

研究了倒刺的年龄与生长规律 ,结果表明 :倒刺鳞片年轮特征主要为疏密切割型。体长与鳞长之间的直线相关L =2 .0 85 4+2 .36 2 0R和幂函数相关L =2 .85 12R0 .952 9中 ,以幂函数相关性密切 ;体重与体长呈指数函数相关W =2 .4 984× 10 - 2 L2 .9936 。 4龄以前生长较快 ,生长指标高 ,体长指标高 ,体长和体重的相对增长率大 ,其生长规律可用VonBertalanffy方程表达 :Lt=[1-e- 0 .132 5(t+ 0 .2 0 95) ],Wt=132 76 .4 0 [1-e- 0 .132 4 (t+ 0 .2 0 95) ]3。体重生长曲线的拐点位于t =8.0 9龄、W8.0 9=3933.6 1g ,即Wt=0 .2 96W∞ 处。倒刺雌性一般在 5龄性成熟 ,雄鱼 3~ 4龄时性成熟。建议江河捕捞对象应为 5 + 龄以上的个体。  相似文献

The distribution of sediment chloroplastic pigments (Chl-a, i.e. chlorophyll a and Pha-a, i.e. phaeophorbide a) in the Southern Yellow Sea of China was studied. Samples were collected from four cruises in January and June 2003, and January and June 2004. The results show that the vertical distribution of Chl-a and Pha-a in the sediment layers 0-2cm, 2-5cm and 5-8cm, follows a stable ratio, 5:3:2. The average ratio of Pha-a to Chl-a in sediment is 2.83. Spearman 2-tailed rank correlation analysis shows that Chl-a and Pha-a contents in each sediment layer have a highly significant correlation. The average contents of Chl-a and Pha-a in the sediment of the 0-8cm layer in the investigated area are 0.31 -0.47μgg-1 and 1.28-1.40 μgg-1 sediment (dry weight), respectively. The average Chl-a and Pha-a contents in sediment are higher in summer than in winter. ANOVA analysis shows that there is a highly significant variation among the Chl-a contents (P = 0.002 <0.01) of the four cruies, but this is not true for the case of Pha-a content (P = 0.766>0.05). The average Chl-a and Pha-a contents in the 2 sediment layers (0-2cm and 2-5cm) have significant or highly significant correlations with organic matter (OM), median diameter (Mdφ), silt plus clay percentage in the January 2003 cruise. In the June 2003 cruise, the average Chl-a content in the 3 sediment layers (0-2cm, 2-5cm, and 5-8cm) has a significant correlation with meiofauna biomass, and Pha-a content has highly significant correlations with water depth, bottom water temperature, OM and Mdφ The contents of Chl-a and Pha-a are lower than those in estuaries and intertidal areas, but close to those in the same area studied previously.  相似文献

【目的】研究热处理温度和时间对凡纳滨对虾肌肉组织蛋白酶L酶活力的影响,并分析其酶活力变化动力学。【方法】从凡纳滨对虾的肌肉组织提取组织蛋白酶L,研究加热温度(35~60℃)和时间(5~60 min)对其酶活力的影响,采用反应动力学模型拟合酶活力的变化过程。【结果】在35℃组织蛋白酶L的酶活力变化适合用一级动力学模型来描述,而在40~60℃组织蛋白酶L的酶活力变化适合用两阶段动力学模型来描述;凡纳滨对虾肌肉组织蛋白酶L存在敏感型和稳定型两种同工酶,敏感型的Z_(T,L)和E_(a,L)分别为158.7℃和5.5k J/mol,稳定型的Z_(T, s)和E_(a, s)分别为71.9℃和12 kJ/mol。【结论】加热处理可以使组织蛋白酶L发生变性而失活,采用合理的加热温度和速度可以降低组织蛋白酶L引起的凝胶劣化。  相似文献

以菠萝 (AnanascomosusMerr)为原料 ,应用改良的方法对菠萝天然酶活力进行测定 ,对菠萝酶活力变化进行了初步探讨。结果表明 :测定未成熟、成熟菠萝酶活力分别为 10 6 .10和 6 5 .85u/mL ;菠萝肉、皮、整果汁酶活力分别为 86 .98、5 0 .72、5 5 .2 1u/mL ;室温贮藏 ,酶活力取样时为 5 4.97u/mL ,第 4天达最高值 72 .78u/mL ,第 8天降至 4 3.6 6u/mL ;4℃贮藏 ,酶活力取样时为 5 4.97u/mL ,第 2天达最高值71.6 8u/mL ,第 4天降至 5 0 .12u/mL ,即不同温度下贮藏不同时间酶活力变化是先升高后降低  相似文献

Ultrasound-assisted solvent extraction was confirmed as a novel,effective method for separating lipid from mackerel pro-tein,resulting in a degreasing rate (DR) of 95% and a nitrogen recovery (NR) of 88.6%.To obtain protein hydrolysates with high ni-trogen recovery and low bitter taste,enzymatic hydrolysis was performed using eight commercially available proteases.It turned out that the optimum enzyme was the ‘Mixed enzymes for animal proteolysis’.An enzyme dosage of 4%,a temperature of 50℃,and a hydrolysis time of 300 min were found to be the optimum conditions to obtain high NR (84.28%) and degree of hydrolysis (DH,16.18%) by orthogonal experiments.Glutamic acid was the most abundant amino acid of MDP (defatted mackerel protein) and MDPH (defatted mackerel protein hydrolysates).Compared with the FAO/WHO reference protein,the essential amino acid chemical scores (CS) were greater than 1.0 (1.0 1.7) in MDPH,which is reflective of high nutritional value.This,coupled with the light color and slight fishy odor,indicates that MDPH would potentially have a wide range of applications such as nutritional additives,functional ingredients,and so on.  相似文献

在室内实验条件下研究了温度对不同规格橄榄蛏蚌耗氧率和排氨率的影响。实验结果表明, 15~30℃下,橄榄蛏蚌耗氧率和排氨率为大规格组均小于小规格组。耗氧率和排氨率与橄榄蛏蚌软体部干重(W)呈现明显的幂函数关系。在实验温度(15~30℃)范围内,随温度的上升,橄榄蛏蚌的耗氧率和排氨率均先逐渐增加,后随之下降,两者的高峰值分别出现在25℃和20℃。耗氧率(O)、排氨率(N)与温度(T)和橄榄蛏蚌软体部干重(W)的二元线性回归方程分别为:O=-0 .651 3+0. 053 2T+0. 107 3W,N=32. 162 6-1. 056 6T+1 .322 2W。二方程复相关系数r分别为0 .964 2和0 .892 1;F检验分别为极显著和显著。  相似文献

A new enzyme (alkaline protease 894) obtained from the marine extremophile Flavobacterium yellowsea (YS-80-122) has exhibited strong substrate-binding and catalytic activity, even at low temperature, but the characteristics of the hydrolysis with this enzyme are still unclear. The pearl oyster Pinctada martensii was used in this study as the raw material to illustrate the kinetic properties of protease 894. After investigating the intrinsic relationship between the degree of hydrolysis and several factors, including initial reaction pH, temperature, substrate concentration, enzyme concentration, and hydrolysis time, the kinetics model was established. This study showed that the optimal conditions for the enzymatic hydrolysis were an initial reaction pH of 5.0, temperature of 30°C, substrate concentration of 10% (w/v), enzyme concentration of 2 500 U/g, and hydrolysis time of 160 min. The kinetic characteristics of the protease for the hydrolysis of P. martensii were obtained. The inactivation constant was found to be 15.16/min, and the average relative error between the derived kinetics model and the actual measurement was only 3.04%, which indicated a high degree of fitness. Therefore, this study provides a basis for the investigation of the concrete kinetic characteristics of the new protease, which has potential applications in the food industry.  相似文献

The effects of different hydrolysis methods on peptidoglycan(PG) were assessed in terms of their impact on the innate immunity and disease resistance of Pacific white shrimp,Litopenaeus vannamei.PG derived from Bifidobacterium thermophilum was prepared in the laboratory and processed with lysozyme and protease under varying conditions to produce several different PG preparations.A standard shrimp feed was mixed with 0.05% PG preparations to produce a number of experimental diets for shrimp.The composition,concentration,and molecular weight ranges of the soluble PG were analyzed.Serum phenoloxidase and acid phosphatase activity in the shrimp were determined on Days 6-31 of the experiment.The protective activity of the PG preparations was evaluated by exposing shrimp to white spot syndrome virus(WSSV).Data on the composition of the PG preparations indicated that preparations hydrolyzed with lysozyme for 72 h had more low-molecular-weight PG than those treated for 24 h,and hydrolysis by protease enhanced efficiency of hydrolysis compared to lysozyme.SDS-PAGE showed changes in the molecular weight of the soluble PG produced by the different hydrolysis methods.Measurements of serum phenoloxidase and acid phosphatase activity levels in the shrimp indicated that the PG preparations processed with enzymes were superior to the preparation which had not undergone hydrolysis in enhancing the activity of the two serum enzymes.In addition,the preparation containing more low-molecular-weight PG enhanced the resistance of the shrimp to WSSV,whereas no increased resistance was observed for preparations containing less low-molecular-weight PG.These findings suggest that the immunity-enhancing activity of PG is related to its molecular weight and that increasing the quantity of low-molecular-weight PG can fortify the effect of immunity enhancement.  相似文献

A dynamic experiment for oil dispersion into a water column was performed with a 21 m long, 0.5 m wide, and 1 m high wind-driven wave tank. At wind velocity between 6-12 m/s and with the oil slide kept constant (about 1 um), the rate of the oil content increase in the water column could be approximated from the difference between the dispersion rate (R) of the oil slick and the coagulation rate (R') of the dispersed oil slick. Assuming the coagulation rate is directly proportional to the concentration of the water dispersed oil slick (i. e. R' =KC),, the integral form of the dynamic model can be expressed as C=R*[1-exp(-K*t)]/K and parameters R and K can be regressed with a computer. The relative deviation of model results from the experimental data was mainly less than 10%. Alpine ecosystems in permafrost region are extremely sensitive to climate changes.To determine spatial pattern variations in alpine meadow and alpine steppe biomass dynamics in the permafrost region of the Qinghai-Tibet Plateau,China,calibrated with historical datasets of above-ground biomass production within the permafrost region's two main ecosystems,an ecosystem-biomass model was developed by employing empirical spatialdistribution models of the study region's precipitation,air temperature and soil temperature.This model was then successfully used to simulate the spatio-temporal variations in annual alpine ecosystem biomass production under climate change.For a 0.44°C decade-1 rise in air temperature,the model predicted that the biomasses of alpine meadow and alpine steppe remained roughly the same if annual precipitation increased by 8 mm per decade-1,but the biomasses were decreased by 2.7% and 2.4%,respectively if precipitation was constant.For a 2.2°C decade-1 rise in air temperature coupled with a 12 mm decade-1 rise in precipitation,the model predicted that the biomass of alpine meadow was unchanged or slightly increased,while that of alpine steppe was increased by 5.2%.However,in the absence of any rise in precipitation,the model predicted 6.8% and 4.6% declines in alpine meadow and alpine steppe biomasses,respectively.The response of alpine steppe biomass to the rising air temperatures and precipitation was significantly lesser and greater,respectively than that of alpine meadow biomass.A better understanding of the difference in alpine ecosystem biomass production under climate change is greatly significant with respect to the influence of climate change on the carbon and water cycles in the permafrost regions of the Qinghai-Tibet Plateau.  相似文献
