Inorganic lead (Pb) is one of the most common environmental pollutants. Much evidence indicates that Pb exposure could directly affect fish growth and development. In this study, we investigated the cytotoxic effects of Pb on cytoskeletal protein stability at both protein and mRNA level in crucian carp Carassius auratus. Pb(NO3)2 treatment in concentration of 100 μmol/L resulted in decreased expression of both α- and β-tubulin but γ-tubulin as assayed with SDS-PAGE, Western Blot, and ELISA. In vivo and in vitro analyses on protein expression of tubulins are consistent. The effect of Pb on mRNA expression varied among different tissues. Our results suggest that cytotoxicity of Pb at protein translation level is stronger than at mRNA expression level.  相似文献

The structural feature of agar polysaccharides fromPorphyra haitanensis grown in south China and transplanted to the north was investigated by fractionation on DEAE—Sephadex A 50, chemical analysis, and infrared and13C-NMR spectroscopy. The agars composed mainly of charged molecules were eluted from DEAE—Sephadex A 50 with 1.0 mol/L NaCl solution from the southernP. haitanensis and with 0.5 mol/L NaCl from the northern one. The13C-NMR spectra showed that agarobiose [(1→3)-β-D-galactopyranosyl-(1→4)-3,6-anhydro-α-L-galactopyranose] and the biological precursor of agarobiose [(1→3)-β-D-galactopyra nosyl-(1→4)-6-sulfate-α-L-galactopyranose] were the major disaccharide repeating units in the charged fractions. The content of the biological precursor in the agar polysaccharides from southernP. haitanensis was higher than that in the northern one, the content of the biological precursor extracted from cold water was higher than that from hot water, and the content of 6-OMe-D-galactose in the southernP. haitanensis polysaccharides was higher than in the northern one. On the basis of scaling analysis,this paper presents a frontal geostrophic dynamic system which canbe used to describe the dynamics of an isolated bottom eddy and shows that five possible dynamicsubregimes are included in the system.A particle-in-cell method is used to investigate the behavior of the eddy in the two dynamic subregimes,with 1)nonlinear effect(ε)relevant to the sloping bottom-inducedbeta effect(β_1)and 2)seamount-induced beta effect(β_2)dominant.In the dynamic subregime of β_1~ ε~10~(-2)and β_2=0,the eddy can keep as a whole for a long time and migrates for a long distance.Inthe dynamic subregime where the seamount-induced beta effect (β_2~10~(-1))is dominant,the seamounttraps the water particles,causing the eddy to break up into a discrete set of eddies moving around the seamount.  相似文献

To investigate the effect of polypeptide fromChlamys farreri (PCF) on NHDFin vitro, we modeled oxidative damage on normal human dermal fibroblasts (NHDF) exposed to ultraviolet B (UVB). In this study, 3-[4,5-Dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide (MTT) and lactate dehydrogenase (LDH) were tested to measure cell viability. Enzymes including superoxide dismutase (SOD), glutathione peroxidase (GSH-PX), catalase (CAT) and xanthine oxidase (XOD) were determined biochemically. Total antioxidative capacity (T-AOC) and anti-superoxide anion capacity (A-SAC) were also determined. Ultrastructure of fibroblasts was observed under transmission electron microscope. The results showed that: UVB (1.176×10−4 J/cm2) suppressed the growth of fibroblasts and the introduction of PCF (0.25%–1%) before UVB reduced the suppression in a concentration-dependent manner. PCF could enhance the activities of SOD, GSH-PX and T-AOC as well as A-SAC. Also PCF could inhibit XOD activity, while it did not affect CAT activity. Ultrastructure of fibroblasts were damaged after UVB irradiation, concentration-dependent PCF reduced the destructive effect of UVB on cells. These results indicated that PCF can protect human dermal fibroblasts from being harmed by UVB irradiation via its antioxidant proerty. The polysaccharides of three species of ChineseGracilaria (Rhodophyta) were extracted by sequential solvent extraction and the chemical structures of the fractions obtained were investigated by using13C-NMR and IR spectroscopy. The13C-NMR spectra showed that the main extract, cold water fraction, fromG. sjeostedtii consisted of repeating disaccharides of agarobiose and (1→3)-β-D-galactopyranosy 1-(1→4)-α-L-galactopyranose 6-sulfate, the latter being a precursor to agarobiose and converted to agarobiose after alkali modification. The component of the 60% ethanol fraction fromG. textorii was composed mainly of agarose 6-sulfate, 6-O-methyl-agarose and some amount of agarose, whse structures remained unchanged after alkali treatment. The autoclave extract fromG. salicornia revealed a complex structure composed of 6-O-methyl-agarose, 2-O-methyl-agarose, branched 4-O-methyl-L-galactose and agarose 4-sulfate. All the substituents in these agaroses were stable against alkali treatment. The alkali extracts fromG. sjeostedtii andG. textorii showed the distinct spectra of amylopectin-like floridean starch, while that fromG. salicornia showed a mixture of agaroses and floridean starch. From the IR spectra the relative contents of total sulfate, 3,6-anhydro-galactose, 4-sulfate and 6-sulfate in some fractions of polysaccharides of the algae were qualitatively estimated by the ratios of absorption bands of 1250/2920, 930/2920, 850/2920 and 820/2920, respectively. A convenient and effective procedure was developed for the conversion of alcohol to olefin by tosylation and subsequent β-elimination promoted by silica gel in this study. Treatment of the alcohols with p-toluenesulfonyl chloride in pyridine at 0℃ affords tosylates which undergo β-elimination with silica gel in dichloromethane or chloroform at room temperature, yielding olefinswith high productivity.  相似文献

Carboxymethyl-chitosan and carboxymethyl-chitin were prepared with the methods developed in our laboratory.The DS(degree of substitution) and DD (degree of deacetylation) of the carboxymethyl-chitosan were 103.14% and 97.18% respectively,while the DS of the carboxymethyl-chitin was 96.37%.Their effects on human fibroblasts,intradermal irritation test,in vitro and vivodegradability,and biocompatibility were evaluated.The results indicate that the polysaccharides at low concentrations can facilitatethe growth of human fibroblasts and the carboxymethyl-chitosan at 100 μg mL<'-1> is the most effective.The polysaccharides at higherconcentrations,however,inhibit the growth of fibroblasts.The PⅡ (Primary Irritation Index) values of CM-chitosan and CM-chitinare both 0.0,which shows that they have no irritation reaction.Both of the polysaccharides show good degradability and biocompati-bility.Carboxymethyl-chitin degrades faster in vitro than carboxymethyl-chitosan.The latter,however,has no inflammatory reactionafter being implanted in vivo for 7 d and shows better biocompatibility.This study may provide a scientific basis for the use of car-boxymethyl-chitosan and carboxymethyl-chitin as biomaterials.  相似文献

Estradiol, or 17-β-estradiol (E2), the most potent naturally occurring estrogen, is involved in the hormone-immune system interaction in both mammals and fish. However, in vivo studies are largely limited, and little is known about whether E2 exerts similar effects on both female and male zebrafish (Danio rerio). Here, we show exposure of both sexes of D. rerio to 20 nmol/L E2 resulted in a significant increase in Vg1 expression, but caused little damage to the hepatocytes, suggesting that this is the optimum E2 concentration. Also, exposure to 20 nmol/L E2 for 20 days caused a marked increase in plasma IgM levels, but had little influence on the peripheral leukocyte density, providing the first evidence of a hormone-immune system interaction in this species.  相似文献

Full maturation of goldfish oocyte was inducedin vitro by 17 α-hydroxy-20β-dihydroprogesterone. The oocyte maturation involves GV migration to the periphery of the oocyte and germinal vesicle breakdown (GVBD). In the experiment, incubation duration for GVBD varied in different broods of oocytes. Generally, if the duration for GVBD was shorter than 6 h, oocytes would have a better chance to survive after maturation and insemination. The maturation of nucleus (GV) and cytoplasm are not synchronous. Cytoplasm maturation occurs several hs after GVBD. Oocytes inseminated 8–9 h after GVBD have the highest fertilizing and hatching rate. Fertilized ova maturedin vitro can develop to sexually mature adults capable of reproduction. To investigate the antitumor effect of bromophenol derivatives in vitro and Leathesia nana extract in vivo, six bromophenol derivatives 6-(2,3-dibromo-4,5-dihydroxybenzyl)-2,3-dibromo-4,5-dihydroxy benzyl methyl ether (1), (+)-3-(2,3-dibromo-4,5-dihydroxyphenyl)-4-bromo-5,6-dihydroxy-1,3-dihydroisobenzofuran (2), 3-bromo-4-(2,3-dibromo-4,5-dihydroxybenzyl)-5-methoxymethyl-pyrocatechol (3), 2,2′,3,3′-tetrabromo-4,4′,5,5′-tetrahydroxy-diphenylmethane (4), bis(2,3-dibromo-4,5-dihydroxybenzyl) ether (5), 2,2′,3-tribromo-3′,4,4′,5-tetrahydroxy-6′-ethyloxymethyldiphenylmethane (6) were isolated from brown alga Leathesia nana, and their cytotoxicity were tested by MTT assays in human cancer cell lines A549, BGC-823, MCF-7, B16-BL6, HT-1080, A2780, Bel7402 and HCT-8. Their inhibitory activity against protein tyrosine kinase (PTK) with over-expression of c-kit was analyzed also by ELISA. The antitumor activity of ethanolic extraction of Leathesia nana (EELN) was evaluated on S180-bearing mice. All compounds showed very potent cytotoxicity against all of the eight cancer cell lines with IC50 below 10 μg/mL. In PTK inhibition study, all bromophenol derivatives showed moderate inhibitory activity and compounds 2, 5 and 6 showed significant bioactivity with the inhibition ratio of 77.5%, 80.1% and 71.4%, respectively. Pharmacological studies reveal that EELN could inhibit the growth of Sarcoma 180 tumor and increase the indices of thymus and spleen to improve the immune system remarkably in vivo. Results indicated that the bromophenol derivatives and EELN can be used as potent antitumor agents for PTK over-expression of c-kit and considered in a new therapeutic strategy for treatment of cancer. The results obtained from the Cu(II)%-pH curve of the system in the presence of added amino acid indicated that the added amino acids of higher concentration can inhibit the Cu(II)-σ-MnO2 ion exchange and that their inhibiting effects decrease in the following sequence when the initial amino acid concentrations are 20.00 ppm: histidine > cystine > glutamic acid > proline > alanine > glycine. The determination of amino acid in solution performed with an amino acid analyser showed that added amino acids react chemically on the surface of the σ-MnO2 According to thermodynamic equilibria relations in the system, amino acids can be changed to amines because of their decarboxylation on the surface of the σ-MnO2. Considering (1) the effect-inhibiting factor of amine FA(β, Ka, pH) representing the complex potential of amine with Cu(II) in solution and (2) the acidic constant of −NH 3 + group in the amine molecule representing the ability of cation exchange of the amine with σ-MnO2, the inhibiting seqence of added amino acid of 20.00 ppm (Eq.(1)) is reasonable. Thus added amino acids do inhibit Cu(II)-σ-MnO2 ion exchange because (1) the amino acid and/ or amine, which is the decarboxylate of the corresponding amino acid, complex with Cu(II) in solution and compete with σ-MnO2 for Cu(II); (2) the cation exchange of the amino acid and/or amine reduce the exchange site on the surface of the σ-MnO2. The results obtained from the isotherms indicated that the theory of interfacial stepwise ion exchange can be extended to the Cu(II) σ-MnO2 ion exchange system in the presence of added amino acid.  相似文献

R-phycoerythrin fromPorphyra haitanensis exists in two aggregation states with different molecular weights. A more highly aggregated form, RPE I, was chromatographed on Bio-Rex 70 column with urea solution (pH 3.0) as eluent, and the molecular weights of the 3 subunits (α, β, γ) obtained were determined on SDS-PAGE at 18000, 19200 and 30000, respectively. α subunit carried two phycoerythrobilin (PEB); β subunit, three PEB and one phycourobilin (PUB); γ subunit, one PEB and three PUB chromophores. The molar ratio of α, β, and γ subunits of RPE I was 6: 6: 1, and their subunit composition was confired to be (αβ)6γ on account of the molecular weight of RPE I, 232000. A lower aggregated form, RPE II, contained α and β subunits similar to those of RPE I, but its subunit composition was the (αβ) monomer of RPE. The antithrombotic and antiplatelet effects of two fucoidan fractions with low molecular weight and different sulfate content from Laminaria japonica were compared in order to examine the influence of chemical character on their antithrombotic activity and the possible mechanism. Both LMW fucoidan fractions exhibited favorable antithrombotic activity in an Fecl3-induced arterial thrombosis. The antithrombotic activity of LMW fucoidan was related with decrease of TXB2 and whole blood viscosity and hematocrit. LMW fucoidan showed a correlation between anticoagulant, antiaggregant and antithrombotic effects in vivo. For LMW fucoidan, antithrombotic activity required high dose of 5-10 nmol kg-1, concomitantly with increase in anticoagulant activity and inhibition of platelet aggregation. Administration of LMW fucoidan significantly promoted the 6-keto-PGF1α content and decreased the TXB2 content, indicating its inhibition of tissue factor pathway and regulation of metabolism of arachidonic acid. By comparison, highly sulfated fucoidan LF2 with Mw 3900 seemed to be a more suitable choice for antithrombotic drug for its antithrombotic activity accompanied with specific inhibitory activity on platelet aggregation, low anticoagulant activity and low hemorrhagic risk in vivo.  相似文献

A laboratory experiment was conducted to assess the bioaccumulation of Pb2+ and its effects on growth, morphology and pigment contents of Spirulina (Arthrospira) platensis. The specimen cultured in Zarrouk liquid medium was treated with various initial metal concentrations (0, 5, 10, 30, 50 and 100 μg mL−1). The growth of S. platensis was adversely affected by Pb2+ at high concentrations (30, 50 and 100 μg mL−1). However, at low concentrations (5 μg mL−1), Pb2+ could stimulate its growth slightly. The pigment contents (chlorophyll α and β carotene) were decreased in a dose-dependent manner. The highest reductions (67% and 53% respectively in chlorophyll α and β carotene) were observed in 100 μg mL−1 treatment group. The LC50 (96 h) of Pb2+ was measured as 75.34 μg mL−1. Apart from a few cases of filament breakages at elevated concentrations (50 and 100 μg mL−1), morphological abnormalities are not specific. Metal bioaccumulation increased with Pb2+ concentrations, but decreased with exposure time. The maximum accumulated amount was 188 mg g−1 dry weight. The bioconcentration factor (BCF) reached to a peak at day 2, followed by a gradual reduction for all the exposure concentrations. S. platensis is able to tolerate considerably high Pb2+ concentrations. Consequently it can be used as a potential species to remove heavy metal from contaminated waters.  相似文献

Retention efficiencies (RE) of scallop (Chlamys farreri), oyster (Crassostrea gigas) and mussel (Mytilus edulis) in a flow-through system were measured to understand the short-term response to various particle and organic matter concentrations. By comparing the RE of C. farreri with that of C. gigas and M. edulis, we gained further knowledge on the feeding physiological characteristics of C. farreri and ascertained the possible cause of high summer mortalities of this species. The experimental feeding conditions included natural differences in the abundance and composition of suspended seston, as well as conditions in which seston abundance and composition were manipulated by adding natural silt or cultured microalgae. The results show that in natural sea water, the minimum particle size for maximal retention in M. edulis, C. gigas and C. farreri was approximately 4, 6, and 8 μm, respectively; the RE of 2-μm (equivalent spherical diameter) particles was 17%, 19%, and 8%, respectively; and the relative standardized RE was 58%, 49%, and 18%, respectively. In C. gigas and M. edulis, the minimal particle size for maximal retention did not change with food quality (organic content). C. farreri was more sensitive to fluctuations in particle concentration and organic content. With particle concentration increase, the minimal particle size for maximal retention in scallop shifted to large particles and the RE for 2-μm particle decreased from 8% in natural seawater to 1.6%–6% in silt-enriched groups. With organic content increase, the minimal particle size for maximal retention shifted from 8 to 5 μm in natural seawater.Variation in RE of C. farreri with food conditions and the relationship between lower RE and smaller particle size may hamper C. farreri from food taking due to the decrease in the size distribution of phytoplankton in Sungo Bay.  相似文献

We extracted marine low-temperature lysozyme (MLTL), a novel lysozyme, from a marine microorganism through fermentation. Our previous study suggested that a low molecular weight (16 kDa) may exert anti-tumor activity through antiangiogenesis. In this study, we extracted a high weight (39 kDa) and investigated its antiangiogenic activity in vivo and in vitro. Using zebrafish embryos as an in vivo study model, we found that treatment with MLTL significantly inhibited the growth of subintestinal vessels (SIVs) in a dose-dependent manner and that 400 μg/ml MLTL was sufficient to block the growth of SIVs. An in vitro study conducted using human umbilical vein endothelial cells (HUVECs) revealed that MLTL suppressed the proliferation, migration and tube formation of HUVECs in a dose-dependent manner. Interestingly, assays by flow cytometry and DNA electrophoresis indicated that MLTL was able to induce apoptosis of HUVECs. Moreover, further study demonstrated that the disruption of intracellular Ca2+ homeostasis may play an important role in MLTL induced apoptosis of HUVECs. Taken together, the results of this study demonstrate for the first time that MLTL inhibits angiogenesis through its pleiotropic effects on vascular endothelial cells and induces apoptosis through regulation of cellular Ca2+ levels. The results of this study also revealed a possible mechanism underlying the antiangiogenic effect of MLTL and suggested that MLTL may be a promising new antiangiogenic agent for use in cancer therapy.  相似文献

The living morphology and infraciliature of two rare marine ciliates, Placus salinus Dietz, 1964 and Strombidium apolatum Wilbert and Song, 2005, collected from the coastal waters near Qingdao, China, were investigated by in vivo observation and protargol impregnation technique. The improved diagnosis for Placus salinus is as follows: medium-sized marine Placus, in vivo (50–60)μm×(30–40)μm; cell elliptical to barrel-shaped; 28–31 somatic kineties; single macronucleus usually ellipsoid and one micronucleus located in the indention of the macronucleus; one contractile vacuole posteriorly positioned. Strombidium apolatum is characterized by: marine strombidium (40–60)μm×(30–45)μmm in vivo, cordiform in shape with somewhat pointed posterior end and conspicuous apical protrusion; extrusomes prominent, about 15μm in length and evenly arranged along the circle kinety; about 16 collar and 5–6 buccal membranelles; one elongate macronucleus and one micronucleus; circle and ventral kineties consisting of about 53 and 45 dikinetids respectively.  相似文献

The effect of Mg2+, NaCl and citric acid on the accumulation of β-carotene inDunaliella Salina was studied. The experimental results showed that 10.5 mmol/L Mg2+, 5 mol/L NaCl, 3 μmol/L citric acid, and CO2 are favorable forDunaliella Salina cell growth and β-carotene accumulation. After 144 h culture under the above conditions, theDunaliella Salina biomass increased by 7.18 times; β-carotene reached 9.61%. 【目的】探究方格星虫酶解物促进伤口愈合的作用。【方法】将小鼠分为空白对照组、方格星虫酶解物组和云南白药组进行止血实验,并建立小鼠全皮层创伤模型。将方格星虫酶解物制成膏状,每天上药一次。于术后3、5、7 d分批各处死5只小鼠,固定7只用于拍照。术后观察创面愈合情况,测定伤口愈合率、炎症因子、转化生长因子-β1和创面羟脯氨酸含量等指标。【结果】相比于空白对照组,方格星虫酶解物组显著缩短止血时间,提高愈合率,缩短掉痂时间、创面宽度,调节炎症因子、转化生长因子-β1表达,促进胶原蛋白沉积,伤口愈合较好,残留疤痕不明显。【结论】方格星虫酶解物具有促进伤口愈合功效,其作用机制与止血、调节炎症因子、转化生长因子-β1水平、促进胶原蛋白沉积有密切联系。  相似文献
