通过对高效液相色谱法(HPLC)测定水产饲料中三聚氰胺含量的方法进行优化,得到最佳色谱条件和前处理方法,建立了水产饲料中三聚氰胺的检测方法。结果表明:以体积比8∶92的乙腈、庚烷磺酸钠离子对试剂为流动相,柱温40℃,流速1.2 mL/min,以波长236 nm检测;以提取液为18 mL50%(φ)乙腈水+1 mL乙酸锌+1 mL1mol/L盐酸,淋洗液3 mL水和3 mL甲醇,洗脱体积6 mL作样品前处理。三聚氰胺加标回收率为85%~100%,相对标准偏差(RSD)小于5%,线性范围为0.2 mg/L~5.0mg/L,相关系数0.999 8,方法检出限为0.2 mg/kg。  相似文献

以菠萝 (AnanascomosusMerr)为原料 ,应用改良的方法对菠萝天然酶活力进行测定 ,对菠萝酶活力变化进行了初步探讨。结果表明 :测定未成熟、成熟菠萝酶活力分别为 10 6 .10和 6 5 .85u/mL ;菠萝肉、皮、整果汁酶活力分别为 86 .98、5 0 .72、5 5 .2 1u/mL ;室温贮藏 ,酶活力取样时为 5 4.97u/mL ,第 4天达最高值 72 .78u/mL ,第 8天降至 4 3.6 6u/mL ;4℃贮藏 ,酶活力取样时为 5 4.97u/mL ,第 2天达最高值71.6 8u/mL ,第 4天降至 5 0 .12u/mL ,即不同温度下贮藏不同时间酶活力变化是先升高后降低  相似文献

为探索城市污泥资源化的途径,以城市污水处理厂的剩余污泥为原料,采用氯化锌活化法制备污泥吸附剂,考察了活化剂浓度、固液比、活化温度、活化时间4种因素对制备污泥吸附剂碘吸附值的影响,并对吸附剂进行了表征分析。实验结果表明:氯化锌浓度3mol/L、固液比1:2、活化温度550℃和活化时间40min为最佳制备条件,此条件下制得的污泥吸附剂碘吸附值为408.94mg/g,比表面积199.196m2/g,总孔体积为0.481cm3/g,微孔体积为0.089cm3/g,平均孔径为4.83nm。与商品活性炭相比,污泥吸附剂的碘吸附值和比表面积较小,孔径较宽,微孔所占比例较小,主要以过渡结构为主。  相似文献

胰蛋白酶的固定化采用吸附-交联法。在单因素试验基础上,通过正交试验,获得了胰蛋白酶固定化的最佳条件:酶与载体体积比为2:1,吸附时间为10h,pH=4.5,缓冲液浓度为0.05mol/L,戊二醛的质量分数为0.1%。此条件下制得的磁性酶活力回收在50%左右,相对酶活力达74%,该磁性酶对啤酒澄清防止冷浑浊有明显效果。  相似文献

柠檬酸/酒石酸可使方解石产生矿物溶蚀及生物矿化等现象,研究它们之间的反应规律对富含有机酸环境中方解石矿物的稳定性评估及相应的地球化学效应研究具有重要意义。通过批量平衡法,研究开放系统条件下方解石与柠檬酸/酒石酸之间的反应规律及对应的溶液化学变化。结果表明:(1)初始浓度为20 mg·L-1条件下,初始pH值由7.7升至9.7,柠檬酸与酒石酸的反应去除量均显著下降,这是由于方解石表面的静电效应及溶液中HCO-3与CO■的竞争效应不断增强,对两种酸的表面吸附反应起抑制作用。(2)初始pH值为7.7及8.3条件下,初始浓度为20 mg·L-1的两种酸与方解石反应7 h后便达到平衡,pH值和Ca浓度较空白值变化不大,辅证了反应的主导机制为表面吸附。(3)在0~900 mg·L-1浓度范围内,柠檬酸和酒石酸在初始pH值为8.3条件下,其反应去除量均能用Langmuir吸附模型很好地拟合,辅证了以上的反应去除机制均为表面吸附;初始pH值为7.7条件下,柠檬酸的反应去除量可用Langmuir...  相似文献

采用批量振荡吸附平衡法设计针铁矿和蒙脱石对菲的吸附解吸试验,对比研究了针铁矿和蒙脱石对菲的吸附解吸行为,并考察了不同K+浓度的溶液对蒙脱石吸附菲的影响,比较分析了线性吸附模型和Freundlich吸附模型描述矿物吸附等温线的准确性,并从吸附热力学角度探讨了矿物的吸附机理。结果表明:针铁矿和蒙脱石对菲的吸附解吸均表现出明显的非线性和解吸滞后现象;相对于线性吸附模型来说,针铁矿和蒙脱石对菲的吸附解吸更符合Freundlich吸附模型;与蒙脱石相比,针铁矿对菲的吸附更为显著,且具有更好的稳定性;溶液中软阳离子K+的存在使蒙脱石对菲的吸附能力得到显著提高;菲在蒙脱石和针铁矿上的吸附过程是一个自发放热,同时伴随着熵值减小的过程;随着温度的升高,蒙脱石和针铁矿对菲的吸附能力均减弱。  相似文献

破碎尼罗罗非鱼(Oreochromis niloticus)头肾组织,用RPMI-1640培养基制成细胞悬液,添加至两层不同浓度的Percoll分离液液面,离心,得非特异性细胞毒性细胞(Non-specific cytotoxic cell,NCC),优化下层Percoll浓度、离心力、离心时间。结果表明,当下层Percoll体积分数为43%,上层Percoll体积分数为34%,离心力为500 g,离心时间为30 min时,尼罗罗非鱼NCC得率最高,为71.28%。  相似文献

研究了木瓜蛋白酶的酶解温度、酶浓度、酶解时间、加水量、pH值对青鳞鱼可溶性蛋白得率的影响 ,探讨了α 氨态氮含量与青鳞鱼可溶性蛋白得率的关系。结果表明 :温度小于 5 0℃ ,随着酶解温度的提高、酶浓度的增加、酶解时间的延长青鳞鱼可溶性蛋白得率及α 氨态氮的含量均增加 ,α 氨态氮含量增加比可溶性蛋白得率的增加明显 ;加水量的增加 ,可溶性蛋白得率增加 ;pH值对水解效果及可溶性蛋白得率影响不大。综合考虑 ,最佳水解条件为 :温度 5 0℃ ,酶浓度为 1 5 0 0u/g ,酶解时间 80min ,m原料 :m水 =1∶2 ,pH 6 8。酶解所得可溶性蛋白经氨基酸分析仪检测 ,其氨基酸总量为 49 47mg/mL ,必需氨基酸占氨基酸总量的 41 0 % ,游离氨基酸含量为 6 3mg/mL ,肽含量为 87 2 %。  相似文献

以小杂鱼或低值鱼为原料 ,采用常温溶剂萃取法进行脱脂 ,研究了不同溶剂、溶剂混合比、流速和萃取时间对鱼类脱脂的影响。结果表明 ,在常温下用等体积的异丙醇和乙酸乙酯混合溶剂 ,以流速 4~ 8mL/min连续萃取 4~ 6h ,脱脂鱼肉的脂肪平均含量w为 0 .19%,脱脂率高达96 .2 6 %。该方法的溶剂损耗量为加热溶剂萃取法的 1/3倍。  相似文献

Polyphenols from the ethanol extracts of Sargassum tenerrimum (ST) with potent antiallergic effects were studied to optimize separation process through column chromatography. The adsorption and desorption characteristics of three widely used adsorbents: macroporous resin, silica gel, and polyvinylpolypyrrolidone (PVPP), were critically evaluated respectively and studied for the optimization of preparative separation of polyphenols. Static operations on these adsorbents showed that macroporous resin had the best adsorption and desorption capability among the three adsorbents. Dynamic adsorption and desorption with macroporous resin packed column were also conducted to optimize the parameters such as: with the optimal values shown in brackets, the concentration of extract solution (4 times diluted), pH value (6–7), adsorption speed (3 BV h−1, bed volumes/per hour), concentration of ethanol (80%), elution speed (3 BV h−1) and elution volume (7 BV). The chromatographic process so optimized gave a purity of 62.43% from the crude polyphenols, providing a promising basis for large scale preparation of bioactive polyphenols upon further scaling up tests.  相似文献

The present study investigated the removal of inorganic arsenic from Pinctada martensii enzymatic hydrolysate through unmodified resin (D296) and Zr(IV)-loaded chelating resin (Zr-D401). By loading Zr to macroporous chelating resin D401, the as exchange adsorption active sites are generated. This transforms D401 from a material that does not have the arsenic adsorption capacity into a material that has excellent arsenic exchange adsorption capacity. The static adsorption experiments were conducted to investigate the optimal removal condition for D296 and Zr-D401. The experimental results show that: the optimum condition for D296 is that T= 25°C, pH = 5, resin additive amount = 1 g (50 mL)?1, and contact time = 10 h, the corresponding arsenic removal rate being 65.7%, and protein loss being 2.33%; the optimum condition for Zr-D401 is that T=25°C, pH = 8, resin additive amount = 1 g (50 mL)?1, and contact time=10 h, the corresponding arsenic removal rate being 70.3%, and protein loss being 4.65%. These results show that both of the two resins are effective in arsenic removal for preserving useful substance. Our research provides scientific evidence and advances in the processing technology for heavy metal removal in shellfish.  相似文献

The adsorption and desorption of algae Cladophora fascicularis and their relation with initial Cd^2+ concentration, initial pH, and co-existing ions were studied. Adsorption equilibrium and biosorption kinetics were established from batch experiments. The adsorption equilibrium was adequately described by the Langmuir isotherm, and biosorption kinetics was in pseudo-second order model. The experiment on co-existing ions showed that the biosorption capacity of biomass decreased with an increasing concentration of competing ions. Desorption experiments indicated that EDTA was efficient desorbent for recovery from Cd^2+. With high capacities of metal biosorption and desorption, the biomass of Cladophora fascicularis is promising as a cost-effective biosorbent for the removal of Cd^2+ from wastewater.  相似文献

研究无乳链球菌(Streptococcus agalactiae)ZQGY08的培养与生长特性,探讨脑心浸液(Brian Heart Infusion,BHI)培养基的营养因子、初始pH值、溶氧、温度等因素对无乳链球菌生长的影响,并采用正交实验法对无乳链球菌培养基营养因子添加量进行优化。结果表明,无乳链球菌生长的最适初始pH为7~8,培养温度为28~37℃,振荡转速为180~200 r/min。在BHI培养基中添加20 mg/mL的酵母粉和2 mg/mL的葡萄糖即可在对数期获得最大细菌量。  相似文献

Batch experiments were conducted to study the effects of low-molecular-weight organic acids(LMWOAs) on desorption of Hg from two kinds of soils in the eastern Shandong Province,China.Of all LMWOAs,oxalic and citric acids were chosen as the representatives in this experiment because they are most common in soils.Desorption of Hg increases with the increase of extracting time,until the highest desorption amount at 6-8 h,and then declines.It indicates that timeliness is very important when organic acids are used for phytoremediation in contaminated soils.When the concentrations of organic acids increase from 1.0 mmol/L to 10.0 mmol/L,Hg desorption increases significantly regardless of oxalic or citric acid.Compared with oxalic acid,citric acid is more effective to enhance the desorption of Hg,especially from fluvo-aquic soil.With the increase of initial pH value from 3 to 8,Hg desorption decreases directly in the presence of oxalic acid.However,Hg desorption first decreases,then increases,and eventually decreases again in the presence of citric acid due to the variation of competitive ability between chelating of citric acid and adsorption of soil in different pH values.Citric acid showed greater ability to release Hg from soils than oxalic acid.According to the results of the present study,it is found that the bioavailabilities of heavy metal can be improved with selecting suitable types and concentrations of organic acid amendment and reasonable soil condition.  相似文献

研究了海豚链球菌(Streptococcus iniae)ZJMX04的培养与生长特性,探讨培养基的pH值、气体环境、温度等因素对海豚链球菌生长的影响,并采用正交实验法对海豚链球菌培养基生长因子添加量进行优化。结果表明,海豚链球菌培养基的最适pH为7~8,最适培养温度为30℃,振荡培养对海豚链球菌的生长影响不明显。培养基优化研究表明,在BHI培养基中添加10mg/mL的酵母粉和2mg/mL的葡萄糖即可在对数期获得最大细菌量。  相似文献

The kinetics of Pb adsorption in seawater by surface sediments from Xiamen Bay was simulated and the effects of seawater conditions were studied . When water temperature changed from 20 to 30℃, pH from 5 to 8 and salinity from 30 to 4 . the adsorption rate increased by about 50% , one order of magnitude and 5 times, respectively . A negative exponential relation between the adsorption rate and salinity has been found . The adsorption processes can be simulated by the exchange- equilibrium kinetic model . Adsorption rate constant, apparent activation energy and equilibrium adsorptive capacity have been quantitatively determined . The adsorption of Pb by sediments is essentially a chemical action .  相似文献

Algal organic materials (AOMs) are one critical factor affecting the efficiency of modified clays used for the mitigation of harmful algal blooms (HABs). This study was conducted to develop a deeper understanding of the mechanisms and factors affecting the adsorption of AOMs onto modified clays. Sodium alginate (polysaccharide) and kaolinite modified with polyaluminium chloride (PACl) were used as AOMs and modified clay model substances, respectively, and the effects of modifier dosage, contact time, solution pH and ionic strength were investigated through batch adsorption experiments. Kinetics revealed that the alginate adsorption rate was described well by a pseudo-second order model. PACl effectively enhanced the adsorption capacity of kaolinite and increased the adsorption rate, and the optimum additive amount of PACl was 5%. The experimental data fitted both the Freundlich and Langmuir adsorption equations well. The adsorption thermodynamics for alginate onto modified clays suggests that alginate adsorption is a spontaneous process. The adsorption of alginate onto modified clays was highly dependent on pH, with a decrease in adsorption observed with increased pH to 9.48, but the opposite was true above pH 9.48. Finally, adsorption increased with increasing ionic strength.  相似文献
