The behaviour of two species of foraminifera (Allogromia laticollaris and Ammonia beccarii) towards living and dead bacteria and inorganic particles was monitored using a cell-permeant fluorescent nucleic acid stain. The foraminifera were studied in seawater containing fluorescently labelled dead and living bacteria, and/or polystyrene particles of the same size as a control. Time-lapse observations under a fluorescence microscope clearly revealed pseudopodial transport of stained bacteria and uptake of bacteria inside the foraminifera. In contrast, no uptake of polystyrene particles was ever observed, although the foraminifera collected these particles and deposited them along the outside of the test. We conclude that foraminifera distinguish food and non-food particles while collecting. There appears to be a variation of uptake rate and final amount of ingested and digested bacteria. These variations occur between individuals of different size and species, and between sampling times (September 2001 and July 2002).  相似文献

The release of enteric pathogenic bacteria in aquatic environments poses the problem of the fate of these bacteria under the effects of environmental factors (solar radiation, salt concentration, temperature, nutrient level, pH, competition). Frequently, these bacterial cells, potentially pathogenic, enter into a non-culturable state on routine bacteriological plating media. However, the use of direct detection methods DAPI stained cells allows the visualization of these Viable but Non Culturable cells VNC. But, beyond the characterization of the viability of the cells electron transport activity, metabolic activity, membrane integrity, structure and/or quantity of DNA, what happens with the virulence of these cells? This problem was experimentally investigated according to the bacterial model Salmonella Typhimurium. The virulence of this strain, which is the agent of the murine typhoid, was evaluated on a mouse model. Experimentally, the effects of some environmental factors on the survival and on the maintenance of virulence of Salmonella Typhimurium were measured in microcosms exposed to UV radiation (four germicidal lamps 8 mW s−1 cm−2, wave length: 254 nm, salt concentration (Sea Salt Sigma, 37, nutrient starvation. The microcosms were simultaneously submitted to these three factors, with variable exposure times. For each of those times, the viability of the nonculturable cells which became nonculturable because of the exposure to the three factors was measured through different physiological states notable in the cells, after using different fluorescent dyes. The stained cells were observed by epifluorescence microscopy and analysed by image cytometry. So, the cellular populations are characterised by enumeration of respiring bacteria CTC, [39], metabolising bacteria YEK, [22] modified, bacteria owning an undamaged cytoplasmic membrane LD, Live/Dead BacLight Viability Kit. Molecular Probes Inc.; we also determined the quantity and/or structure of DNA of the cells fluorescence level of DAPI stained cells), After exposure to the three factors for one hour 13.56 J cm−2, while the plate count cell density rapidly decreased from #107 cells mL−1 to0.1 cell mL−1, physiological states of these viable but non-culturable cells are similar to those of non-exposed cells. On the other hand, after exposure for three hours, only 10 % of the cells deposit a CTC formazan-crystal and 20 % are substrate responsive cells enlarged cells in presence of Yeast Extract and Cephalexin: YEC. Half of the cellular population presents an undamaged cytoplasmic membrane and the level of fluorecence of DAPI stained cells is close to 85 %, which shows that the DNA of these cells is weakly damaged. After exposure to the three experimental factors for24 hours 315 J cm−2, weak replies to the physiological tests used in this study to characterize the viability of the non-culturable cellular population are observed CTC: 4 %; YEC: 2 %; LD: 11.8 % while the fluorescence level of DAPI stained cells remains firm at 80 %. At the same time, the virulence expression of VNC cells of Salmonella Typhimurium, evaluated by intraperitoneal injection to the mouse route which excludes uncontrolled parameters, unlike the per os route does not seem to be correlated with the cellular viability such as it has been evaluated in this study. A 30 min exposure (6.73 J cm−2) to the three environmental factors, leading to the non-culturability of almost the entire exposed cell population 0.08 culturable cell mL−1 whereas the level of viability of those culturable cells is closed to the one of non-exposed cells. The injection of 1000 of those cells 0.001 culturable cells in 100 μL inoculated into the mouse a group of ten mice does not cause any mortality four weeks post-inoculation, whereas the injection of the same dose of non-exposed cells leads to the death of all mice in the group one week post-inoculation. According to our preliminary experiments on Salmonella Typhimurium, the loss of the state of culturability and the loss of virulence towards mice by intra-peritoneal route, because of the exposure to associated effects of UV irradiation 254 nm, salinity 37 and nutrient starvation, seem to be concomitant.  相似文献

本文应用电子显微镜技术观察了从东海(北纬26°30'-30°00',东经124°-129°)海水和海底泥中分离到的8株发光细菌和一株衰失发光性能菌株[#7032(-)]的菌体形态、鞭毛和细胞内部超微结构,结果表明#7032、#7032(-)、#1025菌体有一根粗的端生鞭毛、大量的周生鞭毛和微细的丛生鞭毛.#7032和#7032(-)菌外部形态、鞭毛结构相似,在磷钨酸负染色中鞭毛可显示鞘和核心结构,鞘可呈环饼状或成段脱落.  相似文献

Live (Rose-Vengal stained) benthic foraminifera were studied along a transect across the main area of organic matter deposition in the Cape Blanc upwelling region. The faunal analyses suggest that at the shallowest station (1200 m) the benthic ecosystem is permanently influenced by the upwelling, whereas at the deepest stations (3010 and 2530 m depth) the ocean bottom is subject to significant organic influxes only in summer. The vertical zonation of foraminiferal species in the sediment shows a close correspondence with the depth distribution of oxic respiration, nitrate and sulphate reduction. It is suggested that this linkage is caused by the presence of various stocks of anaerobic and sulphate- and nitrate-reducing bacteria. Deep infaunal foraminiferal species are thought to feed selectively, either on the bacterial stocks or on nutritious particles produced by bacterial degradation of more refractory organic matter. As such, foraminiferal microhabitats are only indirectly controlled by pore water oxygen concentrations.  相似文献

Assemblages of live (Rose-Bengal-stained) and dead benthic foraminifera and stable oxygen and carbon isotopic composition of live benthic foraminifera were studied in and outside a bacterial mat composed of the large sulphur-oxidizing bacteria Thioploca and Beggiatoa from the oxygen-minimum zone off Pakistan (NE Arabian Sea). Two cores from the same Multicorer retrieved a bacterial mat and ambient sediment. The dominant species (Globobulimina affinis, G. turgida, Bolivina pacifica, B. pseudopunctata, Uvigerina peregrina and Buliminella tenuata) in both cores are characteristic for dysoxic oxygen minimum zone conditions. The most significant difference between the two cores is the reduced number of stained benthic foraminifera (SBF) in the top 0.5 cm of the bacterial mat. Faunal densities of stained species are more than four times higher in the sediment surface sample (0–0.5 cm) outside the bacterial mat, at a distance of only 1.5 m. All stained species, however, observed outside the Beggiatoa/Thioploca mat were also observed in the core with the mat. Two species, Virgulinella fragilis and Bolivina dilatata, occur exclusively in the core with the bacterial mat. The diversity within the bacterial mat core is thus slightly higher than outside. Furthermore, the abundances of the species Buliminella morgani, B. tenuata and Alliatina primitiva are substantially higher in the bacterial mat than outside. Globobuliminids, on the other hand, seem to prefer the conditions outside the bacterial mat and are five times more frequent in the core taken outside the bacterial mat. Benthic foraminifers inhabit a broader microhabitat range outside the bacterial mat (∼5 cm) than within (3.5 cm). A marked decrease in SBF abundances was observed at the level of a black sulphur-rich layer which is interpreted to mark the shallow redox front below the bacterial mat. Stable carbon isotope analyses on live benthic foraminifera do not support a relation of the investigated Beggiatoa/Thioploca mat to a constant or seasonal seepage of methane at the continental slope off Pakistan. Surprisingly, however, stable oxygen isotope values of many species and especially of U. peregrina decrease with depth, which calls into question the suitability of U. peregrina as a recorder of bottom-water δ18O.  相似文献

采用光镜技术研究海鳗肝脏、胆囊和胰腺的显微结构。结果表明:海鳗肝小叶不明显,肝门管区不典型。肝细胞较大,呈多边形,糖原和脂滴丰富。核呈圆形或卵圆形,1~2个。胆囊与胆管结构类似其他鱼类。胰腺长条形,紧贴肠左壁,独立存在,少量分布在胆管周围及胆囊壁上。外分泌部由腺泡和各级排泄管组成,内分泌部即胰岛,分散存在于外分泌部之间,胰岛细胞由A,B,D三种细胞组成。A细胞数量较少,位于胰岛周边,夹于B细胞之间,胞质颗粒染成红色;B细胞数量最多,位于胰岛周边,胞质颗粒染成橘黄色;D细胞一般位于胰岛中部染色浅淡的区域,胞质染淡蓝色。  相似文献

Lakes in the beech (Nothofagus sp.) forest foothills between Greymouth and Hokitika, Westland, are characterised by amber‐coloured waters. Microbiological, biochemical, and chemical analyses of samples from nine such lakes during March 1976 indicate that they are unproductive. Microbial biomass was intensively investigated in heavily stained Lakes Haupiri and Lady and in least‐stained Lake Brunner. A combination of adenosine triphosphate assays, Utermöhl phytoplankton counts, and autoradiographic biomass determinations revealed a greater bacterial than algal biomass in heavily stained lakes, a highly unusual phenomenon which suggests that most of the production of organic matter was allochthonous. Autochthonous productivity was low, probably because biologically available phosphorus was very scarce and because discoloration reduced the transparency of the water. Hypqlimnetic oxygen depletion was slight during this period of maximum thermal stratification; decomposition of organic matter was apparently incomplete. Reactive phosphorus did not increase appreciably in partially deoxygenated hypolimnia. Much of the allochthonous organic matter originating from beech forests and their soils may be partially resistant to biodegradation.  相似文献

用5个实验围隔研究了对虾池悬浮颗粒上的附着细菌,结果表明:附着细菌数量波动在0.65×104~23.49×104个/cm3之间,平均为(4.57±3.64)×104个/cm3,占水体细菌总数的1.47%×1.41%(0.28%~8.73%).附着细菌数为1、2、3、4个的附菌颗粒占总附菌颗粒的比数分别为40%、27%、15%及11%,多于4个附着细菌的附菌颗粒数仅占总附菌颗粒数的7%;总悬浮颗粒中的附菌颗粒所占比例为52%;附着细菌数与水体总悬浮颗粒数关系最为密切,与水温及总细菌数也具有显著相关性,但与水层POC及DOC含量却未呈现显著的相关性.  相似文献

Chaetognaths are an abundant component of the plankton and are usually the most important carnivores. Head or tail damage is often found in chaetognath specimens from the laboratory and the sea. Examination by light and scanning electron microscopy (SEM) of specimens with such damage has revealed that bacterial invasion occurs in chaetognaths under laboratory and natural conditions. Living chaetognaths which cease feeding are found to have large numbers of attached bacteria and some parts of their body become opaque. Once they are attacked in this manner the body appears to be a mass of bacteria. Examination of field samples has shown that similar bacterial attacks occur in the sea. Bacterial growth is common inside the muscle tissue of field specimens. Up to 13.3% of two dominant species of chaetognaths in Suruga Bay are attacked by bacteria.  相似文献

Abstract. Three species of Alvinellidae are found on walls of some hydrothermal chimneys at 13o N on the East Pacific Rise (EPR); numerous epibiotic bacteria were associated with two of these polychaetes, Alvinella pompejana and Alvinella caudata , but not with the third, Paralvinella grasslei. Many studies suggest that chemoautotrophic bacteria play a prominent role in deep-sea hydrothermal vent ecosystems. Particularly important are the intracellular bacteria present in the giant pogonophores and bivalve molluscs.
The abundant epibioses observed on these Alvinella species are unusual and the relationships between these bacteria and the worms are unknown. Previous results, including 13C values, morphological insertion of the bacteria on the epidermis, presence of ribulose biphosphate carboxylase, and fixation of 14C bicarbonate in some bacteria and in the dorsal epidermis, suggest that a trophic relationship could exist between Alvinella and its epibiotic bacteria. The present study compared the incorporation area of 14C bicarbonate in the 3 species. Paralvinella , which is devoid of epibiotic bacteria, served as a control.  相似文献

- Abundance of total bacteria, heterotrophic bacteria, coliform and oil degradable bacteria and their relations with salinity and suspended matter in the frontal waters in the Hangzhou Bay are described. The results obtained show that the highest abundances of total bacteria , heterotrophic bacteria are encountered around the front and near the northern shore of the Hangzhou Bay, where a positive correlation ( r =0. 70) between the total bacteria abundance and the total suspended matter (TSM) is persent. Moreover,the abundance of oil degradable bacteria being an indicator of oil pollution, is also high in that region.  相似文献

为探讨环境因素对异养细菌丰度的影响,2016年9月至2017年8月通过月度航次调查对北部湾北部海域异养细菌丰度的时空分布特征进行了系统研究。结果表明,调查海区异养细菌丰度介于(2.75~56.86)×105 cell/mL,平均值为(11.01±6.31)×105 cell/mL。各季节细菌丰度从高至低依次为:夏季、春季、冬季、秋季。异养细菌丰度由近岸海域向西南深水区方向逐渐降低,在近岸浅水区垂直分布均匀,在水深大于20 m的海区出现季节性分层现象:表层细菌丰度较高,底层细菌丰度较低。主成分分析显示温度对异养细菌时空分布有重要影响,秋、冬季异养细菌丰度与温度呈显著负相关,在春、夏季呈显著正相关。细菌丰度与盐度呈显著负相关,说明海水盐度变化是细菌时空分布重要影响因素。异养细菌丰度与叶绿素a和溶解氧含量呈显著正相关,表明浮游植物初级生产过程影响了异养细菌的时空分布。在秋、冬和春3季异养细菌丰度与营养盐水平呈显著负相关,二者关系受浮游植物生物量间接影响。异养细菌时空分布差异取决于环境条件的变化,温度、盐度、叶绿素a和溶解氧含量是影响异养细菌丰度分布的主要因素。  相似文献

The significance of the microheterotrophic community of bacteria and Protozoa in marine systems is reviewed. In surface waters, bacterial production may be linked to cycles of photosynthetically produced dissolved organic matter released from living phytoplankton. The bacteria then form a principal food resource for grazing Protozoa which are themselves potentially available for exploitation by higher trophic levels. Recent data suggest that the interaction between Protozoa and their bacterial prey may be a good deal more complicated than this simple predator-prey relationship implies. Ciliates may show either "steady-state" or strongly cyclical relationships with their bacterial prey, whose community may in turn be supported to a large extent by the release of dissolved organic matter by the ciliate population. This implies that the low standing stocks of bacteria commonly recorded in many pelagic systems may be sustained, as well as exploited, by bacterivorous Protozoa. In other systems, bacterial production may enter the larger zooplankton directly without involvement of the bacteria-Protozoa-bacteria loop. However, the microheterotrophic community of bacteria and Protozoa in general represents a competitive pathway down which a significant proportion of carbon from primary production may be channelled, compared with that flowing directly into the grazing zooplankton and thence to fish. Despite the high primary production which characterizes temporally unstable areas such as the southern Benguela upwelling system, the carrying capacity for fish may therefore be considerably less than that which might be anticipated for a simple herbivore-dominated food chain.  相似文献

海洋浮游异养细菌(以下称浮游细菌)存在个体间的异质性,对浮游细菌异质性的研究是理解细菌生产、代谢及其在生物地球化学循环中重要作用的基础。流式细胞术具有快速分析大量细菌个体的能力,除了用来分析样品中细菌的丰度外,流式细胞术和细胞染色技术结合,被用来研究自然海水中浮游细菌在细胞膜完整性、CTC(5-氰基-2,3-二(4-甲基苯基)四唑氯化物)呼吸功能和核酸含量三个方面的异质性。尽管国外已经有较多研究,但我国在这方面的研究尚比较缺乏,本文综述了自然海区浮游细菌这三项异质性的研究现状,介绍了不同海区(主要是近岸)浮游细菌的异质性及其随环境的变化,以期推动我国在此领域的研究工作。目前对其异质性的变化机制尚没有很好的理论解释,在全球变化的大背景下,针对大洋深海和极区,有关长期变化、全球变暖以及酸化等对浮游细菌异质性的影响研究需要加强。  相似文献

用平皿自然沉降法测定灵山岛空气微生物及肠道致病菌,结果表明该岛处于空气微生物中-轻度污染状态,以轻度污染为主,其中细菌为主要成分,也舍少量肠病菌,空气微生物显出某种时空变化势态,且与气温、人群活动等因素相关,推测谈岛脆弱的生态环境易被外界干扰,应引起关注。  相似文献

采用实时荧光定量PCR、高通量测序等方法对湛江湾沉积物中四个月份的反硝化细菌与厌氧氨氧化细菌的多样性和丰度进行了分析。结果表明:湛江湾沉积物中反硝化细菌和厌氧氨氧化细菌丰度在四个月份的变化和空间分布趋势为:nirS型反硝化细菌在二月份最高,四月份最低,且其平均丰度有从湛江湾湾内向湾口附近呈现先升高再降低的趋势;nirK型反硝化细菌丰度在九月份最高,十一月份最低;nosZ型反硝化细菌在四月份最高,其余月份变化不大;厌氧氨氧化细菌丰度在九月份最高,二月份最低。通过相关性分析结果表明,亚硝酸盐、铵盐等共同调控着湛江湾沉积物中反硝化和厌氧氨氧化细菌丰度变化。系统发育分析表明:湛江湾中存在着一些广泛分布的反硝化细菌,但也生活着一些新奇的nirK型和nosZ型反硝化细菌。对于厌氧氨氧化细菌而言,其主要属于浮霉菌门及Candidatus Scalindua属,具有较高的耐盐性,另外湛江湾海区的厌氧氨氧化细菌也生活着一类在其他地方没有的新分支。典范对应分析分析结果表明:硝酸盐显著影响湛江湾反硝化细菌和厌氧氨氧化细菌的群落结构。湛江湾沉积物中反硝化细菌和厌氧氨氧化细菌存在特殊的竞争与共存的关系,且由亚硝酸盐、硝酸盐、pH等多种环境因子共同驱动。  相似文献

海水中致病菌种类繁多、致病性强,对水产养殖和人体健康存在着潜在危害。传统的检测方法培养过程复杂繁琐、耗时长、效率低,难以满足实际工作的需求。因此,建立海水中致病菌的快速、灵敏、准确的检测技术势在必行。本文综述了流式细胞技术、分子生物学方法、免疫学方法和生物传感器技术等在海水致病菌检测中的应用,总结了上述方法在海水致病菌检测中的研究现状。  相似文献

We present evidence that there are significant interactions between heterotrophic microorganisms, doliolids and Fritillaria within intrusions of nutrient-rich Gulf Stream water stranding on the continental shelf. During the summer of 1981 cold, nutrient-rich water from below the surface of the Gulf Stream was repeatedly intruded and stranded on the continental shelf off northeastern Florida. On August 6 old, stranded Gulf Stream water depleted of nitrate occupied the lower layer on the outer shelf. The upper water was continental shelf water, older but of undefined age. On August 6 free-living bacteria were >106ml−1 everywhere at all depths, an order of magnitude greater than normal bacterial numbers on the northeastern Florida continental shelf. Over 10 days the numbers of free bacteria doubled while bacteria attached to particles increased by a factor of four. The adenylate/chlorophyll ratio showed that phytoplankton dominated the lower layers of intruded water, while the surface water became increasingly dominated by heterotrophic microorganisms (bacteria and protozoa) over 10 days. There were significant, negative correlations between bacteria and doliolids and between bacteria and Fritillaria. Regions of maximum bacterial numbers did not coincide with locations of salp swarms. The increased numbers of bacteria at all depths in a highly stratified system in which most phytoplankton are in the lower layer suggests a diverse source of bacterial growth substrates, some of which involve zooplankton as intermediaries. Production of autotrophs is more than twice that of microheterotrophs on average, but because of their differential distribution, microheterotrophs are the dominant biomass in much of the surface water and may be significant in energy flux to metazoan consumers as well as competitors for mutually useable sources of nutrition.  相似文献

对患弧茵病的养殖斑节对虾作作了细菌学分析,取典型症状的淑死对虾的血淋巴和肝胰腺组织进行细菌分离,纯化,保种,并将分离菌进行回接感染,发现有很强的致病作用,且产生与自然发病相拟的症状,从感染弧菌病的病虾体内进行细菌的分离,纯化和保种,经栓验发现,分离菌和回接感染后的再分离菌均为革兰氏阴性短杆菌,极生单鞭毛,有动力,对O/129弧菌抑制剂敏感,发酵葡萄糖产酸不产气,氧化酶和过氧化氢酶反应呈阳性,不利用乳糖,蔗糖,蜜二糖,能利用柠檬酸盐,甘露醇,果糖和半乳糖,在无盐的蛋白胨水和10%NaCl的蛋白砾水中不生长,在3%,6%,8%NaCl的蛋白砾水中生长良好,吲哚反应呈阳性,V-P反应呈阴性,赖氨酸脱酸酶,鸟氨酸脱羧酶,明胶酶和淀粉酶试验呈阳性,β-半乳糖苷酶,色氨酸脱氨酶,苯丙氨酸脱氨酶和精氨酸双水解酶反应呈阴性,可还原硝酸盐,经鉴定,该病原菌为副溶血弧菌Vibria parahaemolyticus.  相似文献
