Effects of temperature, salinity and light intensity on growth rates of Gracilaria lichenoides and G. tenuistipitata var. liui Zhang et Xia were tested. Eight to ten levels of each factor were first tested separately. The best growth rate was obtained under the conditions of 32°C, 30 and 240 μmol/(m2·s) for G. lichenoides, and 24°C, 20 and 200 μmol/(m2·s) for G. tenuistipitata, respectively. Then a uniform design was used to evaluate the optimal combinations of the three factors. The best conditions for the highest daily specific growth rates (% increase in wet weight) are determined to be 31.30°C, 32.10, and 287.23 μmol/(m2·s) for G. lichenoides (16.26%/d), and 25.38°C, 21.10, and 229.07 μmol/(m2·s) for G. tenuistipitata (14.83%/d), respectively. The effects of temperature, light intensity, salinity, and initial pH on the growth and fatty acid composition of Pinguiococcus pyrenoidosus 2078 were studied for eicosapentaenoic acid (EPA) production potential. The fatty acid composition was assayed by gas chromatography-mass spectrometry, which indicated that the main fatty acids were C14:0, C16:0 and EPA. The highest EPA percentage 20.83% of total fatty acids was obtained at 20℃ with the temperature being set at 20, 24, and 28℃. Under different salinities and light intensities, the highest percentages of total polyunsaturated fatty acids (PUFAs) and EPA were 17.82% and 31.37% of total fatty acids, respectively, which were achieved at salinity 30 and 100μmol photon m-2s-1 illumination. The highest percentages of total PUFAs and EPA were 38.75% and 23.13% of total fatty acids, respectively, which were reached at an initial pH of 6 with the test range being from 5.0 to 9.0.  相似文献

Over the past few years, harmful algal blooms (HABs), such as red tides, have been frequently observed in coastal zones worldwide. The natural allelopathic interactions among macroalgae and red tide microalgae can alter the structure and succession of aquatic ecosystems. We investigated the influence of four environmental factors (temperature, salinity, light, and pH) on the allelopathic effects of the macroalgae Corallina pilulifera on red-tide forming Heterosigma akashiwo under laboratory conditions. Each of the factors had four levels: temperature (15, 20, 25, and 30°C), salinity (10, 20, 30, and 40), light (20, 100, 200 and 400 μmol/(m2?s)), and pH (5.5, 7, 8.5, and 10. Two-factor experiments were designed for each two environmental factors, with six combination treatments (temperature-salinity, temperature-light, temperature-pH, salinity-light, salinity-pH, and light-pH). Results showed that the allelopathic effect was significantly influenced by temperature, salinity, light, and pH. As single factors, the low temperature (15°C), low salinity (10), high-intensity light (400 μmol/(m2?s)), and high pH (10) treatments substantially enhanced the allelopathic effect. The strongest allelopathic effect of C. pilulifera on H. akashiwo was observed under the following treatments: 15°C and salinity of 40, 25°C and pH 10, 25°C with medium- to high-intensity light at 200–400 μmol/(m 2 ?s), 400 μmol/(m2?s) and salinity of 10, 400 μmol/(m2?s) and pH 10, and pH 10 with a salinity of 40.  相似文献

When cultured under certain environmental conditions (25°C, light intensity 80 μmol/m2·s, LD 12/12, in enriched seawater medium with 7×10−4 mol/L NO3−N, 1.56×10−4 mol/L, PO4−P and supplements of other elements like Mn, Fe, I, etc.), male and female gametophytes ofU. pinnatifida kept growing vegetatively and propagated fast at average daily fresh weight increase rate of about 20%. The empirical formulaG m=G o·3m was established to estimate the output of vegetative gametophytes. Vigorous vegetative gametophyte cells began to form reproductive structures (oogonium and spermatangium, when the temperature was lower than 25°C and other environmental factors were kept optimal. The sufficient supply of gametophyte cells provided enough seeds for raisingUndaria sporelings on production scale. Controlled cross-breeding experiments using selected male and female gametophyte clones which increase their cell number by mitosis instead of meiosis were also carried out in vitro. Juvenile sporophytes from the cross-breeding had almost the same length and width increase rates as those of the control. The fact that vegetative gametophytes can be purposely selected, propagated quickly, cross-bred, with the resulting sporophytes having almost the same characteristics leads to a new way to select desiredUndaria strains for long time use without losing the desired economic characteristics. Chondrus is a type of commercially produced red seaweed that widely used for food and carrageen extraction. Although the natural life history of the alga had been well understood, the factors influencing development of the tetraspore and carpospore remain poorly understood. In the perspective of seedling resources, the regulation of early development is crucial for the seedling nursing; therefore, it is necessary to understand the physiological influences during its early development. In this study, we studied the effects of temperature and irradiance on the early development of Chondrus ocellatus Holm under laboratory conditions. The released tetraspores and carpospores were cultivated at different temperatures (10–28°C) and irradiances (10, 60 μmol photons m−2s−1) with a photoperiod of 12L:12D. The results indicate that both tetraspores and carpospores are tolerant to temperatures of 10–25°C, and have the highest relative growth rate at 20°C. Irradiance variances influenced the growth of the discoid crusts, and the influence was more significant with increasing temperature; 60 μmol photons m−2s−1 was more suitable than 10 μmol photons m−2s−1. The optimum temperature and irradiance for the development of seedlings was 20°C and 60 μmol photons m−2s−1, respectively.  相似文献

3-factor experiment was used to study the combined effects of temperature, irradiance and salinity on the growth of an HAB species diatomSkeletonema costatum (Grev.) Cleve. The results showed that temperature (12, 19, 25, 32 °C), irradiance (0.02, 0.08, 0.3, 1.6)×1016 quanta/(s·cm2)) and salinity (10, 18, 25, 30, 35) significantly influenced the growth of this species. There were interactive effects between any two of and among all three physical factors on the growth. In the experiment, the most optimal growth condition forS. costatum was temperature of 25°C, salinity of 18–35 and irradiance of 1.6×1016 quanta/(s·cm2). The results indicatedS. costatum could divide at higher rate and were more likely to bloom under high temperature and high illumination from spring to fall. It was able to distribute widely in ocean and estuary due to its adaptation to a wide range of salinities. The rates of photosynthesis and respiration ofPorphyra, Laminaria andUndaria were determined with an oxygenmeter at different temperatures in an enclosed continuous-flow system. The temperature coefficients of respiration and the net primary productivities for the three species were calculated. The spectacular rates of primary production were found to be as high as 270 g C m−2 forP. yezoensis, 2200 g C m−2 forL. japonica and 160 g C m−2 forU. pinnatifida annually. The low respiratory rates of these three species as well as low light compensation points especially at low temperatures are considered as a reason to save heat energy, resulting in a high productivity, especially in case ofLaminaria. During a survey for marine microalgal resources, we isolated a rare marine euglenoid from the coastal waters of Qingdao, China in 2009, and established a pure culture. Electron microscopic and molecular phylogenetic (18S rDNA and 16S rDNA sequences) analyses revealed a close affinity with Eutreptiella gymnastica, a bloom-forming species. Different culture conditions were monitored to understand optimal E. gymnastica growth characteristics. The optimal growth conditions in a batch culture of this isolate were 20°C, 160 μmol photons/(m2×s) of white light, and a salinity of 10-31. Nutrient experiments demonstrated that growth increased dramatically with a phosphorus concentration greater than 72 μmol/L. Understanding the effect of culture conditions on E. gymnastica may help understanding the blooming mechanism of this alga in its natural environment.  相似文献

A filamentous clone of Polysiphonia urceolata was regenerated from segments cut from the fronds of gametophytes. Unlike wild thalli with short virgate branchlets, the clone was filamentous with few branches. Many transparent trichoblasts arose from pericentral cells during the induction culture, but these were seldom observed during normal growth. The trichoblasts were uniseriate, often colorless, and formed lobed rhizoids rapidly when they came into contact with solid substrates. In addition to morphological characteristics, the photosynthetic properties and growth conditions of the clone differed from those of the mother plant. Cross-gradient light and temperature culture experiments revealed that the most favorable conditions for culture of the filamentous clone were 22°C and 95-120 μE/(m2-s) light intensity. The photosynthetic light saturation value for filaments was approx. 100 μE/(m2-s), which is far lower than that of wild thalli. These results could be used to develop techniques for mass cultures of P. urceolata in photobioreactors for production of seed stock or bioactive products.  相似文献

Oxygen uptake under starvation and short periods of sudden temperature change was measured for larval herring (Clupea harengus L.) reared at average temperature of 7.3, 11 and 12.9°C. Larval stages between first feeding and premetamorphosis were used. For comparison, the routine oxygen uptake (ROU) was also investigated and followed the relationshipQ=0.974+0.174tW −0.210, whereQ is in μg/(mg·h),W is dry body weight in mg andt is temperature in °C. The oxygen uptake under starvation (SOU, deprived of food for 24 h) was different from the routine when the larval dry weight was less than 0.6–0.8 mg; it increased with temperature and body weight giving the relationshipQ=1.568+0.110tW 0.380; if the larval dry weight was more than 0.6–0.8 mg, it reverted to the norm (Q=1.704+0.078tW −0.349). The oxygen uptake was tested in short periods (3 h) of sudden temperature changes in six groups: 7.3 to 11, 7.3 to 12.9, 11 to 7.3, 11 to 12.9, 12.9 to 7.3 and 1.29 to 11°C. The oxygen uptake in the 7.3 to 12.9 and 12.9 to 7.3°C groups varied in a way similar to that of the larvae under starvation. The other four groups' oxygen uptake were more or less normal.  相似文献

The energy budget ofBellamya earuginosa in a shallow algal lake, Houhu Lake (Wuhan, China) was investigated by the measurement of flesh production (32.8 kJ/(m2·a)), egestion (337.7 kJ/(m2·a)), metabolism (246.7 kJ/(m2·a)), and estimation of excretion (21.4kJ/(m2·a)). The net growth efficiency of the species is about 10.9%, which accords with the generally reported value for gastropods. In addition, the relationships between starvation respiration (R, mgO2/(Ind·d)), body weight (Wd, mg in dry wt) and temperature (T, °C) were also determined. The regression equationR=0.044Wd 0.537 e 0.061T was obtained by the least square method, The measured SDA of the species is 26.51% of its gross metabolism. The energy flow ofBranchiura sowerbyi was studied for the first time in China in a shallow macrophytic lake, Biandantang Lake, Hubei Province. The energy flow was calculated from the measurement of flesh production (12.5241kJ/m2a), egestion (517.7302kJ/m2a), metabolism (38.3273 kJ/m2a), and excretion (4.3798kJ/m2a). The net growth efficiency of the species is about 22.7%, which accords well with the generally reported value for oligochaetes. In addition, the relationship between starvation respiration (R, mgO2/ind·d), wet weight (Ww, mg) and temperature (T, °C) were also measured, with the regression function beingR=0.008Ww0.736 e0.050T. Effects of water temperature (17, 21, 25, 30 and 35℃) and body size (14.75-281.41 g initial body weight) on food consumption, growth, feed conversion, and dry matter content in orange-spotted grouper fed to satiation were investigated. The combined effect of temperature (T, ℃) and body weight (W, g) on maximum food consumption (Cmax, g/day) was described as: InCmax=-7.411+0.828 InW+0.317T4).004 7T2, and the optimum feeding temperature was 33.9℃. The combined effect of temperature and body weight on growth (G) was described as: lnG=-4.461-0.2081nW+0.394T-0.006 3T^2. The optimum growth temperature was 31.4℃, whereas overall growth rates were high at 25, 30 and 35 ℃. Feed conversion efficiencies (FCE, %), increasing first and then decreasing with increasing temperature, averaged from 1.8 to 2.1 in terms of dry weight of food fish. The optimum temperature for FCE tended to be lower than that for growth or feeding. Dry matter content increased with both increasing water temperature (17, 25, 30 and 35℃) and body weight, and the combined effect of temperature and body weight on dry matter content (DM, %) was described as: lnDM =3.232+0.01 4 lnW-0.004 4T+0.001 2TInW.  相似文献

1 Introduction It has been reported that habitat nutrient availability frequently limited plant growth and determined species dominance and abundance in natural communities (Miao et al., 2000). Nutrient availability is also a main regulator of aquatic primary production. Human-induced nutrient enrichment results in die-back of native vegetation and alteration of species dominance in various aquatic eco- systems (Miao et al., 2000; Green and Galatowitsch, 2002). Sperm motility is important in reproductive biology. To understand the physiological ecology characteristics of sperm of Sepiella maindroni, we studied the activation rate, active time, and lifespan of sperm to evaluate the influence of external factors, including temperature, salinity, pH, glucose, NaCl, KCl, MgCl2, CaCl2, EDTA-2Na and artificial seawater on sperm motility. The results show that the appropriate activation and motility condition for sperm were: temperature 20?30°C (optimum 25°C), salinity 10.6?30.6 (optimum 15.9) and pH 8.0?8.6 (optimum 8.0-8.4). Sperm activity varied in different concentrations of glucose, NaCl, KCl, MgCl2 and CaCl2. None of the selected concentrations of EDTA-2Na solution could activate the sperm. The activation rate in artificial seawater devoid of Ca2+, Mg2+ or HCO3ˉ was low. The results should help further studies on the preservation and activation of squid sperm.  相似文献
