The present paper evaluates the temporal and spatial impact of heavy metal containing (Cr, Zn, Cu, Ni, Cd and Fe) effluents of brass, electroplating and tannery industries on chemical and microbiological characteristics of affected soil and groundwater. Therefore, samples were drawn from three sites, S1, S2 and S3, with a longitudinally distant from effluent drain of 20, 200 and 700 m, respectively. In general, the metals concentration exceeded the standard limits not only in the discharged effluents but also in the soil and groundwater. The significant reduction of microbial biomass C and N, soil respiration and microbial coefficient with increasing metal content from S3 to S1 was recorded, the effect being more pronounced in summer. However, the Cmic/Nmic ratio decreased whilst the metabolic quotient ($q_{{\rm CO}_{{\rm 2}} } $ ) increased with increasing metal concentration in soil. The content of Zn (11.5 mg/L) and Bacillus sp. was at maximum in groundwater of brass and electroplating industry site, whereas that of tannery site contained maximum Cr (2.34 mg/L) and Enterobacter sp. The toxic metals adversely polluted the groundwater which made it to harbours Escherichia coli beyond the prescribed limit. To check the soil and groundwater pollution, eco‐friendly measures involving improved effluent treatment technology and site‐specific application of treated effluent are recommended.  相似文献

Spatial distribution (horizonal and vertical) of groundwater microbial communities and the hydrogeochemistry in confined aquifers were studied approximately along the groundwater flow path from coteau to plain in the Nomhon area, Qinghai‐Tibet plateau, China. The confined groundwater samples at different depths and locations were collected in three boreholes through a hydrogeological section in this arid and semi‐arid area. The phylogenetic analysis of 16S rRNA genes and multivariate statistical analysis were used to elucidate similarities and differences between groundwater microbial communities and hydrogeochemical properties. The integrated isotopic geochemical measurements were applied to estimate the source and recharge characteristics of groundwater. The results showed that groundwater varied from fresh to saline water, and modern water to ancient water following the flowpath. The recharge characteristics of the saline water was distinct with that of fresh water. Cell abundance did not vary greatly along the hydrogeochemical zonality; however, dissimilarities in habitat‐based microbial community structures were evident, changing from Betaproteobacteria in the apex of alluvial fan to Gammaproteobacteria and then to Epsilonproteobacteria in the core of the basin (alluvial‐lacustrine plain). Rhodoferax, Hydrogenophaga, Pseudomonas, and bacterium isolated from similar habitats unevenly thrived in the spatially distinct fresh water environments, while Sulfurimonas dominanted in the saline water environment. The microbial communities presented likely reflected to the hydrogeochemical similarities and zonalities along groundwater flowpath.  相似文献

This study explores linkages between the microbial composition and hydrochemical variables of pristine groundwater to identify active redox conditions and processes. Two confined aquifers underlying the city of Qianjiang in the Jianghan Plain in China were selected for this study, having different recharge sources and strong hydrochemical gradients. Typical methods for establishing redox processes according to threshold concentration criteria for geochemical parameters suggest iron or sulphate reduction processes. High‐throughput 16S rRNA sequencing was used to obtain diversity and taxonomic information on microbial communities. Instead of revealing iron‐ and sulphate‐reducing bacteria, salt‐ and alkali‐tolerant bacteria, such as the phylum Firmicutes and the class Gammaproteobacteria, and in particular, the family Bacillaceae, were dominant in the downstream groundwater of the first aquifer that had high ion concentrations caused by the dissolution of calcite and dolomite; meanwhile, the heterotrophic microaerophilic families Comamonadaceae and Rhodocyclaceae prevailed in the upstream groundwater of the first aquifer. Sulphate‐reducing bacteria were extremely abundant in the upstream groundwater of the second aquifer, as the SO42? concentration was especially high. Methanogens and methanotrophs were predominant in the downstream groundwater of the second aquifer even though the concentration of SO42? was much higher than 0.5 mg L?1. The microbial communities, together with the geochemical parameters, indicated that the upstream region of the first aquifer was suboxic, that Fe(III) and Mn(IV) reductions were not the main redox processes in the downstream groundwater of the first aquifer with high Fe and Mn concentrations, and that the redox processes in the upstream and downstream regions of the second confined aquifer were SO42? reduction and methanogenesis, respectively. This study expands understanding of the linkages between microbial communities and hydrogeochemistry in pristine groundwaters and provides more evidence for identifying active redox conditions and processes.  相似文献

The performance of one pilot‐scale and two full‐scale membrane bioreactors (MBR) were evaluated based on the control of main operational parameters, composition of microbial community and pathogens concentration in the treated outlet. Plants were designed for 0.75 m3/day (A), 60 m3/day (B) and 30 m3/day (C). Inlet and outlet samples were monitored for chemical oxygen demand (COD), biological oxygen demand, total suspended solids, ammonia nitrogen concentration (NH4–N), nitrate nitrogen concentration, total Kjeldahl nitrogen, total phosphorus and phosphate phosphorus concentration concentrations. Plants showed good COD removal: 91.9% for Plant A, 97.8% for Plant B and 94.2% for Plant C. The targeted nitrogenous ion was NH4–N due to the requirements for outlet limits. NH4–N removal was moderate for Plant A (73.3%) and Plant B (86.1%) and excellent for Plant C (>99%). Excellent phosphorus removal was achieved by Plant A (average outlet concentration was 0.7 mg/L, efficiency 84.7%). Unsatisfactory results for phosphorus removal were achieved at the full‐scale plants due to operational problems. The dependency between the extracellular polymeric substances increase and decreasing mixed liquor volatile suspended solids for both lab and full‐scale plants was confirmed. Soluble microbial product concentrations were reduced by 65–68% after coagulant dosage for Plant A. Outlets from the MBR plants were monitored for the presence of pathogens (thermotolerant coliforms, Escherichia coli, intestinal Enterococci and culturable microorganisms at 22 and 37°C). The treated effluent from Plant A, B and C met Czech national legislation regarding reuse criteria (standards) for environment, irrigation and swimming purposes. Plants B and C were not able to achieve requirements for potable water and personal hygiene quality standards.  相似文献

Understanding basic controls on aquifer microbiology is essential to managing water resources and predicting impacts of future environmental change. Previous theoretical and laboratory studies indicate that pH can influence interactions between microorganisms that reduce ferric iron and sulfate. In this study, we test the environmental relevance of this relationship by examining broad‐scale geochemical data from anoxic zones of aquifers. We isolated data from the U.S. Geological Survey National Water Information System for 19 principal aquifer systems. We then removed samples with chemical compositions inconsistent with iron‐ and sulfate‐reducing environments and evaluated the relationships between pH and other geochemical parameters using Spearman's rho rank correlation tests. Overall, iron concentration and the iron‐sulfide concentration ratio of groundwater share a statistically significant negative correlation with pH (P < 0.0001). These relationships indicate that the significance of iron reduction relative to sulfate reduction tends to increase with decreasing pH. Moreover, thermodynamic calculations show that, as the pH of groundwater decreases, iron reduction becomes increasingly favorable relative to sulfate reduction. Hence, the relative significance of each microbial reaction may vary in response to thermodynamic controls on microbial activity. Our findings demonstrate that trends in groundwater geochemistry across different regional aquifer systems are consistent with pH as a control on interactions between microbial iron and sulfate reduction. Environmental changes that perturb groundwater pH can affect water quality by altering the balance between these microbial reactions.  相似文献

Storm events are major transporters of faecal microbial contaminants, but few studies have reported storm loads or concentration dynamics in relation to discharge or other pollutants, notably fine sediment. Episodically, high loads of faecal contamination during storm flows impact downstream uses of water bodies, particularly contact recreation and shellfish harvesting. We examined the storm dynamics of Escherichia coli, turbidity and discharge in the mixed land use Motueka catchment (2047 km2; 60% forest and 19% pasture) to gain insights into E. coli sources and transport. We also explored different approaches for calculating E. coli loads. Discharge and field turbidity were recorded continuously, and E. coli concentrations were sampled during events, over a 13‐month period near the mouth of the Motueka River. E. coli loads were estimated by interpolation, averaging estimators and by using linear regression with smearing correction of the log‐transformed variables: discharge, turbidity, and both turbidity and discharge. The annual E. coli load was dominated (~98%) by export during events. Comparison of monthly monitoring with the intensive storm monitoring campaign suggests that simple stratification of the sampling into storm and baseflow would greatly improve export estimates. E. coli peak concentrations always preceded discharge and turbidity peaks (which had similar timing). Turbidity can be a useful surrogate for faecal microbes in smaller catchments, but in the Motueka turbidity was no better for predicting E. coli concentration than discharge. Runoff from grazed pasture and direct deposition from livestock are probably the ultimate E. coli sources in the Motueka catchment. However, in‐channel stores seem to dominate E. coli dynamics during events and account for the typical feature of bacterial concentrations peaking ahead of discharge and turbidity. This study demonstrates the importance of storm events to faecal microbial loads and shows that E. coli concentration dynamics may contrast with those of turbidity. A scheme for the fractionation and concentration of suspended particulate matter (SPM) from natural waters has been tested. This scheme involves the sequential use of three fractionation techniques—sieving, continuous flow centrifugation and tangential flow filtration to collect gram amounts of SPM over the entire particulate and colloidal size range. The separation scheme is able to process large samples (ca. 1001), within reasonable times (ca. 1 day) and the apparatus is portable. Reproducibility and potential artifacts introducing during the fractionation and concentration of SPM, particularly when tangential flow filtration is used, are discussed. It has been shown that there is a systematic increase in the content of organic carbon, Mg, Ca, Na, K, Cu and Zn with decreasing particle size, highlighting the importance of the colloidal ( < 1 μm) fraction.  相似文献

Lack of filtration and rapid transport of groundwater and particulate matter make karst aquifers susceptible to bacterial contamination. This study utilized quantitative polymerase chain reaction (qPCR) to examine the transport and attenuation of two nonvirulent isolates of Escherichia coli (E. coli) in relation to traditional groundwater tracers (rhodamine WT dye and 1-µm diameter latex microspheres) in a karst-conduit aquifer in central Kentucky. Bacterial isolates were labeled with stable isotopes (15N and 13C). All tracers were detected more than 6 km downstream from the injection site and demonstrated overlapping breakthrough curves, with differential transport observed between the two bacterial strains. The E. coli isolate containing the kps gene (low attachment) arrived at sampling sites 1.25 to 36 h prior to the bacterial isolate containing the iha gene (high attachment) and was detected in samples collected following storm events in which the iha isolate was not detected. The storage potential of contaminants within karst systems was demonstrated by the remobilization of all tracers during storm events more than 1 month after injection. Bacteria-sized microspheres were more easily remobilized during periods of increased discharge compared to other tracers. The study demonstrated that molecular biology techniques such as qPCR can be utilized as a sensitive analysis of bacterial tracers in karst aquifers and may prove to be a more sensitive analytical technique than stable isotope analysis for field-scale traces.  相似文献

Arsenic in groundwater has been a concern in South and Southeast Asia for more than a decade. We explore here the possibility that hydrogeologic factors recently shown to influence the distribution of arsenic might also affect the level of contamination of shallow (<20 m) wells with microbial pathogens. A total of 96 shallow tube wells in two nearby villages of Bangladesh were surveyed during the wet and dry seasons, along with 55 deeper wells in neighboring villages. One of the two villages is located in a particularly sandy environment where recharge is rapid and shallow wells contain little arsenic. Shallow aquifers in the other village are capped with an impermeable clay layer, recharge is an order of magnitude slower, and arsenic levels are high. The fecal indicator E. coli was detected in 43% of shallow wells, compared with 12% of deeper wells. More shallow wells contained E. coli during the wet season (61%) than during the dry season (9%). In the wet season, a higher proportion of shallow wells in the village with low arsenic levels (72%) contained E. coli compared with the village having high arsenic levels (43%). Differences in arsenic and E. coli distributions between the two sites are likely due to the differences in permeability of near‐surface sediments although differences in average well‐depth between the two villages (9 ± 4 vs. 15 ± 3 m) may play a role as well. Hydrogeologic conditions that favor high levels of fecal contamination but low levels of arsenic in shallow groundwater should be taken into account during arsenic mitigation throughout South and Southeast Asia.  相似文献

Wastewater production in a hospital laundry and the treatment of the most critical wastewater stream, are assessed. Hospital laundry wastewaters are hazardous to the environment due to their high pollutant concentrations and the chemicals added during the clothes washing process. Heterogeneous photocatalysis with UV, O3 and TiO2 and their possible combinations were used for disinfection purposes. A ramp‐type reactor was used for TiO2 (P25 Degussa) fixation and for photochemical diffusion of the ozonized air. After assessing 5‐day biological oxygen demand, chemical oxygen demand, pH, turbidity, and surfactant content, and checking for the presence of thermotolerant coliforms and Escherichia coli, it was concluded that UV/O3/TiO2 was the best process/combination, yielding a 100% disinfection rate and a microbiological inactivation of 0.5070 min–1 for E. coli and of 0.5505 min–1 for thermotolerant coliforms.  相似文献

The detection of microbiological contamination in drinking water from groundwater wells is often made with a limited number of samples that are collected using traditional geochemical sampling protocols. The objective of this study is to examine the variability of fecal indicator bacteria, as observed using discrete samples, due to pumping. Two wells were instrumented as multilevel piezometers in a bedrock aquifer, and bacterial enumeration was conducted on a total of 166 samples (for total coliform, fecal coliform, Escherichia coli, and fecal streptococci) using standard membrane filtration methods. Five tests were conducted using pumping rates ranging from 0.3 to 17 L/min in a variety of purging scenarios, which included constant and variable (incremental increase and decrease) flow. The results clearly show a rapid and reproducible, 1 to 2 log‐unit decrease in fecal indicator bacteria at the onset of pumping to stabilized, low‐level concentrations prior to the removal of three to five well volumes. The pumping rate was not found to be correlated with the magnitude of observed bacterial counts. Based on the results, we suggest sampling protocols for fecal indicator bacteria that include multiple collections during the course of pumping, including early‐time samples, and consider other techniques such as microscopic enumeration when assessing the source of bacteria from the well‐aquifer system.  相似文献

Whilst the potential impact on beach users from microorganisms in water has received considerable attention, there has been relatively little investigation into microbial contaminants in sand. Thirty three beaches across Portugal were analyzed during a five year period (2006-2010) to determine the presence of yeasts, pathogenic fungi, dermatophytes, total coliforms, Escherichia coli and intestinal enterococci in sand.Our results showed that 60.4% of the samples were positive for fungi and that 25.2% were positive for the bacterial parameters. The most frequent fungal species found were Candida sp. and Aspergillus sp., whereas intestinal enterococci were the most frequently isolated bacteria. Positive associations were detected among analyzed parameters and country-regions but none among those parameters and sampling period.Regarding threshold values, we propose 15 cfu/g for yeasts, 17 cfu/g for potential pathogenic fungi, 8 cfu/g for dermatophytes. Eighty four cfu/g for coliforms, 250 cfu/g for E. coli, and 100 cfu/g for intestinal enterococci.  相似文献

Understanding the transport behavior, survival, and persistence of pathogens such as Escherichia coli O157:H7 in the subsurface is essential to protection of public health. In this study, the transport of E. coli O157:H7 in a two‐dimensional bench‐scale sand aquifer system, hereafter referred to as the sandbox, was investigated, with a focus on the impact of biofilm development on E. coli retention and survival. Biofilm growth was initiated through flushing with unsterilized groundwater and addition of glucose, nitrate, and phosphate. Retention of E. coli from an injection test in clean sand, prior to promotion of biofilm growth, was approximately 9%. Subsequent to biofilm growth, 47% of injected E. coli cells were retained under similar flow conditions. After 10 d of no flow, sterile water was flushed through the biofouled sandbox and substantial concentrations (up to 1.5 × 105 cells/mL) of E. coli were measured in the effluent indicating that E. coli had survived the starvation period. Confocal laser scanning microscopy revealed that E. coli were located not only on the surface but also within the biofilm. Imposition of starvation conditions resulted in biofilm sloughing and possible mobilization of biofilm‐associated E. coli.  相似文献

From 2006 to 2008, microbial water quality was monitored along the Georgian coast of the Black Sea. Temperature, pH, salinity, and dissolved oxygen were measured, along with a variety of aquatic microbial parameters, including heterotrophic plate count (HPC), total culturable bacterial count (TCBC), and chlorophyll a (Chl-a) concentration. Total and fecal coliforms and total enterococci counts were recorded as indicators of fecal pollution. Vibrio bacteria, and Escherichia coli- and Vibrio-specific bacteriophages were isolated and enumerated to determine their relationships to standard marine pollution indicators.Persistent microbial pollution was observed, particularly in the summer months, with a higher rate of contamination in estuaries. Microbial indicators generally showed seasonal dependence, suggesting that temperature may influence bacterial dynamics in this environment. No correlation was apparent between fecal pollution indicators and physical-chemical and aquatic microbial parameters, although there were significant relationships amongst the indicators themselves, as well as with the prevalence of Vibrio bacteria and phage.  相似文献

The use of in‐field analysis of vapor‐phase samples to provide real‐time volatile organic compound (VOC) concentrations in groundwater has the potential to streamline monitoring by simplifying the sample collection and analysis process. A field validation program was completed to (1) evaluate methods for collection of vapor samples from monitoring wells and (2) evaluate the accuracy and precision of field‐portable instruments for the analysis of vapor‐phase samples. The field program evaluated three vapor‐phase sample collection methods: (1) headspace samples from two locations within the well, (2) passive vapor diffusion (PVD) samplers placed at the screened interval of the well, and (3) field vapor headspace analysis of groundwater samples. Two types of instruments were tested: a field‐portable gas chromatograph (GC) and a photoionization detector (PID). Field GC analysis of PVD samples showed no bias and good correlation to laboratory analysis of groundwater collected by low‐flow sampling (slope = 0.96, R2 = 0.85) and laboratory analysis of passive water diffusion bag samples from the well screen (slope = 1.03; R2 = 0.96). Field GC analysis of well headspace samples, either from the upper portion of the well or at the water‐vapor interface, resulted in higher variability and much poorer correlation (consistently biased low) relative to laboratory analysis of groundwater samples collected by low‐flow sample or passive diffusion bags (PDBs) (slope = 0.69 to 0.76; R2 = 0.60 to 0.64). These results indicate that field analysis of vapor‐phase samples can be used to obtain accurate measurements of VOC concentrations in groundwater. However, vapor samples collected from the well headspace were not in equilibrium with water collected from the well screen. Instead, PVD samplers placed in the screened interval represent the most promising approach for field‐based measurement of groundwater concentrations using vapor monitoring techniques and will be the focus of further field testing.  相似文献

The impact assessment of molasses‐based distillery‐effluent irrigation on groundwater quality around village Gajraula in the district of Jyotiba Phule Nagar, Uttar Pradesh, India was studied by sampling groundwater on monthly intervals consecutively for summer, winter and monsoon seasons during 2006–2007 and water quality parameters, viz. pH, electrical conductivity (EC), chloride (Cl?), sulphate (SO), nitrate (NO), chemical oxygen demand (COD), total solids (TS), total dissolved solids (TDS), sodium (Na+), potassium (K+), calcium (Ca2+), magnesium (Mg2+), zinc (Zn2+), iron (Fe3+), and total coliforms (TC) were monitored. Results depicted that the values of all parameters decreased with increasing depth of water table. Sulphate, nitrate and potassium contents were maximal in agricultural site during monsoon while EC, Cl?, TS, TDS, Na+, Ca2+, Mg2+, Zn, and Fe were maximal in industrial sites during summer. Groundwater samples of residential site harboured maximum coliforms especially during monsoon, highlighting threat to groundwater. Significant positive correlation matrix between coliforms with nitrate, sulphate and potassium ions explained their survival on these nutrients. To overcome this, important measures emphasizing improvement in effluent treatment technology matching site‐specific characteristics are recommended for eco‐friendly ferti‐irrigation.  相似文献

The expanding use of horizontal drilling and hydraulic fracturing technology to produce oil and gas from tight rock formations has increased public concern about potential impacts on the environment, especially on shallow drinking water aquifers. In eastern Kentucky, horizontal drilling and hydraulic fracturing have been used to develop the Berea Sandstone and the Rogersville Shale. To assess baseline groundwater chemistry and evaluate methane detected in groundwater overlying the Berea and Rogersville plays, we sampled 51 water wells and analyzed the samples for concentrations of major cations and anions, metals, dissolved methane, and other light hydrocarbon gases. In addition, the stable carbon and hydrogen isotopic composition of methane (δ13C‐CH4 and δ2H‐CH4) was analyzed for samples with methane concentration exceeding 1 mg/L. Our study indicates that methane is a relatively common constituent in shallow groundwater in eastern Kentucky, where methane was detected in 78% of the sampled wells (40 of 51 wells) with 51% of wells (26 of 51 wells) exhibiting methane concentrations above 1 mg/L. The δ13C‐CH4 and δ2H‐CH4 ranged from ?84.0‰ to ?58.3‰ and from ?246.5‰ to ?146.0‰, respectively. Isotopic analysis indicated that dissolved methane was primarily microbial in origin formed through CO2 reduction pathway. Results from this study provide a first assessment of methane in the shallow aquifers in the Berea and Rogersville play areas and can be used as a reference to evaluate potential impacts of future horizontal drilling and hydraulic fracturing activities on groundwater quality in the region.  相似文献

The main purpose of this study was to examine the hydrogeochemical factors leading to nitrate contamination of shallow groundwater in an agricultural area. Another purpose was to identify relationships between variations in organic matter levels (as estimated by the BOD and COD parameters) of groundwater that transports effluent from small‐scale livestock holdings. Major cations, anions, BOD and COD of organic matter and total coliforms were analysed. It was found that groundwaters beneath cultivated areas and areas carrying livestock had higher concentrations of calcium, nitrate and chloride than did freshwater. Above all, the nitrification process increased concentrations of nitrate. Nitrate levels were depressed in some places where the groundwater was low in dissolved oxygen. Groundwaters affected by livestock activities showed high concentrations of organic matter (BOD, COD) and high microbial concentrations (as indicated by total coliforms). The COD/BOD ratio increased in the downward direction. It was inferred that this was due to the faster loss of easily biodegradable organic matter compared with non‐biodegradable organic matter proceeding away from a discharge. Accordingly, it is possible to trace effluent in a small area back to a point source by monitoring the COD/BOD ratio of groundwater. Copyright © 2003 John Wiley & Sons, Ltd.  相似文献
