From 2006 to 2008, microbial water quality was monitored along the Georgian coast of the Black Sea. Temperature, pH, salinity, and dissolved oxygen were measured, along with a variety of aquatic microbial parameters, including heterotrophic plate count (HPC), total culturable bacterial count (TCBC), and chlorophyll a (Chl-a) concentration. Total and fecal coliforms and total enterococci counts were recorded as indicators of fecal pollution. Vibrio bacteria, and Escherichia coli- and Vibrio-specific bacteriophages were isolated and enumerated to determine their relationships to standard marine pollution indicators.Persistent microbial pollution was observed, particularly in the summer months, with a higher rate of contamination in estuaries. Microbial indicators generally showed seasonal dependence, suggesting that temperature may influence bacterial dynamics in this environment. No correlation was apparent between fecal pollution indicators and physical-chemical and aquatic microbial parameters, although there were significant relationships amongst the indicators themselves, as well as with the prevalence of Vibrio bacteria and phage.  相似文献

Traditional and molecular methods (PCR) were used to detect, quantify and identify the source of fecal pollution in coastal sites of Puerto Rico and Trinidad. Enterococci and Escherichia coli standard plate counts were used as a general indicator of fecal contamination while the PCR detection of Bifidobacteria adolescentis and human or bovine specific Bacteroidales were used to examine potential sources. Seven of 14 sites in Trinidad including Maracas Bay which is a major public beach contained significant fecal contamination based on enterococci numbers counts exceeding established thresholds for areas of direct contact. Forty six percent of the 27 stations in Puerto Rico were over the established thresholds for enterococci and 49% according to E. coli counts. About 31% of the stations examined in Puerto Rico had evidence of human derived fecal contamination. Human fecal pollution was detected in only one station from Trinidad. Bovine derived contamination was detected only once.  相似文献

The quality of bathing water is of considerable public importance due to the possibility of fecal contamination. In 2009, Croatia implemented the new European Bathing Water Directive (BWD, 2006/7/EC) establishing stricter microbiological standards for new parameters with new reference methods. This study aims to evaluate the equivalence of different methods according to the old and revised BWD and to provide the possibility of data comparison. Furthermore, the directive requires the establishment of the bathing water profile (BWP) for pollution risk assessment. The estimation of consistency of pollution risk assessment with obtained microbiological results was also performed.  相似文献

The increasing demands on recreational waters have made microbial contamination a matter of public and scientific concern. This study aimed to search for Salmonella spp. in waters classified according EU Directive 2006/7/EC, in order to assess associations between its prevalence and the concentration of the non-pathogenic new faecal indicators: Escherichia coli and intestinal enterococci. Although a statistically significant association was observed Salmonella was detected on beaches classified as “Good” (9.3%) and “Excellent” (14.4%) which compromises the idea that faecal indicators can be predictors of pathogens.Attending the high prevalence of Salmonella found (23.1%) it seemed important to improve the efficiency of the conventional analytical method (ISO 6340:1995), comparing its draft with SML-VIDAS Salmonella and two new chromogenic media: AES Salmonella Agar Plate (ASAP) and Simple Method Salmonella (SMS). ASAP showed the higher efficiency and can be recommended for a faster detection and presumptive identification of salmonellae in bathing waters.  相似文献

We evaluated microbiological water quality in the Pettaquamscutt River (Rhode Island, USA), an estuarine river. Fecal coliform (FC) and enterococci (FE) bacteria, presence of Bifidobacterium adolescentis DNA (indicating human fecal contamination), and optical brightener (OB) fluorescence (associated with laundry detergents) were determined for 14 stations from May to September 2010. Six stations had high counts of FE and FC, and the presence of B. adolescentis DNA and high OB fluorescence indicated human fecal contamination - four had septic systems as likely sources of contamination; the others were in sewered areas. The ability of FC and FE to indicate human fecal contamination was assessed against a positive B. adolescentis test. FC and FE had false positive rates of 25% and 17%, respectively, and false negatives of 44% for FC and 63% for FE. Inclusion of molecular and chemical indicators should improve tracking of human fecal contamination sources in the river.  相似文献

The impact of Shokarski stormwater canal on the bathing waters quality of Varna Bay located in the northern Bulgarian Black Sea was investigated during 2007–2014. A total of five monitoring stations were selected: three city beaches near the area of discharge of the stormwater canal and two nearby resorts beaches. The water quality was assessed following the requirements of the old EU Bathing Water Directive (BWD) 76/160/EEC and the new BWD 2006/7/EC. The resort beaches exhibited excellent water quality through the whole period. The situation at the city beaches was worst. The “Officers Beach” situated exactly at the area of discharge of Shokarski stormwater canal exhibited concentrations of ammonium and nitrates significantly above the guide value in most of the cases, the lowest compliance with the total coliforms and faecal coliforms guide values and the highest concentrations of Escherichia coli and intestinal enterococci.  相似文献

Rapid changes in spring water quality in karst areas due to rapid recharge of bacterially contaminated water are a major concern for drinking water suppliers and users. The main objective of this study was to use field experiments with fecal indicators to verify the vulnerability of a karst spring to pathogens, as determined by using a numerical modeling approach. The groundwater modeling was based on linear storage models that can be used to simulate karst water flow. The vulnerability of the karst groundwater is estimated using such models to calculate criteria that influence the likelihood of spring water being affected by microbial contamination. Specifically, the temporal variation in the vulnerability, depending on rainfall events and overall recharge conditions, can be assessed and quantified using the dynamic vulnerability index (DVI). DVI corresponds to the ratio of conduit to diffuse flow contributions to spring discharge. To evaluate model performance with respect to predicted vulnerability, samples from a spring were analyzed for Escherichia coli, enterococci, Clostridium perfringens, and heterotrophic plate count bacteria during and after several rainfall events. DVI was shown to be an indication of the risk of fecal contamination of spring water with sufficient accuracy to be used in drinking water management. We conclude that numerical models are a useful tool for evaluating the vulnerability of karst systems to pathogens under varying recharge conditions  相似文献

Modeling fecal contamination in the Aljezur coastal stream (Portugal)   总被引:1,自引:1,他引:0
This study aims at understanding the fecal contamination behavior in a small coastal stream (Aljezur, Portugal), which has significant economic and ecological values. Like in most small coastal systems, circulation and water renewal in the Aljezur stream exhibit a strong variability due to their dependence on tides, waves, intermittent river flows, and a highly variable morphology. Hence, the problem was approached through a combination of field surveys and the development and application of a hard-coupled three-dimensional hydrodynamic and fecal contamination model. Salinity and temperature results have shown that mixing and transport in the stream are very sensitive to the river flow and wind forcing. The model is able to represent the main patterns and trends observed in Escherichia coli and fecal enterococcus concentrations along the stream, for different environmental and contamination conditions, suggesting die-off rates on the order of 0.50–0.55 day−1. Die-off rate and the representation of the sediment-associated processes were identified as the major remaining sources of uncertainty in the model. Results show that, owing to the processes that occur along the stream, fecal bacteria reach the beaches water in numbers that comply with the European Bathing Waters Directive, even during the summer periods when the upstream concentrations are larger. In particular, results suggest a direct relation between the tidal propagation upstream and the reduction of the fecal bacteria concentrations along the stream that can be relevant for the development of a strategy for the management of the system’s water safety.  相似文献

Whilst the potential impact on beach users from microorganisms in water has received considerable attention, there has been relatively little investigation into microbial contaminants in sand. Thirty three beaches across Portugal were analyzed during a five year period (2006-2010) to determine the presence of yeasts, pathogenic fungi, dermatophytes, total coliforms, Escherichia coli and intestinal enterococci in sand. Our results showed that 60.4% of the samples were positive for fungi and that 25.2% were positive for the bacterial parameters. The most frequent fungal species found were Candida sp. and Aspergillus sp., whereas intestinal enterococci were the most frequently isolated bacteria. Positive associations were detected among analyzed parameters and country-regions but none among those parameters and sampling period.Regarding threshold values, we propose 15 cfu/g for yeasts, 17 cfu/g for potential pathogenic fungi, 8 cfu/g for dermatophytes. Eighty four cfu/g for coliforms, 250 cfu/g for E. coli, and 100 cfu/g for intestinal enterococci.  相似文献

Human fecal contamination of coral reefs is a major cause of concern. Conventional methods used to monitor microbial water quality cannot be used to discriminate between different fecal pollution sources. Fecal coliforms, enterococci, and human-specific Bacteroides (HF183, HF134), general Bacteroides-Prevotella (GB32), and Clostridium coccoides group (CP) 16S rDNA PCR assays were used to test for the presence of non-point source fecal contamination across the southwestern Puerto Rico shelf. Inshore waters were highly turbid, consistently receiving fecal pollution from variable sources, and showing the highest frequency of positive molecular marker signals. Signals were also detected at offshore waters in compliance with existing microbiological quality regulations. Phylogenetic analysis showed that most isolates were of human fecal origin. The geographic extent of non-point source fecal pollution was large and impacted extensive coral reef systems. This could have deleterious long-term impacts on public health, local fisheries and in tourism potential if not adequately addressed.  相似文献

Although urban wet weather discharges may have elevated concentrations of fecal indicator bacteria impacting water quality at swimming beaches, not all of these bacteria may arise from human sources. In this study, the contribution of non-human fecal indicator bacteria was quantified by sampling coastal reference beaches in southern California. Samples were collected at beaches near stormwater discharges from undeveloped watersheds and analyzed for total coliform, Escherichia coli, and enterococci. Surfzone samples exceeded water quality thresholds >10 times more frequently during wet weather than dry weather. Exceedences were greatest <24 h following rainfall, then steadily declined on successive days. Early season storms exceeded thresholds more frequently, and by greater magnitude, compared to late season storms. Large storms exceeded thresholds more frequently than smaller-sized storms, partly due to the breaching of sand berms. When discharges did reach the surf zone, bacterial concentrations in the wave wash were correlated with watershed bacterial flux.  相似文献

The objective of this study was to evaluate whether indicator microbes and physical–chemical parameters were correlated with pathogens within a tidally influenced Estuary. Measurements included the analysis of physical–chemical parameters (pH, salinity, temperature, and turbidity), measurements of bacterial indicators (enterococci, fecal coliform, Escherichia coli, and total coliform), viral indicators (somatic and MS2 coliphage), viral pathogens (enterovirus by culture), and protozoan pathogens (Cryptosporidium and Giardia). All pathogen results were negative with the exception of one sample which tested positive for culturable reovirus (8.5 MPN/100 L). Notable physical–chemical parameters for this sample included low salinity (<1 ppt) and high water temperature (31 °C). Indicator bacteria and indicator virus levels for this sample were within average values typically measured within the study site and were low in comparison with levels observed in other freshwater environments. Overall results suggest that high levels of bacterial and viral indicators were associated with low salinity sites.  相似文献

The objective of this research was to evaluate the groundwater chemical and bacteriological quality in the Barranquita-Knutzen basin (Córdoba, Argentina). The main purpose was to trace contamination through the examination of bacteriological parameters and antibiotic resistance of Escherichia coli, relating them to surface water, hydrogeological features and land use. Thus, 40 water samples were collected. The major chemical components and bacterial indicators were determined and antibiotic resistance was analysed using standard methods. Multivariate factorial analysis showed that the first principal component (PC) reveals the process of water mineralization, while the second and the third PCs explain a low percentage of variance, but collect chemical constituents and total and faecal coliform bacteria, respectively, revealing specific contamination situations. The observed antibiotic resistance profiles of E. coli and their relation with the land uses revealed that the source of faecal contamination in water is mainly from animal residues.  相似文献

The effects of precipitation on the hygienic quality of water and blue mussels collected from five different localities in the urban areas in the Inner Oslofjord were investigated, with samples analysed for Escherichia coli, Salmonella spp., pathogenic Vibrio spp., Norovirus, Sapovirus, Cryptosporidium spp. and Giardia duodenalis. The sampling sites were located at varying distances from the outlet of combined sewer overflows (CSO)-impacted rivers/streams. In general, 1–3 log10 increases in fecal indicator bacteria and human pathogens were observed after heavy rainfalls. Blue mussels appeared to be a useful indicator of the impact of sewage at these sites, and generally a good correlation was identified between concentrations of E. coli and other human pathogens in the mussels. Provision of general advice to the public of avoiding areas near the outlets of CSO-impacted rivers after heavy rainfall may reduce the risk of gastroenteritis by bathers and others that may swallow water during recreational activities.  相似文献

The Dickinson Bayou watershed (near Houston, Texas, Gulf of Mexico) provides habitat for numerous coastally influenced communities of wildlife, including scores of birds and fish. Encroaching development and impervious surfaces are altering the habitat and degrading water quality. Herein we have defined the current health of the bayou using water quality data collected between 2000 and 2006. Elevated bacteria (fecal coliform, Escherichia coli and Enterococcus) and depressed dissolved oxygen concentrations (often <3 mg l−1) are the two major impairments to this ecosystem. While nutrient ratios indicate primary productivity may be nitrogen limited, concerns of eutrophication persist because the bayou has a low intrinsic flushing rate. Consistent with this is the magnitude of algal blooms (ca. 100 μg chl l−1) which often occur in spring/summer. The findings of this study will assist with the understanding of the influence of urban development on small watersheds.  相似文献

Limited by sampling frequency and detection methods, current recreational water monitoring programs often fail to protect public health, especially after heavy rain or flooding, when microbial water quality can change rapidly. In order to assess the variations in the microbial indicators and to develop a scientific warning system, we conducted an intensive sampling project at the No. 1 Bathing Beach. The results show that, during dry weather, the detection rate of Enterococcus was significantly lower than that of Faecal coliform bacteria. On these days, water quality was mainly impacted by pollutants brought in by swimmers than by stormwater outfall. During wet weather, rainfall and microbial bacterial concentrations showed a positive correlation. Trends in the two microbial bacteria were approximately the same. With increasing distance from the shore, the detection rate of microbial bacteria gradually decreased. Microbial bacteria concentrations increased markedly during high tide and under a south wind.  相似文献

This survey was part of a Binational Program (Mexico-United States) in microbiological water quality, with a goal to assess the shoreline bacteriological water quality from Tijuana to Ensenada, Mexico. Samples were collected at 29 sites (19 beaches and 10 outfalls), from the United States border to Punta Banda, Baja California, during summer (1998) and winter (1999). Total coliforms, fecal coliforms and enterococci were used as bacterial indicators. Standard methods were used for total and fecal coliforms, while the Enterolert quick method (IDEXX) was used for the enterococci. Compared with outfalls, the beaches exceeded water quality standards by a small percent, 25.3% in summer and 17% in winter. For outfalls, the percentage of shoreline that exceeded bacterial indicator thresholds had a minor value in summer (32.7%) than in winter (50%). Sites near wastewater discharges had the lowest quality and did not meet the microbiological water quality criteria for recreational use.  相似文献

The influence of seasonal and environmental parameters on the occurrence of bacteria was investigated in two clam species (Venerupis pullastra and Ruditapes philippinarum), water and sediment from the Tagus estuary. Total viable counts (TVC), Escherichia coli, Salmonella spp. and Vibrio spp. were evaluated during one-year. Overall, significant seasonal variations were found in both sampling sites, especially for E. coli and Vibrio spp. levels. In summer, significantly higher Vibrio spp. levels were found in R. philippinarum and sediment samples, but not in V. pullastra clams and water samples. In contrast, significantly higher TVC and E. coli levels were observed in winter months in water and sediment samples. Salmonella spp. was generally isolated when higher levels of E. coli were detected, particularly in R. philippinarum. This study is useful for authorities to develop monitoring strategies for coastal contamination and to estimate human health risks associated with the consumption of bivalves.  相似文献
