Polyphenols from the ethanol extracts of Sargassum tenerrimum (ST) with potent antiallergic effects were studied to optimize separation process through column chromatography. The adsorption and desorption characteristics of three widely used adsorbents: macroporous resin, silica gel, and polyvinylpolypyrrolidone (PVPP), were critically evaluated respectively and studied for the optimization of preparative separation of polyphenols. Static operations on these adsorbents showed that macroporous resin had the best adsorption and desorption capability among the three adsorbents. Dynamic adsorption and desorption with macroporous resin packed column were also conducted to optimize the parameters such as: with the optimal values shown in brackets, the concentration of extract solution (4 times diluted), pH value (6–7), adsorption speed (3 BV h−1, bed volumes/per hour), concentration of ethanol (80%), elution speed (3 BV h−1) and elution volume (7 BV). The chromatographic process so optimized gave a purity of 62.43% from the crude polyphenols, providing a promising basis for large scale preparation of bioactive polyphenols upon further scaling up tests.  相似文献

Ion chromatography-ultra violet-hydride generation-Atomic Florescence Spectrometry was applied to detect 5 arsenic species in seafoods. The arsenic species studied include arsenobetaine(As B), arsenite(As(III)), dimethylarsinic acid(DMA), monomethylarsonic acid(MMA), and arsenate(As(V)), which were extracted from samples using 2% formic acid. Gradient elution using 33 mmol L~(-1) CH_3COONH_4 and 15 mmol L~(-1) Na_2CO_3 with 10 mL CH_3CH_2OH at pH 8.4 allowed the chromatographic separation of all the species on a Hamilton PRP-X100 anion-exchange column in less than 8 min. In this study, an ultrasound extraction method was used to extract arsenic species from seafood. The extraction efficiency was good and the recoveries from spiked samples were in the range of 72.6%–109%; the precision between sample replicates was higher than 3.6% for all determinations. The detection limits were 3.543 μg L~(-1) for As B, 0.4261 μg L~(-1) for As(III), 0.216 μg L~(-1) for DMA, 0.211 μg L~(-1) for MMA, and 0.709 μg L~(-1) for As(V), and the linear coefficients were greater than 0.999. We also developed an application of this method for the determination of arsenic species in bonito, Euphausia superba, and Enteromorpha with satisfactory results. Therefore, it was confirmed that this method was appropriate for the detection of arsenic species in seafood.  相似文献

A new kinetic spectrophotometric method has been developed for the determination of iron (Ⅲ). The method is based on the catalytic effect of iron (Ⅲ) on the oxidation of weak acid brilliant blue dye (RAWL) by KIO4 in acid medium. The advantages of the proposed method are that it is sensitive, accurate, rapid, inexpensive, can be operated under room temperature and has a large determination concentration range compared to other techniques. The obtained optimum conditions are pH 3.15, RAWL (200 mgL-1) 5.00mL, Potassium periodate solution (0.01 molL-1) 0.30mL, phenanthroline (0.02molL-1) 1.00mL, reaction temperature 25℃ and reaction time 7 miu. With this method iron could quantitively be determined in the range 0.00-0.02 mgL-1, the detection limit being 4.10×10-10mL-1. The relative standard deviations (RSD) in five replicate determinations for 3 μgL-1 and 5μgL-1 iron (Ⅲ) are 3.1% and 1.9%, respectively. The method has been applied to the determination of iron (Ⅲ) in tap water samples and seawater samples (from the South China Sea), the recovery rates being 98.0% and 100.5%, respectively.  相似文献

通过静态吸附与解吸实验,比较7种树脂对菠萝皮黄酮类化合物的吸附和解吸特性,筛选出较优的菠萝皮黄酮类化合物吸附剂,并检测吸附流速、上样液浓度、pH值、洗脱流速、洗脱剂体积分数等动态吸附性能。结果表明:AB-8树脂具有较好的吸附和解吸效果,吸附流速1mL/min、上样液浓度2.25mg/mL、pH值6、洗脱流速0.5mL/min、洗脱剂体积分数90%条件下,AB-8型大孔树脂可用于菠萝皮黄酮类化合物的分离纯化。  相似文献

A new kinetic spectrophotometric method has been developed for the determination of vanadium (V). The method is based on the catalytic effect of vanadium (V) on the oxidation of weak acid brilliant blue dye (RAWL) by KBrO3 using the citric acid as activation reagent. The obtained optimum conditions are: c (RAWL) = 1×10−4 molL−1, c (KBrO3) = 3×10−2 molL−1, c (citric acid) = 9×10−3 molL−1, pH = 2.50, the reaction time being 7.0 min and the temperature being 25.0°C. Under the optimum conditions, the proposed method allows the determination of vanadium (V) in the range of 0–70.0 ng mL−1 and the detection limit is down to 0.407 ng mL−1. For standard vanadium (V) solution determination, the recovery efficiency is in the range of 98.5%–102% and the RSD ranges from 0.76%–1.25%. Moreover, it is demonstrated that most cations and anions do not interfere with the determination of vanadium (V) under the analytical condition. The new method was successfully applied in the determination of vanadium (V) in fresh water and seawater samples with satisfactory results. Vanadium (V) in the seawater samples from Qingdao offshore was determined using the method and the distribution of vanadium (V) was mapped. Compared with other instrumental analytical methods, the proposed method shows good selectivity, sensitivity, simplicity, lower cost and rapidity. It can be employed on shipboard easily.  相似文献

The aim of this study is to isolate protoplasts from Undaria pinnatifida. Protoplasts of the alga were isolated enzymatically by using alginate lyase, which was prepared by fermenting culture of a strain Vibrio sp. 510. Monofacterial method was applied for optimizing digestion condition. The optimum condition for protoplast preparation is enzymatic digestion at 28°C for 2h using alginate lyase at the concentration of 213.36 U (8 mL) every 0.5g fresh thalline with NaCl 50 and at the shaking speed of 150 r min−1 during digestion. The protoplast yield can reach 2.62±0.09 million per 0.5 g fresh leave under the optimum condition. The enzyme activity is inhibited by Ca2+ and slightly enhanced by Fe2+ and Mn2+ at concentrations of 0.05, 0.08 and 0.10 mol L−1.  相似文献

Printing and dyeing industry is a considerable source of environmental contamination. In this study treatment of printing and dyeing wastewater with a new type of sewage treatment agent, fly-ash coated with chitosan particles (FCCP), was examined. The effects of pH, stirring time, sedimentation time and temperature on color, COD, turbidity and NH3-N removal were determined. The optimum dosage of FCCP and the influence of individual factors on removal efficiency were tested. The optimum parameters determined using the L16(45) orthogonal experiment were as follows: FCCP (weight ratio of chitosan to fly-ash 1:6) dosage, 4 g·L−1; temperature, 35°C; pH, 4. The stirring time and sedimentation time were 20 min and 5 h, respectively. Under these optimum conditions, the color, COD and NH3-N removal ratios were 97%, 80% and 75%, respectively.  相似文献

Preparation of Fe^2+ chelate of fish protein hydrolysate (Fe-FPH) obtained from low value fish proteins was introduced and its bioactivity was studied by compound enzymolysis. The optimum conditions for hydrolysate chelating Fe^2+ are DH (degree of hydrolysis) at 5%, pH 7.0, 20℃ and 15 min chelating time for FM (material not being defatted). Four types of Fe-FPH including CA (deposit after chelating), CB (deposit in 50% of absolute ethanol solution), CC (suspended deposit in 80% of absolute ethanol solution), and CD (bottom deposit in 80% of absolute ethanol solution) were fractionated with absolute ethanol from FM. Structural analysis through infra-red spectrum revealed that Fe^2+ was combined strongly with amino-group and carboxyl-group in each chelate and each Fe^2+ could form two five-member ring structures. All of the four chelates were shown more significant antioxidative activity and can be used as natural hydrophobic and hydrophilic antioxidant. Among all the chelates, the CB possesses the most effective antioxidative activity at 92% as high as that of a-tocopherol. Among all Fe-FPHs, only CD showed the most effective antibacterial activity against Escherichia coli, Staphylococcus aureus, Salmonella typhi, and Bacillus subtilis and can be used as natural antibacterial. It provides a more effective way for utilization of low value fish proteins and key information of Fe-FPH as additive in food industry.  相似文献

通过对高效液相色谱法(HPLC)测定水产饲料中三聚氰胺含量的方法进行优化,得到最佳色谱条件和前处理方法,建立了水产饲料中三聚氰胺的检测方法。结果表明:以体积比8∶92的乙腈、庚烷磺酸钠离子对试剂为流动相,柱温40℃,流速1.2 mL/min,以波长236 nm检测;以提取液为18 mL50%(φ)乙腈水+1 mL乙酸锌+1 mL1mol/L盐酸,淋洗液3 mL水和3 mL甲醇,洗脱体积6 mL作样品前处理。三聚氰胺加标回收率为85%~100%,相对标准偏差(RSD)小于5%,线性范围为0.2 mg/L~5.0mg/L,相关系数0.999 8,方法检出限为0.2 mg/kg。  相似文献

A new kinetic-spectrophotometric method is proposed for the determination of copper ( Ⅱ ). The method is based on the catalytic effect of copper ( Ⅱ ) on the oxidation of weak acid brilliant blue dye (RAWL) by hydrogen peroxide. The copper ( Ⅱ ) can be determined spectrophotometrically by measuring the decrease of absorbance of RAWL at λ = 626 nm using the fix-time method. The optimum reaction conditions are as follows: pH 7.20, buffer solution NaOH-KH2PO4, RAWL (200 mgL-1) 5.00 mL, H2O2 (30%) 0.50 mL, reaction temperature 80 ?? and reaction time 20 min. The linear range of this method is between 0 μg L^-1 and 12 μg L^-1 and the limit of detection is 0.011 μg L-1, the relative standard deviation (RSD) in five replicate determinations for 2 and 8 μg L-1 copper ( Ⅱ ) are 3.2% and 2.3%, respectively. Twenty ions do not interfere in the determination of copper ( Ⅱ ). The method has been applied satisfactorily to the determination of copper ( Ⅱ ) in freshwater samples (tap water and Yellow River water from Lijin, Shandong, China) and seawater samples (from the South China Sea), the recovery rates are 98.0%, 102.5% and 96.0%, respectively.  相似文献

Tolerance of a facultative marine fungus Aspergillus flavus towards As (V) was tested. Luxuriant growth of the test isolate was observed in culture media with As (V) concentrations of 25 mg L−1 and 50 mg L−1, indicating its tolerance to the metal. Accumulation rate of arsenic was always higher when exposed to As (V) at 50 mg L−1 than at 25 mg L−1. The study reveals Aspergillus flavus as a promising candidate for environmental bioremediation. Arsenic contents (mg g−1) in the fungus when exposed to 50 mg L−1 As (V) were measured as 11.1773, 4.0983, and 8.0000 mg g−1 on day 3, 6 and 9, respectively. The highest content was observed initially, i.e. on day 3, followed by a decline and a rise again. These results provide baseline information for further explorations regarding the exploitation of the fungus for arsenic removal.  相似文献

A new kinetic spectrophotometric method has been developed for the determination of trace Ni (II) in natural water.The method is based on the catalytic effect of Ni (II) on the oxidation of weak acid brilliant blue dye (RAWL) by KIO4 in acid medium.The concentration of nickel (II) can be determined spectrophotometrically by measuring the decrease of absorbance of RAWL at λ= 626 nm using the fix-time method.The influencing factors are investigated by the orthogonal experimental design.The obtained optimum analytical conditions are:pH=2.00,c RAWL=5.00×10-5 mol L-1,cKIO 4= 2.00×10-5 mol L-1,the reaction time t=10 min and the temperature T=25℃..Under the optimum conditions,the developed method allows the measurement of Ni (II) in a range of 0-40.0 ng mL-1.The standard deviation of eleven independent measurements of blank reaction is S=3.08×10-3 and the limit of detection is 2.20 ng mL-1.The relative standard deviations (RSDs) in six replicate determinations of 5 ng mL-1 and 8 ng mL-1 Ni (II) are 2.87% and 1.11%,respectively.Moreover,the experiments show few cations and anions can interfere with the measurement of Ni (II).The recovery efficiencies of this method are in a range of 97.0%-102.5% in freshwater samples.But there is a decreasing effect,which is about 0.2 times the added Ni (II) in seawater medium.After reasonable calibration this processing method is used for the determination of Ni (II) in seawater samples successfully.The results show this developed method has high accuracy and precision,high sensitivity,large range of linearity and high speed.The method can,therefore,be employed at room temperature.  相似文献

In Wuxi Wastewater Treatment Plant, the Anaerobic, anoxic and oxic (A2/O) process was employed to remove the nitrogen and phosphorus, which exhibited the positive results of the high removal efficiency for phosphorus with a range of 67.7% to 89.9% and an average value of 78.0. The effluent of phosphorus met the national discharge standard. The removal of TN was effected by both BOD variation of influent and wastewater temperature. TN removal was in the range of 28.5% to 55.8% with an average value of 39.4%. The energy cost was 0.15 kWh(m3d)-1 or 1.35 kWh(kgBOD·d)-1. The annual average sludge production was 46.3 m3d-1, the annual average dosage for the dewatering was 40 kgd-1.  相似文献

A method for the analysis of selenium in marine aquatic products by HG-AFS has been investigated. The method is based on the reduction of inorganic selenium to volatile SeH2 which is bubbled out by carrier gas of pure argon, and then swept to Ar-H2 flame quarts atomizer to measure its fluorescence intensity. The hydride generation, transportation, atomization and some instrumental parameters were studied by a kind of orthogonal design. The optimum conditions selected are as follows: reactive acidity, 20% HC1; the amount of NaBH4, 4.9mL; gas flow of argon, 600mLmin^-1; atomizing temperature, 200 ℃ ; negative high voltage, - 300V; light current, 100 mA; integral time, 7s. The detection limit of the presented method is 0.072μgL^-1 for selenium. The calibration curve shows a satisfactory line inthe concentration range from 0.000 to 1.000μgL^-1 Se. The recovery is 95.8%-102.2%.  相似文献

Information on phosphorus (P) adsorption and its impacts on the redistribution of the P fraction in soil profiles are important for environmental management under intensive agricultural practices. To clarify the dominant factors influencing soil phosphorus adsorption in an Entisol (locally known as purple soil), P adsorption experiments were conducted in Sichuan Basin of southwestern China for cropland and woodland soils with acidic, neutral and calcareous origins throughout their profile. After various doses of P were added during incubation experiments, soil P fractions were also analyzed. The results showed that there were no significant differences in Fe-oxides and P adsorption along the vertical gradients. Agricultural practices and lower pH conditions reduced the P adsorption capacity of purple soils throughout the soil profiles. For acidic and neutral purple soil profiles, the P adsorption capability was mainly influenced by Fe-oxides and soil texture. Ca-bound P and Fe-Al-bound P represented the majority of the total inorganic P of calcareous soils. There was a saturation of adsorption capacity by sesquioxide and a high risk of dissoluble reactive P (NH4Cl-P) being released out of the soil profile in acidic and neutral purple soils after the greatest P addition, indicated by the higher proportions of NH4Cl-P (over 40%) and decreasing Fe-Al-P fraction. P fractions migrated with greater difficulty in calcareous purple soil profiles as Ca-P fraction peaked over 65% when adding a P dose at or greater than 80 g P kg-1, indicating the high potential of P adsorption. The X-Ray Diffraction analysis also verified the formation of brushite. Adaptive management practices should be designed to alleviate P losses for acidic and neutral purple soils.  相似文献

In this work,response surface methodology(RSM)was employed to model and optimize electrodialysis process for mercury(Hg(II))removal from seaweed extracts.Box-Behnken design(BBD)was utilized to evaluate the effects and the interaction of influential variables such as operating voltage,influent flow rate,initial concentration of Hg(II)on the removal rate of Hg(II).The developed regression model for removal rate response was validated by analysis of variance,and presented a good agreement of the experimental data with the quadratic equation with high value coefficient of determination value(R2=0.9913,RAdj 2=0.9678).The optimum operating parameters were determined as 7.17V operating voltage,72.54L h−1 influent flow rate and 5.04mgL−1 initial concentration of mercury.Hg(II)removal rate of 76.45%was acquired under the optimum conditions,which showed good agreement with model-predicted(75.81%)result.The results revealed that electrodialysis can be considered as a promising strategy for removal of Hg(II)from seaweed extracts.  相似文献

Algal organic materials (AOMs) are one critical factor affecting the efficiency of modified clays used for the mitigation of harmful algal blooms (HABs). This study was conducted to develop a deeper understanding of the mechanisms and factors affecting the adsorption of AOMs onto modified clays. Sodium alginate (polysaccharide) and kaolinite modified with polyaluminium chloride (PACl) were used as AOMs and modified clay model substances, respectively, and the effects of modifier dosage, contact time, solution pH and ionic strength were investigated through batch adsorption experiments. Kinetics revealed that the alginate adsorption rate was described well by a pseudo-second order model. PACl effectively enhanced the adsorption capacity of kaolinite and increased the adsorption rate, and the optimum additive amount of PACl was 5%. The experimental data fitted both the Freundlich and Langmuir adsorption equations well. The adsorption thermodynamics for alginate onto modified clays suggests that alginate adsorption is a spontaneous process. The adsorption of alginate onto modified clays was highly dependent on pH, with a decrease in adsorption observed with increased pH to 9.48, but the opposite was true above pH 9.48. Finally, adsorption increased with increasing ionic strength.  相似文献

为了改善海上油田深部调驱效果,研制一种易于配制、施工简便的由聚合物FP、交联剂和助剂组成的冻胶体系.基于合适的成冻时间和成冻强度,优选适合该油藏条件的冻胶调驱体系配方,即聚合物FP(1 500~2 000mg.L-1)+交联剂(750~1 250mg.L-1)+助剂(1 000~1 500mg.L-1).利用三层非均质岩心进行调驱体系用量和调驱时机对调驱效果影响的评价,优化调驱体系的注入用量和注入时机参数.结果表明:当水驱至目标油藏含水率70%时,随着注入调驱体系用量的增加,采收率增加值越高;基于采出程度和经济效益双重考虑,优选调驱体系的用量为0.2PV;当注入用量为0.2PV调驱体系,油藏综合含水率低于50%条件下,早期注入调驱体系进行深部调驱,采收率增加值高,能够达到很好的调驱效果.  相似文献
