2,4-二氯苯氧乙酸对细基江蓠孢子生长和存活的影响   总被引:1,自引:0,他引:1  
以细基江蓠(Gracilaria tenuistipitata)的果孢子和四分孢子为实验材料,用不同浓度的(0,0.1,0.5,1,2,4,6 mg/L)生长激素2,4-二氯苯氧乙酸(2,4-D)处理刚刚放散的孢子和附着2d孢子,研究其对果孢子和四分孢子附着和生长及存活的影响。与对照组相比,2,4-D使两种孢子的附着量明显降低;2.0~4.0mg/L的2,4-D可以极显著的促进孢子的生长(P<0.01);0.1~2.0mg/L的2,4-D提高孢子的存活率(P<0.01);6.0mg/L的2,4-D使得孢子的生长和存活率降低。综合以上指标,在细基江篱采孢子育苗时,孢子附着2~3d后用2,4-D处理孢子萌发体,以1.0~2.0mg/L为宜。  相似文献

江蓠(Gracilaria verrucosa)生活史中有三个世代,即孢子体、配子体和果孢子体(囊果)。其中:孢子体和配子体均为单生、同形的多分枝藻体,果孢子体则附生在雌配子体上。在生殖期间,江蓠的孢子体,除基部老枝和初生的新枝不形成四分孢子囊外,藻体的各部分分枝,均可形成四分孢子囊,放散出四分孢子。雌配子体也是除基部老枝和初生新枝外,藻体的各部分分枝均可附生果孢子体(囊果)。果孢子体成熟放散出果孢子。关于江蓠四分孢子和果孢子放散方式和放散过程的研究,国内外只有零星记载。曾呈奎等1959年,在进行与江蓠幼苗培育试验时,仅记载着:“经过干燥刺激15分钟的藻体放回海水中,发现20~30分钟后,孢子就大量放出,但四分孢子囊是孢子成团冲破表皮脱离母体,  相似文献

舌状蜈蚣藻(Grateloupia livida)是一种具有较高食用和药用价值的经济红藻。本文比较了不同温度(10,15,20,25,30℃)、光照强度(0,20,60,120,200μmol.m-2.s-1)及盐度(8,16,24,32,40)条件对舌状蜈蚣藻果孢子的放散及附着的影响。实验结果表明,舌状蜈蚣藻果孢子放散的最适宜温度为20℃,光照强度为120μmol.m-2.s-1,适宜盐度24~40;果孢子附着的最适宜温度为20℃,光照强度为60μmol.m-2.s-1,适宜盐度为24~40。温度及光照条件对果孢子的放散具有显著影响,在20℃、120μmol.m-2.s-1条件下放散量最大,低温(10℃)以及黑暗条件下可以显著抑制果孢子的放散;中、高盐度(24,32,40)条件下,果孢子放散没有显著差异,而在低盐度(8,16)条件下,果孢子的放散量显著减少。温度和光照对舌状蜈蚣藻果孢子的附着均存在显著影响,在20℃和60μmol.m-2.s-1下果孢子附着数量最多,在10℃,30℃和黑暗条件下附着数量最少;在低盐度(8,16)海水中果孢子的附着受到抑制,在中、高盐度(24,32,40)海水中果孢子的附着没有明显差异。  相似文献

海洋污损生物附着是增加海上作业成本、影响设施安全和缩短其使用年限的重要因素,掌握海洋污损生物幼虫和孢子的采集与培养技术是开展防除研究工作的基础。网纹藤壶Balanus reticulatus、缘管浒苔Enteromorpha linza、石莼Ulva lactuca和细基江蓠Gracilaria tenuistipitata是华南沿海主要的污损生物种类。研究上述4种污损生物幼虫和孢子的采集与培养技术,结果显示,网纹藤壶无节幼虫于30℃黑暗环境中培养,5d左右即可发育为金星幼虫,可将其在4℃条件下储存备用。强度为4000lx的光照射缘管浒苔和石莼,可刺激其游孢子大量释放;细基江蓠经阴干处理再浸泡海水,可诱导其果孢子的放散。每天接受900—1300lx光照12h,室温(21—26℃)下培养4d,是观察缘管浒苔和石莼游孢子附着萌发的最佳条件,而细基江蓠果孢子的附着萌发观察则以室温(18—23℃)下培养2d为宜。  相似文献

为研究单条胶黏藻(Dumontia simplex Cotton)的孢子发育类型和生活史,探明其孢子发育的最适环境因子,在实验室条件下,通过培养观察,研究不同温度、光照强度和盐度对其孢子(果孢子和四分孢子)早期发育的影响。结果表明:(1)孢子发育过程中无丝状体出现,孢子的发育类型为直接盘状体型。(2)温度、光照强度和盐度对其孢子的早期发育均有影响,温度6—30°C范围内,果孢子发育的最适温度为9°C,四分孢子为9—12°C;光强80—400μmol/(m2·s)范围内,果孢子发育的最适光强为160μmol/(m2·s),四分孢子为160—200μmol/(m2·s);盐度15—50范围内,孢子(果孢子和四分孢子)发育的最适盐度为30。(3)单条胶黏藻生活史由单倍的雌、雄配子体和二倍的果孢子体及四分孢子体构成,配子体和四分孢子体外形一致,交替出现,属于典型的同型世代交替。  相似文献

英国江蓠 Gracilaria verrucosa(Huds.)Papenfuss在外形上变异幅度很大,地区性海藻志或海藻名录很少不包含本种在内;有少数专论性的著作对本种均有详细的记述,因此,被认为是一个世界广布种。本文通过囊果被构造及其他特征,将中国和日本产、曾定名为“Gracilaria verrucosa(Huds.)Papenfuss”的标本与英国Devon模式标本产地的标本进行了比较研究,结果证明:模式标本的囊果被构造与中国、日本产的完全不同,因此,中国和日本产的应为一新种——真江蓠Gracilaria asiatica sp.nov.  相似文献

本研究探究了白、蓝、红、绿光4种光质对坛紫菜(Pyropia haitanensis)果孢子和壳孢子萌发的影响。结果表明,与白光相比,用绿光培养时,果孢子萌发管分枝形成速度、萌发管生长速度,以及壳孢子的细胞分裂速度均相对较快,用红光与蓝光培养时则相对较慢。不同光质对坛紫菜果孢子和壳孢子的存活率无显著影响,但能造成萌发体色泽呈现明显差异。刚从孢子囊释放出来的两种孢子均呈黄褐色,经白光培养的两种孢子以及蓝光培养的果孢子,其萌发体颜色加深至鲜红色,蓝光培养的壳孢子萌发体颜色加深尤为明显,最终呈深紫红色,而红光和绿光培养的两种孢子颜色变浅,最终呈浅黄褐色。研究表明,绿光培养可能不利于坛紫菜孢子萌发过程的色素积累,但有利于孢子萌发及萌发体的生长。  相似文献

在实验室条件下,以细弱蜈蚣藻(Grateloupia tenuis Wang et Luan)为材料,进行了不同温度和光照强度等主要环境因子对其发育影响的研究,同时对其生活史进行了详细观察。结果表明:(1)在4—30°C范围内,盘状体形成与直立枝生长的最适温度为24°C;(2)在光照强度10—200μmol范围内,盘状体形成和直立枝生长的最适光照强度为120μmol/(m2s);(3)孢子(果孢子和四分孢子)发育类型为间接盘状体类型;(4)生活史由单倍体的雌、雄配子体,双倍体的果孢子体(囊果)及四分孢子体构成,四分孢子体和配子体在形态上完全相同,属于典型的同型世代交替;与属模蜈蚣藻(G.filicina)一致。  相似文献

Comparison of growing thalli in alternating haploid and diploid phases of Grateloupia subpectinata (Rhodophyta) was studied. Fertile thalli from gametophyte and tetrasporophyte of G. subpectinata were collected from Yangyang, on the eastern coast of Korea. The size of the released tetraspores and carpospores was measured; the spores were then incubated at the temperature of 20°C, irradiance of 40 μmol photon m-2s-1 and photoperiod of 12L and 12D. Carpospores were also cultivated in the same conditions as the tetraspores culture. The crusts were subsequently transferred to a tank culture after six months. The specific growth rate (SGR) was measured by observing 50 crusts and 30 thalli. The released carpospores had a larger diameter (9.98 μm) than the tetraspores (9.38 μm). The crusts from the carpospores also show a higher specific growth rate (14.04% d-1) than tetraspores (13.39% d-1). After being transferred and cultured in a tank, the upright thalli grew slowly in May-June (13–15°C) and rapidly in July–September (17–22°C). The length of growing thalli of sporophyte from carpospores also revealed a higher specific growth rate (2.83% d-1) than gametophytic thalli (2.38% d-1). The specific growth rate of crusts and thalli developed from carpospores was higher than that of the crusts developed from tetraspores. This result suggests that the cultivation of sporophytes may be more profitable than gametophytes because harvesting can be done more efficiently.  相似文献

On the basis of morphological observations, life history and molecular phylogeny, Grateloupia yangjiangensis, which is similar to G. filicina, G. orientalis, G. catenata, and G. ramosissima in appearance, was re-examined. The results are as follows:(1) the auxiliary-cell ampullae of G. yangjiangensis were of Grateloupia type, thalli was fleshy and gelatinous in texture, and the erect axes were compressed; the cortex was 0.25–0.30 mm thick, consisting of five to seven outer layers, and there were five inner layers of triangular or stellate cells;(2) there was no filamentous stage in the development of the carpospores;(3) the ribulose-1,5-bisphosphate carboxylase/oxygenase gene(rbcL) sequence of four G. yangjiangensis examined showed that there was no intergeneric divergence among them, and for the phylogenetic tree, four sequences of G. yangjiangensis formed a single monophyletic subclade within the large Grateloupia clade of Halymeniaceae. In conclusion, G. yangjiangensis was a single species within the genus Grateloupia. This research provided criterion for identification and cultivation of G. yangjiangensis.  相似文献

丝状藻体是紫菜生活史中的重要阶段,其生长增殖与环境条件密切相关.本研究考察了与坛紫菜丝状藻体生长增殖相关的多个影响因子,探讨了可促进坛紫菜丝状藻体生长、达到生长速率快的优化调控培养条件.具体操作实施过程为:初始丝状藻体来自成熟紫菜叶状体的果孢子释放,基于8因子3水平的正交实验设计方案[采用正交表L27(313)],确定了丝状藻体生长对多个环境因子的响应以及优化的调控培养条件.结果表明,除开pH、复合维生素浓度和NaHCO3浓度外,其他因子对丝状藻体生长均有显著的影响(p≤0.043 8).获得的优化调控培养条件为:温度18℃、光强27μmol/m2·s-1、盐度25、pH值8.5、复合维生素浓度15μg/dm3、NaHCO3浓度2 mg/dm3、每天光照时数18 h、营养液浓度为f培养介质.优化调控培养条件下的丝状藻体生长速率高达12.07%,平均值±标准偏差SD为(10.70±0.88)%/d.  相似文献

以坛紫菜(Porphyra haitaneusis)叶状体为材料,利用单细胞克隆技术,研究记录坛紫菜体细胞的发育途径。本文选择介绍了坛紫菜体细胞3种途径8种发育方式:1)多数体细胞的叶状体途径:A通过极性分裂直接形成单细胞苗;B通过叶状分裂成苗;C先形成细胞团后形成畸形苗;2)来自雌、雄叶片未成熟区域体细胞的性细胞途径:D发育形成精子囊并放散精子;E单细胞发育形成类似果孢子囊结构再放散果孢子,果孢子萌发成丝状体;3)来自雄叶片体细胞的丝状体途径:F单细胞发育至多细胞团,放散后萌发成丝状体;G经过不断分裂形成愈伤组织,直接形成丝状体;H分裂成2细胞后,一个细胞侧面突起发育成丝状体。  相似文献

丝状藻体在紫菜养殖的人工苗种生产中非常重要,在利用于分离提取高价值的藻红蛋白方面也具有潜在的利用价值。本研究采集了成熟紫菜叶状体的果孢子释放的初始丝状藻体,基于3因子3水平的正交实验设计方案[采用具有分析交互作用的正交表L27(313)],确定了丝状藻体藻红蛋白含量对环境因子的响应以及优化的调控培养条件。结果表明,温度、光照强度和盐度因子对丝状藻体藻红蛋白含量均有显著影响(P≤0.048 1),并且温度和光照强度或盐度因子间存在交互作用(P ≤0.043 1),但光照强度和盐度因子间的交互作用不显著(P=0.469 8)。获得的优化调控培养条件为:温度18 ℃、光强1 500 lx、盐度20或25。优化调控培养条件下的丝状藻体藻红蛋白含量高达64.73 mg/g.dw(平均值±标准偏差SD为58.24±5.07 mg/g.dw)。  相似文献

以长江水系大规格中华绒螯蟹野生群体和养殖群体为选育基础群,以降低一龄性早熟率、提前或延后二龄成熟期和提高成蟹规格为主要选育目标,进行中华绒螯蟹良种培育,综合比较野生和养殖群体选育子一代(野选G1,养选G1)的养殖性能和性腺发育情况,结果显示:(1)扣蟹养殖阶段,野选和养选G1的早熟率分别比对照组降低36.89%和15.25%,养选G1早熟率略高于野选G1(P0.05);野选G1在9—11月平均体重略低于养选G1和对照组,且8—9月野选G1的增重率和特定生长率显著低于对照组;野选和养选G1的正常扣蟹产量显著高于对照组,且两选育群体G1早熟蟹的产量显著低于对照组(P0.05);(2)成蟹养殖阶段,野选G1的生殖蜕壳时间和性腺发育略晚于对照组,养选G1的生殖蜕壳时间和性腺发育略早于对照组;(3)三群体中华绒螯蟹在成蟹养殖阶段的早期(3—6月)生长速度接近,6—8月野选G1雄体的增重率和特定生长率明显低于其它两组,8—9月则明显高于其它两群体雄体;野选G1雌体的生长速度仅在6—7月明显低于其它两组,7—9月则反之;(4)野选G1的平均产量显著高于对照组,两选育群体最终成蟹平均规格均高于对照组,仅野选G1雌体成蟹体重显著大于对照组(P0.05);无论雌体还是雄体,野选和养选G1成蟹平均体重均比对照组大10%以上;两选育群体雄体和雌体的大规格成蟹(雄蟹≥175g,雌蟹≥125g)比例均高于对照组,但差异不显著(P0.05)。综上,野选G1具有一龄扣蟹早熟率低、二龄成蟹规格大和性腺成熟时间略有推迟等特点,养选G1具有扣蟹养殖阶段饲料系数低、性腺发育时间提前和成蟹规格大等特点,两群体G1均具有进一步选育的潜力。  相似文献

The invasive red alga Grateloupia turuturu Yamada could turn Vibrio parahaemolyticus into nonculturable state in live algal culture. In order to elucidate the mechanism of such an effect, a series of culture experiments were performed in this investigation based on three hypothesized causes, namely bacterial attachment, production of reactive oxygen species (ROS) and the discharge of water soluble secondary metabolic compounds. The results reveal that attachment to the thallus surface of G. turuturu was the major reason for the decrease of V. parahaemolyticus in seawater. Further investigations show that V. parahaemolyticus attachment to the surface of algal thallus in live cultures of seaweeds was a common phenomenon. However, the disappearance of the culturability of V. parahaemolyticus occurred only on the thallus of G. turuturu over 72 h among all six algal species tested. Electron microscopic scanning shows that most of V. parahaemolyticus attached to G. turuturu changed from the initial normal bacilli to coccoid-shape after 72 h. The enclosure experiments by enclosing the algal thallus in tubes demonstrate that the nonculturability of V. parahaemolyticus in the water of live culture of G. turuturu occurred after the physical contact of the V. parahaemolyticus to the alga. The capacity of G. turuturu in affecting the culturability of V. parahaemolyticus was not influenced after inhibition of photosynthesis by treatment of 3''-(3,4-dichlorophenyl)-1'',1''-dimethyl urea (DCMU) at non-lethal levels. Production of reactive oxygen species after addition of live culture of bacteria was excluded by on-line analyzing the oxidation of dichlorohydrofluorescein (DCFH) to dichlorofluorescein (DCF) in the presence of peroxidase on a VersaFluor fluorometer.  相似文献
