Trare amounts of benzene hydrocarbons obtained in Jiaozhou Bay (Qindao) were enriched bysorption on a GDX-102 column and eluted by carbon disulfide. Giant red sea cucumber (Parastichopus californicus) is an under-utilized species due to its high tendency to autolysis.The aim of this study was to evaluate the functional properties of collagen hydrolysates from this species.The degree of hydrolysis (DH),amino acid composition,SDS-PAGE,emulsion activity index (EAI),emulsion stability index (ESI),foam expansion (FE),and foam stability (FS) of hydrolysates were investigated.The effects of pH on the EAI,ESI FE and FS of hydrolysates were also inves-tigated.The results indicated that the β and α1 chains of the collagen were effectively hydrolyzed by trypsin at 50℃ with an En-zyme/Substrate (E/S) ration of 1:20 (w:w).The DH of collagen was up to 17.3% after 3 h hydrolysis with trypsin.The hydrolysates had a molecular weight distribution of 1.1 17 kDa,and were abundant in glycine (Gly),proline (Pro),glutamic acid (Glu),alanine (Ala) and hydroxyproline (Hyp) residues.The hydrolysates were fractionated into three fractions (< 3 kDa,3 10 kDa,and > 10 kDa),and the fraction of 3 10 kDa exhibited a higher EAI value than the fraction of > 10 kDa (P<0.05).The fraction of > 10 kDa had higher FE and FS values than other fractions (P<0.05).The pH had an important effect on the EAI,ESI,FE and FS.All the fractions showed undesirable emulsion and forming properties at pH 4.0.Under pH 7.0 and pH 10.0,the 3 10 kDa fraction showed higher EAI value and the fraction of > 10 kDa showed higher FE value,respectively.They are hoped to be utilized as functional ingredients in food and nutraceutical industries.  相似文献

A new kinetic spectrophotometric method is developed for the measurement of manganese (II) in water. The method is based on the catalytic effect of manganese (II) with the oxidation of weak acid brilliant blue dye (RAWL) by KIO4 using the Nitrilo triacetic acid (NTA) as an activation reagent. The optimum conditions obtained are 40 mgL−1 RAWL, 1×10−4molL−1 KIO4, 2×10−4 molL−1 Nitrilo triacetic acid (NTA), pH = 5.8, the reaction time of 3.00 min and the temperature of 20.0 °C. Under the optimum conditions, the proposed method allows the measurement of manganese (II) in a range of 0–50.0 ng mL−1 and with a detection limit of down to 0.158 ng mL−1. The recovery efficiency in measuring the standard manganese (II) solution is in a range of 98.5%–102%, and the RSD is in a range of 0.76%–1.25%. The new method has been successfully applied to the measurement of manganese (II) in both fresh water and seawater samples with satisfying results. Moreover, few cations and anions interfere with the measurement of manganese (II). Compared with other kinetic catalytic methods and instrumental methods, the proposed method shows fairly good selectivity and sensitivity, low cost, cheapness, low detection limit and rapidity. The SCENTO-System was used to study the carbon dynamics between phytoplankton primary production and heterotrophic bacterial secondary production. Most of the methods used nowadays in situ for limnological synecology studies were applied. Primary production measurement showed an increasing tendency with increasing content of chlorophylla. It provided a true photosynthetic rate lying within the range of eutrophic lakes. Net EOC released from the algae ranged from 8.5 to 27.5 μg C l−1(6h)−1. Accompanying the algal products the number of bacteria increased from 1.475 ×109 to 8.074×109 cells l−1. The bacterial mean cell volume was small, between 0.0315 and 0.0548μm3. Bacterial carbon production from direct growth estimates was compared with independent calculations of bacterial growth from EOC uptake and3H-thymidine incorporation. Direct estimates were 2.97–10.0 μg Cl−1 (24h)−1 with the exception of a zero-growth on the third day. EOC uptake was 123.5–191.0 μg Cl−1 (6h)−1. That calculated from3H-thymidine incorporation was 0.2–0.5 μg Cl−1 (6h)−1.14C-glucose dark uptake ran parallel to the increasing bacterial biomass. The respiration of glucose was 6.5% (avg.) of the gross uptake. Since the system operated without grazing pressure, a real carbon flow from primary production to bacterial secondary production could be observed.  相似文献

The investigation shows that the concentrations of nutrients are high in estuarine and coastal waters and low in offshore waters. The concentration of nitrate in estuaries is controlled through a physical mixing process and is also affected by biotic activity. The annual transport of total inorganic nitrogen and dissolved phosphate-phosphorus from the Huanghe River water to the sea is about 8.45 ×104 and 1.45×103 tons respectively. The distributions of inorganic nitrogen and silicate in interstitial water of surface sediments are similar to those in surface and bottom seawater. Their contents in interstitial water are 227–552 μmol/l (average375) for ammonia, 0.31–9.0 μmol/l (average 1.6) for nitrite, 0–41 μmol/l (average6.0) for nitrate, and 41–139 μmol/l (average 77) for silicate. The average concentrations of phosphate in the surveyed area are 0.64 μmol/l for seawater and 1.2 μmol/l for interstitial water. A cycle of phosphate in the estuary is also suggested in this paper. This study deals with contribution of artificial food pellet and natural food to Chinese prawn (Penaeus orientalis) growth in a semiintensive culture pond. The prawn carbon consumption, budget, and the effects of some factors on the budget were investigated. The results showed that 26.2% of P. orientalis growth carbon came from formulated feed at the initial culture stage (when the prawns were 0.06±0.01 g in wet weight), and was 62.5% when the prawns were 9.56±1.04 g. The remaining part of the growth carbon was derived from organic fertilizer and natural food. The highest growth rate occurred at 20×10-3 salinity. Suitable salinity for culturing Chinese prawn was (20-28)×10-3.  相似文献

Graded levels of manganese were supplemented to a semi-purified diet containing 45% crude protein, to provide six levels of manganese (i. e. containing 5, 25, 50, 70, 140 and 210×10−6, respectively) for two experiments with these experimental diets. The initial weight of shrimp used in the 35–day experiment I was 0.30±0.04 g, and that in the 70–day Experiment II was more than one gram. The results showed that optimum content in the semi-purified diet for the more than 1 gram shrimp ranged from 70 ×10−6, to 140×10−6, but supplementation of Mn was not necessary for the small shrimp. A method is described for determining dimethylI sulfide (DMS) in seawater. DMS was first extractedfrom the seawater using organic reagent, then reverse-extracted by 5% HgCl2. In the laboratory DMS wasreleased by concentrated HCI and finally measured by GC-FPD. The limit of detection me O.05 ng ofS. IAA 3-Indolylacetic acid, NAA a-Naphthylacetic acid and cytokinins in PESI culture medium were used in a study on the effects of plant hormones on the growth of free-living conchocelis ofPorphyra yezoensis which showed that its growth in medium with cytokinins, IAA and NAA was more rapid than that in medium with non—phytohormones; that the optimal concentrations for promoting growth were 10 μg/L for IAA and ZA (Zeatin), and 0.1 μg/L for BA 6-Benzyl amino purine and KIN 6-Furfurylamino- purine. Mix use of NAA, IAA and cytokinins, NAA/ZA 1–1000/1 μg/L, NAA/BA 10/1-1000 μg/L, NAA/KIN 1/1-1000 μg/L promoted growth. A new kinetic spectrophotometric method has been developed for the determination of iron (Ⅲ). The method is based on the catalytic effect of iron (Ⅲ) on the oxidation of weak acid brilliant blue dye (RAWL) by KIO4 in acid medium. The advantages of the proposed method are that it is sensitive, accurate, rapid, inexpensive, can be operated under room temperature and has a large determination concentration range compared to other techniques. The obtained optimum conditions are pH 3.15, RAWL (200 mgL-1) 5.00mL, Potassium periodate solution (0.01 molL-1) 0.30mL, phenanthroline (0.02molL-1) 1.00mL, reaction temperature 25℃ and reaction time 7 miu. With this method iron could quantitively be determined in the range 0.00-0.02 mgL-1, the detection limit being 4.10×10-10mL-1. The relative standard deviations (RSD) in five replicate determinations for 3 μgL-1 and 5μgL-1 iron (Ⅲ) are 3.1% and 1.9%, respectively. The method has been applied to the determination of iron (Ⅲ) in tap water samples and seawater samples (from the South China Sea), the recovery rates being 98.0% and 100.5%, respectively.  相似文献

Twenty-four compounds including eight steroids (1–8), nine triterpenoids (9–16, 24), three flavonoids (20–22), and four benzenecarboxylic derivatives (17–19, 23) were isolated and identified from stems and twigs of medicinal mangrove plant Sonneratia caseolaris. The structures of the isolated compounds were determined by extensive analysis of their spectroscopic data. Among these metabolites, compounds 1, 4–20 and 22–24 were isolated and identified for the first time from S. caseolaris. In the in vitro cytotoxic assay against SMMC-7721 human hepatoma cells, compound 21 (3′,4′,5,7-tetrahydroxyflavone) exhibited significant activity with IC50 2.8 μg/mL, while oleanolic acid (14), 3,3′-di-O-methyl ether ellagic acid (18), and 3,3′,4-O-tri-O-methyl ether ellagic acid (19) showed weak activity. None of these compounds displayed significant antibacterial activites. Effects of temperature on oxygen consumption rate (OCR) and ammonia—N excretion rate of scallopChlamys farreri (1.7–6.2 cm in shell height) were studied in laboratory from Dec. 30,1996 to Jan. 28,1997. Under the controlled conditions of ambient water temperature 10–31°C and salinity 32, the concentrations of dissolved oxygen and ammonia—N were determined by the Winkle method and the hypobromite method, respectively. Results showed that the OCR ranged from 1.20 mg/g (DW) · h to 5.76 mg/g (DW) · h. The OCR increased with temperature from 10°C to 23°C, but at 28°C the OCR of mature individuals decreased, and that of different size scallops reduced at 31°C. The ammonia—N excretion rate ranged from 113.03 μg NH4-N/g (DW) · h to 486.63 μg NH4-N/g (DW) · h, and increased with temperature from 10°C to 31°C. The size-fractionated phytoplankton biomass, and the spatial and temporal variations in abundance of Synechococcus (SYN) and picoeukaryotes (PEUK) were measured in the Taiwan Strait during three cruises (August 1997, February–March 1998, and August 1998). The results show that picophytoplankton and nanophytoplankton dominate the phytoplankton biomass, in average of 38% and 40%, respectively. SYN and PEUK varied over time in abundance and carbon biomass, greater in summer than in winter, in range of (7.70–209.2)×106 and (0.75–15.4)×106 cells/cm2 in the abundance, and 1.93–52.3 and 1.57–32.4 μgC/cm2 in the carbon biomass, for SYN and PEUK, respectively. The horizontal distributions of both groups were diurnal but heterogeneous in abundance, depending on the groups and layer of depths. The ability of several ab initio models to predict experimental 29Si-NMR chemical shift is examined. The shielding values of trimethylsilyl chloride (A), t-butyldimethylsilyl chloride (B) and allytrimethylsilane (C) are calculated by GIAO, CSGT and IGAIM methods, using HF/6–31G*, B3LYP/6-31G*, HF/6–311+G**, B3LYP/6–311+G** and MPW1PW91/6–311+G** models respectively. The 29Si chemical shifts calculated by GIAO method using HF/6–311+G** model are highly in agreement with those obtained experimentally. All of the models above reproduce the trends of chemical shifts in all cases studied, suggesting that the models are of practical value.  相似文献
