Crossbreeding is an effective approach to manage the genetic decline in aquaculture.One-way hybrids of Crassostrea sikamea(♀)and Crassostrea gigas(♂)have advantages in growth traits and adaptation to high temperature.Here,we used high-throughput sequencing to analyze the molecular processes in the hybrids under and after thermal stress.The hybrids were cultured in the seawater with an increasing temperature from 25℃to 40℃during 10 hours,which is regarded as the thermal stress stage.Then the temperature decreased from 40℃to 25℃within 2 h,which is regarded as the recovery stage.In this study,1293 significant diffe-rentially expressed genes(DEGs)were obtained under thermal stress,of which 576 were upregulated and 717 were downregulated,and 740 significant differentially expressed genes(DEGs)were obtained in the recovery stage,of which the number of upregulated and downregulated genes was 409 and 331,respectively.The antigen processing and presentation,NOD-like,and NF-kappa B path-ways were significantly enriched during the thermal stress stage.The MAPK and PPAR signaling pathways were significantly enrich-ed during the recovery stage.The HSP70,HSP90,and CANX genes were strongly and rapidly upregulated in the control/thermal stress groups but were slightly less upregulated in the thermal stress/recovery group.These results indicate that the innate immune system or nonspecific immunity was deployed to protect interior tissues from thermal stress.In addition,85%of the mutual DEGs were involved in bidirectional regulation(up/down or down/up)when the oysters were removed from the thermal stress to recover.This study provides preliminary insight into the molecular response of C.sikamea(♀)and C.gigas(♂)hybrids to thermal stress and provides a basis for future studies on temperature-adaptation and the possible expansion of hybrid breeding.  相似文献

To elucidate the molecular response of shrimp hepatopancreas to white spot syndrome virus(WSSV) infection, microarray was applied to investigate the differentially expressed genes in the hepatopancreas of ‘Huanghai No. 2'(Fenneropenaeus chinensis). A total of 59137 unigenes were designed onto a custom-made 60 K Agilent chip. After infection, the gene expression profiles in the hepatopancreas of the shrimp with a lower viral load at early(48–96 h), peak(168–192 h) and late(264–288 h) infection phases were analyzed. Of 18704 differentially expressed genes, 6412 were annotated. In total, 5453 differentially expressed genes(1916 annotated) expressed at all three phases, and most of the annotated were either up-or down-regulated continuously. These genes function diversely in, for example, immune response, cytoskeletal system, signal transduction, stress resistance, protein synthesis and processing, metabolism among others. Some of the immune-related genes, including antilipopolysaccharide factor, Kazal-type proteinase inhibitor, C-type lectin and serine protease encoding genes, were up-regulated after WSSV infection. These genes have been reported to be involved in the anti-WSSV responses. The expression of genes related to the cytoskeletal system, including β-actin and myosin but without tubulin genes, were down-regulated after WSSV infection. Astakine was found for the first time in the WSSV-infected F. chinensis. To further confirm the expression of differentially expressed genes, quantitative real-time PCR was performed to test the expression of eight randomly selected genes and verified the reliability and accuracy of the microarray expression analysis. Peptides in shrimp hemolymph play an important role in the innate immune response.Analysis of hemolymph will help to detect and identify potential novel biomarkers of microbial infection.We used magnetic bead-based purification(ClinProt system) and matrix-assisted laser desorption/ionization time of flight mass spectrometry(MALDI-TOF MS) to characterize shrimp hemolymph peptides.Shrimp serum and plasma were used as the source of samples for comparative analysis,and it was found that serum was more suitable for shrimp hemolymph peptidomic analysis.To screen potential specific biomarkers in serum of immune-challenged shrimps,we applied magnetic bead-based MALDI-TOF MS to serum samples from 10 immune-challenged and 10 healthy shrimps.The spectra were analyzed using FlexAnalysis 3.0 and ClinProTools 2.1 software.Thirteen peptide peaks significantly different between the two groups were selected as candidate biomarkers of lipopolysaccharide(LPS)-infection.The diagnostic model established by genetic algorithm using five of these peaks was able to discriminate LPS-challenged shrimps from healthy control shrimps with a sensitivity of 90% and a specificity of 100%.Our approach in MALDITOF MS-based peptidomics is a powerful tool for screening bioactive peptides or biomarkers derived from hemolymph,and will help to enable a better understanding of the innate immune response of shrimps.  相似文献

This study assesses potential effects of adaption to climate change in the future as a carbon related value using a baseline and credit approach, considering the implementation of the Reducing Emissions from Deforestation and forest Degradation(REDD) mechanism. Basic data were obtained for implementing the REDD mechanism in the Democratic People's Republic of Korea(DPRK) for scientific decision-making to prevent deforestation and forest degradation. The potential effects according to the implementation of the REDD mechanism in the DPRK based on forest status data(the latest) are as follows. If the deforestation rate is reduced to a level below 6% through a 20-year REDD mechanism beginning in 2011, 0.01–11.64 C-tons of carbon credit per ha could be issued for DPRK. Converted into CO?-tons per ha, this amounts to 0.03–42.68 CO?-tons, which translates to a minimum of 226,000 CO?-tons and a maximum of 289,082,000 CO?-tons overall for forests in DPRK. In terms of carbon price, this measures up to 1.10 million USD–1.4 billion USD, considering that the REDD carbon price in voluntary carbon markets in 2010 was around 5 USD.  相似文献

Iwagaki oyster,Crassostrea nippona,widely distributes along the seashore of Eastern Asia,and has been considered to be a potential breeding species due to its delicious taste and high commercial value.In order to study its genetic background and population structure,we developed 46 novel polymorphic microsatellite markers using next-generation sequencing technique and characterized them in 30 individuals.The number of alleles ranged from 3 to 22,while the observed and expected heterozygosities varied from 0.133 to 1.000 and 0.455 to 0.949,respectively.Fifteen microsatellite markers were selected and grouped into five highly informative multiplex PCRs for C.nippona.We evaluated and validated these multiplex PCRs in a cultured population including 173 candidate parents and 486 offspring.In actual parentage analysis,80%of the offspring were correctly assigned to their parental pairs using three multiplex PCRs.Furthermore,the success rate of parentage assignment reached 96%when the other two multiplex PCRs were added.These 46 microsatellite loci with high variability and the five multiplex PCRs described here provide a powerful tool for pedigree reconstruction,resource conservation and selective breeding program of C.nippona.  相似文献

Ballast water has been a topic of concern for some time because of its potential to introduce invasive species to new habitats. To comply with the International Convention for the Control and Management of Ships' Ballast Water and Sediments, members of the International Maritime Organization (IMO) must equip their ships with on-board treatment systems to eliminate organism release with ballast water. There are many challenges associated with the implementation of this IMO guideline, one of which is the selection of species for testing the ecological impacts of the treated ballast water. In the United States, ballast water toxicity test methods have been defined by the United States Environmental Protection Agency. However, the test methods had not been finalized in China until the toxicity test methods for ballast water were established in 2008. The Chinese methods have been based on species from three trophic levels: Skeletonema costatum, Neomysis awatschensis, and Ctenogobius gymnauchen. All three species live in broad estuarine and open sea areas of China; they are sensitive to reference toxicants and acclimatize easily to different conditions. In this paper, the biological characteristics, test processes and statistical analysis methods are presented for the three species. Results indicate that the methods for evaluating these three organisms can be included in the ecological toxicity tests for treated ballast water in China.  相似文献

The growth, biochemical content and bioaccumulation quantity of 2,2’,4,4’-tetrabromodiphenyl ether (BDE-47) in Skeletonema costatum were studied under different N:P ratios (1, 4, 16, 64 and 128). All cellular biochemical contents of S. costatum presented decreasing trend over cultivation time. At early stage of cultivation, the cellular protein, carbohydrate and lipid in S. co-statum presented higher values in treatments of N:P=4 and 16. However, they were lower in these treatments at the late stage, but higher in treatments N:P=1 and 128. Similarly, BDE-47 levels per cell of S. costatum were higher in treatments of N:P=4 and 16 at early stage of cultivation, which were 3.8 and 3.7 ng (106 cells)-1, respectively. At the middle stage of cultivation, the BDE-47 level per S. costatum cell lowered;and it further reduced in the treatments of N:P=4 and 16 at the late stage with the values 0.6 and 0.5 ng (106 cells)-1, respectively. However, it rose in N:P=128, reaching up to 2.3 ng (106 cells)-1. Compared with BDE-47 per cell, BDE-47 per algal volume under different N:P ratios did not present obvious difference. The quantity BDE-47 accumulated per cell of S. co-statum was positively correlated with protein, carbohydrate and lipid per cell; meanwhile, the BDE-47 per volume had a positive correlation with biochemical content per volume. The variation of bioaccumulation ability of BDE-47 in S. costatum can be ex-plained by biochemical changes due to N:P ratios.  相似文献

In this study,the influences of immersion bathing in different concentrations of Bacillus velezensis DY-6 on the body weight gain rate and non-specific immune enzyme activities of the coelom fluid of sea cucumber(Apostichopus japonicus)were determined in order to obtain the optimum bacterial concentration.The gut microbiota change in A.japonicus was then analyzed through high-throughput sequencing during the immersion bathing in B.velezensis DY-6 at the optimum concentration for 49 d.The results illustrate that the body weight growth rate of all bathing groups was higher than that of the control.The highest growth rate(25.3%)was achieved when the bacterial concentration was 1 × 10~3 CFU/mL.The activities of non-specific immune enzymes(ACP,AKP,SOD and LZM)of all bathing groups increased,and the activities of the enzymes of groups bathed with the bacterium at 1×10~3 and 1×10~4 CFU/mL reached the highest on day 21 and day 28.Taking the growth rate and economic cost into consideration,the optimum concentration of B.velezensis DY-6 was 1×10~3 CFU/mL.The influences of immersion bathing in B.velezensis DY-6 at 1×10~3 CFU/mL on the gut microbiota of A.japonicus were then evaluated through 16 S rDNA sequencing analysis.Results showed that the gut microbiota changed with the addition of B.velezensis DY-6,and the richness and diversity of the gut microbiota peaked twice on day 14 and day 21,respectively.In association with the non-specific immune enzyme activities and if day 28 was selected as the dividing point,the community structure of the gut microbiota could be obviously divided into two types.The correlation analysis revealed that the non-specific immune enzyme activities were correlated significantly to some gut bacteria(in the phyla Firmicutes,Proteobacteria,and Bacteroidetes)after immersion bathing in B.velezensis DY-6.Our finding s will provide the theoretical foundation for probiotic application in sea cucumber farming.  相似文献

The current testing for paralytic shellfish poisoning(PSP) in shellfish is based on the mouse bioassay(MBA).To alleviate animal welfare concerns,we evaluated the utility of using sublethal indicators of toxicity as an alternative to measuring time to death.Live mice were injected with a PSP congener and the changes in neurotransmitter levels were measured 60,90,and 120 min after injection.Acetylcholine(ACh) was the most sensitive marker for PSP toxicity.The changes in neurotransmitter levels were most pronounced in the blood.Thus,measurement of Ach levels in the blood may serve as a sensitive predictor for PSP that would not require sacrifice of the mice.This method was relatively simple,sensitive(less than 1 μg/kg weight,equivalent to 20 ng/mL),low maintenance,and rapid(less than 60 min).  相似文献
