We collected nine Enteromorpha specimens from the coast of Yantai and evaluated their diversity based on analyses of their ITS (internal transcribed spacer) and 5S rDNA NTS (non-transcribed spacer) sequences. The ITS sequences showed slight nucleotide divergences between Enteromorpha linza and Enteromorpha prolifera. In contrast, multiple highly variable regions were found in the ITS region of Enteromorpha flexuosa. In general, there were more variable sites in the NTS region than in the ITS region in the three species. The variations in 5S rDNA NTS sequences indicated that the molecular diversity of Enteromorpha from the coast of Yantai is very high. However, a phylogenetic tree constructed using 5S rDNA NTS sequence data indicated that genetic differences were not directly related to geographical distribution.  相似文献

Anthropogenic disturbances influence plant regeneration and species diversity, which impact the conservation status of protected areas. A study was conducted in the Sitakund Botanical Garden and Eco-park (SBGE), Chittagong, Bangladesh to analyze the natural regeneration and tree species diversity in disturbed and less disturbed zones. Stratified and systematic random sampling was used to select 50 sample plots from each of the two zones. A total number of 109 plant species from 43 families were recorded in the study; of which 93 species were of natural origin while the rest were planted. From the species with natural origin 66 were tree species, 9 were shrub species and 28 were climbers. Species richness, density of regeneration and disturbance index in the height classes (0- 0.5 m) and {dbh 〉 6 cm) indicated significant differences between the zones. The study analyzed how disturbances affect species diversity in the area. It was found that species richness and basal area are negatively related with disturbances. Moreover, density (N/ha) of trees was more likely to decrease with increasing tree height that reflects the huge demand of local people to harvest large trees as part of their income generating activities. The study findings have implications for future managementdecisions of the SGBE. To restore these ecosystems, management should focus on both biodiversity eonservation and providing benefits to local people without hampering forest ecosystems.  相似文献

The wild Porphyra yezoensis collected from the Qingdao coast was used to prepare protoplasts by enzyme digestion. The pure line was constructed by cultivating the protoplasts. The 18S rDNA of the P. yezoensis pure line was cloned and sequenced. Sequence analysis was executed for this sequence and other 22 sequences retrieved from GenBank. A phylogenetic tree was constructed using the neighbor-joining method. The results revealed a high diversity of 18S rDNA sequences in genus Porphyra and the considerable variation of 18S rDNA sequences in different strains of the same species P. yezoensis and P. tenera. Significant difference of 18S rDNA sequence was observed between P. yezoensis from Qingdao, China, and the two strains of P. yezoensis from Japan, but the three strains of P. yezoensis formed a stable clade in the phylogenetic tree. These results indicate the possibility of interspecies and intraspecies discrimination of Porphyra using the 18S rDNA sequences.  相似文献

The 18S ribosomal DNA gene (18S rDNA) sequences (approxtmately 1300 bp in length) were amplified from the DNA extracted from the free-living marine nematodes collected from the inter-tidal sediment of Qingdao coast in bulk with nematode specific primers. The PCR products were cloned, re-amplified, digested with Rsa I and Hin61 restriction endonucleases and separated in agarose gel. Among 17 restriction fragment length types, types 1, 2 and 6 covered 61.2%, 14.4% and 9.3% of the clones analyzed, respectively, while the remaining 14 only covered 21 clones, which accounted for 15.1% of the total. Twenty-four representative clones were sequenced and phylogenetically analyzed by referring to those currently available in RDP and GenBank databases. Although it was hard to assign these sequences to known species or genera due to the lack of the 18S rDNA sequence data of known marine free-living nematodes, the obtained sequences were assigned to the nematodes of Adenophorea. Among them, twelve sequences were close to Pontonema vulgate and Adoncholaimus sp., four to Daptonema procerus and two (identical) to Enoplus brews. Our results showed that free-living marine nematode diversities could be determined by PCR retrieving and analysis of the 18S rDNA sequences and an 18S rDNA sequence could be assigned to a species or a genus only if the 18S rDNA sequences of the free-living marine nematodes were accumulated to some extent.  相似文献

The phylogenetic diversity of cuhurable psychrophilic bacteria associated with sea ice from the high latitude regions of Canadian Basin and Chukchi Sea, Arctic, was investigated. A total of 34 psychropilic strains were isolated using three methods of ( Ⅰ ) dilution plating ( at 4 ℃ ), ( Ⅱ ) bath culturing ( at - 1 ℃ ) and dilution plating, and ( Ⅲ) cold shock ( -20 ℃ for 24 h), bath culturing and dilution plating under aerobic conditions. Sea-ice samples were exposed to -20 ℃ for 24 h that might reduce the number of common microorganisms and encourage outgrowth of psyehrophilic strains. This process might be able to be introduced to isolation psychrophilic bacteria from other environmental samples in future study. 16S rDNA nearly full-length sequence analysis revealed that psychrophilic strains felled in two phylo- genetic divisions, γ-proteobacteria (in the genera ColweUia,Marinobaeter、Shewanella,Glaciecola,Marinomortas and Pseudoalteromortas ) and Cytophaga-Flexibacter-Baeteroides ( Flavobacterlum and Psyehroflexus). Fifteen of bacterial isolates quite likely represented novel species (16S rDNA sequence similarity below 98% ). One of strains (BSi20002) from Canadian Basin showed 100% sequence similarity to that of Marinobacter sp. ANT8277 isolated from the Antarctic Weddell sea ice, suggesting bacteria may have a bipolar distribution at the species level.  相似文献

The systematic classification of the Eucheumatoideae is difficult because of their variable morphology and interpretation of reproductive structures. Kappaphycus and Eucheuma specimens cultivated on the Hainan and Fujian coast of China were introduced from Vietnam, the Philippines and Indonesia. Combined with morphological characteristics, all Kappaphycus and Eucheuma cultivated strains were identified by internal transcribed spacer (ITS) sequences. The phylogenetic tree was constructed using neighbor-joining and maximum likelihood methods. The results indicate that different ITS sequence lengths occurred in the different genera and species. An obvious difference in morphology could be found in the protuberance shape between Kappaphycus and Eucheuma. The protuberance in Eucheuma was thorn-like and in Kappaphycus was wartlike or papillate. Their ITS sequence lengths differed significantly in nucleotide variation rates up to 58.55%-63.90%. All nucleotide variations occurred in the ITS1 and ITS2 regions except for five nucleotide transversions in the 5.8S rDNA region. In addition, the difference was at the branches among congeneric species. Kappaphycus sp. had branches with small buds, while K. alvarezii did not have such a feature. The nucleotide variation rates varied from 7.02% to 7.48% among species; within the same species of the clades it was <1.20%. Eucheumatoideae algae cultivated in China consisted of three clades, K. alvarezii, Kappaphycus sp., and E. denticulatum. The results indicate that ITS sequence analysis was an effective way for identification of interspecies and intraspecies phylogenetic relationships and might provide a clue for molecular identification of algal Eucheumatoideae.  相似文献

The pathogenic species of genus Vibrio cause vibriosis, one of the most prevalent diseases of maricultured animals and seafood consumers. Monitoring their kinetics in the chain of seafood production, processing and consumption is of great importance for food and mariculture safety. In order to enrich Vibrio-representing 16S ribosomal RNA gene (rDNA) fragments and identify these bacteria further real-timely and synchronously among bacterial flora in the chain, a pair of primers that selectively amplify Vibrio 16S rDNA fragments were designed with their specificities and coverage testified in the analysis of seawater Vibrio community. The specificities and coverage of two primers, VF169 and VR744, were determined theoretically among bacterial 16S rDNAs available in GenBank by using BLAST program and practically by amplifying Vibrio 16S rDNA fragments from seawater DNA. More than 88.3% of sequences in GenBank, which showed identical matches with VR744, belong to Vibrio genus. A total of 33 clones were randomly selected and sequenced. All of the sequences showed their highest similarities to and clustered around those of diverse known Vibrio species. The primers designed are capable of retrieving a wide range of Vibrio 16S rDNA fragments specifically among bacterial flora in seawater, the most important natural environment of seafood cultivation.  相似文献

Eighteen gametophytes including L.japonica ,L.ochotensis and L.longissima,were verified with random amplified polymorphic DNA (RAPD) technique.Eighteen ten-base primers were chosen from 100 primers selected for final amplification test.Among the total of 205 bands amplified ,181(88.3%) were polymorphic ,The genetic distance among different strains ranged from 0.072 to 0.391,The dendrogram constructed by unweighted pair -group method with arithmetic(UPGMA) method showed that the female and male gamtophytes of the same cell lines could be grouped in pairs respectively,It indicated that RAPD analysis could be used not only to distin-gusih different strains of Laminariz ,but also to distinguish male and female gametophyte within the same cell lines,There is ambiguous systematic relationship if judged mereely by the present data It seems that the use of RAPD marker is limited to elucidstion of the phylogenetic relationship among the species of Laminaria.  相似文献

Acila divaricata (Hinds, 1843) and A. mirabilis (Adams and Reeve, 1850) are common benthic bivalves in China. A number of researchers have proposed that the latter species is a junior synonym of the former species. Because of morphological similarities, it is difficult to distinguish these two species based on visual examination only. For better understanding of their taxon-omy, the mitochondrial CO1 gene fragments of five individuals of A. divaricata from the East China Sea and six individuals of A. mirabilis from the Yellow Sea were sequenced in this study. The phylogenetic relationships of the obtained COI sequences, together with nineteen sequences of three species of the genus Nucula, were analyzed. The pairwise intra- and inter-specific distances for the CO1 sequences ranged from 0.002 to 0.017 and from 0.128 to 0.134, respectively, and no overlap was found. Phylogenetically, A. divaricata and A. mirabilis form distinct clades and cluster into a sister to all other Nucula species. The results indicated that A. di-varicata and A. mirabilis are two distinct species. The differences in the morphology and distribution between the two species were briefly discussed.  相似文献

Previous studies found intraspecific diversity in Scrippsiella trochoidea A. R. Loeblich III, a widely distributed calcareous cyst-producing dinoflagellate. In this study, three strains (ST-1, ST-D6 and ST-K) of S. trochoidea isolated from the East Asian waters were studied, together with other geographical strains, to resolve their phylogenetic relationships. For the three East Asian isolates, two highly diverse regions of nuclear-encoded ribosomal DNA (rDNA), the 5.8S rDNA and its flanking internal transcribed spacers 1 and 2, and the 5' portion of the large-subunit rDNA (encompassing the "D1" and "D2" domains) were sequenced. Homologous sequences from other geographical isolates were selected from the GenBank database and the phylogenetic relationships were inferred from the molecular data of these strains. Strains of S. trochoidea were found to cluster into three major clades (STR1, STR2 and STR3), as reported in previous studies. Two of the three strains ST-1 and ST-K, were grouped in clade STR2, the other strain, ST-D6, belonged to clade STR3. The intraspecific diversity of S. trochoidea in East Asian waters makes it necessary to carry out phylogenetic investigations in combination with data of biogeography, population dynamics, and life cycle on the ecophysiology of a region.  相似文献

The nuclear-encoded small subunit ribosomal RNA gene (18S rDNA) of 16 isolates of Chondrus from 8 countries were sequenced. A total of 1796 nucleotides were obtained and aligned with the phylogenetic analysis conducted. The results suggest that the entity from Dalian, China, regarded as C. sp1 is C. pinnulatus. The C. sp2 previously depicted as C. yendoi or Mazzaella japonica may belong to genus Chondrus. So, 4 Chondrus species, i.e. C. ocellatus, C. nipponicus, C. armatus, and C. pinnulatus are distributed in China. However, the entity from Connemara, Ireland, named C. crispus, is not a Chondrus species but that of Mastocarpus stellatus, although it is morphologically similar to C. crispus. Phylogenetic analysis based on complete 18S rDNA sequence data shows that genus Chondrus includes 3 main lineages: the Northern Pacific lineage, containing C. ocellatus, C. yendoi, and C. nipponicus; C. armatus, and C. pinnulatus form the sub-North Pacific lineage; and the Northern Atlantic Ocean lineage, comprising samples of C. crispus from Canada, Portugal, Ireland, Germany and France. The phylogenetic relationships indicate that genus Chondrus might have a North Pacific ancestral origin, radiated to North Atlantic area, and then formed the species C. crispus.  相似文献

Although snow cover plays an important role in structuring plant diversity in the alpine zone, there are few studies on the relationship between snow cover and species diversity of alpine meadows on the eastern Qinghai-Tibetan Plateau. To assess the effect of snow cover on plant species diversity of alpine meadows, we used ten parallel transects of 60 m × 1 m for this study and described the changes in species diversity and composition associated with snow depth. With the division of snow depth into six classes, the highest species richness (S) and species diversity (H′) occurred with an intermediate snow depth, i.e., class Ⅲ and class Ⅳ, showing a unimodal curve with the increase in snow depth. The relationship between snow depth and plant diversity (both richness and Shannon index) could be depicted by quadratic equations. There was no evident relationship between diversity (both S and H′) and soil water content, which implied that other more important factors influenced species diversity. The patterns of diversity found in our study were largely attributed to freeze-thaw alteration, length of growing season and disturbances of livestock grazing. Furthermore, snow depth affected species composition, as evaluated by the Sorensen's index of similarity. In addition, almost all species limited to one snow depth class were found only in class Ⅲand class Ⅳ, indicating that intermediate snow depth was suitable for the survival and growth of many alpine species.  相似文献

The complete mitochondrial cytochrome oxidase subunit Ⅱ (COⅡ) gene of Penaeinae shrimp Fenneropenaeus chinensis was cloned and sequenced. The gene is 688 bp in length and codes for 229 amino acids. It shows 83.2%, 87.0% and 83.8% sequence similarity to Marsupenaeus Japonicus, Penaeus monodon and Farfantepenaeus notialis, respectively. The A+T content of the whole gene and that at the third position of codons are 64.7% and 78.2%, respectively. The phylogenetic relationship between F. chinensis and three other species representing genera Farfanatepenaeus, Marsupenaeus and Penaeus was analyzed. Results showed that the genetic distances among the four taxa ranged from 0.144 0 to 0.200 5, exceeding those estimated with COⅠ and partial 16S rRNA gene sequences among Marsupenaeus, Litopenaeus and Melicertus, and being therefore larger than the value among subgenera. It has been suggested that the COⅡ gene has a faster evolutionary rate than that of the COⅠ gene and partial 16S rRNA gene and could be used for phylogenetic analysis at genus or species level. The results of the present study indicated that Farfantepenaeus, Fenneropenaeus, Marsupenaeus and Penaeus are at a higher phylogenetic level than subgenus, which supports the opinion of the elevation of phylogenetic status of the four subgenera to genus level.  相似文献

Over the past decades,jellyfish occurred increasingly and abundantly in coastal areas worldwide.Usually,biomass of jellyfish,especially when they bloom,can be determined by visual counting.However,tiny individuals of jellyfish(e.g.,planulae,polyps,and ephyrae) are difficult to detect in the field.In this study,species-specific quantitative real-time PCR assays(qPCR)(SYBR Green I) targeting the mitochondrial 16 S rDNA(mt-16 S rDNA) of jelly fish were developed and were used to estimate the distribution and seasonal fluctuations of four jelly fish species(Nemopilema nomurai,Cyanea nozakii,Rhopilema esculentum,and Aurelia coerulea) in Jiaozhou Bay(JZB),China in 2013.The mt-16 S rDNA of A.coerulea and N.nomurai was detected in most of the survey months and it peaked in July(1.03×10~4 copies/L) and September(1.08×10~6 copies/L),respectively.The mt-16 S rDNA of C.nozakii occurred from August to October only with monthly mean values of 7.18-46.17 copie s/L and was mainly located from the middle part to the outer part of the bay.The mt-16 S rDNA of R.esculentum was the least abundant among the four species and was detected in only one sample(B2 station in March),with a value of 88.49 copies/L.The Spearman correlation test revealed that phytoplankton biomass was significantly and positively correlated with the mt-16 S rDNA abundance of A.coerulea(R=0.37,P0.01) and negatively with the mt-16 S rDNA of N.nomurai(R=-0.36,P0.01).The qPCR assay will enable the identification and quantification of jellyfish species in their whole life history and can be used as an approach in combination of the traditional j elly fish survey.  相似文献

The mountain ecosystem is a hotspot of biodiversity and provides a significant contribution to the humans' livelihoods. However, there is still a lack of information regarding floral diversity and how it has been affected due to a logging operation in Sarawak. A study was conducted to determine vegetation recovery at the Heart of Borneo conservation area at different elevations: 750–1200 m(S1), 1200–1500 m(S2) and 1500 m(S3). A total of 891 trees were enumerated. The trees comprised 55 families, 98 genera and 203 species. The lowest elevation site, S1, contained the highest number of species(117) and families(42), while the highest elevation site, S3, contained the lowest diversity(25 families and 59 species). A similar forest structure was recorded, whereby diameter at breast height(dbh), basal area and height decreased as elevation increased. The most speciose families in S1 were Dipterocarpaceae(11 species) and Lauraceae(10 species), while Lauraceae and Fagaceae were the dominant tree families at S2 and S3. Secondary tree species are still abundantly found in the logged plots, especially at S2(45.5%), indicating that after 30 years, these stands are still in the recovery stages. The logged plots retained typical secondary forest appearances with lower canopy diameters and heights and dbh than the unlogged forest. Although the species richness approached that of the unlogged plots, especially at S1(90.9%) and S3(94.9%), there were still major differences in the floristic compositions between logged and unlogged plots(35.4%, 21.5% and 44.7% for S1, S2 and S3, respectively). Although potential seed sources were in close proximity, succession at S2 has proceeded much slower than at S1 and S3. Lower concentrations of soil nutrients, especially N, may be the main reason for the slow recovery. In conclusion, the results suggested that the selectively logged forest at the Payeh Maga Highland required more than three decades to recover and achieve a climate forest stage.  相似文献

Phylogenetic analysis based on 16S rDNA of 8 strains of cultivable bacteria isolated from Arctic sea-ice was studied.The results showed that strain BJ1 belonged to genus Planococcus,which was a genus of low mole percent G C gram-positive bacteria;strain BJ6 belonged to genus Burkholderia of β-proteobacteria and the rest 6 strain all belonged to γ-proteobacteria,of which strain BJ8 was a species of Pseudoalteromonas,strain BJ2-BJ5 and BJ7 were members of genus Psychrobacter.Phylogenetic analysis also indicated that bacteria of genus Psychrobacter of the isolates formed a relatively independent phylogenetic cluster in comparison with other bacteria belonged to genus Psychrobacter.  相似文献

Willow species(Salix fragilis L.and S.alba L.) are important elements of cold desert agroforestry systems in the Lahaul valley,northwestern Himalaya.Their ability to grow through shoot-cuttings plantations under extreme and xeric climatic conditions of cold deserts,makes them ecologically suited and socially accepted for forestry programmes.Willow species in cold deserts may combat desertification and can thrive well in these areas as compared to any other species.Salix fragilis L.and S.alba L.are under cultivation in the Lahaul valley.However,S.fragilis is widely cultivated under the agroforestry and plantation forestry systems.In the Lahaul valley,willow species are used as subsistence resources and for socio-religious purposes.The present study was carried out to examine the vegetation analysis,density,diversity and distribution of willow species,present status and potential willow plantation sites and uses.The results of present study revealed that a higher species density was recorded at middle altitude villages(Jahlma-3,000 m asl and Hinsa-2,700 m asl),except,a higher density of S.fragilis was at Khoksar(3,200 m asl) in plantation forestry on south-facing slopes.In forests,S.fragilis was planted mainly along the water channels,resulted a low density.The shrubby willow species occurred naturally in the entire Lahaul valley up to an altitude of 3,850 m asl.81% of households felt the scarcity of willow trees,whereas,19% of households were satisfied with the willow trees they owned.The majority of willow species were planted in middle altitudes on privately owned irrigated lands.It was rated most potential prospective willow planting site by the farmers.Small size of land-holdings was one of the main constraints for establishing a large number of willow plantations.Commercial aspects of willow species need to be investigated to encourage the farmers to plant more willows further.  相似文献

A new record of Sebastes koreanus(Kim and Lee,1994) was documented based on morphological characters and DNA barcoding. Fifty-six S ebastes specimens were collected from the coastal waters of northern China. Samples were identified as S. koreanus based on morphological characters. The coloration and morphometric measurements were consistent with those described from specimens collected in South Korea. In this study,specimens had the following morphological characteristics:light brown body with dark stripes and tiny dark spots,4–5 wide indistinct vertical patterns on the side,2 radial stripes behind and below the eyes,1 large dark blotch on the opercle. Additionally,the following meristic characters were recorded:dorsal fin XIV-13,pectoral fin 16,anal fin III-6–7,pelvic fin I-5,lateral line scales 29–30,and vertebrae 26. The fragment of cytochrome oxidase subunit I(C OI) gene of mitochondrial DNA was sequenced for phylogenetic analysis. The mean genetic distance within the species was 0.3%. Net genetic distances between S. koreanus and other S ebastes species ranged from 3.1% to 7.6%,which was greater than the threshold for species delimitation. The phylogenetic analysis strongly supports the validity of S. koreanus in China at the genetic level. The origion,evolution,patterns of speciation and unique features in genome divergence among primate lineages of this species still need future directions of research.  相似文献
