Heat shocks effectively produced triploids inPenaeus chinensis. Fertilized eggs heat shocked (28–32°C) for 8 to 16 minutes, starting from 8 to 20 minutes after spawning, resulted in triploidy induction rates of 39%–75%. Several triploid populations were cultured to 10 cm. In a triploid population, two kinds of ovaries were observed. Histological examination showed apparent differences between these two kinds of ovaries, whereas among male shrimp, there were no such differences. 牡蛎是我国出口量较大的水产品之一,由于产品本身的特殊性,在生长过程中易产生腹泻性贝毒(DSP).本实验中原料牡蛎DSP生物法检测结果为阴性,冷冻贮存4个月后检测结果即为阳性.本研究以分析这一变化原因为目的,采用LC-MS法对DSP生物检测的阳性样品进行确认,结果表明,生物法DSP检测的结果为假阳性,造成生物法DSP检测中小白鼠死亡的是牡蛎在贮藏过程中产生的脂溶性有毒物质.进一步分析确定该有毒物质为牡蛎脂肪水解物--游离脂肪酸.同时本研究对牡蛎在不同贮存温度和贮存时间下产生的游离脂肪酸含量进行了测定,在-10 ℃贮存温度下,贮存0,2,4,6,8,10个月,游离脂肪酸的变化范围为5.2~788.6 mg/kg;在-10 ℃,-18 ℃,-25 ℃贮存温度下,贮存4个月,游离脂肪酸的变化范围为92.5~389 mg/kg.  相似文献

This paper presents a three‐dimensional elastoplastic constitutive model for predicting the hydraulic and mechanical behaviour of unsaturated soils. It is based on experimental results obtained from a series of controlled‐suction triaxial tests on unsaturated compacted clay with different initial densities. Hydraulic hysteresis in the water‐retention behaviour is modelled as an elastoplastic process, with the elastic part modelled by a series of scanning curves and the elastoplastic part modelled by the main drying and wetting curves. The effect of void ratio on the water‐retention behaviour is studied using data obtained from controlled‐suction wetting–drying cyclic tests on unsaturated compacted clay with different initial densities. The effect of the degree of saturation on the stress–strain‐strength behaviour and the effect of void ratio on the water‐retention behaviour are considered in the model, as is the effect of suction on the hydraulic and mechanical behaviour. The initial density dependency of the compacted soil behaviour is modelled by experimental relationships between the initial density and the corresponding yield stress and, thereby, between the initial density and the normal compression line. The model is generalized to three‐dimensional stress states by assuming that the shapes of the failure and yield surfaces in the deviatoric stress plane are given by the Matsuoka–Nakai criterion. Model predictions of the stress–strain and water‐retention behaviour are compared with those obtained from triaxial tests with different initial densities under isotropic compression, triaxial compression and triaxial extension, with or without variation in suction. The comparisons indicate that the model accurately predicts the hydraulic and mechanical behaviour of unsaturated compacted soils with different initial densities using the same material constant. A new damage-locating method for bridges subjected to a moving load is presented, and a new ‘moving load damage-locating indicator' (MLDI) is introduced. A vehicle is modeled as a moving load, the bridge is simplified as an Euler-Bernoulli beam, and the damage is simulated by a reduction of stiffness properties of the elements. The curvature and MLDI values at each node of the baseline model (undamaged) and the damage model are computed respectively. Then the damage or damages can be located from a sudden change of the MLDI value. The feasibility and effectiveness of the proposed method are validated by numerical simulation. The results indicate that the method is effective, being able to not only locate a single damage accurately, but also locate multiple damages in simply-supported bridges, including multiple damages in continuous bridges. The results also indicate that the MLDI can accurately locate damages under 5% measurement noise.  相似文献

The spatial distribution of stage-specific abundance and reproduction of the copepod Paracalanus parvus were studied from October 2005 to September 2006 in the Jiaozhou Bay. This copepod occurred continuously in this bay throughout the year. The species reached the lowest abundance in April and peaked in June. From October to December, distribution center mainly occurred in offshore water and at the mouth of the bay. In winter, early copepodites and adults gradually decreased and till February, most of the population was only comprised of CIV–CV stages. Overwintering copepodites matured in March and males tended to mature before female. From May to September, each stage occurred in the population and gradually reached high abundance. Temperature and chlorophyll a (Chl-a) concentration in the three stations can't clearly explain the seasonal variation in stage-specific abundance, so we surmised the important effect of the Yellow Sea. Egg production rate (EPR) reached its lowest in winter and peaked in June at 60.8 eggs female−1 day−1 in nearshore water. In the warming period, EPR in nearshore water was statistically higher and EPR > 10 eggs female−1 day−1 lasted longer than that in offshore water, showing the importance of nearshore water for recruitment of P. parvus. Our study showed that EPR was positively related to temperature and total chlorophyll a in offshore water and mouth of the bay. In nearshore water, the relationships between EPR and temperature and Chl-a in three size fractions were not the same as those in offshore water, suggesting complicated ecosystem in such a eutrophic area in warming period.  相似文献

We extracted marine low-temperature lysozyme (MLTL), a novel lysozyme, from a marine microorganism through fermentation. Our previous study suggested that a low molecular weight (16 kDa) may exert anti-tumor activity through antiangiogenesis. In this study, we extracted a high weight (39 kDa) and investigated its antiangiogenic activity in vivo and in vitro. Using zebrafish embryos as an in vivo study model, we found that treatment with MLTL significantly inhibited the growth of subintestinal vessels (SIVs) in a dose-dependent manner and that 400 μg/ml MLTL was sufficient to block the growth of SIVs. An in vitro study conducted using human umbilical vein endothelial cells (HUVECs) revealed that MLTL suppressed the proliferation, migration and tube formation of HUVECs in a dose-dependent manner. Interestingly, assays by flow cytometry and DNA electrophoresis indicated that MLTL was able to induce apoptosis of HUVECs. Moreover, further study demonstrated that the disruption of intracellular Ca2+ homeostasis may play an important role in MLTL induced apoptosis of HUVECs. Taken together, the results of this study demonstrate for the first time that MLTL inhibits angiogenesis through its pleiotropic effects on vascular endothelial cells and induces apoptosis through regulation of cellular Ca2+ levels. The results of this study also revealed a possible mechanism underlying the antiangiogenic effect of MLTL and suggested that MLTL may be a promising new antiangiogenic agent for use in cancer therapy.  相似文献

国家海洋环境预报中心建立了中国海高分辨率风暴潮数值预报模式,模式在水平分辨率和网格的嵌套方面都较以往的业务化模式有改进和提高.自2003年起将模式投入业务化运行以来,连续三年共对11个台风风暴潮过程进行了跟踪预报,并将数值预报结果与实测资料相对比.本文将对预报模式三年来的预报结果进行检验.  相似文献

An open reading frame (lcn61) of lymphocystis disease virus China (LCDV-cn), probably responsible for encoding putative zinc-finger proteins was amplified and inserted into pET24a (+) vector. Then it expressed in E. coli BL21 (DE3), and His-tag fusion protein of high yield was obtained. It was found that the fusion protein existed in E. coli mainly as inclusion bodies. The bioinformatics analysis indicates that LCN61 is C2H2 type zinc-finger protein containing four C2H2 zinc-finger motifs. This work provides a theory for functional research of lcn61 gene.
