本文以长牡蛎为酶源提取材料,用正丁醇抽提,硫酸铵分级分离,DEAE-52和Sephadex G-100柱层析,得到一定纯度的碱性磷酸酶,提纯倍数为61.96,比活力达1.326U/mg.该酶的最适pH值9.6,最适温度38℃,初速度6.8μmol/(dm~3·min),米氏常数Km值为1.30mmol/dm~3.实验结果表明,Mg~(2+)对该酶有明显的激活作用;Zn~(2+)、Cu~(2+)、Hg~(2+)对该酶有明显的抑制作用.  相似文献   

合浦珠母贝26S蛋白酶体S4,S7亚基基因片段的分离与分析

26S蛋白酶体是真核生物中一种具有ATP依赖性的蛋白酶复合体,主要通过泛肽途径选择性降解细胞内与代谢调控、细胞周期有关的功能蛋白及异常蛋白,参与多种细胞活动的调控过程。26S蛋白酶体由具有催化活性的20S亚复合体和一个具有调节作用的19S亚复合体组成,其中19S亚复合体中的ATP酶亚基是调节26S蛋白酶体活性的重要组件。本通过简并引物PCR手段,从软体动物合浦珠母贝(Pinctada fucata)中扩增到参与构成19S亚复合体的S4和S7(MSS1)两个亚基的基因片段。这两个基因片段所编码的ATP酶组件包含有Gx4GKT,DEID,SAT和H／QRxGRxxR等26S蛋白酶体ATP酶亚基的共同功能基序。这是首次在软体动物中报道26S蛋白酶体的ATP酶亚基基因序列,为研究软体动物中26S蛋白酶体的结构与功能奠定了分子基础。  相似文献   

基于非期望产出的中国滨海旅游生态效率评价——以我国沿海城市为例

旅游生态效率是反映旅游经济与旅游环境双向效应(bi-effect)的重要判断指标。本文基于非期望产出视角,采用超效率SBM模型,评价了我国东部沿海18个主要滨海旅游城市的旅游生态效率,并分析了其时空演变特征。研究结果表明:(1)中国东部沿海18市滨海旅游生态效率整体呈中等水平,平均值处于0.727～0.894,阶段特征明显,呈现前期平稳发展,中期下降,后期回升的发展趋势;(2)各城市滨海旅游生态效率水平差异显著,分为下行风险型,持续成长型,稳定型和波动型4种发展类型。平均效率值最高为天津,最低为三亚;(3)分解效率均值中纯技术效率较高,规模效率次之,技术进步是影响综合效率的主要因素;(4)投入冗余和非期望产出冗余已经成为阻碍滨海旅游生态效率进步的关键因素。  相似文献   

Some stability properties of large-scale baroclinic flows with nonlinear zonal current profile

The stability properties of a baroclinic zonal current with nonlinear velocity profile are investigated. The integral method is applied to the governing eigenvalue equation having the vertical velocity as the dependent variable. Expressed in terms of the Rossby number and the Richardson number, stability criteria, unstable regions in the complexc plane, and the upper bound of the unstable wave growth rate are found. Some differences in the results are noted between the present model and the quasi-geostrophic streamfunction model, particularly in connection with the effect of the velocity profile curvature termU _zz . It is conjectured in the present model that, depending on extreme behaviors ofU _zz , the propagation speed of unstable waves can be greater thanU _max or smaller thanU _min .  相似文献

- The data of landsat TM of multi-temporal for Lingdingyang Estuary, Pearl River in China is firstly used with suspended sediment concentration of field measurement to establish a correlative model equation. After the ratio processing of TM data and atmospheric correction, the images of suspended sediment concentration of different temporals are exported from the image processing systems AREIS II and III. These images express the characteristics of suspended sediment distribution, the mode of sediment transport and the extent of dispersion under the actions of tidal current and wind condition of different seasons.  相似文献

Formation of Equilibrium Beach Profile of the Abandoned Yellow River Delta Coast in North Jiangsu

The abandoned Yellow River Delta coast is a typical erodible silty and muddy coast in China. The paper analyses the marine dynamic characteristics and the mechanism of beach erosion of this area. Analysis and calculation show that in this sea area wave and tidal current action should be considered. Based on the above analysis, an equilibrium beach profile calculation model is developed, in which the wave-current interaction is considered while sediment supply and sediment re-deposition are neglected. The model consists of four parts: (1) calculation of wave parameters, (2) calculation of velocity due to wave-current interaction at different water depth, (3) calculation of friction velocity and shear stress at different water depths, and (4) calculation of the amount of sediment erosion, erosion intensity and variation of beach profile. Calculated results are in good agreement with observed data. Finally, the evolution tendency is discussed and the equilibrium beach profile of this coast is calculated. B  相似文献
