Endocrine disrupting chemicals (EDCs) are increasingly viewed as persistent pollutants, similar to natural hormones in function. This paper describes the expression profiles of 7 genes (DMRT, VTG, GnRHR, FSHR, CYP17A, CYP19A, and CYP19B) involved in sex steroid synthesis and action as well as sexual development in adult male and female Cynoglossus semilaevis, after exposure to different concentrations of Bisphenol A (BPA) and 17β-estradiol (E2). Both BPA (1, 10, 50, 125, and 250 mg/kg) and E2 (0.5, 5, and 10 mg/kg) induced changes in target gene expression, although the estrogenic effects of E2 as a model estrogen were stronger. Among the 7 genes, VTG, CYP17A and CYP19 responded strongly to BPA or E2 exposure and can thus serve as reference biomarkers for estrogenic EDCs exposure in marine teleosts. These data will provide a window to establish a hypothalamic-pituitary-gonadal model in C. semilaevis to better understand the effect pathways of EDCs. 相似文献

Pectenotoxins (PTXs) are a group of marine algal toxins. In this study, the accumulation and depuration of pectenotoxins in
brown crab Cancer pagurus were investigated. Crabs were fed with toxic blue mussels Mytilus edulis for 21 days and then depurated for 42 days. Toxins were extracted with methanol from the digestive glands of contaminated
crabs, uncontaminated crabs (control group) and from blue mussels for comparison. Extracts were analyzed by liquid chromatograph
coupled with tandem mass spectrometry (LC-MS-MS). The concentrations of PTX-2, PTX-2 SA, 7-epi-PTX-2 SA, and PTX-12 were analyzed in two batches of toxic blue mussels and the crabs. A one-compartment model was applied
to describe the depuration of PTXs. The half-life of PTXs was estimated to be 6–7.5 days. After depuration for 42 days, the
amount of PTXs measured in the crab digestive glands was less than 1 μg/kg.

采用同源克隆和末端快速扩增(RACE)方法, 克隆了半滑舌鳎中 GnRHR 全长 cDNA。通过基因全长以及推断的氨基酸序列分析, 得知该序列包含一个 7tm-1 保守结构域, 为 G 蛋白偶联受体家族成员。利用 PHYLIP 3.5c 邻位相连法以及 DNAstar 中的 CLUSTAL W 方法对相应的氨基酸序列进行了聚类分析和序列相似度分析。结果表明, 半滑舌鳎 GnRHR 鳉 与鲈鱼、琥珀鱼、虹鳟以及青 等鱼类中的 GnRHR聚为一支, 亲缘关系较近, 且相似度较高, 分别为 90.1%、 89.7%、 79.0%以及 78.3%, 而与高等哺乳动物人、小鼠相似性仅分别为 18.8%和 16.2%, 亲缘关系较远。应用半定量 RT-PCR 技术分析其组织表达, 发现 GnRHR 广泛表达于各个组织, 但表达量差异较大, 在性腺、脑和肾中表达量较高, 其它组织较弱。 相似文献

为明确环境新型污染物--氨基脲(semicarbazide,SEM)在贝类体内的存在形态,本文建立了一种SEM在扇贝中形态测定的液相色谱-串联质谱(liquid chromatography-tandem mass spectrometry,LC-MS/MS)分析方法。扇贝中SEM总量测定方法为:样品用盐酸水解,以2-硝基苯甲醛为衍生剂衍生,乙酸乙酯提取,超滤离心净化,进LC-MS/MS分析。结合态SEM测定方法为:样品分别采用50%甲醇水溶液、75%甲醇水溶液、甲醇和水洗涤后,按照SEM总量测定方法处理,采用甲醇和2 mmol/L乙酸铵作为流动相进行梯度洗脱,电喷雾离子源正离子(electrosprary ionization,ESI+)选择反应监测模式对SEM进行定性和定量测定。本方法在1~20 μg/L范围内线性关系良好,相关系数大于0.999。SEM在1.00、5.00、10.0 μg/kg添加水平的回收率在84.2%~118%,相对标准偏差均<15%。本方法扇贝中SEM的定量限为1.0 μg/kg,本方法灵敏、准确、操作简便,适用于扇贝中SEM存在形态的测定。 相似文献
